Description
45 Die Erfindung stellt sich das Ziel, neue Verbindungen der allgemeinen Formel I bereitzustellen, die auf das Zentralnervensystem eine stärkere Wirkung ausüben als das bisher einzige bekannte und im Handel befindliche 5H-2,3--Benzodiazepin-Derivat, das Grandaxin (Tofizopam; 1-/3,4-50 -Dimethoxyphenyl/-4-methyl-5-äthyl-7,8-dimethoxy-5H-2,3--benzodiazepin1; ungarische Patentschrift Nr. 155 572, US-Pattentschrift Nr. 3 736 315, schweizerische Patentschrift Nr. 519 507).
Die Verbindungen der allgemeinen Formel I beziehungs-55 weise 'ihre Säureadd'itionssalze werden erfindungsgemäss hergestellt, indem man ein 1,5-Diketon der allgemeinen Formel II

Die Erfindung betrifft neue 5H-2,3-Benzod'iazepin-De-rivate, deren Säureadditionssalze sowie ein Verfahren zur 65 Herstellung dieser Verbindungen. Die Erfindung betrifft ferner Arzneimittelpräparate, die die neuen Verbindungen enthalten.

in einem polaren Lösungsmittelmedium, vorzugsweise in Wasser, Alkoholen mit 1-4 Kohlenstoffatomen, Carbonsäuren mit 1-3 Kohlenstoffatomen, Dioxan, Dimethylformamid, Pyridin oder Gemischen der aufgeführten Lösungsmittel, im allgemeinen bei —20 bis +120°C, vorzugsweise bei 60-100°C, mit dem l-5fachen der äquimolaren Menge von Hydrazin, Hydrazinhydrat oder einem mit einer Carbonsäure mit 1-3 Kohlenstoffatomen gebildeten Hydrazinsalz umsetzt, die erhaltene Verbindung der allgemeinen Formel I gewünschtenfalls zu einer anderen Verbindung der allgemeinen Formel I umsetzt und/oder in an sich bekannter Weise ihre pharmakologisch verwendbaren Säureadd'itionssalze bildet beziehungsweise aus den Säureadditionssalzen der Verbindungen der allgemeinen Formel I in an sich bekannter Weise die Base freisetzt oder andere Säure-addit'ionssalze bildet.
Bei als Substituenten R eine Arylgruppe oder heterocyclische Gruppe enthaltenden Ausgangsverbindungen der allgemeinen Formel II wird dem Reaktionsgemisch nach der Umsetzung mit dem Hydrazin eine Mineralsäure, zum Beispiel Salzsäure oder Schwefelsäure, zugesetzt, um d'ie Cyclokondensation zu beschleunigen.
Für 'die Umwandlung einer Verbindung d'er allgemeinen Formel I in eine andere Verbindung der allgemeinen Formel I kommen zum Beispiel folgende Reaktionen in Frage:
