Description
Die Erfindung betrifft vorzugsweise diejenigen Verbindungen der Formel I, worin X] Fluor oder Chlor, X2 Chlor, R] Wasserstoff und R2Niederalkanoyl,z. B. Propionyl, darstellen, sowie die entsprechenden 1,2-Dihydroderivate.
Die Erfindung betrifft in erster Linie Verbindungen der Formel I, worin Xt Fluor oder Chlor, X2 Chlor, Rj Chlor und R2 Niederalkanoyl, z. B. Propionyl, darstellen.
Die Erfindung betrifft vor allem insbesondere die in den Beispielen genannten Verbindungen der Formel I.
Erfindungsgemäss werden Verbindungen der Formel I, bzw. ihre 1,2-gesättigten 2-unsubstituierten Analogen in an sich bekannter Weise hergestellt, indem man eine Verbindung der Formel ch2X2

worin Ri, R2 und X2 die obgenannten Bedeutungen haben, bzw. ein 1,2-gesättigtes, 2-unsubstituiertes Derivat davon, mit einem Halogenwasserstoff der Formel H-X, (III) behandelt.
Die Aufspaltung der 9ß,llß-Oxidogruppe in einem Ausgangsmaterial der Formel II (einschliesslich entsprechender 1,2-gesättigter 2-unsubstituierter Derivate) durch Behandeln mit einem Halogenwasserstoff der Formel III, d. h. mit Chlorwasserstoff oder Fluorwasserstoff, wird in an sich bekannter Weise vorgenommen, wobei man zweckmässig einen wasserfreien Halogenwasserstoff, gegebenenfalls in Gegenwart eines inerten Lösungsmittels, wie Chloroform, Tetrahydrofuran oder insbesondere Dimethylformamid, Fluorwasserstoff auch in wässriger Lösung, verwendet.
Anstelle des Halogenwasserstoffs der Formel III kann man auch ein Fluorwasserstoff- oder Chlorwasserstoff-abgebendes Mittel, wie das Salz einer solchen Säure mit einer tertiären organischen Base, z. B. Pyridin, oder, insbesondere im Fall von Fluorwasserstoff, eine ähnliche geeignete Additionsverbindung verwenden. Ein besonders günstiges Verfahren wird im US-Patent 3 211758 beschrieben, wonach man Fluorwasserstoff in Form eines Adduktes mit einem Carbaminsäure- oder Thiocar-baminsäure-Derivat, insbesondere mit Harnstoff, zur Anwendung bringt.
Die Ausgangsstoffe der Formel II können in an sich bekannter Weise erhalten werden, z.B. durch Abspalten von Wasser aus einer 6a-Fluor-llß-hydroxy-16ß-methyl-2-Rr17a-OR2-21-X2-pregna-l,4-dien-3,20-dion-Verbindung,z. B. durch Behandeln mit einem geeigneten Säurechlorid, wie Phosphoroxychlorid oderMethansulfonsäurechlorid, in Gegenwart einer Base, z. B. Pyridin, Anlagern von unterbromiger Säure (die z. B. in Form von N-Bromacetamid oder N-Bromsuccinimid verwendet wird) an die 9,11-Doppelbindung der so erhältlichen 6a-Fluor-16ß-methyl-2-Ri-17a-OR2-21-X2-pregna-l,4,9(ll)-trien-3,20-dion-Verbindung und Abspalten von Bromwasserstoff aus der entsprechenden 9a,llß-Bromhydrin-Verbindung durch Behandeln mit einer Base, z. B. einem alkalimetallcarbonat oder -hydroxid, z.B. Kaliumcarbonat oder Kaliumhydroxid, unter Bildung des gewünschten Ausgangsmaterials der Formel II. In den obigen Zwischenprodukten haben Rj, R2 und X2 die vorstehend gegebenen Bedeutungen .Analog können auch die entsprechenden 1,2-gesättigten 2-unsubstituierten Ausgangsstoffe enthalten werden.
