Description
55 Die Erfindung betrifft neue Cephalosporine sowie Verfahren zur Herstellung dieser Verbindungen.
Die erfindungsgemässen Verbindungen sind Zwischenprodukte bei der Herstellung von 7a-Methoxycephalosporine der Hauptanmeldung und haben ein breites antibakterielles 60 Spektrum gegen Gram-positive und Gram-negative Bakterien; im besonderen sind sie wirksam gegen Pseudomonas aeruginosa, Klebsiella pneumoniae und Proteus species, welche sich als Ursachen für klinisch äusserst ernste Infektionskrankheiten herausgestellt haben. Darüber hinaus sind 65 die erfindungsgemässen Verbindungen gegenüber ß-Lacta-masen von Bakterien stabil. Sie sind daher äusserst brauchbar als Zwischenprodukte und zur Behandlung verschiedenster Infektionskrankheiten.

Der Ausdruck «Acyl» bedeutet im Sinne der Beschreibung eine Acyl-Gruppemit 1 bis 10 Kohlenstoffatomen, wie Acetyl, Pro-pionyl, Buturyl, Benzoyl, Naphthoyl, Bentancarbonyl, Cyclohexancarbonyl, Furuyl, Thenoyl oder dgl. Der Ausdruck « Acyloxy» bedeutet im Sinne der Beschreibung eine einwertige Acyl-O-Gruppe, in der die Acyl-Gruppe die oben definierte Bedeutung hat und die über ein Sauerstoffatom gebunden ist. Das heisst, es handelt sich um Acyloxy-Gruppen mit 1 bis 10 Kohlenstoffatomen. Wenn der Ausdruck «Acyl» oder «Acyloxy» sich auf eine Gruppe mit einem heterocyclischen Ring, welcher N, O und/oder S in jeder beliebigen Anzahl in jeder beliebigen Position des Rings enthält, so werden die Heteroatome in die Anzahl der Kohlenstoffatome eingerechnet.
In den allgemeinen Formeln bedeutet R1 ein Wasserstoffatom oder eine auf dem Gebiet der Penicilline und Cephalosporine bekannte carboxyl-schützende Gruppe. Als solche carboxyl-schützende Gruppe kommen in Sinne der Erfindung solche in Frage, welche auch bisher schon auf dem Gebiet der Penicilline und Cephalosporine eingesetzt wurden, insbesondere esterbildende Gruppen, welche durch katalytische Reduktion, chemische Reduktion oder andere Behandlungen unter milden Bedingungen entfernt werden können; esterbildende Gruppen, welche leicht im lebenden

Körper entfernt werden können; andere bekannte esterbildende Gruppen, welche leicht durch Behndlung mit Wasser oder einem Alkohol entfernt werden können, z.B. organische Silyl-Gruppen, organische phosphorhaltige Gruppen, organische zinnhaltige Gruppen oder dgl.
Beispiele geeigneter carboxyl-schützender Gruppen sind
(a) Alkyl-Gruppen,

Die aufgezählten carboxyl-schützenden Gruppen sind typische Beispiele der verwendbaren Gruppen. Allgemein kann man alle Gruppen verwenden, welche in den US-Patentschriften 3 499 909; 3 573 296; 3 641 018 und in den DT-OSS 2 301 014; 2 253 287 und 2 337 105 beschrieben sind.
R2 bedeutet in den allgemeinen Formeln eine Acetoxygruppe, eine Carbamoyloxygruppe oder eine mit einem substituierten oder unsubstituierten heterocyclischen Ring verknüpfte Thiogruppe. Als Salze der erfindungsgemässen Cephalosporine der allgemeinen Formel (I) kommen Salze der Säure-Gruppe in Frage sowie Salze der basischen Gruppe. Beide Salz-Typen sind auf dem Gebiet der Penicilline und der Cephalosporine bekannt. Unter diesen Salzen sind pharmazeutisch verträgliche Salze bevorzugt. Salze der Säure-Gruppe umfassen Salze mit Alkalimetallen, wie Natrium, Kalium oder dgl.; mit Erdalkalimetallen, wie Calcium, Magnesium oder dgl.; mit Ammonium; mit stickstoffhaltigen organischen Basen, wie Procain, Dibenzylamin, N-Benzyl-ß-phenäthylamin, 1-Ephenamin, N,N-Dibenzyläthylendiamin, Trimethylamin, Triäthylamin, Tributylamin, Pyridin, Dimethylanilin, N-Methylpiperidin, N-Methylmorpholin, Diäthylamin und Dicyclohexylamin.
Salze an der Basen-Gruppe umfassen Salze mit Mineralsäuren, wie Salzsäure, Schwefelsäure oder dgl.; mit organischen Carbonsäuren, wie Oxalsäure, Ameisensäure, Trichlor-essigsäure, Trifluoressigsäure oder dgl.; mit organischen Sul-fonsäuren, wie Methansulfonsäure, Toluolsulfonsäure, Naphthalinsulfonsäure oder dgl.
Von vorliegender Erfindung werden alle optischen Isomeren und Racemate umfasst sowie alle Kristallformen und Hydrate der Cephalosporine der allgemeinen Formel (I) sowie Salze derselben.
Die Verbindungen der allgmeinen Formel (I) oder die Salze derselben können nach bekannten Verfahren hergestellt werden, z.B. nach dem unten beschriebenen Verfahren.

