Description
Die vorliegende Erfindung betrifft ein Verfahren zum Herstellen eines optisch aktiven Hemiesters. Hintergrund der Erfindung
Ein optisch aktiver Hemiester, beispielsweise ein Imidazolidin-2-on-Hemiester-Derivat, war als Zwischenprodukt zum Synthetisieren von d-Biotin (Vitamin H) und ähnlichem bekannt. Es war ein Verfahren zum Herstellen von Hemiester-Zwischenprodukt-Derivaten durch Ringöffnungsreaktionen eines zyklischen Säureanhydrids unter Verwendung einer Komplexverbindung von Alkaloiden und Dialkylzink, wie in Bull. Chem. Soc. Jpn., 1993, 66, 2128 beschrieben, und ein Verfahren der Verwendung eines optisch aktiven Diisopropoxytitans TADDOLates, wie in J. Org. Chem., 1998, 63, 1190 beschrieben, bekannt. Zusammenfassung der Erfindung
Erfindungsgemäss kann ein optisch aktiver Hemiester mit guter optischer Aktivität industriell vorteilhaft hergestellt werden.
Der erste Aspekt der vorliegenden Erfindung betrifft ein Verfahren zum Herstellen eines optisch aktiven Hemiesters der Formel (1):

Zunächst wird der erste Aspekt der vorliegenden Erfindung erläutert.
Bei dem zyklischen Säureanhydrid der Formel (2) umfassen Beispiele des Halogenatoms, welches durch R<1> oder R<2 >dargestellt ist, ein Fluoratom, ein Chloratom, ein Bromatom und ein Jodatom.
Beispiele der Alkylgruppe, welche wahlweise mit einer Alkoxylgruppe oder einem Halogenatom substituiert ist, dargestellt durch R<1> oder R<2>, umfassen eine lineare, verzweigte oder zyklische Alkylgruppe mit 1 bis 10 Kohlenstoffatomen, wobei diese Alkylgruppe wahlweise mit einem Halogenatom, einer Alkoxylgruppe (beispielsweise einer [C1-C5]-Alkoxylgruppe, wie etwa einer Methoxy-, Ethoxy-, n-Propoxy- oder i-Propoxygruppe, n-Butoxygruppe, s-Butoxygruppe, t-Butoxygruppe, n-Pentyloxygruppe, i-Pentyloxygruppe oder Neopentyloxygruppe) und ähnlichem substituiert ist.

Beispiele der Alkenylgruppe, welche wahlweise mit einer Alkoxylgruppe oder einem Halogenatom substituiert ist, umfassen eine (C2-C3 )-Alkenylgruppe, welche wahlweise mit einer (C1-C5 )-Alkoxylgruppe oder einem Halogenatom substituiert ist. Spezifische Beispiele davon umfassen eine Vinylgruppe, eine Propen-1-yl-Gruppe, eine Propen-2-yl- Gruppe, eine 2-Methylpropen-l-yl-Gruppe, eine 2,2-Dichlor-vinylgruppe, eine 2,2-Dibromvinylgruppe, eine 2-Methoxyvinylgruppe und Ähnliches.
Beispiele der Aralkylgruppe, welche wahlweise mit einer Alkylgruppe, einer Alkoxylgruppe oder einem Halogenatom substituiert ist, umfassen eine (C7-C11)-Aralkylgruppe, welche wahlweise mit einer (C1-C5)-Alkylgruppe (beispielsweise einer Methylgruppe, einer Ethylgruppe, n-Propylgruppe, i-Propylgruppe oder n-Butylgruppe, s-Butylgruppe, t-Butylgruppe, n-Pentylgruppe, i-Pentylgruppe oder Neopen-tylgruppe), einer (C1-C5 )-Alkoxylgruppe oder einem Halogenatom substituiert ist.
Spezifische Beispiele davon umfassen eine Benzylgruppe, eine 1-Phenethylgruppe, eine 2-Phenethylgruppe, eine alpha -Naphthylmethylgruppe, eine beta -Naphthylmethylgruppe und ähnliches, wobei aromatische Ringe davon wahlweise mit mindestens einer Gruppe, welche aus dem Halogenatom, der Alkoxylgruppe oder der Alkylgruppe, wie oben beschrieben, ausgewählt ist, substituiert sind.
Beispiele der Arylgruppe, welche wahlweise mit einer Alkylgruppe, einer Alkoxylgruppe oder einem Halogenatom substituiert ist, umfassen eine (C6-C18)-Arylgruppe (beispielsweise eine Phenyl-, Naphthyl-, Anthranyl- oder Phenanthrylgruppe), welche wahlweise mit dem Halogenatom, der (C1-C5 )-Alkyl- oder der (C1-C5 )-Alkoxylgruppe, wie oben beschrieben, und ähnlichem substituiert sein kann.
Beispiele der Alkylgruppe, Aralkylgruppe, Silylgruppe oder Acylgruppe, welche durch R<a> dargestellt sind, umfassen jeweils eine (C1-C5 )-Alkylgruppe, wie etwa eine Methylgruppe, eine Ethylgruppe, eine Propylgruppe, eine n-Butylgruppe, t-Butylgruppe, eine Pentylgruppe oder ähnliches, eine (C7-C8)-Aralkylgruppe, wie etwa eine Benzylgruppe oder eine Phenethylgruppe, eine Silylgruppe mit drei (C1-C4)-Alkylgruppen, wie etwa eine Trimethylsilylgruppe, eine t-Butyldimethylsilylgruppe oder ähnliches, eine (C2-C6)-Acylgruppe, wie etwa eine Acetylgruppe, eine Benzoylgruppe oder ähnliches.
Beispiele der Acylgruppe, welche durch R<b> dargestellt ist, umfassen die gleichen Acylgruppen, wie oben beschrieben. Beispiele der Alkylgruppe in der Alkoxycarbonylgruppe oder der Alkylsulfonylgruppe und Beispiele der Aralkylgruppe in der Aralkyloxycarbonylgruppe umfassen jeweils die gleichen Gruppen, wie oben für R<a> definiert.
Spezifische Beispiele der Arylsulfonylgruppe umfassen eine p-Toluolsulfonylgruppe oder ähnliches. Spezifische Beispiele der Alkylsulfonylgruppe umfassen eine Methylsulfonylgruppe. Spezifische Beispiele der Haloalkylsulfonylgruppe umfassen eine Trifluormethansulfonylgruppe und ähnliches .
Beispiele der Gruppe, welche durch R<1>-Gruppen oder R<2>-Gruppen gebildet wird, umfassen eine Gruppe der folgenden Formel (9a) bis (9d):