5 Verfahrensgemäss erhältliche Verbindungen der Formel I (einschliesslich ihrer 1,2-Dihydroanalogen) mit salzbildenden Eigenschaften, d. h. Verbindungen, worin R2 einen Carboxy-niederalkanoylrest darstellt, können in an sich bekannter Weise durch Behandeln mit einer geeigneten Base, wie einem Alkali-io metall-, wie Natrium- oder Kaliumhydroxid, -carbonat oder hydrogencarbonat (wobei man vorzugsweise die äquivalente Menge der Base verwendet), üblicherweise in einem wässrigen oder wässrig-alkoholischen Medium, in ihre Salze überführt werden.
15 Umgekehrt kann man ein verfahrensgemäss erhältliches Salz einer Verbindung der Formel I mit salzbildenden Eigenschaften in die freie Verbindung überführen, z. B. ein entsprechendes Salz mit einer Säure, wie einer anorganischen Säure behandeln (z.B. durch Ansäuern eine Lösung des Salzes in einem wässrigen oder 20 wässrig-organischen Medium auf einen pH-Wert von etwa 1—2 bringen.
Vorzugsweise setzt man in den obigen Verfahrensschritten diejenigen Ausgangsstoffe ein, die zu vorstehend als besonders bevorzugt beschriebenen Verbindungen führen.
Die gemäss der vorliegenden Erfindung hergestellten Endstoffe können als pharmazeutische Präparate enthaltend eine erfindungsgemässe Verbindung der Formel I oder ein 1,2-gesät-tigtes 2-unsubstituiertes Derivat einer solchen Verbindung als Wirkstoff, sowie zur Herstellung von solchen pharmazeutischen 30 Präparaten, Verwendung finden.
Dabei kommen in erster Linie topisch anwendbare pharmazeutische Präparate, wie Cremen, Salben, Pasten, Schäume, Tinkturen und Lösungen, in Frage, die von etwa 0,005 % bis etwa 0,1 % des Wirkstoffs enthalten, ferner Präparate zur oralen 35 Verabreichung, z. B. Tabletten, Dragées und Kapseln, und parenteralen Verabreichung. Die Herstellung solcher Präparate erfolgt in an sich bekannter Weise.
Die gemäss der vorliegenden Erfindung hergestellten neuen Verbindungen verwendet man vorzugsweise zur Behandlung von 40 Entzündungen, in erster Linie als lokal anzuwendende antiinflammatorische Glucocorticoide, üblicherweise in Form von pharmazeutischen Präparaten, besonders in Form von topisch anwendbaren pharmazeutischen Präparaten.
Die nachfolgenden Beispiele illustrieren die oben beschrie-45 bene Erfindung; sie sollen jedoch diese in ihrem Umfang in keiner Weise einschränken. Temperaturen werden in Celsiusgraden angegeben.

Beispiel 1
a) Eine Suspension von 1,0 g 21-Chlor-9ß, 1 lß-epoxy-16a-hydroxy-16ß-methyl-pregn-4-en-3,20-dion-17-propionat in 5,4 ml Dioxan und 1,4 ml Orthoameisensäuretriäthylester wird mit 0,9 ml einer Lösung, bestehend aus 277 mg p-Toluolsulfonsäure, 2,7 ml Dioxan und 0,55 ml Äthylalkohol, versetzt und 90 min bei 55 Zimmertemperatur gerührt. Nach Zugabe von 3,8 ml Pyridin wird das Reaktionsgemisch mit Äthylacetat verdünnt, dreimal mit gesättigter Natriumchlorid-Lösung gewaschen, getrocknet und im Wasserstrahlvakuum eingedampft. Der so erhaltene rohe 3-Äthylenoläther(3-Äthoxy-21-chlor-9ß,llß-epoxy-17a-60 hydroxy-16ß-methylpregna-3,5-dien-20-on-17-propionat) wird in einem Gemisch von 50 ml Tetrahydrofuran und 10 ml Wasser gelöst und während 40 min bei Zimmertemperatur mit gasförmigem Perchlorylfluorid behandelt. Nachdem man anschliessend während 10 min Stickstoff durchgeleitet hat, wird die Reaktions-65 lösung mit Äthylacetat verdünnt, nacheinander mit 10 % Kaliumjodid-Lösung, mit 10 % Natriumthiosulfat-Lösung und dreimal mit gesättigter Natriumchlorid-Lösung gewaschen, getrocknet und im Wasserstrahlvakuum eingedampft. lende Rohprodukt wird über eine 100-fache Gewichtsmenge Kieselgel (Stufensäule) chromatographiert. Die mit einem Gemisch von Toluol-Äthylacetat (90:10) eluierten Fraktionen ergeben, aus Methylenchlorid/Äther umkristallisiert, das 21-Chlor-6a-fluor-9ß,llß-epoxy-17a-hydroxy-16ß-methyl-pregn-4- 5 en-3,20-dion-17-propionat, Smp. 174—177°C.
