Dai dati di letteratura è noto che la L-alanina è importante per la funzione dei T linfociti, perchè è essenziale per la risposta in vitro di queste cellule agli stimoli mitogenici (Rotter V. et al.: J. Immunol. 123, 1726,1979), La L-alanina inoltre è anche essenziale alla crescita in vitro di linfociti (Nordlind K. et al.: Int. Archs Allergy Appi. Immunol. 59, 215, 1979). Dati dai nostri laboratori, comunque, documentano che questo amminoacido ha solamente una attività immunostimolante molto debole, come è mostrato nel test in vitro di induzione di Thy 1.2 in cellule T immature di topi normali.
Da dati di letteratura risulta che anche la L-arginina è dotata di attività immunostimolante sia in vitro che in vivo, in particolare in animali stressati o nei quali siano state provocate lesioni (Barbul A. et ài, J. Surg. Res. 29, 228,1980; J. Parenteral Enterai Nutr. 4, 446,1980; ibid. 5,492,1981) ed in tumori indotti sperimentalmente (Rettura G.: J. Parenteral Enterai Nutrition 3,409,1979).
Nel nostro test di induzione di Thy 1.2, anche la L-arginina mostra una attività scarsa, benché statisticamente significativa.
Abbiamo sintetizzato un tripeptide avente la sequenza Arg-Ala-Arg ed abbiamo confrontato la sua attività nel nostro test con quella della miscela dei due singoli amminoacidi nel rapporto molare 2Arg:1 Ala. II tripeptide risulta essere di gran lunga più attivo della miscela dei due amminoacidi, infatti esso induce il 13% di cellule (P<0.01), mentre la miscela solo il 5% (statisticamente non significativo).
La scelta di mettere la L-arginina in entrambe le posizioni terminali del tripeptide è basata sul fatto che, in molti peptidi immunostimolanti noti, questo amminoacido occupa o la posizione N-terminaie (come nel caso della timopentina) o la C-terminale (per es. in tuftsina, ubiquitina, timopoietina).

ANALISI HPLC
Il tripeptide è stato analizzato mediante HPLC a coppia ionica, secondo le condizioni di separazione qui descritte:
Eluente: NaH2P04 0.05M pH 4.3 + sds 5x10-4 m, MeOH; 50 :50.
Flusso: 1 ml/min.
Lunghezza d'onda: 225 nm.
Volume iniettato: 20 mei.
Campione: 20 mcg.
Colonna: u Bondapack C18 (WATERS), 300 x 3.9 mm.
La strumentazione usata è stata la seguente:
Cromatografo liquido: SERIES 4 (PERKIN ELMER).
Valvola di iniezione: Reodyne mod. 7125-075 con loop da 20 ul.
Rivelatore: Spettrofotometro LC 95 (PERKIN ELMER).
Integratore computerizzato: Data Station 3600 (PERKIN ELMER).
Il profilo del tripeptide in HPLC è riportato nella figura.
RESISTENZA ALL'AMBIENTE GASTRICO SIMULATO IN VITRO.
Il tripeptide è risultato resistente all'ambiente gastrico simulato in vitro, in cui si è utilizzata la soluzione di fluido gastrico simulato USP XXI (HCl + pepsina) a T 37 per 5 ore.
METODO DI SINTESI

Boc-Ala-Arg-OBe (1)
Alla Boc-AIa (0.1 mole) disciolta in cloruro di metilene e raffreddata a 0°C +/-1 è stato aggiunto isobutil-cloroformiato (0.1 mole) sotto agitazione ed abbassando la temperatura a -15°C. Dopo aver mantenuto la miscela di reazione in agitazione a -15°C per 15 minuti, è stata aggiunta lentamente una soluzione preraffreddata di NG-Nitro-Arginina-benzil estere di-p-tosilato (0.1 mole) e N-metil-morfolina (NMM) (0.2 mole) in dimetiiformamide. La miscela di reazione è stata lasciata sotto agitazione per una notte.
[ solventi sono stati rimossi per evaporazione a pressione ridotta ed il residuo ottenuto ripreso in acetato di etile.
L'acetato di etile è stato lavato, in fasi successive, con acqua, con acido cloridrico 1N, con una soluzione acquosa di sodio bicarbonato al 5%, e ancora con acqua, quindi anidrificato su sodio solfato. Il solvente è stato rimosso per evaporazione a pressione ridotta ed il prodotto ottenuto, un liquido molto den2

