Abstract
Die Erfindung betrifft ein Verfahren für die Herstellung leicht zu spaltender Enzymsubstrate für die Quantifizierung von Proteasen mit der allgemeinen Fornel R, - p-Glu - A-j - A, - NH - R2
Description
Die Erfindung betrifft ein Verfahren zur Herstellung neuartiger, leicht zu spaltender Enzymsubstrate, die für den Einsatz als Reaktionsinittel bei der quantitativen Bestimmung von Serinproteasen und SH-Proteasen, oder zur Untersuchung von Reaktionen, bei denen Serinproteasen gebildet, inhibiert oder verbraucht werden, oder zur Bestimmung von Paktoren, die derartige Reaktionen beeinflussen oder an ihnen beteiligt sind, vorgesehen sind·
Charakteristik der bekannten technischen Lösungen
Kürzlich wurde eine Reihe synthetischer Peptidsubstrate beschrieben. Die Mehrzahl von ihnen besitzt eine Tripeptidkette, bei der entv/eder die Aminogruppe der N-terminalen Aminosäure durch eine Acylgruppe blockiert ist oder frei ist, dann allerdings die D-Konfiguration hat. Die C-terminale Aminosäure wird durch eine Chromophor- oder Fluorophorgruppe blockiert, die bei der Reaktion des. Enzyms mit dem Substrat abgespalten wird. Die auf diese Weise gebildete, freigesetzte chromophore oder fluoreszierende Gruppe kann quantitativ durch photometrische oder photometrische Fluoreszenzmethoden bestimmt werden. Die kurze Peptidkette ist für die Affinität des Substrates hinsichtlich des Enzyms von Bedeutung. In diesem Zusammenhang scheint es wichtig zu sein, daß die H-terminale Amino-'säure entweder die L-Form mit einer acylierten Aminogruppe oder die D-Form mit einer freien Aminogruppe aufweist.
Ziel der Erfindung
Ziel der Erfindung ist die Bereitstellung von leicht su spaltenden Enzymsubstraten, deren Reaktionsgeschwindigkeit mit den zu bestimmenden Proteasen und deren Sensibilität wesentlich erhöht ist.
Der Erfindung liegt die Aufgabe zugrunde, Enzympräparate mit leicht abspaltbaren Gruppen zu versehen. Es wurde ermittelt, daß Substrate mit einer U-terminalen Pyroglutaminsäure durch eine Reihe von Serinproteasen sehr leicht gespalten v/erden. Pyroglutaminsäure, deren qü-Aminogruppe nicht frei, sondern durch Ringschluß acyliert ist, d. h. Lactam der Glutaminsäure, wurde vorher nicht bei Peptiden verwendet, die für den

Einsatz als Substrate bei der Bestimmung von Enzymen vorgesehen waren* Erfindungsgemäß v/erden neuartige chromophote Substrate oder die Salze davon hergestellt, die der folgenden allgemeinen Formel:
H-j - g-Glu -A-J-A2-entsprechen, v/orin
R·} Wasserstoff oder eine Schutzgruppe, vorzugsweise t-Butyloxycarbonyl oder Benzyloxycarbonyl sein kann;
A-J unter den Aminosäuren Glys Ala, VaI, Leu, lie, Sei·, Thr, Pro, Pip, Phe oder Tyr ausgewählt sein kann;
A2 Arg oder Lys sein kann;
R2 eine aromatische, möglichst substituierte, Kohlenwasserstoffgruppe sein kann, worin -KFI-R2 eine, physikalisch-chemisch bestimmbare Gruppe darstellt, vorzugsweise eine Chromophor- oder Fluorophorgruppe, die durch das genannte Enzym gespalten wurde und dann ein Spaltprodukt der Formel H2N - R2 bildet, deren Menge quantifiziert werden kann·
Bei der Synthese der neuartigen erfindungsgemäßen Substrate werden in der Peptidchemie allgemein bekannte, herkömmliche Schutzgruppen und Kopplungsniethoden angewandt·
Das Prinzip der Synthese kann in einer stufenweisen Kopplung der Aminosäuren an den C-terminalen Arginyl- oder Lysyirest bestellen, der entweder anfangs mit einer angekoppelten bestimmbaren Gruppe (-"HXT9-Rp entsprechend der Formel), die dann als Carboxyischutzgruppe dient, oder mit einer Carboxyl-