Im folgenden soll das Herstellungsverfahren näher erläutert werden.
Die Verbindung (II) kann beispielsweise in einem inerten Lösungsmittel, wie in Wasser, Aceton, Tetrahydrofuran, 5 Dioxan, Acetonitril, Dimethylforamid, Dimethylacetamid, Methanol, Äthanol, Methoxyäthanol, Diäthyläther, Diiso-propyläther, Benzpl, Toluol, Methylenchlorid, Chloroform, Äthylacetat oder Methylisobutylketon, und zwar in einem dieser Lösungsmittel oder in einem Gemisch von zweien oder 10 mehreren dieser Lösungsmittel aufgelöst oder suspendiert werden. Die erhaltene Lösung oder Suspension wird sodann mit der Verbindung (III) oder mit einem reaktiven Derivat der Carboxylgruppe der Verbindung (III) versetzt. Das Gemisch wird beispielsweise in Anwesenheit einer Base bei 15 —60 bis 80°C, vorzugsweise —40 bis 30°C zur Reaktion gebracht. Eine Reaktionszeit von 5 min bis 5 h reicht im allgemeinen aus.
Als Basen bei obiger Reaktion kommen anorganische Basen in Frage, z.B. Alkalihydroxide, Alkalihydrogencarbo-20 nate, Alkalicarbonate und Alkaliacetate; tertiäre Amine, wie Trimethylamin, Triäthylamin, Tributylamin, Pyridin, N-Methylpiperidin, N-Methylmorpholin, Lutidin, Collidin usw. und sekundäre Amine, wie Dicyclohexylamin, Diäthyl-amin usw. Eine Base muss nicht unbedingt vorhanden sein. 25 Wenn man die Verbindung (III) oder ein Salz derselben als Ausgangsmaterial einsetzt, so kann die Reaktion in Anwesenheit eines dehydratisierenden Kondensationsmittels durchgeführt werden, z.B. von N,N'-Dicyclohexylcarbo-diimid, N-Cyclohexyl-N' -morpholinoäthylcarbodiimid, 30 N,N'-Diäthylcarbodiimid, N,N'-Carbonyl-bis(2-methylimi-dazol), Trialkylphosphite, Äthylpolyphosphat, Phosphor-oxychlorid, Phosphortrichlorid, 2-Chlor-l,3,2-dioxapho-spholan, Oxazolylchlorid, Dimethylchlorforminiumchlorid und Dimethyläthoxyforminiumchlorid.
35 Man kann eine herkömmliche Verfahrensweise anwenden, wenn man eine Verbindung der allgemeinen Formel (I), in der R' eine die Carboxyl-Gruppe schützende Gruppe ist, in eine Verbindung der allgemeinen Formel (I) umwandeln will, in der R1 ein Wasserstoffatom ist, oder in ein Salz der letz-40 teren Verbindung oder wenn man eine Verbindung der allgemeinen Formel (I), in der R1 ein Wasserstoffatom ist, in ein Salz derselben umwandeln will oder in eine Verbindung der allgemeinen Formel (I), in der R1 eine die Carboxyl-Gruppe schützende Gruppe ist oder wenn man ein Salz einer Verbin-45 dung der allgemeinen Formel (I) in die freie Säure umwandeln will.
Wenn man eine Verbindung umsetzen will, in der die Gruppe A, B oder R2 reaktiv ist, so kann man die reaktive Gruppe mit einer Schutzgruppe schützen, welche gewöhnlich 50 zum Schutz von Carboxyl-Gruppen, Amino-Gruppen oder Hydroxyl-Gruppen eingesetzt wird. Nach der Reaktion kann eine solche Schutzgruppe in herkömmlicher Weise entfernt werden, um die Gruppe A, B oder R2 wieder zu regenerieren. Die Herstellu.ngsbedingungen sind nicht auf die oben ange-55 gebenen Bedingungen beschränkt.
Sie können vielmehr je nach dem speziellen Typ der Reaktionskomponenten modifiziert werden.
Die Isolierung des Cephalosporins (I) oder des Salzes desselben vom Reakationsgemisch kann in üblicher Weise 60 erfolgen.
Unter den verschiedenen Cyclosporinen dieser Erfindung sind diejenigen bevorzugt, welche eine Acetoxy-, eine substituierte 5-(l,2,3,4-Tetrazolyl)-tio- oder 5-(l,3,4-Thiadia-zolyl)thiogruppe an der Stelle des Restes R2 der Formel (I) 65 tragen.
Die Cephalosporine dieser Erfindung finden Verwendung als Zwischenprodukte bei der Herstellung entsprechender 7a-Methocycephalosporine und zur Behandlung von