b) 720 mg der letztgenannten Verbindung werden in einem Kunststoffgefäss mit 14,5 ml eines Reagens, zubereitet durch Vermischen von 10 g Harnstoff mit 13,25 g wasserfreiem flüssigem Fluorwasserstoff, Übergossen und während 3 h unter Eis- 10 kühlung gerührt. Das Reaktionsgemisch wird auf 55 ml eiskalte gesättigte Ammoniak-Lösung gegossen, mit Essigsäure schwach angesäuert und zweimal mit Chloroform extrahiert. Die organischen Phasen werden vereinigt, mit eiskalter verdünnter Natronlauge gewaschen, getrocknet und am Wasserstrahlvakuumeinge-15 dampft. Das Rohprodukt wird über eine 100-fache Gewichtsmenge Kieselgel (Stufensäule) chromatographiert. Die mit einem Gemisch von Toluol-Essigester (90:10) eluierten Fraktionen ergeben, aus Methylenchlorid/Äther umkristallisiert, das 21-Chlor-6a,9a-difluor-llß,17a-dihydroxy-16ß-methyl-pregn-4- 20 en-3,20-dion-17-propionat, Smp. 212—213° C.

Beispiel 2
In der im Beispiel lb beschriebenen Weise erhält man aus 21 Chlor-6a-fiuor-9ß, 1 l-epoxy-17-hydroxy-16ß-methyl-pregna-1,4-dien-3,20-dion-propionat (hergestellt nach Beispiel 4b) das 21-Chlor-6a,9a-difluor-llß,17a-dihydroxy-16ß-methyl-pregna-l,4-dien-3,20-dion-17-propionat, Smp. 220—221°C (aus Methy-lenchlorid-Äther), bzw. aus dem entsprechenden 2,21-Dichlor-analogdas2,21-Dichlor-6a,9a-difluor-llß,17a-dihydroxy-16ß- 30 methyl-pregna-1,4-dien-3,20-dion-17-propionat, Smp.
202—204°C (aus Methylenchlorid-Äther).
Beispiel 3
In eine Lösung von 400 mg21-Chlor-7a-fluor-9ß,ll-epoxy-17-hydroxy-16ß-methyl-pregn-4-en-3,20-dion-propionat in 20 ml

Chloroform wird während 30 min bei 0° C Chlorwasserstoffgas eingeleitet. Das Gemisch wird weitere 30 min bei 0°C stehen gelassen, mit Chloroform verdünnt, mit einer eiskalten gesättigten Natriumhydrogencarbonat-Lösung gewaschen, getrocknet und im Vakuum eingedampft. Das erhaltene rohe 9a,21-Dichlor-6a-fluor-llß,17-dihydroxy-16ß-methyl-pregn-4-en-3,20-dion-17-propionat wird in der üblichen Weise (s. Beispiel 5a) chromatographisch gereinigt, womit ein Endprodukt vom Smp. 198° C (Zersetzung) resultiert.
Beispiel 4
a) In eine Lösung von 2,8 g 21-Chlor-6a-fluor-9ß, 11-epoxy-17-hydroxy-16ß-methyl-pregna-l ,4-dien-3,20-dion-propionat in 140 ml Chloroform wird während 30 min bei 0°C Chlorwasserstoffgas eingeleitet. Das Gemisch wird weitere 30 min bei 0° C stehen gelassen, mit Chloroform verdünnt, mit einer eiskalten gesättigtenNatriumhydrogencarbonat-Lösung gewaschen, getrocknet und im Vakuum eingedampft. Das erhaltene Rohprodukt wird an 100-facher Gewichtsmenge Silicagel (Stufensäule) chromatographiert. Das gewünschte Produkt wird mit einem
99:1-Gemisch von Toluol-Methanol eluiert. Nach Kristallisation aus Methylenchlorid-Äther erhält man 9cc,21-Dichlor-6a-fluor-llß, 17-dihydroxy-16ß-methyl-pregna-l ,4-dien-3,20-dion-17-propionat, Smp. 212—213°C (Zersetzung).