schutzgruppe, die entfernbar ist, versehen sein kann, und in diesem Fall ist die bestimmbare Gruppe an das geschützte Tripeptidderivat gebunden, oder die N-terminale geschützte Dipeptidfraktion kann auch getrennt synthetisiert und dann an den Arginyl- oder Lysylrest mit oder ohne bestimmbare Gruppe nach dem obigen Vorgang gebunden werden. Gleich, welche Synthesewege gewählt werden, die Zwischen- und Endprodukte v/erden durch Rekristallisation und/oder Filtrationschromatographie gereinigt.
Insbesondere werden erfindungsgemäß leicht spaltbare Enzymsubstrate für Proteasen der allgemeinen Formel
R-J - p-Glu - A-J - A2 - UH - R2
in der Weise hergestellt, daß die Synthese entweder durch eine schrittweise Kopplung an A2, das R2 oder eine entfernbare Carboxylschutzgruppe trägt, die dann durch R2 substituiert wird, durchgeführt wird, oder von R- -p-Glu-A, worin R^ eine Schutzgruppe ist, vorzugsweise t-Butyloxycarbonyl oder Benzyloxycarbonyl, ausgegangen wird, und an A2 - IiH .- R2-gekoppelt wird, wobei R2 getrennt als Ersatz für eine entfernbare Carbonylschutzgruppe eingebaut werden kann. , .

Die Erfindung betrifft weiterhin ein Verfahren für die Labordiagnostik von Proteasen, das dadurch gekennzeichnet ist, daß als Substrat eine Verbindung der allgemeinen Formel

Ausführungsbeispiel
Die Erfindung wird anhand folgender Beispiele, erläutert.
die Dünnschichtchromatographieanalyse der Eluate und des Produktes v/erden Glasplatten mit Silicagel P254 (Merck) als Absorptionsmedium verwendet. Bei den benutzten Lösungsmittelsystemen handelt es sich um:
A: n-Butanol:HOAc:Wasser 3:2:1 (Volumen) P1: Chloroform:HeOH 9:1 (Volumen)