Verbindungen der allgemeinen Formel I, in denen R, R3 und R4 für Hydroxylgruppe und/oder R1 für Hydroxy-methylgruppe steht, werden in an sich bekannter Weise al-kyliert, aralkyliert oder acyliert; die 5H-2,3-Benzodiazepin--4-carbonsäureester können mit Metallhydrin, zum Beispiel mit Natriumborhydrid, zu den entsprechenden 4-Hydroxy-methyl-5H-2,3-benzodiazepin-Derivaten reduziert werden; die 5H-2,3-Benzodiazepin-4-carbonsäuren sind durch Verseifen ihrer Ester zugänglich, und diese Säuren können zu in 4-Stellung unsubstituierten 5H-2,3-Benzod'iazepinen dfe-carboxyliert werden; die in 4-Stellung eine Methylgruppe enthaltenden 5H-2,3-Benzodiazepine können mit Selendioxyd zu den entsprechenden Formylderivaten oxydiert werden.
Die physiologisch verträglichen Salze der Verbindungen der allgemeinen Formel I können zum Beispiel mit Salzsäure, Bromwasserstoffsäure, Phosphorsäure, Schwefelsäure oder Perchlorsäure bereitet werden. Weist wenigstens einer der Substituenten ein basischeres Stickstoffatom auf, so können zur Salzbildung auch organische Säuren, zum Beispiel Essigsäure, Weinsäure, Milchsäure, Maleinsäure, Fumarsäure, verwendet werden.
Die 5H-2,3-Benzodiiazepin>-Derivate der allgemeinen Formel I werden im allgemeinen vor der Salzbildung gereinigt, jedoch kann zur Salzbildung auch das Rohprodukt eingesetzt werden. Schlecht kristallisierende Verbindungen der allgemeinen Formel I werden zweckmässig zu einem gut kristallisierenden Salz, zum Beispiel dem Rhodanat oder dem Hydrochlorid, umgesetzt, aus welchem dann gewünschtenfalls die Base in einer Form freigesetzt werden kann.
Geht man von 2-Benzopyriliumsalzen der allgemeinen Formel III aus, so kann als Säureakzeptor ein Überschuss der Hydrazinkomponente dienen, jedoch können als Säureakzeptor auch Alkalihydroxyde, -carbonate oder -hydrogen-carbonate, ferner geeignete organische Basen, zum Beispiel Pyridin oder Triäthylamin, verwendet werden. Die Ausgangsstoffe der allgemeinen Formeln II, III, IV und V können notwendigenfalls vor der Umsetzung mit der Hydrazinkomponente, zweckmässig ohne Isolierung, ineinander übergeführt werden.
Die als Ausgangsstoffe verwendeten Verbindungen der allgemeinen Formeln II, III und IV sind in folgenden Literaturstellen beschrieben beziehungsweise können aufgrund der dort gegebenen Hinweise hergestellt werden: Ber. Auf die Herstellung der neuen Ausgangsverbindun'gen der allgemeinen Formel V wird im Beispielteil näher eingegangen. Die Ausgangsverbindungen der allgemeinen Formel IV können in zwei Formen (als 6H-2-benzopyran-2-one und als 2-Benzopyrilium-6-oxyde) geschrieben werden. Jede der beiden Schreibweisen ist kennzeichnend für die Verbindungen.
Die erfindungsgemässen 5H-2,3-Benzodiazepine der allgemeinen Formel I weisen bedeutende Wirkungen auf das Zentralnervensystem auf, so verringern sie zum Beispiel die