Substituenten, welche durch R<21> dargestellt sind, werden im Folgenden erläutert.
Die Alkylgruppe, welche wahlweise mit einer Alkoxylgruppe oder einem Halogenatom substituiert ist, umfasst eine lineare, verzweigte oder zyklische (C1-C10)-Alkylgruppe, welche wahlweise mit einem Halogenatom oder einer (C1-C5 )-Alkoxylgruppe substituiert ist.
Die Aralkylgruppe, welche wahlweise mit einer Alkylgruppe, einer Alkoxylgruppe oder einem Halo-genatom substituiert ist, umfasst eine (C7-C11)-Aralkylgruppe, welche wahlweise mit einem Halogen-atom, einer (C1-C5 )-Alkoxylgruppe oder einer (C1-C5 )-Alkylgruppe substituiert ist (beispielsweise eine Benzylgruppe oder ähnliches).
Die Arylgruppe, welche wahlweise mit einer Alkylgruppe, einer Alkoxylgruppe oder einem Halogen-atom substituiert ist, umfasst eine (C6-C18)-Arylgruppe, welche wahlweise mit einem Halogenatom, einer (C1-C5 )-Alkylgruppe oder einer (C1-C5 )-Alkoxylgruppe substituiert ist.
Die oben beschriebene Aralkylgruppe ist bevorzugt. Insbesondere ist eine Benzylgruppe bevorzugt.
Bei den R<1>- und R<2>-Gruppen sind sowohl R<1>- als auch R<2>-Gruppen vorzugsweise Wasserstoffatome. Die Gruppe, welche durch R<1>- oder R<2>-Gruppen der oben erwähnten Formel (9c) gebildet wird, ist bevorzugt.

Die R<3>-Gruppe der Hydroxyverbindung der Formel (3) wird im Folgenden erläutert.
Beispiele der Alkylgruppe, welche wahlweise mit einer Alkoxylgruppe, einer Phenoxygruppe, einer Dialkylaminogruppe oder einem Halogenatom substituiert ist, umfassen eine (C1-C8)-Alkylgruppe, welche wahlweise mit einer (C1-C5 )-Alkoxylgruppe, einer Phenoxygruppe, einer Di(C1-C3)alkylaminogruppe oder einem Halogenatom substituiert ist.

Beispiele der Hydroxyverbindung der Formel (3) umfassen einen Alkylalkohol mit der Alkylgruppe, wie oben definiert, einen Aralkylalkohol mit der Aralkylgruppe, wie oben definiert, einen Arylalkohol mit der Arylgruppe, wie oben definiert, und ähnliches.
Spezifische Beispiele davon umfassen Methylalkohol, Ethylalkohol, n-Propylalkohol, i-Propylalkohol, n-Butylalkohol, s-Butylalkohol, t-Butylalkohol, n-Pentylalkohol, Neopentylalkohol, Amylalkohol, Hexylalkohol, Octylalkohol, 2-Methoxyethanol, 2-Phenoxyethanol, 2-(Dimethylamino)ethanol, 2-Chlorethanol,
Benzylalkohol, 2-Methylbenzylalkohol, 4-Methylbenzylalkohol, 2-Methoxybenzylalkohol, 4-Methoxybenzylalkohol, 2-Phenoxybenzylalkohol, 4-Phenoxybenzylalkohol, 4-Nitrobenzylalkohol, 4-Chlorbenzylalkohol,
Phenol, 2-Methylphenol, 4-Methylphenol, 2-Methoxyphenol, 4-Methoxyphenol, 4-Nitrophenol, 4-Chlorphenol und ähnliches.
Unter den R<3> der Hydroxyverbindung der Formel (3) sind die Alkylgruppe, welche wahlweise mit einer Alkoxylgruppe, einer Phenoxygruppe, einer Dialkylaminogruppe oder einem Halogenatom substituiert ist, und die Aralkylgruppe, welche wahlweise mit einer Alkylgruppe, einer Alkoxylgruppe, einer Phenoxygruppe, einer Nitrogruppe oder einem Halogenatom substituiert ist, bevorzugt.
Eher bevorzugt sind primäre Alkohole der Formel R<33>CH 2 OH, worin R<><33> eine (C1-C7)-Alkylgruppe, welche wahlweise mit einer (C1-C5 )-Alkoxylgruppe, einer Phenoxygruppe, einer Di(C1-C3)alkylaminogruppe oder einem Halogenatom substituiert ist, oder eine Phenylgruppe, welche wahlweise mit einer (C1-C5 )-Alkylgruppe, einer (C1-C5 )-Alkoxylgruppe, einer Nitrogruppe oder einem Halogenatom substituiert ist, darstellt.
Die Menge der Hydroxyverbindung (3) ist nicht besonders kritisch, und die Verbindung kann als Lösungsmittel in Überschuss gegenüber dem zyklischen Säureanhydrid (2) verwendet werden und kann nach Vollendung der Reaktion wiedergewonnen werden, beispielsweise durch Destillation oder ähnlichem. Die Menge der Hydroxyverbindung (3) ist typischerweise 1 Mol oder mehr pro Mol des zyklischen Säureanhydrids (2).
Der asymmetrische Katalysator, welcher bei dem vorliegenden Verfahren zu verwenden ist, kann durch Zusammenbringen einer Lewis-Säure-Verbindung, welche aus einem Halogenid, Alkoxid oder Trifluormethansulfonat eines Elements der Gruppe 3, 4, 13 oder 14 des Periodensystems der Elemente mit einem optisch aktiven Liganden, welcher aus einer Diol-, Aminoalkohol- und Bisoxazolidinverbindung ausgewählt ist, ausgewählt ist, hergestellt werden.
Beispiele der Lewis-Säure-Verbindung, welche aus einem Halogenid, Alkoxid oder Trifluormethansulfonat eines Elements der Gruppe 3, 4, 13 oder 14 des Periodensystems der Elemente ausgewählt ist, umfassen ein Borhalogenid, wie etwa Bortrifluorid, Bortrichlorid, ein Aluminiumhalogenid oder -alkoxid, wie etwa Aluminiumtrichlorid, Aluminiumtriisopropoxid oder ähnliches, ein Titanhalogenid oder -alkoxid, wie etwa Titantetrachlorid, Titantetraisopropoxid oder ähnliches, ein Zinnhalogenid, wie etwa Zinntetrachlorid, Zinndichlorid oder ähnliches, ein Lanthanalkoxid, wie etwa Lanthantriisopropoxid oder ähnliches, und Scandiumtriflat (-trifluormethansulfo nat) oder ahnliches. Insbesondere sind Zinndichlorid und Titantetraisopropoxid bevorzugt.