Ia Cbo-Gly-Arg(N02)pNa (Molvolumen = 530,5)
175 ml konzentrierte EOAc und 105 ml 5,6H-HBr in HOAc werden zu 35 g (0,074 Mol) Cbo-Arg(N02)-pNA gegeben. Das Gemisch wird zur Reaktion stehen gelassen und nach dem Klären der Reaktionslösung 30 Minuten lang gerührt. Das Gemisch wird anschließend unter kräftigem Rühren in etwa 2 1 trockenen Äther gegossen. Das entstandene Präzipitat wird filtriert, mit trockenem Äther gewaschen und danach über NaOH vakuumgetrocknet. Das gewonnene HBr-SaIz von H-Arg(N02)-pNA wird in 150 ml trockenem destilliertem DMP gelöst und bei niedriger Temperatur (-10 0C) mit Ät-,Η neutralisiert, bis eine schwach basische Reaktion auf einem angefeuchteten pH-Papier über dem Reaktionsgemisch erzielt wird. (Gewöhnlich v/erden etwa 1,5 Äquivalente HBr neutralisiert.) Das entstandene At.,NHBr wird abfiltriert. Unter kalten Bedingungen wird 1,1 Äquivalent = 0,81 Mol = 26,8 g Cbo-Gly-OpNP dem Reaktionsgemisch zugesetzt; nach 30 Minuten wird ein weiteres 1/2 Äquivalent = 5,2 ml Ät-alT zugegeben. Das Gemisch läßt man zur Reaktion über Nacht bei Raumtemperatur stehen, und anschließend wird es zur Gev/innung eines Öls unter Vakuum eingedampft. Das Öl wird in einer wäßrigen Lösung von 2 % NaHCOo trituriert und anschließend in heißem MeOH gelöst und unter Rühren und Kühlen auskristallisiert. Die entstandenen Kristalle werden abfiltriert und mit kaltem MeOH gewaschen. Die Dünnschichtchromatographie zeigt nur geringfügige Verunreinigungen, und daher wird das Produkt aus dem gleichen Lösungsmittel rekristallisiert. Ib1 Cbo-p-Glu-OH: (relative Molekülmasse = 263,3) 56,3 g (0,20 Mol) Cbo-Glu-OH werden in 400 ml ÄtOAc gelöst und 39,4 g (0,24 Mol) DCCI, in 60 ml ÄtOAc gelöst, werden zugegeben, und es wird in einem Eisbad gerührt. Nach zwei Stunden, wird der entstandene DCU filtriert, und die zurückbleibende Lösung wird zur Trocknen eingedampft. Das Produkt (Anhydrid von Cbo-G-lu) wird aus ÄtOAc und Petroläther auskristallisiert. Das Anhydrid wird in 120 ml Dioxan plus 260 ml Äther gelöst, und anschließend werden 48 ml in Äther gelöstes DCHA bis zu einem Volumen von 100 ml zugegeben. Uach kurzer Zeit setzt sich Cbo-p-Glu.DCHA ab. Ss wird etwa 1 Stunde lang gerührt, und danach wird das gebildete DCHA-SaIz abfiltriert und mit Äther und ÄtOAc gewaschen. Das DCHA-SaIz wird in ÄtOAc suspendiert, und mit 25 g (0,18 Mol) in Wasser gelöstem KHSO. geschüttelt, und das dadurch gebildete Cbo-p-Glu-OH geht dann in die ÄtOAc-Phase über. Die ÄtOAc-Phase wird mit 10 % NaCl in Wasser gewaschen und über Na2SO. getrocknet (wobei die Gefahr der Präzipitation des Produktes besteht). Die ÄtOAc-Phase wird zu einem kleinen eingedampft und mit Petroläther ausgefällt; dadurch ergeben sich schwere Kristalle von Ib1, die gemäß TLC rein sind.

10,25 g (18,3 inHol) H-GIy- Arg(HH2^ ^ gewonnen durch Entfernung des Schutzes von Cbo-Gly-ArgCiFC^pNA mit HBr nach der in Ia beschriebenen Verfahrensweise, v/erden in 35 ml trockenem DIvIP gelöst und bei einer niedrigen Temperatur (-1G 0C) mit At-JJ neutralisiert, bis eine basische Reaktion auf angefeuchtetem pH-Papier erfolgt» Die gewonnene ÄtNHBr wird abfiltriert· 2,47 g (18,3 mMol) HOBT und 5,26 g (20 mMol) Cbo-p-Glu-OH (Ib') v/erden zu dem Reaktionsgemisch gegeben, und danach v/erden 4,53 g (22 mMol) in einer geringen Menge DMF gelöstes DGCI bei einer niedrigen Temperatur zugesetzt. Das Gemisch läßt man zur Reaktion über Nacht bei Raumtemperatur stehen; anschließend wird die Reaktionslösung zu einem Öl eingedampft. Das Öl wird mit 2 % HaHCOo in Wasser und mit reinem Wasser trituriert. Die gewonnene feste Verbindung wird in einer geringen Menge Aceton gelöst und danach werden heißes MeOH und eine kleine Menge V/asser zugesetzt. Das Produktv-wird unter Kühlen und Rühren auskristallisiert, und die entstandenen Kristalle v/erden abfiltriert. Das gewonnene Ib ist gemäßTLC rein. Nach sorgfältiger Vakuumtrocknung werden 8,90 g (76 %) Ib gewonnen.