In Tierversuchen gewonnene allgemeine Verhaltensbeobachtungen sowie die ED50-Werte nach der «Fighting behaviour» -Untersuchung sind für einige kennzeichnende Vertreter der neuen Verbindungen in der Tabelle 1 zusammengefasst. Daten zu der narkosepotenzieren>-den Wirkung sind in Tabelle 2 angegeben.

Aus den Daten der Tabellen 1 und 2 sind die vorteilhaften Eigenschaften der erfindungsgemässen neuen Verbindungen eindeutig ersichtlich.
Die Untersuchung der narkosepotenzierenden Wirkung wurde an Mäusen vorgenommen, denen die Verbindungen in Dosen von 12,5, 25, 50, 100 mg/kg p.o. 30 Minuten vor der Messung verabreicht wurden. Es wurde festgestellt, um wieviel Prozent die Verbindungen die mit 50 mg/kg i.v. Hexobarbital-Na hervorgerufene Narkose verlängern. Als

Die Verbindungen wurden als mit Tween-80 bereitete Suspension verabreicht (Tween 80: Polyoxyäthylen-sorbitan--monooleat) [L. Petocz, I. Kosóczky: Therapia Hungarica 23, 143 (1974)]. Die Verbindung hat in therapeutischer Dosis keinerlei Muskelrelaxationswirkung und beeinflusst die Bewegungskoordination erst in grossen Dosen. Letzteres wurde mit dem bekannten Rotarod-Test bestimmt (bei i.p. Applikation ist ED50 = 87 mg/kg).
Die Verbindung gemäss Beispiel 63 kann als Minortran-qu'illizer bei unterschiedlichen Neurosen und psychovege-tativen Störungen angewendet werden.
Die Erfindung wird anhand der folgenden Beispiele näher erläutert.
Beispiel 1
l-(4-Chlorphenyl)-4-methyl-7,8-dimethoxy-5H-2,3-benzo-
diazepin
1,13 g (2,73 mMol) l-(4-Chlorphenyl)-3-methyl-6,7-di-methoxy-2-benzopyrilium-perchlorat werden in 10 ml Methanol suspendiert, bis zum Sieden erhitzt und mit 1,0 ml 98%igem Hydrazinhydrat versetzt. Das Gemisch wird eingedampft und der Rückstand mit Wasser auf ein Filter gespült. 0,87 g (2,65 mMol) getrocknetes Produkt werden erhalten, das bei 188-198°C schmilzt. Durch Umkristallisieren aus 30 ml Äthanol erhält man 0,6312 g (1,92 mMol, 72,5 %) einer weissen, kristallinen Substanz, die bei 209 bis 211°C schmilzt.

Man geht von den entsprechenden 2-Benzopyriliumsal-zen aus, arbeitet auf die im Beispiel 1 beschriebene Weise und verwendet als Reaktionsmedium Methanol, Äthanol oder Isopropanol. Bei den Schmelzpunkten ist jeweils angegeben, welches Lösungsmittel zum Umkristallisieren verwendet wuide.

5 Zu der Suspension von 17,77 g (28,5 mMol) l-(3-Meth-oxy-4-benzyloxy-phenyI)-3-methyl-4-äthyl-6-methoxy-7-ben-zyIoxy-2-benzopyrilmm-perchlorat in 30 ml Bisessig werden unter Rühren bei 80-88°C 8,4 ml 100%iges Hydrazinhydrat tropfenweise zugegeben1. Das Gemisch wird bei io 95-100°C eine Stunde lang gerührt, dann lässt man -80 ml 2% ige Natronlauge einfliessen. Das ausgeschiedene rohe 5H-2,3-Benzodîazep'in-Derivat wird mit 80 ml Chloroform ausgeschüttelt. Die chloroformische Phase wird zuerst mit 50 ml l%iger Natriumhydrogencarbonatlösung, dann mit 15 50 ml Wasser gewaschen, über Natriumsulfat getrocknet und dann zur Trockne eingedampft. Der Rückstand wird aus 30 ml Methanol umkristallisiert. 12,57 g (82%) weisse, schwach cremefarbene Kristalle werden erhalten.

Das Reaktionsgemisch wird auf Siedetemperatur erhitzt, eine Stunde lang gerührt, dann auf 60°C gekühlt, mit 0,41 ml (8 mMol) 98%igem Hydrazinhydrat versetzt und erneut eine Stunde lang gerührt. Dann werden portionsweise 0,154 g Natriumhydrogencarbonat zugesetzt. 65 Das Reaktionsgemisch wird zum Sieden erhitzt, mit Aktivkohle geklärt und dann filtriert Das Filtrat wird eingedampft und mit insgesamt 25 ml Wasser auf ein Filter aufgebracht. 0,835 g (60,5%) Produkt werden erhalten, das

Ein Gemisch aus 10,37 g (28,9 mMol) 2-(l-Äthylaceto-nyl)-4,5-d'imethoxy-2'-chlorbenzophenon, 10,5 ml Eisessig und 2,65 konzentrierter Salzsäure wird unter Rühren auf 95°C erhitzt. Nach dem Abkühlen auf 60°C werden 2,14 ml 98%iges Hydrazinhydrat in Portionen zugesetzt. Zu der Lösung, deren Temperatur auf 85°C angestiegen ist, werden nach 30 Minuten die Lösung von 1,44 g Natriumhydroxyd in 4,5 ml Wasser und anschliessend 10 ml Methanol gerührt. Dann wird die Lösung in 130 ml Wasser eingegossen, d:as sich ausscheidende 5H-2,3-Benzod'iazepin- Derivat wird isoliert. 9,04 g (88%) Produkt werden erhalten, das bei 129-131°C schmilzt. C20H21ClN2O2 = 356,9. Das Rohprodukt kann aus Äthanol umkristallisiert werden und schmilzt dann bei 147-149°C.
Das aus Isopropanol umkristallisierte Rhodanid (C20H22-C1N202 . SCN = 425,95) schmilzt bei 169-171°C.