Beispiele der optisch aktiven Diolverbindung umfassen optisch aktives 1,1-Binaphthanol, 1,2-Diphenylethan-1,2-diol und ähnliches.
Die R<6> des optisch aktiven Aminoalkohols der Formel (7) werden im Folgenden beschrieben.
Die wahlweise substituierte Alkylgruppe umfasst eine Alkylgruppe, welche wahlweise mit einer Alkoxylgruppe oder einem Halogenatom substituiert ist (beispielsweise eine lineare, verzweigte oder zyklische Alkylgruppe mit 1 bis 10 Kohlenstoffatomen, welche wahlweise mit einem Halogenatom oder einer (C1-C5 )-Alkoxylgruppe substituiert ist), und eine Aralkylgruppe, welche wahlweise mit einer Alkylgruppe, einer Alkoxylgruppe oder einem Halogen-atom substituiert ist (beispielsweise eine (C7-C11)-Aralkylgruppe, welche wahlweise mit einer (C1-C5 )- Alkylgruppe, einer (C1-C5)-Alkoxylgruppe oder einem Halogenatom substituiert ist).
Die wahlweise substituierte Alkenylgruppe umfasst eine Alkenylgruppe, welche wahlweise mit einer Alkoxylgruppe oder einem Halogenatom substituiert ist (beispielsweise eine (C2-C3 )-Alkenylgruppe, welche wahlweise mit einer (C1-C5 )-Alkoxylgruppe oder einem Halogenatom substituiert ist).
Die wahlweise substituierte Arylgruppe umfasst eine Arylgruppe, welche wahlweise mit einer Alkylgruppe, einer Alkoxylgruppe oder einem Halogenatom substituiert ist (eine (C16-C18)-Arylgruppe (beispielsweise eine Phenyl-, Naphthyl-, Anthranyl- oder Phenanthrylgruppe), welche wahlweise mit einem Halogenatom, einer (C1-C5)-Alkyl- oder (C1-C5 )-Alkoxylgruppe substituiert sein kann).
Beispiele der wahlweise substituierten Alkylengruppe umfassen eine Ethylen- oder Trimethylengruppe, welche mit einem Benzolring annelliert werden kann (beispielsweise eine Indanylgruppe oder ähnliches).
Hinsichtlich der spezifischen Beispiele dieser Gruppen kann auf die oben beschriebenen für R<1> und R<2> verwiesen werden.
Bei dem optisch aktiven Bisoxazolin der Formel (8) haben die wahlweise substituierte Alkylgruppe und die wahlweise substituierte Arylgruppe jeweils die gleiche Bedeutung, wie oben in Verbindung mit Formel (7) definiert.
Spezifische Beispiele des optisch aktiven Liganden umfassen optisch aktives 1,1-Binaphthol, 1,2-Diphenylethan-1,2-diol,
2-Amino-1,1-diphenyl-3-phenylpropan-1-ol, 1-Amino-2-indanol, 2-Amino-1-indanol, 2-Amino-1,1-diphenyl-2-phenylethan-1-ol, 2-Amino-1,1-diphenyl-3-methylbutan-1-ol, 2-Amino-1,1-bis[2-butoxy-4-(2-methylpropan-2-yl)phenyl]-3-phenylpropan-1-ol, 2-Amino-1,1-bis[2-butoxy-4-(2-methylpropan-2-yl)phenyl]-2-phenylethan-1-ol, 2-Amino-1,1-bis[2-butoxy-4-(2-methylpropan-2-yl)phenyl]-3-methylbutan-1-ol, 2-Aminocyclopentanol, 2-Aminocyclohexanol, 2,2'-Isopropylidenbis(4-t-butyl-2-oxazolin), 2,2'-Isopropylidenbis(4-phenyl-2-oxazolin), 2,2'-Metylenbis(4-phenyl-2-oxazolin) und ähnliches.
Bevorzugt sind optisch aktiver Aminoalkohol und Bisoxazolin, vermehrt bevorzugt sind 2-Amino-1,1-bis[2-butoxy-4-(2-methylpropan-2-yl)-phenyl]-3-phenylpropan-1-ol und 2,2'-Isopropylidenbis(4-t-butyl-2-oxazolin).
Die optisch aktiven Liganden sind im Handel erhältlich, oder das optisch aktive Oxazolin kann gemäss einer Quelle, wie etwa EP-A-0 895 992 oder ähnliches, hergestellt werden.
Die Menge der zu verwendenden Lewis-Säure-Verbindung ist nicht besonders kritisch, ist jedoch gewöhnlich eine katalytische Menge, etwa 0,00001 bis weniger als 1 Mol, vorzugsweise etwa 0,0001 bis 0,5 Mol pro Mol des zyklischen Säureanhydrids (2).
Die Menge des optisch aktiven Liganden ist nicht besonders kritisch, beträgt aber gewöhnlich 1 bis 10 Mol, vorzugsweise etwa 1 bis 2 Mol pro Mol der Lewis-Säure-Verbindung. Der asymmetrische Katalysator kann durch ein Verfahren hergestellt werden, welches ein Zusammenbringen der Lewis-Säure-Verbindung mit dem optisch aktiven Liganden umfasst, und der derart gebildete Katalysator kann vor einer Verwendung isoliert werden, alternativ kann der Katalysator in situ hergestellt werden. Beispielsweise kann der Vorgang des Zusammenführens der Lewis-Säure-Verbindung mit dem optisch aktiven Liganden gleichzeitig in einem Reaktionssystem der Umsetzung des zyklischen Säureanhydrids (2) mit der Alkoholverbindung (3) durchgeführt werden.
Das Umsetzen des zyklischen Säureanhydrids (2) mit einer Alkoholverbindung (3) kann in Anwesenheit einer Base, welche aus einer anorganischen Base oder einer aromatischen tertiären Aminverbindung ausgewählt ist, ausgeführt werden.
Beispiele der anorganischen Base umfassen ein Carbonat oder Bicarbonat eines Alkalimetalls und ein Carbonat eines Erdalkalimetalls (beispielsweise Lithiumcarbonat, Kaliumcarbonat, Natriumcarbonat, Lithiumhydrogencarbonat, Kaliumhydrogencarbonat, Natriumhydrogencarbonat, Magnesiumcarbonat) und Ammoniumcarbonat, und
Beispiele der aromatischen tertiären Aminverbindung umfassen N-Methylimidazol, eine Pyridinverbindung (beispielsweise eine Pyridinverbindung, welche wahlweise mit einem Alkyl oder einem Halogen-atom substituiert ist, wie etwa Pyridin, 2-Methyl-5-ethylpyridin, 2,6-Dichlorpyridin, Picolin oder ähnlichem) und ähnliches.
Bevorzugt sind ein Carbonat oder Bicarbonat eines Alkalimetalls oder ein Carbonat eines Erdalkalimetalls, wie etwa Natriumcarbonat, Natriumhydrogencarbonat, Kaliumhydro gencarbonat oder Magnesiumcarbonat und eine Pyridinverbindung, wie etwa 2,6-Dichlorpyridin.
Die Menge der zu verwendenden Base ist nicht besonders kritisch, beträgt jedoch gewöhnlich etwa 10 Mol oder weniger, vorzugsweise etwa 2 Mol oder weniger pro Mol des zyklischen Säureanhydrids (2).
Die Reaktion des zyklischen Säureanhydrids (1) mit der Hydroxyverbindung (3) wird gewöhnlich in einer inerten Gasatmosphäre, wie etwa Argon, Stickstoff und ähnlichem, durchgeführt. Die Reaktion kann unter einem normalen, erhöhten oder erniedrigten Druck durchgeführt werden.
Die Reaktion kann ohne Lösungsmittel oder in einem Lösungsmittel durchgeführt werden.
Beispiele des zu verwendenden Lösungsmittels umfassen einen halogenierten aliphatischen Kohlenwasserstoff, wie etwa Dichlormethan, Chloroform, 1,2-Dichlorethan und ähnliches, einen aliphatischen Kohlenwasserstoff, wie etwa Hexan, Heptan, Octan, Nonan oder ähnliches, einen aromatischen Kohlenwasserstoff, wie etwa Benzol, Toluol, Xylol, einen halogenierten aromatischen Kohlenwasserstoff, wie etwa Chlorbenzol oder ähnliches, ein Etherlösungsmittel, wie etwa Diethylether oder Tetrahydrofuran, und ein gemischtes Lösungsmittel davon.
Die Reaktionstemperatur ist nicht besonders kritisch, befindet sich jedoch gewöhnlich in einem Bereich von -80 bis 100 DEG C, vorzugsweise -50 bis 50 DEG C.
