Abstract
Die Erfindung betrifft ein Verfahren zur Herstellung von Verbindungen der allgemeinen Formel I, worin X die Gruppe > CO oder > CH(OH), R tief 2 Wasserstoff oder eine C tief 1-C tief 6-Alkylgruppe und R tief 3 Wasserstoff oder eine Hydroxygruppe ist und R tief 1 den Adamantylrest oder einen gesaettigten oder einfach ungesaettigten C tief 3-C tief 16-Cycloalkylrest bedeutet, wobei dieser C tief 3-C tief 16-Cycloalkylrest auch durch eine C tief 1-C tief 4-Alkylgruppe oder ein Halogenatom substituiert sein kann und deren Saeureadditionssalzen durch Umsetzung entsprechender N,N'-Methylen-bis-oxazolidine mit Ketonen der Formel R tief 1-CO-C(R tief 2)H tief 2 im sauren Medium und gegebenenfalls anschliessender Reduktion.

<p>Die erfindungsgemäß hergestellten Verbindungen werden als Arzneimittel angewandt. Sie sind pharmakodynamisch wirksam und bewirken beispielsweise eine Verbesserung der cerebralen oder peripheren Durchblutung·</p>

<p>Durch die DE-OS 2 919 495 sind verschiedene Verfahren zur Herstellung von Verbindungen der allgemeinen Formel</p>

Im einzelnen handelt es sich zum Beispiel um organische Ionenaustauscher, deren aktive, das heißt, ionenbildende Gruppe die Carboxylgruppe oder eine organische saure Gruppe wie zum Beispiel die SuIfonsäuregruppe (SO<sub>3</sub>H) oder Phosphorsäur egruppe ist, während das hochmolekulare Gerüst (Matrix) aus einem Kunstharz (Acrylharz, Polystyrolharz, Styrol-Divinylbenzol-Copolymerisat, Styrol-Acrylsäure-Copolymerisat, Divinylbenzol-Polymerisat, Vinylbenzol-Polymerisat, Kondensationsprodukte aus Phenol und Formaldehyd) besteht. Weiterhin kommen in Betracht: saure Cellulose-, Stärke- und Dextran-Ionenaustauscher, Kohle-Ionenaustauscher (aus sulfonierten hochmolekularen Humuskohlen) sowie saure anorganische Ionenaustauscher wie Zeolithe und Aluminiumsilikate.</p>
<p>Der pH-Wert der Reaktionslösung beziehungsweise der Reaktionsmischung soll zwischen 1 - 6, vorzugsweise</p>
<p>20 2-3 liegen.</p>
<p>Die Ausgangsstoffe der Formel. II können beispielsweise dadurch erhalten werden, daß man ein Phenyläthylamin der allgemeinen Formel</p>

Diese Umsetzung erfolgt in einem Lösung's- oder Suspensionsmittel bei Temperaturen zwischen 20 und 180° C, insbesondere zwischen 30° C und 150° C. Als Lösungs- beziehungsweise Suspensionsmittel kommen in Betracht: Wasser, niedere gesättigte</p>
<p>aliphatische C ..-C ,--Alkohole, niedere aliphatische gesättigte Ether mit Alkylresten aus 1-5 C-Atomen,</p>
<p>' aromatische Kohlenwasserstoffe wie Benzol, Methylbenzole (Toluol), Dimethylbenzole (Xylol). Im allgemeinen arbeitet man bei Siedetemperatur des verwendeten Lösungsmittels. Das bei der Reaktion entstehende Wasser kann beispielsweise durch Zugabe von Trocknungsmitteln wie Natriumsulfat, Kaliumcarbonat, Kaliumchlorid, Molekularsieben (zum Beispiel Typ 4A) oder durch azeotrope Destillation (zum Beispiel bei Verwendung aromatischer Kohlenwasserstoffe als Lösungsmittel) entfernt werden.</p>
<p>Das Verfahren muß unter Ausschluß von Säuren durchgeführt werden, da die Verfahrensprodukte säurelabil sind.</p>
<p>Im allgemeinen verwendet-man pro 1 Mol Phenyläthylamin.</p>
<p>'5 der Formel IV 1,5 - 5 Mol Formaldehyd. Vorzugsweise verwendet man 3 Mol Formaldehyd beziehungsweise das Äquivalent eines Formaldehyd liefernden Stoffes pro 2 Mol Phenyläthylamin der Formel IV. Als Formaldehyd liefernde Stoffe kommen beispielsweise in Frage:</p>
<p>^<sup>w</sup> Polyformaldehyd, Paraformaldehyd. Bei der Verwendung von Formaldehyd liefernden Stoffen, bei denen der Formaldeyhd im sauren Medium freigesetzt wird, sollte die Formaldehydfreisetzung der eigentlichen Reaktion vorgeschaltet und überflüssige Säure vorsichtig neu-</p>