l-(2-Jodphenyl)-4-methyl-7,8-dimethoxy-5H-2,3-benzo-diazepin
Ein Gemisch aus 2,26 g (5,32 mMol) 2-Acetonyl-4,5-di-methoxy-2'-jodbenzophenon, 20 ml Methanol und 0,75 ml ml 100%igem Hydrazinhydrat wird 20 Minuten lang gekocht und dann eingedampft. Der Rückstand wird mit Wasser auf ein Filter aufgebracht. 2,2 g Monohydrazon werden erhalten. Das l-(2-Hydrazono-propyl)-4,5-dimethoxy-2'--jodbenzophenon wird in 20 ml methanolischer Salzsäure gelöst, die Lösung wird eingedampft. Der Rückstand wird mit 5 %iger Natronlauge alkalisch gemacht, wobéi das 5H--2,3-Benzodiazepin-Derivat ausfällt. Das Produkt wird abfiltriert und mit 5 X 3 ml Wasser gewaschen. Ausbeute: 1,57 g (71%).

Die orangefarbene Lösung wird eingedampft, der Rückstand wird in 5 ml Wasser gelöst und mit 0,5 g Ammoniumrhodanid versetzt, wobei sich das Salz der Rhodanwassestoffsäure ausscheidet. Es wird abfiltriert, mit 6 X 2 ml Wasser gewaschen und dann getrocknet. 1,48 g Rohprodukt werden erhalten, das bei 132-134°C schmilzt. Nach dem Umkristallisieren aus Isopropanol liegt der Schmelzpunkt bei 142-144°C. Aus dem umkristallisierten Rhodanid kann das reine 5H-2,3-Benbodiazepin-Derivat mit Alkalien oder Ammoniumhydroxyd freigesetzt werden.

Ein Gemisch aus 1,11 g (4,1 mMol) l-Benzal-3,4-dime-thyl-6,7-dimethoxy-dsochromen, 5,5 ml Isopropanol und 0,25 ml (5,2 mMol) 98%igem Hydrazinhydrat wird drei Stunden lang gekocht und dann abgekühlt, wobei sich das Produkt in reiner Form abscheidet.

Das als Ausgangsstoff dienende Isochromen-Derivat wird folgendermassen hergestellt: ein Gemisch aus 2,08 g (10 mMol) a-Methyl-3,4-dimethoxy-phenylaceton. 1,36 g (10 mMol) Phenylessigsäure und 20 g Polyphosphorsäure wird 10 Minuten lang bei 100°C gehalten. Dann wird das viskose Reaktionsgemisch in 100 ml Wasser gelöst, der pH-Wert der gelbbraunen Lösung mit festem Natriumcar-bonat auf 6 eingestellt und dadurch das rohe l-Benzal-3,4--dimethyl-6,7-dimethoxy-isochomen ausgefällt. 2,16 g (73%) Produkt werden erhalten, das nach Umkristallisieren aus Äthanol bai 158-160°C schmilzt.

l-Benzyl-4-methyl-5-äthyl-7,8-dimethoxy-5H-2,3-benzo--diazepin
Man arbeitet auf die im Beispiel 36 angegebene Weise und verwendet als Ausgangsstoff l-Benzal-3-methyl-4-äthyl--6,7-d'imethoxy-isochromen.

