Verfahren zur Herstellung von Purinderivaten
Die Anwendung der vorliegenden Erfindung erfolgt auf dem Gebiet der Arzneimittel, insbesondere der antiviralen Mittel.
Virale Infektionen sind weit verbreitet und resultieren in einer Vielzahl von Symptomen. Einige Virusinfektionen können leicht durch das körpereigene Abwehrsystem bekämpft werden, wenn dieses Abwehrsystem jedoch beeinträchtigt ist, kann dies bleibende Schäden, z.B. Blindheit, oder sogar , den Tod zur Folge haben. Eine Virusart, die derart ernsthafte Infektionen verursachen kann, ist die Gruppe der Herpes-Viren. .

Die derzeit zur Behandlung viraler Infektionen verwendeten Arzneistoffe sind in vielen Fällen unwirksam und selbst bei bestehender Wirksamkeit sind große und/oder kontinuierliche Gaben erforderlich, die ernsthafte Nebenwirkungen und/oder toxische Effekte ergeben. Es besteht daher Bedarf für wirksame antivirale Mittel, die in niedrigeren Dosen als herkömmliche Arzneistoffe wirksam und dementsprechend hinsichtlich möglicher Nebenwirkungen und der Toxizität sicherer sind.
In der US-PS 4 199 574 sind Verbindungen der folgenden allgemeinen Formel beschrieben:

Ziel der Erfindung ist es, ein Verfahren zur Herstellung neuer Purinderivate bereitzustellen.

Aufgabe der Erfindung ist es, ein Verfahren zu-r Herstellung neuer substituierter 9-(1- oder 3-Monoacyloxy- oder 1 ,S-Diacyloxy-^-propoxymethyl)-purinderivate und deren SaI zen bereitzustellen, die besonders wirksame antivirale Mittel darstellen.

In einer Ausführungsform betrifft die Erfindung ein Verfahren zur Herstellung dieser Verbindungen der Formel (I), bei dem man eine Verbindung der nachstehenden Formel (X) oder (XIV) verestert.
In einer anderen Ausführungsform betrifft die Erfindung ein Verfahren zur Herstellung von neuen Verbindungen der nachstehenden Formeln (IXa) und (XIV), wobei A Wasserstoff ist, die sich als Zwischenprodukte und antivirale Mittel eignen.
Sofern nichts anderes angegeben ist, haben die verwendeten Ausdrücke folgende Bedeutung:
"Alkyl" bedeutet einen geradkettigen oder verzweigten einwertigen Substituenten, der nur aus Kohlenstoff und Wasserstoff besteht, nicht ungesättigt ist und 1 bis 19 Kohlenstoff atome enthält. Beispiele für Alkyl sind Methyl, Ethyl, Propyl, n-Butyl, 2-Methyl-2-propyl, n-Octyl, n-Decyl, n-Tetradecyl und n-Nonadecyl. "Nieder-alkyl" bezieht sich auf die genannten Alkylgruppen mit 1 bis 6 Kohlenstoffatomen "Alkenyl" bezieht sich auf geradkettige oder verzweigte einwertige Substituenten, die nur aus Kohlenstoff und Wasserstoff bestehen, ein oder mehr Doppelbindungen aufweisen und 2 bis 19 Kohlenstoffatome enthalten. Beispiele für"Alkenyl" sind Propenyl, Pentenyl, Heptenyl, Dodecenyl, Dodecadienyl, Pentadecenyl und Pentadecadienyl. "1-Adamantyl" bezieht sich auf die folgende Ringstruktur:

"Nieder-alkoxy" bedeutet "Nieder-alkyl-O-", wobei "Niederalkyl" wie oben definiert ist. Beispiele für "Nieder-alkoxy" sind Methoxy, Ethoxy, i-Butoxy und n-Hexyloxy. "Alkoxyalkyl" bedeutet Alkyl-O-alkylen, wobei Alkyl wie oben definiert ist und Alkylen eine zweiwertige Alkylgruppe darstellt. Beispiele für Alkoxyalkyl sind Methoxymethyl, i-Propoxymethyl, n-Octanyloxymethyl und Ethoxypropyl. "Nieder-alkylthio" bedeutet "Nieder-alkyl-S-", wobei "Nieder-alkyl" wie oben definiert ist. Beispiele für "Nieder-alkylthio" sind Methylthio, n-Propylthio und n-Pentylthio. "Thio" bedeutet -SH. "Amino" bedeutet -NH2-•"Azido" bedeutet N . "Halogen" bedeutet z.B. Fluor, Chlor und Brom. "2-Carboxyethyl" bedeutet
"Pharmazeutisch verträgliche Additionssalze" sind Salze, die die biologische Wirksamkeit und sonstigen Eigenschaften der freien Verbindung besitzen und biologisch oder in anderer Weise nicht unerwünscht sind. Geeignete Säuren für die Salzbildung sind anorganische Säuren, wie Chlorwasserstoff-, Bromwasserstoff-, Schwefel- und Phosphorsäure, sowie organische Säuren, wie Trifluoressigsäure, Methansulfonsäure, Ethansulfonsäure und p-Toluolsulfonsäure. "Pharmazeutisch verträgliche Alkalimetallsalze" sind die Metallsalze der freien Carboxyl- oder Carboxyethylgruppe.
Beispiele für Alkalimetalle sind Natrium und Kalium.