I. Von 0,8 g (1,25 mMol) Ib wird mit 30 ml EF bei 0 0C in einem Zeitraum von 1 Stunde in Gegenwart von 0,6 ml Anisol der Schutz entfernt. Nach dein Eindampfen unter Vakuum wird das Produkt in etwa 30 ml 2/oiger HOAc gelöst und mit einer geringen Menge Äther gewaschen. Die HpO-Phase wird auf einer Säule mit Sephadex G-15

(Pharmacia Pine Chemicals) in 2 %iger HOAc mit dem gleichen Sluierungsniittel chromatografiert. Die Fraktion mit dem reinen Acetat von I wird lyophylisiert und auf einer Säule mit QAE-25 (Pharmacia Pine Chemicals) in Form von Chlorid mit AtOH-H2O (1:1) mit dem gleichen Eluierungsmittel dem Ionenaustausch unterzogen.
Die Fraktion mit dem reinen Hydrochlorid von I wird lyophilisiert.
Ausbeute: 485 mg (78 %) von I.
TLC Rf = 0,29 (A) zeigt nur einen Fleck.

Die in Tabelle I beschriebenen Beispiele II bis XIII werden im Prinzip wie die vorstehenden Beispiele durchgeführt, daher sind nur die physikalischen Werte, die Ausbeuten sowie die Kopplungs- und Reinigungsmethoden in dieser Tabelle aufgeführt.
p-Glu-Gly-Arg-4-MeO-ß-NA.HCl (relative Molekülmässe = 534,0) XIVa p-Glu-Gly-Arg-OH.HCl (relative Molekülmasse = 378,8)
Trypsin (Νονό, 400 nl einer lösung von 2,0 mg Trypsin pro ml 1 mM HCl) wird zu einer Lösung aus* 250 mg (0,50 mlvlol) p-Glu-Gly-Arg-pNA.HCl (S-2444) in 100 ml 0,1M NaHCO3 in H2O gegeben. Die Freisetzung von pNA wird mit Hilfe eines Spektrophotometers bei 405 nm verfolgt. Die Freisetzung ist nach etwa 45 Minuten beendet, und die Lösung wird anschließend mit HOAc bis auf einen pH-Wert von annähernd 4 angesäuert.

Sie wird dann zur Trockne eingedampft, in MeOH gelöst und auf einer Säule mit LH-20 (Pharmacia Fine Chemicals) in MeOH mit dem gleichen Eluierungsmedium chromatorgrafiert. Die Fraktion mit der reinen Säure XVIa wird eingedampft, in H2O gelöst und lyophilisiert,
Ausbeute: 167 mg (88 %) XIVa, das gemäß TLC rein ist. Rf = 0,08 (A)
XIV:
Eine Lösung von 27 /Ul (0,25 mlvlol) PCl3 in 450 ul trockenem Pyridin wird unter trockenen Bedingungen und in einem Eisbad zu einer Lösung von 105 mg (0,50 rnMol) 4-MeO-B-NA.HCl in 2,25 ml trockenem Pyridin gegeben. Nach etwa 45 Minuten Stehen bei Raumtemperatur wird das gesamte nach obiger Beschreibung hergestellte XIVa zugegeben. Wenn die Reaktion aufhört (nach etwa 1 Stunde) wird das Reaktionsgemisch zu einem dunkelroten Öl eingedampft (unter reduziertem Druck). Das Öl wird in einer geringen Menge AtOH-H2O (1:1) gelöst und auf eine Säule mit QAE-25 (Pharmacia Fine Chemicals) in Form von Chlorid in XtOH-HpO (1:1) mit dem gleichen Eluierungsmedium gegeben. Die Fraktion mit dem nahezu reinen Hydrochlorid von XIV wird eingedampft, wieder in einer geringen Menge AtOH-H2O (1:1) gelöst, und diese Verfahrenswelse wird wiederholt. Das dadurch gewonnene Hydrochloric von XIV ist rein und wird nach dem Verdampfen des gesamten Alkohols lyophilisiert.
Ausbeute: 85 mg (36 %) von XIV.