<p>tralisiert werden, damit die gewünschten dimeren Oxazolidine nicht dem sauren Milieu ausgesetzt sind.</p>
<p>Beispielsweise kann das N ,N'-Methylen-bis-(4-methyl-5-phenyl-oxazolidin) wie folgt erhalten werden:</p>

<p>4 00 g (2,6 Mol) <i> £ </i>-Norephedrin-Base werden in 800 ml Wasser in der Hitze gelöst und 330 g 35%ige wässrige Formalinlösung tropfenweise zugesetzt. Die Reaktionsmischung wird 4 Stunden im siedenden Wasserbad erwärmt.</p>
<p>Anschließend wird mehrmals mit Methylenchlorid extra-35</p>
<p>hiert, mit K<sub>2</sub>CO<sub>3</sub> getrocknet und das Lösungsmittel im Vakuum entfernt. Der feste Rückstand wird aus Diisopropyläther umkristallisiert</p>
<p> Ausbeute: 423 g (94,5%) .5 F.: 98 - 99°C.</p>
<p>Die<sup>1</sup>Herstellung dieser Verbindung kann auch in folgender</p>
<p>abgeänderter Weise erfolgen:</p>
<p>10 g (0,066 Mol) €-Norephedrin-Base, 4 g (0,132 Mol) i« Paraformaldehyd, 6,6 g Natriumsulfat wasserfrei,</p>
<p>werden in 100 ml getrocknetem Xylol suspendiert</p>
<p>und unter Rühren 5 Stunden unter Rückfluß erhitzt.</p>
<p>Die anorganischen Bestandteile werden abfiltriert,</p>
<p>das Lösungsmittel im Vakuum eingeengt und das feste ir Rohprodukt aus Diisopropyläther umkristallisiert.</p>
<p>Ausbeute: 6,1 g = 55 %</p>
<p>F. : 98 - 99° C.</p>
<p>10 g (0,066 Mol) £ -Norephedrin-Base, 4 g (0,132 Mol) Paraformaldehyd, werden in 150 ml Xylol suspendiert und im Wasserabscheider für 5 Stunden unter Rückfluß erhitzt. Das Lösungsmittel wird im Vakuum eingeengt und der feste Rückstand aus Diisopropyläther umkristallisiert</p>
<p> Ausbeute: 7,6 g = 68 % F.: 100° C.</p>

<p>"i Ausgangsstoffe der Formel II, worin R<sub>3</sub> eine Hydroxygruppe bedeutet, können eine übliche Schutzgruppe enthalten, die nach der Umsetzung abgespalten wird. Es handelt sich hierbei insbesondere um Reste, die durch Hydrolyse in nicht saurem Medium leicht abspaltbar sind und gegebenenfalls bereits während der Reaktion abgespalten werden. Falls solche Schutzgruppen bei der Verfahrensreaktion nicht abgespalten werden, erfolgt eine Abspaltung nach der Reaktion. Häufig enthalten die Ausgangsverbindungen aufgrund ihrer Herstellung bereits derartige Schutzgruppen.</p>
<p>Bei diesen Schutzgruppen handelt es sich beispielsweise um leicht solvolytisch abspaltbare Acylgruppen Die solvolytisch abspaltbaren Schutzgruppen werden beispielsweise durch Verseifung mittels basischer Substanzen (Pottasche, Soda, wässrige Alkalilösungen, alkoholische Alkalilösungen, wässriges NHJ bei Temperaturen zwischen TO und 150° C, insbesondere 20 100° C, abgespalten. Als Lösungs- beziehungsweise Suspensionsmittel hierfür kommen beispielsweise in Betracht: Wasser, niedere aliphatische Alkohole, cyclische Ether wie Dioxan oder Tetrahydrofuran, aliphatische Ether, Dimethylformamid und so weiter . sowie Mischungen dieser Mittel.</p>
<p><i>* ·> </i>Beispiele für hydrolytisch abspaltbare Reste sind:</p>
<p>Trifluoracetylrest, Phthalylrest, Tritylrest, p-Toluolsulfonylrest und ähnliche sowie niedere Alkanoylreste wie Acetylrest, Formylrest, tert.-Butyloxicarbonylrest und ähnliche. Auch die Carbalkoxygruppe (zum Beispiel</p>
<p>niedrigmolekulare) kommt in Frage.</p>