Erkrankungen auf dem Gebiet der Humanmedizin und dem Gebiet der Tiermedizin. Die erfindungsgemässen Cephalosporine der allgemeinen Formel (I) werden bei Menschen und Tieren in Form der freien Säuren oder in Form von pharmazeutisch verträglichen Salzen oder Estern verabreicht. Die
Verbindungen können in verschiedenen Applikationsformen hergestellt und verabreicht werden, welche auf dem Penicillin- und Cephalosporingebiet üblich sind.
In Tabelle 1 sind die antibakteriellen Aktivitäten der reprä-s sentativen Verbindungen dieser Erfindung angegeben.

Die Zahlen in Tabelle 1 geben die minimalen Hemmkonzentrationen (MIC, angegeben in mcg/ml) der jeweiligen Verbindung an. Diese wird bestimmt nach dem Verfahren in "Chemotherapy (Society of Chemotherapy, Japan), Band 16, 98-99 (1968)". Hierzu wird das Testbakterium in einer Heart-Infusionsbrühe (Eiken Kagaku Co.) kultiviert und mit der erhaltenen Kultur wird ein Heart-Infusionsagarmedium (Eiken Kagaku Co.) geimpft. Nach einer 20stündigen Inku-20 bation bei 37°C wird der Wuchszustand der Bakterien untersucht, um die minimale Hemmkonzentration (MIC angegeben in mcg/ml) festzustellen. Die Impfrate beträgt 104 Zellen/Platte (106 Zellen/ml).

Im folgenden wird die Herstellung der erfindungsgemässen Verbindungen anhand von Beispielen näher erläutert. Prozent-Angaben beziehen sich, wenn nichts anderes angegeben ist, auf das Gewicht.
Beispiel 1
(1) In 15 ml wasserfreiem Methylenchlorid werden 2,0 g a-(4-Äthyl-2,3-dioxo-l-piperazincarboxamido)-a-(2-trichlor-
äthoxycarbonylamino-4-thiazolyl)-essigsäure aufgelöst. Zu der Lösung gibt man bei —20°C 0,43 ml N-Methylmorpholin.
Zu dem erhaltenen Gemisch gibt man tropfenweise über eine Zeitdauer von 5 min eine Lösung von 0,41 ml Äthyl-65 chlorcarbonat in 2 ml Methylenchlorid, wobei die Temperatur auf —20 bis —25°C gehalten wird. Die Reaktion wird während 1,5 h bei der gleichen Temperatur durchgeführt. Zum Reaktionsgemisch gibt man tropfenweise bei -15 bis

-20°C über eine Zeitdauer von 15 min eine Lösung von 2,0 g stellt. Die organische Phase wird abgetrennt, mit Sole gewa-Diphenylmethyl-7-amino-3-[5-(l -methyl-1,2,3,4-tetra- sehen, über Magnesiumsulfonat getrocknet und unter ver-

