Verfahren zur Isolierung von Colchicin. Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Isolierung von Colchi- cin aus pflanzlichem Ausgangsmaterial, z. B. der Droge, wie z. B. Samen oder Knollen von Colchicum autumnale L. oder einer Mi schung von daraus bereiteten geeigneten Ex trakten und natürlicher Zellsubstanz, z. B. Blattpulver.
Der geringen Basizität des Colchicins, das in wässeriger Lösung gegen Lakmus neutral reagiert, wird in dem Verfahren Rechnung getragen. Der Zusatz von sauer wirkenden Reagentien zur Fixation des Alkaloides in natürlicher Zellsubstanz beschränkt sich auf die Anwendung von stark sauren Mitteln, d. h. von Mineralsäuren, z. B. Schwefelsäure, deren Menge erst ausreicht, wenn sie in schwachem Überschuss angewandt werden.
Der geeignete Überschuss von Mineralsäure ist vorhanden, wenn aus einer Probe der Zellsubstanz mit einem Gemische von 4 Tei len Alkohol und 6 Teilen Benzol freie Mi neralsäure, z. B. Schwefelsäure, gerade ex trahiert werden kann.
Das Verfahren beruht demnach darauf, dass man durch Zusatz von Mineralsäuren in geringem Cberschuss das Alkaloid iii natürlicher Zellsubst.anz gegen über mit Wasser nicht mischbaren, niedrig siedenden Fettlö sungsmitteln fixiert, worauf man deni Material durch erschöpfende Vor- cxtraktion mit einem dieser Fettlösungsmit- tel die löslichen,
nicht basischen Begleit stoffe des Alkaloides entzieht, dann das so voi-gereinig(,e Extraktionsgut in neuen Men gen Fettlösungsmittel suspendiert und alka lisch macht., worauf man das in Freiheit ge setzte Alkaloid aus der Zellsubstanz heraus löst und den so gewonnenen alkaloidhal- tigen Extrakt bei niedriger Temperatur zur Trockne verdampft. Das hinterbleibende :
iocl)prozentige Colch.icin wird zweckmässig auf bekannte Weise, z. B. über die kristalli sierende Chloroformverbindung, von Neben alkaloiden und geringen Mengen nicht ba sischer Stoffe befreit.
Bei clei@ geringen Absorptionsfähigkeit, wie sie selbst stark angesäuerter Colchicum- samen bei der Filation des Colchicins zeigt. ist es vorteilhaft, den Samen vor der Ver arbeitung mit Blattpulver zu vermengen, um alsdann durch Ansäuren dieser Mischung eine vollständige Fixation des Alkaloides gegenüber Fettlösungsmitteln zu erreichen.
<I>Beispiel 9:</I> Eine feinpulverige Mischung von 1 kg Herbstzeitlosensamen und '/_ kg Blattmehl, das mit. Alkohol und Benzol vorextrahiert worden war, wird mit so viel 50 %iger Schwefelsäure in einer säurebeständigen Mischtrommel behandelt, dass eine Lösung voll 4 Teilen Alkohol und 6 Teilen Benzol g erzade kleine Mengen freier Schwefelsäure zu extrahieren vermag.
Alsdann befeuchtet Tran das Pflanzenpulver mit 0,'7 Liter Benzol und extrahiert es im Perkolator erschöpfend reit Benzol. Das auf diese Weise von nicht basischen Stoffen befreite Material wird nun in # Liter Benzol suspendiert und die be wegte Suspension mit gasförmigem Ammo niak bis zur deutlich alkalischen Reaktion behandelt. Nach zweistündiger Einwirkung von Ammoniak und Lösungsmittel saugt man auf der 'rutsche ab und wäscht mit Benzol in Portionen von 1/.. Liter erschöp fend nach.
Der Extrakt wird unterhalb 40" zur Trockne verdampft, wobei ein liellgelb- geUrbtes sprödes, in verdünnter Säure nahe-. zii restlos lösliches Produkt hinterbleibt. Aus diesem hochprozentigen Rohcolchiciii wird das reine Alkaloid auf bekannte Weise von Begleitstoffen befreit, als Chloroformverbin- dung, kristallisiert und in einheitlichem Zu stande erhalten.