<p> ' Die sich als Gegebenenfalls-Reaktion anschließende Reduktion der Ketogruppe von Verbindungen, worin X die Gruppe ^CO bedeutet zu Verbindungen, worin X die Gruppe <i>y </i>CHOH ist, sowie die Reduktion einer Doppelbindung des Restes R. wird im allgemeinen durch katalytische Hydrierung durchgeführt. Als Katalysatoren kommen beispielsweise die üblichen feinverteilten Metallkatalysatoren wie Edelmetallkatalysatoren zum Beispiel Raney-Nickel, Platin oder insbesondere Palladium in Frage. Das Verfahren kann bei normalen Temperaturen oder erhöhten Temperaturen durchgeführt werden. Zweckmäßig arbeitet man in einem Temperaturbereich von etwa 40 bis 200° C, gegebenenfalls unter erhöhtem Druck (1 - 100, insbesondere 1-50 bar). Enthält die</p>
<p><i>^5 </i>phenolische Hydroxylgruppe die Benzylschutzgruppe, so wird diese bei der katalytischen Hydrierung gleichzeitig abgespalten, wenn zum Beispiel ein Palladiumkatalysator verwendet wird.</p>
<p>Die Reduktion der Ketogruppe ist jedoch auf andere Weise ebenfalls möglich, beispielsweise mittels komplexen Metallhydriden (zum Beispiel Lithiumaluminiumhydrid, Natriumborhydrid, Cyanoborhydrid, Lithiumtri-tert.-butoxy-aluminiumhydrid) oder mittels</p>

<p>Das verwendete Ausgangsamin der Formel IV kann der diastereomeren Reihe des Ephedrins (Erythro-Reihe) oder des Pseudoephedrins angehören. Es können sowohl die reinen Enantiomeren als auch die entsprechenden Racemate eingesetzt werden. Dementsprechend werden Bis-Oxazolidine der Formel II erhalten, die entweder der Erythro-Reihe oder der Threo-Reihe angehören. Die Bis-Oxazolidine der beiden Reihen unterscheiden sich in der Stellung von CH^-Gruppe und Phenylring. Die Lage der beiden Substituenten geht aus den beiden untenstehenden Stereoformeln hervor:</p>