Beispiel 38
l-(3,4-Dimethoxyphenyl)-4-methyl-5-äthyl-7-hydroxy-8--methoxy-5H-2,3-benzodiazepin a) Zu der Lösung von 115 g (0,325 Mol) l-(3,4-Dimeth-oxyphenyl)-3-methyl-4-äthyl-7-mehtoxy-6H-2-benzopyran--6-on in 115 ml Eisessig werden bei 80-100°C innerhalb von 5 Minuten unter Rühren 34,5 ml 98%iges Hydrazinhydrat zugetropft. Die Lösung wird auf dem Wasserbad eine Stunde lang bei 95-100°C gehalten, dann mit 140 ml 2%iger Natronlauge verdünnt und abgekühlt. Das 5H-2,3-Benzodia-zepin-Derivat fällt in Form cremefarbener Kristalle aus. Diese werden abfiltriert, mit 4 X 50 ml Wasser gewaschen und d'ann getrocknet. 111,7 g Rohprodukt werden erhalten, das bei 210-212°C schmilzt. Zwecks Reinigung wird das Produkt in 223 ml heissem Dimethylformamid gelöst und die Lösung mit 2 g Aktivkohle geklärt. Nach dem Filtrieren s wird die Aktivkohle mit 3 x 50 ml Dimethylformamid gewaschen und die Waschflüssigkeit der Lösung zugesetzt. Die Lösung wird mit 1300 ml destilliertem Wasser verdünnt, wobei sich d'as reine Produkt kristallin abscheidet. Ausbeute: 110,35 g (94%), Schmp.: 210-212°C. Gaschromatogra-io phisch gemessene Reinheit: über 99%. C21H24N204 = 368,4. das aus Isopropanol umkristallisierte Hydrochlorid (C21H25-N204CI) hat einen Zersetzungspunkt von 218-220°C.
b) Zu der auf 80-100°C erwärmten Suspension von i5 4,35 g (0,01 Mol) l-(3,4-Dimethoxyphenyl)-3-methyl-4-äthyl--6-hydroxy-7-methoxy-2-benzopyriliumbromid in 15 ml 50%iger Essigsäure werden unter Rühren 1,2 ml 98%iges Hydrazinhydrat zugetropft. Dann werden dem Reaktionsgemisch bei 90-100°C 200 ml 10%ige Kochsalzlösung zuge-20 setzt, worauf das Rohprodukt ausfällt. Es wird zur Reinigung in Dimethylformamid oder Äthanol gelöst und dann mit Wasser ausgefällt. Die Ausbeute beträgt 92-95%, der Schmelzpunkt liegt bei 210-212°C. Die Substanz zeigt mit dem gemäss a) hergestellten Produkt keine Schmelzpunktsde-25 pression.
c) Man arbeitet wie unter a) beschrieben, verwendet jedoch statt Eisessig das zehnfache Volumen Methanol und dampft die Lösung nach einstündigem Kochen ein. Der
30 Rückstand wird gegebenenfalls mit Wasser behandelt und dann auf die unter b) beschriebene Weise mit Wasser aus seiner mit Dimethylformamid bereiteten Lösung ausgefällt. Ausbeute: 95%, Schmelzpunkt: 210-212°C.

Aoetylierun'g von l-Aryl-5H-2,3-benzodiazepineö der allgemeinen Formel I, die wenigstens eine phenolische Hydroxylgruppe enthalten.
5 mMol d'er ©ine oder mehrere phenolische Hydroxylgruppen enthaltenden Verbindung der allgemeinen Formel I werden in 5 ml wasserfreiem Pyridin suspendiert. Bei 20-25°C wird zu der Suspension die eineinihalbfache Menge an auf die phenolischen Hydroxylgruppen bezogen notwendigem Essigsäureanhydrid zugegeben. Das Reaktionsgemisch erwärmt sich kaum und wird schnell zu einer klaren Lösung. Diese wird 5-20 Stunden l'ang bei 20-25°C stehen gelassen, dann wird das acetylierte Derivat mit 50 ml Wasser ausgefällt. Das Rohprodukt wird zweckmässig aus Isopropanol oder wässrigem Isopropanol umkristallisiert.
Statt Essigsäureanhydrid kann auch Acetylchlorid verwendet werden, statt Pyridin als Reaktionsmedium kann auch in Triäthylamin oder in Alkalicarbonat enthaltendem Benzol, Aceton oder Dimethylformamid gearbeitet werden.