Sofern nichts anderes angegeben ist, liegen die beschriebenen Monoester der Formel (I) in razemischer Form vor. Die Erfindung ist jedoch nicht auf diese razemische Form beschränkt, sondern umfaßt die einzelnen optischen Isomeren der Monoester (I) .
Die Definition, daß R zusammen mit R eine Ketogruppe bildet, schließt auch die tautomere Hydroxyform ein, jedoch wird der Einfachheit halber die Keto-Form zur Darstellung beider tautomeren Formen verwendet.

Die Verbindungen der Formel (I) und ihre pharmazeutisch verträglichen Salze zeigen starke antivirale Aktivität bei der Verabreichung an Warmblüter und Kaltblüter, insbeson-

dere Säugetiere, Vögel und Fische, besonders aber beim Menschen. Beispielsweise zeigen die erfindungsgemäßen Verbindungen ausgezeichnete Wirkung gegen Herpes Simplex-Virus I und II und verwandte Viren, wie Cytomegalo-Virus, Epstein-Barr-Virus und Varicella-Zoster-Virus sowie Virus-
heptatitis, beispielsweise Hepatitis B. Verbindungen der Formel (I), bei denen R1 und R2 -C(O)R7 (R7 ist Ethyl) sind, d.h. die Dipropanoatester, zeigen Eigenschaften, die sie besonders zur Behandlung von Virusinfektionen geeignet machen. Diese Ester sind in wäßrigen Lösungen löslicher, als anhand der Löslichkeit verwandter Homologer zu erwarten gewesen wäre, so daß sie insbesondere zur parenteralen Applikation geeignet sind. Das Dipropanoat ist auch eher bioverfügbar.
Antiviral wirksame pharmazeutische Zusammensetzungen, sowohl für die Tiermedizin als auch die Humanmedizin, die die erfindungsgemäßen Verbindungen enthalten, werden nach bekannten Methoden hergestellt und enthalten bekannte Excipienten. Die erfindungsgemäßen Verbindungen können parenteral (z.B. intravenös, subkutan, intraperitoneal oder intramuskulär), oral, topisch, rektal oder intranasal angewandt werden.

Die Zusammensetzungen werden oral oder parenteral in Dosen (berechnet als freie Base) von etwa 0,1 bis 300 mg/kg, vorzugsweise 1,0 bis 30 mg/kg Körpergewicht des Säugetiers verabreicht und werden beim Menschen als Dosierungseinheit von 1 bis 500 mg ein bis fünf mal täglich angewandt. Für die orale Applikation können Feinpulver oder Granulate Verdünnungs-, Dispergier- und/oder oberflächenaktive Mittel enthalten und können als Arzneitränke in Wasser oder einem Sirup, in Kapseln oder Sachets in trockenem Zustand oder einer nicht-wäßrigen Lösung oder Suspension, die geeignete Suspendiermittel enthalten kann, in Tabletten, die Bindemittel und Gleitmittel enthalten können, als Suspensionen in Wasser oder einem Sirup oder als Aerosole vorliegen. Wo erwünscht oder notwendig, können Geschmacks-, Konservierungs-, Suspendier-, Verdickungs- oder Emulgiermittel eingesetzt werden. Tabletten und Granulate sind bevorzugt und können beschichtet sein. Die Menge der Verbindung (I) in der Formulierung kann 0,1 bis 99 Gewichtsprozent oder mehr, bezogen auf die gesamte Formulierung, betragen, die des Excipienten etwa 1 bis 99,9 Gewichtsprozent. Vorzugsweise ist die Verbindung (I) in einer Menge von 10 bis 95 Gewichtsprozent vorhanden.
Für die parenterale Applikation oder in Form von Tropfen, z.B. gegen Augeninfektionen, können die Verbindungen in wäßrigen Lösungen in einer Konzentration von etwa 0,1 bis 10, vorzugsweise etwa 0,1 bis 7 %, angewandt werden. Die Lösung kann Antioxidantien, Puffer und andere geeignete Additive enthalten.
Alternativ können für Infektionen des Auges oder anderer äußerer Gewebe, z.B. des Mundes oder der Haut, die Mittel auf den infizierten Körperteil mit Vorteil topisch aufgetragen werden, z.B. als Salben, Cremes, Aerosole oder