Nach Vollendung der Reaktion kann der Katalysator von optisch aktiven Hemiestern durch Spülen mit Wasser oder saurem Wasser entfernt werden, und das Produkt kann einfach von der Reaktionsmischung durch gewöhnliche Nachbehandlung, wie etwa Extraktion, Phasentrennung, Destillation oder ähnliches, getrennt werden, und dieses kann weiter durch Säulenchromatographie oder ähnliches gereinigt werden, wenn dies notwendig ist. Die organische Base kann beispielsweise durch Zugeben einer starken Base zu der getrennten wässrigen Phase wiedergewonnen werden.
Als nächstes erfolgt eine Beschreibung des zweiten Aspekts der vorliegenden Erfindung, welcher ein Verfahren zum Herstellen eines optisch aktiven Imidazolidin-2-ons der Formel (4) betrifft, welches ein Umsetzen eines zyklischen Säureanhydrids der Formel (5) mit einer Hydroxyverbindung der Formel (3) in Anwesenheit eines optisch aktiven Alkaloids, welches aus Chinin, Epichinin, Cinchonin und Cinchonidin ausgewählt ist, betrifft.
In Formel (4) und (5) ist R<21> das gleiche, wie oben in Verbindung mit Formel (9c) definiert. R<21> ist vorzugsweise die Aralkylgruppe, welche wahlweise mit einer Alkylgruppe, einer Alkoxylgruppe oder einem Halogenatom substituiert ist, und eher bevorzugt eine Benzylgruppe.
Beispiele des zyklischen Säureanhydrids der Formel (5) umfassen 4,6-Dimethyl-2,4,6,3a,6a-penta-hydro-4,6-diaza-2-oxapentalen-1 ,3,5-trion, 4,6-Bisbenzyl-2,4,6,3a,6a-penta hydro-4,6-diaza-2-oxapentalen-1,3,5-trion, 4,6-Diphenyl-2,4,6,3a,6a-pentahydro-4,6-diaza-2-oxapentalen-1,3,5-trion und ähnliches.
Bei dem optisch aktiven Alkaloid ist Chinin bevorzugt.
Die Menge des zu verwendenden optisch aktiven Alkaloids ist nicht besonders kritisch und beträgt gewöhnlich etwa 0,00001 Mol bzw. kann eine katalytische Menge sein, beispielsweise weniger als 1 Mol pro Mol des zyklischen Säureanhydrids (2).
Die Reaktion wird vorzugsweise in Anwesenheit einer Base ausgeführt. Beispiele der Base umfassen eine anorganische Base (beispielsweise ein Carbonat oder Bicarbonat eines Alkalimetalls oder ein Carbonat eines Erdalkalimetalls, wie etwa Lithiumcarbonat, Kaliumcarbonat, Natriumcarbonat, Lithiumhydrogencarbonat, Kaliumhydrogencarbonat, Natriumhydrogencarbonat, Magnesiumcarbonat oder ähnliches) und Ammoniumcarbonat, und eine aliphatische oder aromatische tertiäre Aminverbindung, wie etwa Triethylamin, Ethyldiisopropylamin, Pyridin, 1,2,2,6,6-Pentamethylpiperidin, 2-Methyl-5-Ethylpyridin, 2,6-Dichlorpyridin, Picolin, N-Methylimidazol und ähnliches. Insbesondere ist 1,2,2,6,6-Pentamethylpiperidin bevorzugt.
Die Menge der zu verwendenden Base ist nicht besonders kritisch, beträgt aber gewöhnlich etwa 0,1 bis 10 Mol, vorzugsweise etwa 0,1 bis 2 Mol pro Mol des zyklischen Säureanhydrids (5).
Die Reaktion des zyklischen Säureanhydrids (5) mit der Hydroxyverbindung (3) wird in Anwesenheit des optisch aktiven Alkaloids und gewöhnlich in einer inerten Gasatmosphäre, wie etwa Argon, Stickstoff und ähnlichem durchgeführt. Die Reaktion kann unter einem normalen, erhöhten oder erniedrigten Druck durchgeführt werden.
Die Reaktion kann ohne Lösungsmittel oder in einem Lösungsmittel durchgeführt werden.
Beispiele des zu verwendenden Lösungsmittels umfassen einen halogenierten Kohlenwasserstoff, wie etwa Dichlormethan, Chloroform, 1,2-Dichlorethan, Tetrachlorkohlenstoff und ähnliches, einen aliphatischen Kohlenwasserstoff, wie etwa Hexan, Heptan, Octan und Nonan, einen aromatischen Kohlenwasserstoff, wie etwa Benzol, Toluol, Xylol oder ähnliches, einen halogenierten aromatischen Kohlenwasserstoff, wie etwa Chlorbenzol und ähnliches, ein Etherlösungsmittel, wie etwa Diethylether und Tetrahydrofuran, und ein daraus gemischtes Lösungsmittel.
Die Reaktionstemperatur ist nicht besonders kritisch und befindet sich gewöhnlich in einem Bereich von -80 bis 100 DEG C, vorzugsweise -60 bis 50 DEG C.
Nach Vollendung der Reaktion kann die Base von den optisch aktiven Imidazolidin-2-on-Verbindungen (4) durch Spülen mit Wasser oder saurem Wasser entfernt werden. Das erwünschte Produkt kann einfach von der Reaktionsmischung durch eine Nachbehandlung, wie etwa Extraktion, Phasentrennung, Konzentration und/oder ähnliches getrennt werden. Das Alkaloid und die aliphatische oder aromatische tertiäre Aminverbindung können beispielsweise durch Zugeben einer starken Base zu einer wässrigen Phase, welche durch Spülen der Reaktionsmischung erhalten wird, wiedergewonnen werden. Die vorliegende Erfindung wird durch Beispiele genauer beschrieben, ist jedoch nicht auf diese begrenzt. Beispiel 1
17,5 mg (0,01 mmol) Titantetraisopropoxid und 38,1 mg (0,06 mmol) (R)-2-Amino-1,1-bis[2-butoxy-4-methylpropan-2-yl]phenyl]-3-phenylpropan-1-ol wurden in ein mit Stickstoff gespültes 20-ml-Gefäss gegeben und 5 ml Toluol wurden dazu gegeben, um diese zu lösen, und die resultierende Lösung wurde auf 0 DEG C gekühlt. Danach wurden 200 mg (0,6 mmol) Cis-4,6-bisbenzyl-2,4,6,3a,6a-pentahydro-4,6-diaza-2-oxapentalen-1,3,5-trion, 66 mg (0,6 mmol) Benzylalkohole auf einmal bzw. tropfenweise dazugegeben und die resultierende Mischung wurde bei der gleichen Temperatur 13 Stunden gerührt.
2N Salzsäure wurde zu der Reaktion zugegeben, und das Produkt wurde mit Diethylether extrahiert. Nachdem die getrennte organische Phase mit einer gesättigten wässrigen Natriumchloridlösung gespült wurde, wurde diese durch HPLC (Hochdruckflüssigchromatographie) analysiert (interner Standard: Diisopropylphthalat). (4R,5S)-1,3-bisbenzyl-2-oxo-5-[benzyloxycarbonyl]imidazolidin-4-carbonsäure wurde in einer Ausbeute von 43,2% erhalten, und die optische Reinheit wurde mit einer optisch aktiven HPLC-Säule (hergestellt durch Sumika Chemical Analysis Service: SUMICHIRAL OA-3300) gemessen und zu 23,7%ee bestimmt. Beispiel 2
17,5 mg (0,06 mmol) Titantetraisopropoxid und 38,1 mg (0,06 mmol) (R)-2-Amino-1,1-bis[2-butoxy-4-methylpropan-2-yl]phenyl]-3-phenylpropan-1-ol wurden in ein mit Stickstoff gespültes 20-ml-Gefäss gegeben und 5 ml Toluol wurden dazugegeben, um diese zu lösen, und auf 0 DEG C gekühlt. Danach wurden 200 mg (0,6 mmol) Cis-4,6-bisbenzyl-2,4,6,3a,6a-pentahydro-4,6-diaza-2-oxapentalen-1,3,5-trion, 66 mg (0,6 mmol) Benzylalkohol und 64,4 mg (0,6 mmol) Natriumkarbonat dazugegeben, und die resultierende Mischung wurde bei der gleichen Temperatur 13 Stunden gerührt.
Nachbehandlung und Analyse wurden wie bei Beispiel 1 durchgeführt. (4R, 5S)-1,3-Bisbenzyl-2-oxo-5-[benzyloxycarbonyl]imidazolidin-4-carbonsäure wurde in einer Ausbeute von 78,7% erhalten, und die optische Reinheit betrug 50,6%ee. Beispiele 3-9
(4R, 5S)-1,3-Bisbenzyl-2-oxo-5-[benzyloxycarbonyl]imidazolidin-4-carbonsäure wurde in einer ähnlichen Weise wie bei Beispiel 2 erhalten, ausser, dass eine in Tabelle 1 dargestellte Base anstelle von Natriumcarbonat bei Beispiel 2 verwendet wurde. Die Ergebnisse sind in Tabelle 1 dargestellt. Tabelle 1