Ein Gemisch aus 0,44 g l-(4-Methoxy-3-acetoxyphenyl)--4-methyl-5-äthyl-7-acetoxy-8-methoxy-5H-2,3-benzodiaze-pin (Verbindung gemäss Beispiel 47), 8 ml Äthanol, 8 ml Wasser und 0,4 g Kaliumhydroxyd wird 15 Minuten lang unter Stickstoffatmosphäre gerührt, dann werden 50 ml 20%ige Kochsalzlösung und 0,6 g Ammoniumchlorid zugegeben. Der entstehende Niederschlag wird abgetrennt und aus 7,5 ml 20%igem Dimethylformamid umkristallisiert. 0,23 g reines, sich bei 210-212°C zersetzendes l-(4-Methoxy--3-hydroxyphenyl)-4-methyl-5-äthyl-7-hydroxy-8-methoxy--5H-2,3-benzodiazepin werden erhalten. Das Produkt ist mit dem Produkt von Beispiel 41 identisch und zeigt mit diesem keine Schmelzpunktsdepression.

l-(3,4-Dimethoxyphenyl)-5-äthyl-7,8-dimethoxy-5H-2,3--benzodiazepin
Die aus der Verbindung gemäss Beispiel 16 durch Hydrolyse leicht gewinnbare l-(3,4-Dimethoxyphenyl)-5-äthyl--7,8-dimethoxy-2,3-benzodiazepin-4-carbonsäure (Zersetzungspunkt: 133-135°C) wird in kochendem Dimethylformamid schnell decarboxyliert, wobei sich obige Verbindung bildet.

I-(3,4-Dimethoxyphenyl)-4-formyl-5-äthyl-7,8-dimethoxy--5H-2,3-benzodiazepin
Zu der Suspension von 19,1 g (0,05 Mol) l-(3,4-Dimeth-oxyphenyl)-4-methyl-5-äthyl-7,8-dimethoxy-5H~2,3-benzo-dliazepin in 150 ml 80%igem Dioxan werden bei einer In-nentemperatur von 84-87°C unter Rühren innerhalb von 90 Minuten 6,05 g Selendioxyd gegeben. Das metallische Selen wird durch Klären entfernt, die weingelbe Lösung im Vakuum eingedampft und der viskose Rückstand mit 100 ml Wasser vermischt. Dabei fällt das Produkt als gelbes Pulver aus. Es wird mit 5 X 10 ml Wasser gewaschen und im Vakuumschrank getrocknet. 19,3 g (96%) Rohprodukt werden erhalten, das bei 103-105°C schmilzt.

Aus l-(3,4-Dimethoxyphenyl)-4-methyl-5-äthyl-7-hy-droxy-8-methoxy-5H-2,3-benzodiazepin (Beispiel 38) werden durch in an sich bekannter Weise vorgenommenes Alkylieren beziehungsweise Aralkylieren der phenolischen Hydroxylgruppe folgende Verbindungen hergestellt.

Der Dragéekern wird in der üblichen Weise mit Zucker und Talkum überzogen und dann mit Bienenwachs poliert. Das fertige Dragée wiegt 70 mg.
Wie daraus hervorgeht, sind von der erfindungsgemäs-5 sen Verbindung 10 mg als Wirkstoff menge für eine Dosvereinheit ausreichend. Die einzige bekannte ähnliche Verbindung, das Grandaxin, kommt in Form von Tabletten mit 50 mg Wirkstoffgehalt in den Handel.