Bestimmung; von Proteasen mit Hilfe von Substraten mit einer entfernbaren und auf physikalisch-chemischem Wege bestimmbaren Gruppe . .
Me nach den Beispielen erzeugten Substrate werden zur Bestimmung der verschiedenen Enzyme in folgender Weise verwendet.
Das Prinzip der Bestimmung basiert auf der Tatsache, daß das durch die enzymatische Hydrolyse gewonnene Spaltprodukt ein UV-Spektrum zeigt, das sich ganz wesentlich von dem des Substrates unterscheidet. So haben alle erfindungsgemäßen p-Nitroanilin-Substrate zum Beispiel ihre Absorptionsmaxima bei 310 nm bei einem molaren Extinktionskoeffizienten von annähernd 12000. Bei 405 nm hat die Absorption dieser Substrate fast gänzlich aufgehört. p-Nitroanilin, das bei der enzymatischen Hydrolyse von dem Substrat freigesetzt wird, hat sein Absorptionsmaximum bei 380 nm mit einem molaren Extinktionskoeffizienten von 13200, der bei 405 nm nur auf 9620 abgesunken ist. Durch spektrophotometrische Messung bei 405 nm ist es daher leicht, die gebildete Menge p-Nitroanilin zu verfolgen, die der Geschwindigkeit der enzymatischen Hydrolyse proportional ist, die ihrerseits durch die aktive Menge des Enzyms bestimmt wird.
Um die Menge des gebildeten fluoreszierenden Amins zu messen, werden die Proben in einem Fluoreszenzspektrophotometer mit einem Licht, das eine Anregungswellenlänge (etwa 350 nm für ß-Naphthylamin und 4-Methoxy-ßnaphthylamin, und 380 nm für 7-Amino-4-methyi-coumarin) hat, bestrahlt, und die Menge des abgespaltenen Pro-

duktes wird dann durch Messen des emittierten Lichtes (etwa 420 nm für ß-Naphthylamin und 4-Methoxyß-naphthylamin, und 440 nm für 7-Amino-4-methylcoumarin) ermittelt.
In Tabelle II ist die Reaktionsgeschwindigkeit der synthetischen Substrate mit verschiedenen Enzymen aufgeführt. Die Reaktionsgeschwindigkeit wird auf ein Bezugssubstrat (Reaktionsgeschwindigkeit = 100) für jedes Enzym bezogen. Einige der Bezugssubstrate sind bereits bekannt und im Handel erhältlich.
Die Nützlichkeit der neuartigen erfindungsgemäßen Substrate und die Tatsache, daß sie durch verschiedene Enzyme schneller hydrolysiert v/erden als die bisher zur Verfügung stehenden Substrate, gehen aus dieser Tabelle II hervor.
Claims (2)
1. Diagnostisches Mittel mit guter löslichkeit und hoher Spezifität gegenüber Serinproteasen und SH-Proteasen, gekennzeichnet dadurch, daß es ein chrcmophores Substrat der allgemeinen Formel

R2 β eine aromatische, möglichst substituierte, Kohlenwasserst off gruppe, worin -NH-H2 eine physikalischchemisch bestimmbare Gruppe ist, vorzugsweise eine Chromophor- oder Fluorophorgruppe, die durch ein , anwesendes Enzym gespalten wird und dann ein Spaltprodukt der Formel H2H-Rp bildet, dessen Menge quantifiziert werden kann;
neben üblichen Hilfsstoffen enthält.
2« Mittel nach Punkt 1, gekennzeichnet dadurch, daß R2 = pHA, ß-2»TA, 4-MeO-B-IU, 7-A-4-M-C ist.