so, è stato cromâtografato mediante TLC con il sistema CHC13:MeOH:AcOH (90:8:2). La purezza era del 95% e le rese deli'80%.
Il gruppo Boc è stato rimosso mediante trattamento con la miscela acido trifluoroacetico 50%: cloruro di metilene 1:1,10 mi per g, per 30 min. La miscela di reazione è stata evaporata a pressione ridotta, ed il residuo ottenuto è stato triturato in etere etilico, filtrato, lavato con etere ed essicato sotto vuoto. Le rese sono state del 98%.
Il prodotto ottenuto TFA-Ala-Arg-OBe è stato neutralizzato con NMM e fatto reagire con Z3-Arg disciolta in dimetilformamide-tetraidrofurano, usando NMM e isobutilcloroformiato e operando nelle stesse condizioni descritte per (1). Rese 60%.
Da un controllo fatto con cromatografia su strato sottile con il sistema cloroformio:metanolo (92:8) è risultata una sola macchia principale.
Il tripeptide è stato idrogenato con Pd/C in acido acetico-acqua-metanolo. A fine reazione, la sospensione è stata filtrata per rimuovere il catalizzatore ed il filtrato è stato evaporato sotto vuoto.
Il prodotto ottenuto, il tripeptide, è stato purificato mediante distribuzione in contro corrente nel sistema n-butanolo: acido acetico: acqua (4:1:5) con rese del 50%.
Dalla cromatografia su strato sottile con sistema butanolo:acido acetico:acqua:piridina (32:6:22:20) è risultato una macchia principale e dal controllo HPLC una purezza del 97%.
ATTIVITÀ' BIOLOGICHE
1. a induzione in vitro dell'antigene thy 1.2
La capacità di Arg-Ala-Arg di indurre in vitro il differenziamento dei precursori delle cellule T del topo in linfociti esprimenti markers T cellulari è stata provata evidenziando l'induzione dell'antigene di membrana Thy 1.2.
MATERIALI E METODI
TOPI: topi nudi (nu/nu) SPF dell'età di 8 settimane.
PREPARAZIONE DELLE CELLULE: le milze sono state rimosse in condizioni asettiche, sminuzzate e passate attraverso un setaccio in acciaio inossidabile a maglie fini in una soluzione salina bilanciata di Hank (HBSS).
Gli splenociti, lavati e risospesi in mezzo di coltura 199 (Gibco Ltd) supplementato con l'1% di BSA (Boehringer Mannheim) e con gentamicina (100 mcg/ml), sono stati incubati per 45 min. in colonne di lana di nylon equilibrate secondo il metodo di Julius et al. (Eur. J. Immunol: 3,645,1973).
Sono state usate le popolazioni cellulari effluenti arricchite in precursori delle cellule T.
INDUZIONE: 0,5x106 cellule effluenti in 0,1 mi di mezzo di coltura, sono state incubate a 37°C per 18 ore, con o senza il tripeptide. Le colture sono state eseguite in duplicato.
Al termine dell'incubazione, le cellule sono state lavate con Io 0,87% di ammonio cloruro per lisare gli eritrociti, poi con HBSS.
L'induzione dell'antigene di membrana Thy 1.2 è stata determinata mediante immunofluorescenza diretta.
IMMUNOFLUORESCENZA DIRETTA: le cellule sono state incubate a 4°C per 20 min. con anticorpo monoclonale anti Thy 1.2 coniugato con fluoresceina (Bio-Yeda) alla diluizione di 1:200. La miscela è stata centrifugata a 300 g per 5 min., lavata 2 volte in HBSS e poi sospesa per il conteggio al microscopio a fluorescenza (Leitz Orthoplan).
La differenza in percentuale di cellule fluorescenti tra colture con e senza tripeptide, ha dato l'attività inducente del prodotto,
RISULTATI: come mostrato nella tabella, il tripeptide induce la comparsa del marker Thy 1.2 in cellule T immature con una risposta ottimale a 10 mcg/ml. La curva dose-risposta è a forma di campana, poiché concentrazioni maggiori e minori di 10 mcg/ml provocano una Induzione inferiore.