- 6438352PATENTANSPRÜCHE 1. 5H-2,3 -B enzodiazepin-Deri vate der allgemeinen Formel I(I)worinR für Wasserstoff, Alkylgruppe mit 1-5 Kohlenstoffatomen, Aminogruppe, Dialkylaminoalkyl-, Alkylamino-, Dialkylaminogruppe mit 1-5 Kohlenstoffatomen in jedem Alkylteil, für unsubst'ituierte oder durch 1-3 Halogenatome, Hydroxyl-, Acyloxy-, Methyl-, Trifluormethyl-, Nitro-, Amino-, Methylend'ioxygruppe, Alkoxygruppe mit 1-4 Kohlenstoffatomen und/oder Aralkoxygruppe substituierte Aryl-gruppe, Styrylgruppe oder Phenylalkylgruppe mit 1-3 Kohlenstoffatomen im Alkylteil oder für 1 oder 2 Stickstoff-, Sauerstoff- und/oder Schwefelatome enthaltende heterocy-clische Gruppe steht,R1 Wasserstoff, Alkylgruppe mit 1-4 Kohlenstoffatomen, Hydroxymethyl-, Formyl-, Carboxyl-, Carbalkoxy-, Arylgruppe oder heterocyclische Gruppe bedeutet,R2 für Wasserstoff, Alkylgruppe mit 1-4 Kohlenstoffatomen, Dialkylaminoalkyl-, Alkylamino- oder Dialkylaminogruppe m!it 1-4 Kohlenstoffatomen in jedem Alkylteil oder für Arylgruppe steht,R3 und R4 zusammen eine Methylendioxygruppe bilden oder aberR3 und R4 unabhängig voneinander für Wasserstoff, Halogen, Nitro-, Amino-, Hydroxyl-, Acyloxygruppe, für Alkylgruppe mit 1-3 Kohlenstoffatomen, Alkoxygruppe mit 1 bis 5 Kohlenstoffatomen, für Dialkylaminoalkylgruppe oder Aralkoxygruppe stehen,mit der Einschränkung, dass, für den Fall R = 3,4-Dimeth-s oxyphenylgruppe, R1 = Methylengruppe und R2 = Äthylgruppe, R3 und R4 eine andere Bedeutung als Methoxy-gruppe haben, sowie ■die Säureadditionssalze dieser Verbindungen.
- 2. Verfahren zur Herstellung von 5H-2,3-Benzodiaze-10 pin-Derivaten nach Anspruch 1 sowie der Säureadditionssalze dieser Verbindungen, dadurch gekennzeichnet, dass man ein 1,5-Diketon der Formel II(II)oder e'in 2-Benzopyriliumsalz der Formel III25 2(IH)woranX- für Chlorid-, Bromîd-, Jodid-, Tetrafluorborat-, Te-tfachlorferrat-, Hexachlorstannat-, Hydrogensulfat-, Dihy-drogenphosphat- oder Perchlorat-Anion steht, oder « ein 6H-2-Benzopyran-6-on der Formel IV(IV)Roder ein Isochromen der Formel V60(V)3643835worinR5 und R6, unabhängig voneinander, für Alkylgruppe mit 1-3 Kohlenstoffatomen oder eine beliebige der für R angegebenen Bedeutungen stehen,in einem polaren Lösungsmittelmedium mit dem 1 bis 5fachen der äquimolaren Menge von Hydrazin, Hydrazin-hydrat oder einem C1_3-Carbonsäurehydrazinsalz umsetzt, im Falle von als Substituenten R eine Arylgruppe oder heterocyclische Gruppe enthaltenden Ausgangsverbindungen der Formel II dem Reaktionsgemisch noch eine Mineralsäure zusetzt, und die erhaltene Verbindung in ihre physiologisch verträglichen Säureadditionssalze überführt bzw. aus den Säureadditionssalzen der Verbindungen der Formel I die Base freisetzt oder andere Säureadditionssalze bildet.
- 3. Verfahren nach Anspruch 2, dadurch gekennzeichnet, dass man das Ci_3-Carbonsäurehydrazinsalz für die Umsetzung in Situ bildet.
- 4. Verfahren zur Herstellung von Verbindungen der Formel I gemäss Anspruch 1, worin R und/oder R3 und R4 Alkoxy oder Aralkoxy bedeuten, dadurch gekennzeichnet, dass man nach dem Verfahren gemäss Anspruch 2 oder 3 eine Verbindung der Formel I hergestellt, in der R und/oder R3 und R4 für die Hydroxylgruppe steht und diese al-kyliert oder aralkyliert.