<p>Je nachdem, ob man von entsprechenden reinen Enantiomeren oder Racematen der Ausgangsstoffe IV ausgeht, erhält man die Bis-Oxazolidine der Formel II ebenfalls in Form der reinen Enantiomeren oder der Racemate. Bei der Spaltung der Bis-Oxazolidine unter sauren Bedingungen werden die Asymmetriezentren nicht berührt, so daß diese unter Erhaltung der jeweils vorliegenden Konfiguration erfolgt</p>
<p> Diejenigen Verfahrensprodukte der Formel I sowie auch die Ausgangs-Bis-oxazolidine der Formel II, die als Racemate anfallen, können in an sich bekannter Weise,</p>
<p>zum Beispiel mittels einer optisch aktiven Säure unter schonenden Bedingungen bei tiefen Temperaturen in die optisch aktiven Isomeren gespalten werden. Es ist natürlich auch möglich, von vornherein optisch aktive beziehungsweise auch diastereomere Ausgangsstoffe einzusetzen, wobei dann als Endprodukt I eine entsprechende reine optisch aktive Form beziehungsweise diastereomere Konfiguration erhalten wird. Beispielsweise handelt es sich um Verbindungen der Norephedrin-Konfiguration (Erythro-Reihe) und der Pseudonorephedrin-Konfiguration (Threo-Reihe). Es können auch diastereomere Racemate auftreten, da in den hergestellten Verbindungen zwei oder mehr asymmetrische Kohlenstoff-. atome vorhanden sind. Trennung ist auf üblichem Weg,</p>
<p>15 zum Beispiel durch Umkristallisieren möglich.</p>
<p>Je nach den Verfahrensbedingungen und Ausgangsstoffen erhält man die Endstoffe der Formel I in freier Form oder in Form ihrer Salze. Die Salze der Endstoffe können in an sich bekannter Weise, beispielsweise mit Alkali oder Ionenaustauschern, wieder in die Basen übergeführt werden. Von den letzteren lassen sich durch Umsetzung mit organischen oder anorganischen — Säurerr,-""insbesondere solchen, die zur Bildung von therapeutisch verwendbaren Salzen geeignet sind, Salze gewinnen. Als solche Säuren seien beispielsweise genannt: Halogenwasserstoffsäuren, Schwefelsäure, Phosphorsäuren, Salpetersäure, Perchlorsäure, organische Mono-, Di- oder Tricarbonsäuren der aliphatischen, alicyclischen, aromatischen oder heterocyclischen Reihe sowie Sulfonsäuren. Die Reaktionsmischung wird über Nacht bei Raumtemperatur belassen- Das auskristallisierte Produkt wird abgenutscht und mit 5 ml Isopropanol· und 20 ml Aceton gewaschen. Es vlrd 8 Stunden unter Rückfluß erhitzt, mit 5° nil Aoeton versetzt und das Lösungsmittel im Vakuum entfernt. Nach Zugabo von 100 ml Wasser wird mehrfach mit Chloro form extrahiert. Der nach Trocknung mit Natriumsxilfat und anschließender Entfernung des Llisungamittela Im Vakuum erhältliche Rückstand vlrd aus laopropanol kriatalliaiert.</p>

<p>3 <i>S </i>J^-/3-Hydroxy-3-phenyl-propyl-(-2<sub><</sub>}<sub>i</sub>7'-(3-adamantyl-3-oxopropyl)-amin-hydrochlorid werden in verdünntem Ammoniak suspendiert. Die Suspension vird mehrmals mit Chloroform ausgeschüttelt. Nach Trocknung und Entfernung des Lösungsmittels im Vakuum vird der Rückstand aus Isopropanol umkristallisiert. Ausbeute: 82 <i>$></p>

Claims (2)
- Erf indungsanspruch1. Verfahren zur Herstellung von Verbindungen der allgemeinen formelR1 -X-CH-CH0-KH-CH(CHO-CH(Oh) -// -^T Iι ι C- Jworin X die Gruppe > CO oder > CH(OH), Rp Wasserstoff oder eine C.-Cg-Alkylgruppe und R-, Wasserstoff oder eine Hydroxygruppe ist und R. den Adamantylrest oder einen gesättigten oder einfach ungesättigten C^-C.g-Cycloalkylrest bedeutet, wobei dieser Cn-C1g-Cycloalkylrest auch durch eine Cj-C^-Alkylgruppe oder ein Halogenatom substituiert sein kann und deren Säureadditionssalzen, gekennzeichnet dadurch, daß man ein NjlF-Methylen-bis-oxazolidin der Formel IIα ι nno/ Q59 890 11 - 2Λ- .worin R- die angegebenen Bedeutungen hat mit einem Keton der formel IIIR1-GO- GH2 (III)worin R1 und Rp die oben angegebenen Bedeutungen haben in Gegenwart einer Säure oder saurer Ionenaustauscher umsetzt und gegebenenfalls in den erhaltenen Verbindungen der Formel I, worin R1 eine Doppelbindung enthält "und/oder X die GO-Gruppe bedeutet, gegebenenfalls diese Doppelbindung und/oder diese •''GO-Gruppe reduziert», wobei die Ausgangsverbindungen Schutzgruppen enthalten1/und diese Schutzgruppen wähimd oder nach der Verfahrensreaktion abgespalten werden, und man weiterhin, gegebenenfalls die erhaltenen Verfahrensprodukte in ihre Salze überführt.
- 2. Verfahren gemäß Punkt 1, ,gekennzeichnet dadurch, daß Verbindungen der Formel I hergestellt werden, worin Rp und R_ Wasserstoff bedeuten und R1 ein gesättigter oder einfach ungesättigter Gg-Gg-Cycloalkylrest ist.