b) Das Ausgangssteroid wird folgendermassen erhalten: Ein Gemisch von 3 g21-Chlor-6a-fluor-9ß, ll-epoxy-17-hydroxy-16ß-methyl-pregn-4-en-3,20-dion-propionat (s. Beispiel la) und 3 g 2,3-Dichlor-5,6-dicyan-l ,4-benzochinon in 150 ml Dioxan wird 20 h in einer Stickstoffatmosphäre unter Rückfluss gekocht. Das abgekühlte Gemisch wird genutscht, das Filtrat eingedampft, in Methylenchlorid gelöst und durch die 15-fache Gewichtsmenge neutrales Aluminiumoxid (Aktivität II) filtriert. Das Filtrat wird eingeengt, womit das rohe 21-Chlor-6a-fluor-9ß,ll-epoxy-17-hydroxy-16ß-methyl-pregna-l,4-dien-3,20-dion-propionat resultiert, welches ohne weitere Reinigung zur Behandlung mit Fluor- bzw. Chlorwasserstoff geeignet ist.

- 3. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man mit wasserfreiem Chlorwasserstoff behandelt.
- 4. Verfahren nach einem der Ansprüche 1—3, dadurch gekennzeichnet, dass man eine Verbindung der Formel 1, worin Ri Wasserstoff oder Chlor, R2 Alkanoylmit2—5 Kohlenstoffatomen, Xj Fluor oder Chlor und X2 Chlor bedeutet, herstellt.
- 5. Verfahren gemäss Anspruch 1 oder 2, dadurch gekennzeichnet, dass man 2,21-Dichlor-6a,9a-difluor-llß,17a-dihy-droxy-16ß-methyl-pregna-l,4-dien-3,20-dion-17-propionat herstellt.
- 6. Verfahren gemäss Anspruch 1 oder 3, dadurch gekennzeichnet, dass man 9a,21-Dichlor-6a-fluor-llß,17a-dihydroxy-16ß-methyl-pregna-l ,4-dien-3,20-dion-17-propionat herstellt.
- 7. Verfahren gemäss Anspruch 1 oder 2, dadurch gekennzeichnet, dass man 21-Chlor-6a,9a-difluor-llß,17a-dihydroxy-16ß-methyl-pregna-l ,4-dien-3,20-dion-17-propionat herstellt.
- 8. Verfahren gemäss Anspruch 1 oder 2, dadurch gekennzeichnet, dass man21-Chlor-6a,9a-difluor-llß,17a-dihydroxy-16ß-methyl-pregn-4-en-3,20-dion-17-propionat herstellt.

Dabei ist das Natriumsalz einer Verbindung der Formel I besonders bevorzugt, worin R2 für 3-Carboxypropionyl steht.Die erfindungsgemässen Verbindungen besitzen wertvolle pharmakologische Eigenschaften. So zeichnen sie sich durch eine hervorragende antiinflammatorische Wirksamkeit bei einer verminderten systemischen Wirkung aus. So z. B. bei lokaler Anwendung im Rohwattegranulomtest an der Ratte kann man die Hemmung der entzündlichen Vorgänge im Dosisbereich von etwa 0,001 bis etwa 0,10 mg/Pellet feststellen, wobei aber die ersten Anzeichen einer systemischen Wirkung erst an oder sogar oberhalb der oberen Grenze dieses Dosisbereichs bemerkbar sind, und zwar die Gewichtsabnahme des Thymus oberhalb 0,03 mg/Pellet und des gesamten Körpers erst bei 1,0 mg/Pellet. Bei der topischen Verabreichung (Rattenohr-Test nach Tonelli) liegt ED5o sogar bei etwa 10—30 meg/ ml. Wegen diesen biologischen Eigenschaften sind die neuen Verbindungen in allen Indikationen, für die sich Glucocorticoid-Steroide mit entzündungshemmenden Eigenschaften eignen, insbesondere jedoch als topisch anzuwendende antiinflammatorische Glucocorticoide, z.