11,1 mg (0,06 mmol) Zinnchlorid(II) und 18 mg (0,06 mmol) 2,2'-Isopropylidenbis((4S)-4-t-butyl-2-oxazolin) wurden in ein mit Stickstoff gespültes 20-ml-Gefäss gegeben und 5 ml Toluol wurden dazu gegeben, um diese zu lösen, und auf 0 DEG C gekühlt. Danach wurden 200 mg (0,6 mmol) Cis-4,6-bis-benzyl-2,4,6,3a,6a-pentahydro-4,6-diaza-2-oxapentalen-1,3,5-trion, 61,9 mg (0,6 mmol) Benzylalkohol auf einmal bzw. tropfenweise dazugegeben, und die resultierende Mischung wurde bei der gleichen Temperatur 13 Stunden gerührt.
Nachbehandlung und Analyse wurden wie bei Beispiel 1 durchgeführt. (4R, 5S)-1,3-Disbenzyl-2-oxo-5-[benzyloxycarbonyl]imidazolidin-4-carbonsäure wurde in einer Ausbeute von 39,94% erhalten, und die optische Reinheit betrug 55,2%ee. Beispiel 11
199 mg (0,06 mmol) Cis-4,6-bisbenzyl-2,4,6,3a,6a-pentahydro-4,6-diaza-2-oxapentalen-1,3,5-trion wurden in ein mit Stickstoff gespültes 20-ml-Gefäss gegeben, und 5 ml Toluol wurden dazugegeben und auf -50 DEG C gekühlt. Danach wurden 212,7 mg (0,06 mmol) Chinin, 53,1 mg (1,7 mmol) Methanol auf einmal bzw. tropfenweise dazugegeben, und die resultierende Mischung wurde bei der gleichen Temperatur 21 Stunden gerührt. 2N Salzsäure wurde der Reaktionsmischung zugegeben, um die Reaktion zu beenden, und das Produkt wurde mit Diethylether extrahiert. Die getrennte organische Phase wurde mit einer gesättigten wässrigen Natriumchloridlösung gespült und einer quantitativen Analyse durch HPLC (interner Standard: Diisopropylphthalat) unterzogen. (4S, 5R)-1,3-Bisbenzyl-2-oxo-5-[methoxycarbonyl]imidazolidin-4-carbonsäure wurde in einer Ausbeute von 49,1% erhalten.
Die optische Reinheit wurde mit einer optisch aktiven HPLC-Säule (hergestellt durch Sumika Chemical Analysis Service: SUMICHIRAL OA-3300) gemessen und zu 73,3%ee bestimmt. Beispiel 12
199 mg (0,6 mmol) Cis-4,6-bisbenzyl-2,6,6,3a,6a-pentahydro-4,6-diaza-2-oxapentalen-1,3,5-trion wurden in ein mit Stickstoff gespültes 20-ml-Gefäss gegeben, und 2,5 ml Toluol und 2,5 ml Tetrachlorkohlenstoff wurden dazugegeben. Die resultierende Lösung wurde auf -20 DEG C gekühlt. Danach wurden 213,9 mg (0,06 mmol) Chinin, 64,0 mg (0,6 mmol) Benzylalkohol auf einmal bzw. tropfenweise zugegeben, und die resultierende Lösung wurde bei der gleichen Temperatur 16 Stunden gerührt. Danach wurde die Reaktionsmischung wie bei Beispiel 11 behandelt. (4S, 5R)- 1,3-Bisbenzyl-2-oxo-5-[methoxycarbonyl]imidazolidin-4-carbonsäure wurde in einer Ausbeute von 76,8% erhalten. Die optische Reinheit des Produkts betrug 73,6%ee. Beispiel 13
Nach der gleichen Weise wie der bei Beispiel 12, ausser, dass Cinchonidin anstelle von Chinin bei Beispiel 12 verwendet wurde, wurde (4S, 5R)-1,3-Bisbenzyl-2-oxo-5-[methoxycarbonyl]imidazolidin-4-carb onsäure in einer Ausbeute von 81,4% erhalten. Die optische Reinheit des Produkts betrug 25,9%ee. Beispiel 14
202 mg (0,69 mmol) Cis-4,6-bisbenzyl-2,6,6,3a,6a-pentahydro-4,6-diaza-2-oxapentalen-1,3,5-trion wurden in ein mit Stickstoff gespültes 20-ml-Gefäss gegeben und 5 ml Toluol wurden dazugegeben, und die resultierende Lösung wurde auf -50 DEG C gekühlt. Danach wurden 19,8 mg (0,06 mmol) Chinin und 106,0 mg (0,68 mmol) 1,2,2,6,6-Pentamethylpiperidin dazugegeben, und 55,0 mg (1,7 mmol) Ethanol wurden danach tropfenweise zugegeben, und die resultierende Lösung wurde bei der gleichen Temperatur 21 Stunden gerührt. Die Reaktionsmischung wurde wie bei Beispiel 11 behandelt. (4S, 5R)-1,3-Bisbenzyl-2-oxo-5-[methoxycarbonyl]imidazolidin-4-carbonsäure wurde in einer Ausbeute von 56,0% erhalten. Die optische Reinheit des Produkts betrug 59,8%ee