Bei der Verbindung der oberen Zone handelt es sich vermutlich um Diphenylmethyl-7-[D-a-(2-Trichloräthoxycarbo- (3) 0.2 g Diphenylmethyl-7-[D-a-(2-amino-4-thiazolyl)-a-(4-nylamino-4-thiazolyl)-a-(4-äthyl-2,3-dioxo-1 -piperazincar- äthyl-2,3-dioxo-1 -piperazincarboxamido)acetamido]-3-[5-(l -boxamido)-acetamido]-3-f5-(l-methyl-1,2,3,4-tetrazolyl)thio- methyl-1,2,3,4-tetrazolyl)thiomethyl]-A3-cephem-4-carbo-methyl]-A3-cephem-4-carboxylat. 35 xylat aus (1) werden in 2.0 ml Anisol gelöst. (1) 0,8 g der in Beispiel 1-(1) erhaltenen oberen Verbindung werden in 15 ml 85%iger Ameisensäure gelöst.
Nachdem man unter Eiskühlung 0,4 g Zn-Pulver hinzugefügt IR (KBr) cm-1; v C = O 1765, 1700,1670 hat, rührt man noch 3 Stunden bei dieser Temperatur. Nach NMR(DMSO-d6) S-Werte: 1.11 (3H, t, -CH3), 3.30-4.10 Abfiltrieren des Ungelösten wird das Filtrat am Vakuum ein- 65 (8H, m, ^CHa x 4), 3.94 (3H, s, -CH3), 4.28,4.33 (2H, geengt. Zum erhaltenen Rückstand werden 10 ml Methy- ABq, Z>CH2), 5.08 (1H, d, ^CH), 5.48 (1H, d, SSsCH), lenchlorid, 5 ml Aceton und 10ml Wasser hinzugefügt und 5.65(lH,dd, 5&CH), 6.60 (1H, s, ^^CH), 6.80 (1H, bs, der pH mit gesättigter Natriumbicarbonatlösung auf 7 einge- -COOH), 9.26(1 H,d, ^>NH), 9.64 (1H, d, ^>NH)

(5) 0.15 g des in (3) erhaltenen Trifluoressigsäuresalzes von 7-[D-cc-(2-Amino-4-thiazolyl)-a-(4-äthyl-2,3-dioxo-1 -piperazincarboxamido)acetamido]-3-[5-(l-methyl-l, 2,3,4-tetrazolyl)thiomethyl]-A3-cephem-4-carbonsäure werden in 3 ml 50% wässrigem Aceton gelöst und mit 0.018 g (äquimo-lare Menge) Pyridin versetzt. Nach Abdampfen des Lösungsmittels wird Wasser zugegeben und filtriert. Man erhält so kristallines 7-[D-a-(2-Amino-4-thiazolyl)-a-(4-äthyl-2,3-dioxo-1 -piperazincarboxamido)acetamido]-3-[5-( 1 -methyl-1,2,3,4-tetrazolyl)thiomethyl]-A3-cephem-4-carbonsäure.
Auf ähnliche Weise erhält man die folgenden Verbindungen:
7-[D-a-(2- Amino-4-thiazolyl)-a-(4-äthyl-2,3-dioxo-1 -piper-azincarboxamido)acetamido]-3-[5-(l-carbamoylmethyl-s 1,2,3,4-tetrazolyl)thiomethyl]-A3-cephem-4-carbonsäure. 7-[D-a-(2-Amino-4-thiazolyl)-a-(4-äthyl-2,3-dioxo-l-piper-azincarboxamido)acetamido]-3-acetoxymethyl-A3-cephem-4-carbonsäure.
7-[D-a-(2-Amino-4-thiazolyl)-a-(4-äthyl-2,3-dioxo-l-piper-lo azincarboxamido)acetamido]-3-[2-(5-methyl-1,3,4-thiadia-zolyl)thiomethyl]-A3-cephem-4-carbonsäure.

Cephalosporine der folgenden allgemeinen Formel (I) oder Salze derselbenO 0A-N N-CONH-CH-CONH 1—^ B T1»(R )n COOR1ch2r(I)wobei R1 ein Wasserstoffatom oder eine auf dem Gebiet der Penicilline und Cephalosporine bekannte carboxyl-schüt-zende Gruppe; R2 eine Acetoxygruppe, eine Carbamoyloxy-gruppe oder eine mit einem substituierten oder unsubstitu-ierten heterocyclischen Ring verknüpfte Thio-Gruppe; R3 eine niedere Alkylgruppe; n 0,1 oder 2; A ein Wasserstoffatom oder eine substituierte oder unsubstituierte Alkylgruppe is und B eine Gruppe der FormelN4R NIUJoder deren TautomeresRHN-I'tTJl bedeutet, wobei R4 ein Wasserstoffatom oder eine auf dem oder eine 2-(5-Methyl-l,3,4-thiadiazolyl)thio-GruppeGebiet der Penicilline und Cephalosporine bekannte amin- bedeutet.schützende Gruppe und R5 ein Wasserstoffatom oder eine auf 3. Cephalosporine nach Anspruch 2, wobei R4 und R5 ein dem Gebiet der Penicilline und Cephalosporine bekannte 25 Wasserstoffatom bedeuten.imin-schützende Gruppe bedeuten. 4.