- 5. Verfahren nach einem der Ansprüche 2 bis 4, dadurch gekennzeichnet, dass man die Umsetzung der Aus-gangsverbindüngen mit Hydrazin oder Hydrazinhydrat in einem polaren Lösungsmittel, vorzugsweise in Wasser, Alkoholen mit 1 bis 4 Kohlenstoffatomen, Carbonsäuren mit 1 bis 3 Kohlenstoffatomen, Dioxan, Dimethylformamid, Pyridin oder Gemischen der aufgeführten Lösungsmittel vornimmt.
- 6. Verfahren nach einem der Ansprüche 2 bis 5, dadurch gekennzeichnet, dass man die Umsetzung der Ausgangsstoffe der allgemeinen Formeln II-V mit Hydrazin, Hydrazinhydrat oder einem C^-Carbonsäurehydrazinsalz bei Temperaturen zwischen —20 und + 120°C, vorzugsweise 60-100°C, vornimmt.
- 7. Verfahren nach einem der Ansprüche 2 bis 6, dadurch gekennzeichnet, dass man bei Ausgangsverbindungen der allgemeinen Formel III als Säureakzeptor Alkalihydroxyde, Alkalicarbonate, Alkalihydrogencarbonate oder organische Basen, vorzugsweise Pyridin oder Triäthylamin, verwendet.
- 8. Verfahren nach einem der Ansprüche 2 bis 7, dadurch gekennzeichnet, dass man zur Reinigung der Verbindungen der allgemeinen Formel I deren gut kristallisierende Salze, vorzugsweise die mit Rhodanwasserstoffsäu-re oder Salzsäure gebildeten Salze, herstellt.
- 9. Verfahren n'ach einem der Ansprüche 2 bis 8, dadurch gekennzeichnet, dass man die Ausgangsverbindungen der allgemeinen Formeln II-V vor dem Umsetzen mit der Hydrazinkomponente vorzugsweise ohne Isolierung ineinander überführt.
- 10. Pharmazeutische Präparate, dadurch gekennzeichnet, dass sie Verbindungen der allgemeinen Formel I, worin die Bedeutung von R, R1, R2, R3, R4 die gleiche wie in Anspruch 1 ist, oder deren physiologisch verträgliche Säureadditionssalze enthalten.Die erfindungsgemässen 5H-2,3-Benzdiazepin-Derivate entsprechen der allgemeinen Formel IIa15 worinR für Wasserstoff, Alkylgruppe mit 1-5 Kohlenstoffatomen, Aminogruppe, Dialkylaminoalkyl-, Alkylamino-, -Dialkylamin'ogruppe mit 1-5 Kohlenstoffatomen in jedem Alkylteil, für unsubst'ituierte oder durch 1-3 Halogenatome, 2o Hydroxyl-, Acyloxy-, Methyl-, Trifluormethyl-, Nitro-, Amino-, Methylendioxygruppe, Alkoxygruppe mit 1-4 Kohlen-stoffatomen und/oder Aralkoxygruppe substituierte Arylgruppe, Styrylgruppe oder Phenylalkylgruppe mit 1-3 Kohlenstoff atomen im Alkylteil oder für 1 oder 2 Stickstoff-, 25 Sauerstoff- und/oder. Schwefelatome enthaltende heterocyclische Gruppe steht,R1 Wasserstoff, Alkylgruppe mit 1-4 Kohlenstoffatomen, Hydroxymethyl-, Formyl-, Carboxyl-, Carbalkoxy-, Arylgruppe oder heterocyclische Gruppe bedeutet, 30 R2 für Wasserstoff, Alkylgruppe mit 1-4 Kohlenstoffatomen, Dialkylaminoalkyl-, Alkylamino- oder Dialkylami-nogruppe mit 1-4 Kohlenstoffatomen in jedem Alkylteil oder für Arylgruppe steht,R3 und R4 zusammen eine Methylendioxygruppe bilden 35 oder aberR3 und R4 unabhängig voneinander für Wasserstoff, Halogen, Nitro-, Amino-, Hydroxy- oder Acyloxygruppe, für Alkylgruppe mit 1-3 Kohlenstoff atomen, Alkoxygruppe mit 1 bis 5 Kohlenstoffatomen, für Dialkylaminoalkylgrappe oder 40 Aralkoxygruppe stehen,mit der Einschränkung, dass für den Fall R = 3,4-Dimeth-oxyphenylgruppe, R1 = Methylgruppe und R2 = Äthylgruppe, R3 und R4 eine andere Bedeutung als Methoxy-gruppe haben.