Pulver, vorzugsweise als Salben oder Cremes. Die Verbindungen können in einer Salbe, z.B. einer wasserlöslichen Salbengrundlage, oder einer Creme, z.B. einer Öl-in-Wasser-Cremegrundlage, in einer Konzentration von etwa 0,01 bis 10, vorzugsweise 0,1 bis 7 und insbesondere etwa 3,0 % G/V enthalten sein. Außerdem können Virusinfektionen unter Anwendung eines Systems mit verzögerter Wirkstoffreigäbe, wie es in der US-PS 4 217 898 beschrieben ist, behandelt werden.
Für die Verwendung als Aerosol wird der Wirkstoff vorzugsweise in feinverteilter Form zusammen mit einem Tensid und einem Treibmittel angewandt. Typische Wirkstoffanteile sind 0,01 bis 20, vorzugsweise 0,04 bis 1,0 Gewichtsprozent. Die Tenside müssen selbstverständlich nicht-toxisch sein und sollten vorzugsweise in dem Treibmittel löslich sein. Spezielle Beispiele für verwendbare Tenside sind Ester oder Partialester von Fettsäuren mit 6 bis 22 Kohlenstoffatomen, wie Capron-, Octan-, Laurin-, Palmitin-, Stearin-, Linol-, Linolen-, Oleostearin- und Ölsäure, mit aliphatischen mehrwertigen Alkoholen oder deren cyclischen Anhydriden, wie Ethylenglykol, Glycerin, Erythrit, Arabit, Mannit, Sorbit, von Sorbit abgeleiteten Hexitanhydriden (Sorbitanester) sowie die Polyoxyethylen- und Polyoxypropylenderivate dieser Ester. Gemischte Ester, z.B. gemischte oder natürliche Glyceride, können ebenfalls angewandt werden. Bevorzugte Tenside sind die Oleate und Sorbitan, z.B. die Handelsmarken "ARLACEL C" (Sorbitansesguioleat), "Span 80" (Sorbitanmonooleat) und "Span 85" (Sorbitantrioleat).
Das Tensid kann 0,1 bis 20, vorzugsweise 0,25 bis 5 Gewichtsprozent, der Zusammensetzung ausmachen. Der Rest besteht gewöhnlich aus dem Treibmittel. Verflüssigte Treib-

mittel sind gewöhnlich Gase bei Normalbedingungen und werden unter Druck kondensiert. Beispiele für geeignete verflüssigte Treibmittel sind niedere Alkane mit bis zu 5 Kohlenstoffatomen, wie Butan und Propan, und vorzugsweise fluorierte oder fluorchlorierte Alkane, wie sie z.B. unter dem Warenzeichen "Freon" vertrieben werden. Auch entsprechende Gemische können angewandt werden.
Zur Herstellung des Aerosols wird ein mit einem geeigneten Ventil versehener Behälter mit dem entsprechenden Treibmittel gefüllt, das den feinverteilten Wirkstoff und das Tensid enthält. Die Bestandteile werden bei erhöhtem Druck gehalten, bis sie unter Wirkung des Ventils freigesetzt werden.
Die erfindungsgemäßen Verbindungen und Zusammensetzungen eignen sich nicht nur zur Behandlung des Menschen, sondern auch anderer Säugetiere, Vögel, z.B. Hühner und Truthähne, sowie Kaltblüter, z.B. Fische. Viruserkrankungen bei Vögeln, z.B. die Marek-Krankheit, werden mit den erfindungsgemäßen Verbindungen nach be-.kannten Methoden verhindert und/oder bekämpft, indem man den Vögeln z.B. eine die Verbindung enthaltende Zusammen-Setzung injiziert oder die Verbindung dem Futter oder Trinkwasser zusetzt.
Fische in eingegrenzten Bereichen, z.B. Becken, Aquarien, oder Tanks, können ebenfalls gegen Virusinfektionen behandelt werden, z.B. Herpes-ähnliche Viren, wie den Kanal-Katzenfisch-Virus (CCV), Kerpes-Virus-Salomones und Nerka-Virus, indem man die Verbindung direkt dem Wasser des Beckens, Aquariums oder Tanks zusetzt öder die Verbindungen dem Futter zugibt.
Die genauen Einzelheiten der Applikation der erfindungsgemäßen Verbindungen und Zusammensetzungen richten sich natürlich nach den Anforderungen des zu behandelnden Individuums, der Behandlungsart und dem Urteil des Behandelnden.