1.B INDUZIONE DELL'ANTIGENE THY 1.2 IN VIVO
Il tripeptide è stato somministrato per 4 giorni consecutivi e i topi sono poi stati sacrificati 24 ore dopo l'ultima somministrazione. Le milze sono state rimosse e le cellule esaminate per l'espressione dell'antigene Thy 1.2 mediante fluorescenza. I topi di controllo hanno ricevuto il Medium 199 (M199), il mezzo in cui il farmaco è stato dissolto.
I topi avevano un peso medio di circa 24 g.

I dati mostrano che il tripeptide ELS2 è in grado di indurre la maturazione degli splenociti sia dopo somministrazione orale che intraperitoneale.
II dosaggio ottimale è di 1055 ug/kg mentre con dosaggi più aiti si osserva un andamento a plateau della curva dose-risposta.
2. STIMOLAZIONE IN VITRO DELLA PRODUZIONE DI LINFOCHINE MATERIALI E METODI
PREPARAZIONE DELLE CELLULE MONONUCLEATE DI SANGUE UMANO PERIFERICO (PBMC). Il sangue venoso periferico viene ottenuto da volontari sani.
Gli eritrociti sono separati dalle cellule bianche mediante gradienti Ficoll-Hipaque. Il «buffy eoat» (PBMC) viene rimosso e lavato, e le cellule sono risospese ad una concentrazione di 1x106 celtule/ml in RPMI, addizionato con I'1% di penicillina/streptomicina, 1*1% di glutamina e l'1% di siero di feto di vitello inattivato riscaldato (FCS, 56 C, 30 min.).
PREPARAZIONE DEL FATTORE Di CRESCITA.
Le PBMC, alla concentrazione di 1x106 cellule /mi in FCS 1% inattivato riscaldato, vengono incubate con o senza fitoemaggiutinina (PHA) alla concentrazione di 0.75% (v/v).