einer Alkoxylgruppe oder einem Halogen-atom substituiert ist, eine Arylgruppe, welche wahlweise mit einer Alkylgruppe, einer Alkoxygruppe oder einem Halogenatom substituiert ist, oder eine Gruppe der Formel (10): R<a>O-, R<b>NH oder R<a>R<b>N darstellen, worin R<><a> eine Alkylgruppe, eine Aralkylgruppe, eine Silylgruppe oder eine Acylgruppe darstellt, und R<><b> eine Acylgruppe, eine Alkoxycarbonyl- oder Aralkyloxycarbonylgruppe oder eine Alkylsulfonyl-, Halogenalkylsulfonyl- oder Arylsulfonylgruppe darstellt, oder die R<1>- oder die R<2>-Gruppen an deren Endstellen derart gebunden sein können, dass ein Ring gebildet wird, und R<><3> eine Alkylgruppe, welche wahlweise mit einer Alkoxylgruppe, einer Phenoxygruppe, einer Dialkylaminogruppe oder einem Halogenatom substituiert ist, eine Aralkylgruppe, welche wahlweise mit einer Alkylgruppe, einer Alkoxylgruppe,
einer Phenoxygruppe, einer Nitrogruppe oder einem Halogenatom substituiert ist, eine Arylgruppe, welche wahlweise mit einer Alkylgruppe, einer Alkoxylgruppe, einer Nitrogruppe oder einem Halogenatom substituiert ist, darstellt.