<I>Beispiel</I> 2.- 2 kg staubfein gepulverte Radix Colchici werden in einer Mischtrommel mit 240 gr Schwefelsäure r1 <I>:
</I><B>1)</B> während einer halben Stunde innig vermischt, dann mit 0,8 Liter Benzol befeuchtet und im Perkolator mit Benzol erschöpfend vorextrahiert. Die in diesem Versuche zur Fixation des Alkaloides angewandte Säuremenge ist noch zu gering, als dass mit einem Gemische von 6 Teilen Benzol und 4 Teilen Alkohol freie Schwefel säure aus der Droge hätte extrahiert werden können. Der Vorextrakt zeigt denn auch schwache Alkaloidreaktionen, Evas natürlich einen kleinen Verlust an Alkaloid zur Folge hat.
Trotzdem lohnt sieh die Weiterverarbei tung des Material; es wird in S Liter Ben zol suspendiert, worauf man durch Einlei ten von gasförmigem Ammoniak in die be wegte Suspension bis zu deutlich alkalischer lleaktion (las Colchicin in Lösung bringt.
Man saugt alsdann auf der Nutsche ab, N,r,--äsclit mit Benzol portionenweise erschöp fend nach und verarbeitet den colchirciii- haltigen Extrakt, Wie im Beispiel 1 ange geben.
Method for isolating colchicine. The present invention relates to a method for isolating colchicin from plant starting material, eg. B. the drug, such as. B. seeds or tubers of Colchicum autumnale L. or a Mi mixture of suitable extracts prepared therefrom and natural cell substance, eg. B. leaf powder.
The method takes into account the low basicity of colchicine, which reacts neutrally against lacmus in an aqueous solution. The addition of acidic reagents to fix the alkaloids in natural cell matter is limited to the use of strongly acidic agents, i. H. of mineral acids, e.g. B. sulfuric acid, the amount of which is only sufficient if it is used in a slight excess.
The appropriate excess of mineral acid is present when mineral acid from a sample of the cell substance with a mixture of 4 parts of alcohol and 6 parts of benzene free mineral acid, e.g. B. sulfuric acid, just ex can be extracted.
The method is based on the fact that by adding a small excess of mineral acids, the alkaloid of natural cell substance is fixed against water-immiscible, low-boiling fat solvents, whereupon the material is fixed by exhaustive pre-extraction with one of these fat solvents the soluble ones

With clei @ low absorption capacity, as demonstrated by even strongly acidified colchicine seeds during the filation of colchicine. It is advantageous to mix the seeds with leaf powder before processing, in order to then achieve a complete fixation of the alkaloids against fatty solvents by acidifying this mixture.
<I> Example 9: </I> A finely powdered mixture of 1 kg autumn crocus seeds and 1 / _ kg leaf flour, which is mixed with Alcohol and benzene had been pre-extracted, is treated with enough 50% sulfuric acid in an acid-resistant mixing drum that a solution full of 4 parts of alcohol and 6 parts of benzene is able to extract even small amounts of free sulfuric acid.
Then Tran moistened the plant powder with 0.7 liters of benzene and extracted it exhaustively in the percolator with benzene. The material freed from non-basic substances in this way is then suspended in # liters of benzene and the suspension is treated with gaseous ammonia until it has a clearly alkaline reaction. The extract is evaporated to dryness below 40 ", leaving behind a slightly yellow, brittle product that is almost completely soluble in dilute acid. The pure alkaloid is freed from accompanying substances in a known manner from this high-percentage crude colic, as a chloroform compound and received in a uniform condition.
<I> Example </I> 2.- 2 kg of finely powdered Radix Colchici are mixed with 240 g of sulfuric acid r1 <I> in a mixing drum:

The amount of acid used to fix the alkaloids in this experiment is still too small for free sulfuric acid to be extracted from the drug with a mixture of 6 parts of benzene and 4 parts of alcohol. The pre-extract shows weak alkaloid reactions, which Evas naturally results in a small loss of alkaloid.
Nevertheless, it is worth processing the material; it is suspended in 5 liters of benzene, whereupon it is brought into solution by introducing gaseous ammonia into the moving suspension until it becomes a distinctly alkaline reaction (as in colchicine.
It is then suctioned off on the suction filter, N, r, -asclit exhaustively with benzene in portions, and the extract containing colchircii is processed, as indicated in Example 1.