II tripeptide viene aggiunto alla concentrazione di 1 ug/ml alle colture. Il periodo di incubazione è di 18— 24 ore a 37°C in atmosfera umidificata. Le colture vengono poi filtrate attraverso filtri con porosità 0.22 nM e i supernatanti vengono esaminati per la presenza dei fattori di crescita.
MISURAZIONE DEI FATTORI DI CRESCITA NEI SUPERNATANTI.
A. Cellule usate nei test.
Le cellule B usate per esaminare la presenza del fattore di crescita delle cellule B (BCGF) appartengono a culture di linee cellulari a lungo termine, mantenute con BCGF e sono EBV-negatìve. Queste cellule sono cresciute in mezzo privo dì siero usando Nutridoma (Boehringer Mannheim Biochemicals), e non rispondono alla IL—2.
Le cellule T usate per esaminare la presenza di IL—2 sono isolate di fresco. Esse vengono inizialmente stimolate con PHA (0.75%) e vengono mantenute in cultura almeno per 10 giorni prima dell'uso (per ridurre il «background» e stabilire la loro dipendenza alla IL—2).
B. Preparazione delle cellule per l'utilizzazione nel saggio.
1. Le cellule B sono di solito utilizzate 4 giorni dopo l'ultimo trattamento con BCGF. Vengono lavate 4 volte con RPM11640 per rimuovere ogni traccia di BCGF e sospese alla concentrazione di 15x104 cellule/m! in RPM11640 e Nutridoma (alla concentrazione finale dell'1%,)
2. Le cellule T vengono utilizzate 4 giorni dopo l'ultimo trattamento con IL—2. Vengono lavate 4 volte e sospese alla concentrazione di 50x104 cellule/ml in RPMI coi 5% di FCS.
C. Procedura del saggio.
1. Le cellule B coltivate a lungo termine vengono incubate con varie concentrazioni di supernatante proveniente dalle colture di PBMC in piastre a 96 pozzetti a fondo piatto.
Ciascun pozzetto ha un volume totale di 200 ul, costituito da 100 ul di cellule B (15x103 cellule) e 100 ui di supernatante. Si esamina l'efficacia delle cellule B incubandole con varie concentrazioni di BCGF purificato (Cellular Products, Inc. Buffalo, N.Y.).
Le colture vengono incubate per 24 ore, dopo di che 1 uCi di [3 H-Tdr] viene aggiunto e si incuba nuovamente per 12 ore.
Le colture sono poi raccolte e contate in uno scintilloscopio.
2. Le cellulte T vengono incubate in pozzetti a fondo piatto. Il volume totale in ciascun pozzetto è di 200 ul contenente 50x103 cellule, li periodo di incubazione è di 72 ore, comprese 12 ore di marcature con [3 H-Tdr].

5. AUMENTO IN VITRO DEL NUMERO DI CELLULE
Il tripeptide, aggiunto sia a culture di linfociti T che di miscele di linfociti B e T ogni quattro giorni alla concentrazione di 5 ug/ml per un periodo di 30 giorni, è in grado di aumentare il numero di cellule con un massimo di + 50% rispetto alle culture di controllo osservato tra il decimo e il quindicesimo giorno dell'esperimento.
TOSSICOLOGIA
TOSSICITÀ' ACUTA
Gli studi di tossicità acuta condotti sul topo e sul ratto hanno mostrato che fino alla dose di 10Q0 mg/kg i.m. il tripeptide è totalmente privo di effetti collaterali.

TÖLLERABILITA'
Studi su conigli e topi hanno mostrato che il prodotto, alla dose di 100 mg/kg rispettivamente i.v. e ì.p., non provoca alcuna modificazione emodinamica ne effetti comportamentali.
In particolare, il tempo di sonno indotto da pentobarbital mostra solo un lieve aumento.
ATTIVITÀ' ALLERGIZZANTE
Il prodotto, alla dose di 100 mg/kg i.m., non induce alcun fenomeno di sensibilizzazione nella cavia.
SALI DEL TRIPEPTIDE
Le ricerche sopra menzionate sono state condotte con l'acetato del tripeptide; comunque è ben noto allo stato dell'arte che risultati simili possono essere ottenuti usando altri sali per esempio il trìfluoroace-tato, il cloridrato, il solfato.
Claims (5)
1. Un tripeptide costituito da L-Ala (alanina), e 2 L-Arg (argìnina) e avente la seguente struttura: Arg-Ala-Arg
2. Un tripeptide in accordo alia rivendicazione 1 quale immunistimolante.
3. Composizione farmaceutica per il trattamento terapeutico di patologie caratterizzate da deficienze primarie e secondarie dei sistema immunitario, contenente il tripeptide secondo la rivendicazione 1.
4. Composizione farmaceutica ad attività immunostimolante per somministrazione parenterale e orale, contenente il tripeptide secondo la rivendicazione 1.
5. Composizione farmaceutica secondo la rivendicazione 3, in cui il tripeptide è un suo sale scelto fra i sali:acetato, trifluoroacetato, cloridrato e solfato.