Verfahren nach Anspruch 1, wobei R<1> und R<2> verschieden sind und unabhängig voneinander ein Wasserstoffatom, ein Halogenatom, eine lineare, verzweigte oder zyklische Alkylgruppe mit 1 bis 10 Kohlenstoffatomen, welche wahlweise mit einem Halogenatom oder einer (C1-C5)-Alkoxylgruppe substituiert sein kann, eine (C2-C3)-Alkenylgruppe, welche wahlweise mit einer (C1-C5)-Alkoxylgruppe oder einem Halogen-atom substituiert ist, eine (C7-C11)-Aralkylgruppe, welche wahlweise mit einer (C1-C5)-Alkylgruppe, einer (C1-C5)-Alkoxylgruppe oder einem Halogenatom substituiert ist, eine Phenyl-, Naphthyl-, Anthranyl- oder Phenanthrylgruppe,
welche wahlweise mit einem Halogenatom, einer (C1-C5)-Alkyl- oder einer (C1-C5)-Alkoxylgruppe substituiert sein kann, darstellen, R<><a> eine (C1-C5)-Alkylgruppe, eine (C7-C8)-Aralkylgruppe, eine Silylgruppe mit drei (C1-C4)-Alkylgruppen, eine (C2-C6)-Acylgruppe darstellt. R<b> eine (C2-C6)-Acylgruppe, eine (C1-C5)-Alkoxycarbonylgruppe oder eine (C1-C5)-Alkylsulfonylgruppe, eine (C7-C8)-Aralkyloxycarbonylgruppe darstellt, oder die R<1>- oder die R<2>-Gruppen eine Gruppe der Formeln (9a) bis (9d) bilden:

3. Verfahren nach Anspruch 1 oder 2, wobei entweder die R<1>-Gruppen oder die R<2>-Gruppen Wasserstoffatome sind.

Verfahren nach Anspruch 1 oder 2, wobei die R<2>-Gruppen eine Gruppe der Formel (9c) bilden: -NR<21>CON(R<21>)- worin R<><21> eine Alkylgruppe, welche wahlweise mit einer Alkoxylgruppe oder einem Halogenatom substituiert ist, eine Aralkylgruppe, welche wahlweise mit einer Alkylgruppe, einer Alkoxylgruppe oder einem Halogen-atom substituiert ist, eine Arylgruppe, welche wahlweise mit einer Alkylgruppe, einer Alkoxylgruppe oder einem Halogenatom substituiert ist, darstellt, und die R<1>-Gruppen Wasserstoffatome sind.
5. Verfahren nach Anspruch 4, wobei die R<21>-Gruppen eine Benzylgruppe sind.
6. Verfahren nach Anspruch 1 oder 2, wobei die Menge des asymmetrischen Katalysators eine katalytische Menge ist.

Verfahren nach Anspruch 1 oder 2, wobei die Umsetzung des zyklischen Säureanhydrids der Formel (2) mit der Hydroxyverbindung der Formel (3) in Anwesenheit einer Base, welche aus einer anorganischen Base oder einer aromatischen tertiären Aminverbindung ausgewählt ist, durchgeführt wird.
8. Verfahren nach Anspruch 7, wobei die anorganische Base ein Carbonat eines Alkalimetalls oder eines Erdalkalimetalls oder ein Bicarbonat eines Alkalimetalls ist.
9. Verfahren nach Anspruch 7, wobei die aromatische tertiäre Aminverbindung eine Pyridinverbindung ist.

Verfahren nach einem der Ansprüche 1 oder 2, wobei der optisch aktive Ligand ein optisch aktiver Aminoalkohol der Formel (7) ist:

worin R<8> und R<9> verschieden sind und R<5>, R<7>, R<8>, R<9>, R<10 >und R<11> unabhängig voneinander ein Wasserstoffatom, eine wahlweise substituierte Alkylgruppe, eine wahlweise substituierte Alkenylgruppe oder eine wahlweise substituierte Arylgruppe darstellen, oder R<9> und R<10> derart gebunden sein können, dass eine wahlweise substituierte Alkylengruppe gebildet wird.

Verfahren nach Anspruch 10, wobei R<8> und R<9 >verschieden sind und R<6>, R<7>, R<8>, R<9>, R<10> und R<11> unabhängig voneinander ein Wasserstoffatom, eine lineare, verzweigte oder zyklische Alkylgruppe mit 1 bis 10 Kohlenstoffatomen, welche wahlweise mit einem Halogenatom oder einer (C1-C5)-Alkoxylgruppe substituiert sein kann, eine (C2-C3)-Alkenylgruppe, welche wahlweise mit einer (C1-C5)-Alkoxylgruppe oder einem Halogen-atom substituiert ist, eine (C7-C11)-Aralkylgruppe, welche wahlweise mit einer (C1-C5)-Alkylgruppe, einer (C1-C5)-Alkoxylgruppe oder einem Halogenatom substituiert ist, eine Phenyl-, Naphthyl-, Anthranyl- oder Phenanthrylgruppe, welche wahlweise mit einem Halogenatom, einer (C1-C5)-Alkyl- oder (C1-C5)-Alkoxylgruppe substituiert ist, darstellen, oder R<9> und R<10> derart gebunden sein können,
dass eine Ethylen- oder Trimethylengruppe gebildet wird, welche mit einem Benzolring anneliiert sein kann.
12. Verfahren nach Anspruch 1 oder 2, wobei die Lewis- Säure-Verbindung ein Titanalkoxid ist.
13. Verfahren nach Anspruch 1 oder 2, wobei der optisch aktive Ligand ein optisch aktives Bisoxazolin der Formel (8) ist:

worin R<12> ein Wasserstoffatom, eine wahlweise substituierte Alkylgruppe, eine wahlweise substituierte Arylgruppe darstellt und die zwei geminalen Alkylgruppen an deren Endstellen derart gebunden werden können, dass ein Ring gebildet wird, R<13> eine wahlweise substituierte Alkylgruppe, eine wahlweise substituierte Arylgruppe darstellt, und R<><14> ein Wasserstoffatom, eine wahlweise substituierte Alkylgruppe, eine wahlweise substituierte Arylgruppe darstellt, und die durch "*" bezeichneten Kohlenstoffatome asymmetrische Kohlenstoffatome mit einer S- oder R-Konfiguration darstellen.

Verfahren nach Anspruch 13, wobei R<<>12> ein Wasserstoffatom, eine lineare, verzweigte oder zyklische Alkylgruppe mit 1 bis 10 Kohlenstoffatomen, welche wahlweise mit einem Halogenatom oder einer (C1-C5)-Alkoxylgruppe substituiert sein kann, eine (C7-C11)-Aralkylgruppe, welche wahlweise mit einer (C1-C5)-Alkylgruppe, einer (C1-C5)-Alkoxylgruppe oder einem Halogenatom substituiert ist, eine Phenyl-, Naphthyl-, Anthranyl- und Phenanthrylgruppe, welche wahlweise mit einem Halogenatom, einer (C1-C5)-Alkylgruppe oder einer (C1-C5)-Alkoxylgruppe substituiert sein kann, darstellt, R<13> eine lineare, verzweigte oder zyklische Alkylgruppe mit 1 bis 10 Kohlenstoffatomen, welche wahlweise mit einem Halogenatom oder einer (C1-C5)-Alkoxylgruppe substituiert sein kann, eine (C7-C11)-Aralkylgruppe, welche wahlweise mit einer (C1-C5)-Alkylgruppe, einer (C1-C5)

Verfahren nach Anspruch 1 oder 2, wobei die Lewis- Säure-Verbindung Zinnchlorid ist.
16. Verfahren zum Herstellen eines optisch aktiven Imidazolidin-2-ons der Formel (4):

18. Verfahren nach Anspruch 16 oder 17, wobei die Umsetzung des zyklischen Säureanhydrids der Formel (5) mit der Hydroxyverbindung der Formel (3) in Anwesenheit der optisch aktiven Alkaloidverbindung und einer Base durchgeführt wird.

Verfahren nach Anspruch 16, wobei R<21> eine Aralkylgruppe ist, welche wahlweise mit einer Alkylgruppe, einer Alkoxylgruppe oder einem Halogenatom substituiert ist.
20. Verfahren nach Anspruch 17, wobei R<21> eine Benzylgruppe ist.
21. Verfahren nach Anspruch 20, wobei das Alkaloid Chinin ist.
22. Verfahren nach Anspruch 18, wobei die Menge des optisch aktiven Alkaloids eine katalytische Menge ist.
23. Verfahren nach Anspruch 18, wobei die Base ein aliphatisches oder aromatisches tertiäres Amin ist.
24. Verfahren nach Anspruch 18, wobei die Base eine Piperidinverbindung ist.