Im Reaktionsschema Ia wird die Verbindung der Formel III dadurch hergestellt, daß man Epichlorhydrin (II) zu einer Lösung eines Alkalimetallsalzes, vorzugsweise Natriumsalzes, von gegebenenfalls substituiertem Benzylalkohol in einem Lösungsmittel, wie Dimethylformamid, Dimethylacetamid, Hexamethylphosphoramid, Dimethylsulfoxid, SuIfolan, Tetrahydrofuran oder Dioxan, bei einer Temperatur von etwa 0 bis 1000C, vorzugsweise etwa 15 bis 400C, tropft. Das Reaktionsgemisch wird etwa 10 bis 24 Stunden, vorzugsweise etwa 12 bis 18 Stunden, bei einer Temperatur von etwa 0 bis 1000C, vorzugsweise etwa 20 bis 50°C, gerührt.
Die Verbindung der Formel (III) wird zu der Verbindung (IV) chlormethyliert, indem mam trockenes Chlorwasserstoffgas in eine Lösung der Verbindung und von Paraformaldehyd in einem Halogenkohlenwasserstoff, wie Dichlorethan, Chloroform, Dichlormethan oder 1,1,2-Trichlorethan, die auf eine Temperatur von etwa 0 bis 250C, vorzugsweise etwa 00C, gekühlt ist, einleitet. Das Chlorwasserstoffgas wird innerhalb 30 Minuten bis 3 Stunden, vorzugsweise 1 bis 2 Stunden, zugeführt, bis sich der Paraformaldehyd löst.
Die Lösung wird etwa 12 bis 48 Stunden bei etwa 0 bis 100C, vorzugsweise etwa 16 bis 24 Stunden bei etwa 0 bis 5°C, gehalten.
Die Verbindung der Formel (V) wird hergestellt, indem man ein Alkalimetallacetat, wie Natriumacetat, mit der Verbindung der Formel (IV), gelöst in einem Lösungsmittel, wie Dimethylformamid, Tetrahydrofuran, Dimethylacetamid, Hexamethylphosphoramid, Dimethylsulfoxid, SuIfolan oder Dioxan, bei einer Temperatur von etwa 0 bis 450C, vorzugsweise etwa 0 bis 25°C, umsetzt. Die Lösung wird etwa 5 bis 24 Stunden, vorzugsweise etwa 10 bis 18 Stunden, bei einer

Temperatur von etwa 10 bis 300C, vorzugsweise etwa 15 bis 25°C, gerührt.
In Reaktionsschema Ib wird die Verbindung der Formel (VII) dadurch hergestellt, daß man Guanin (VI) etwa 10 bis 24 Stunden, vorzugsweise etwa 12 bis 18 Stunden, mit Acetanhydrid unter Rückfluß erhitzt.
N ,9-Diacetylguanin der Formel (VII) wird mit der Verbindung der Formel (V) unter Bildung der Verbindung der Formel (VIII) umgesetzt und zwar als solches oder in einem Lösungsmittel, wie Dioxan oder SuIfolan, in Gegenwart einer katalytischen Menge einer Säure, wie Bis-(p-nitrophenyl)-phosphat, Toluolsulfonsäure, Methylphosphonsäure oder Dichloressigsäure, vorzugsweise Bis-(p-nitrophenyl)-phosphat, bei einer Temperatur von etwa 75 bis 2000C, vorzugsweise etwa 110 bis 1800C. Die Umsetzung erfolgt im allgemeinen mit 0,8 bis 1,2 Mol der Verbindung (V) pro 1 Mol der Verbindung (VII).
Die Benzyl-Schutzgruppen der Verbindung (VIII) werden durch katalytische Hydrierung abgespalten, wobei man die Verbindung (IX) erhält. Hierzu gibt man einen Katalysator, wie Palladium-auf-Kohlenstoff, als Aufschlämmung zu einer Lösung der Verbindung (VIII), die in einem Lösungsmittel, wie wäßrigem Methanol, gelöst ist. Wasserstoff wird auf die Lösung mit einem Druck von 1 bis 13,8 bar, vorzugsweise 2 bis 5,5 bar, aufgepreßt.
Die Verbindung (X) wird dadurch hergestellt, daß man eine Verbindung (IX) mit einer Base, wie Ammoniak, gelöst in einem Alkohol, wie Methanol, desacetyliert. Eine Lösung der Verbindung (IX) und der Base wird etwa 5 bis 36 Stunden, vorzugsweise etwa 10 bis 24 Stunden, bei einer Tem-

peratur von etwa 10 bis 300C, vorzugsweise etwa 15 bis 250C, gerührt.
Die Verbindung (X) kann zu dem Diacetat (XI) verestert werden, indem man die Verbindung (X) mit überschüssigem Acetanhydrid entweder als solches oder in einem Lösungsmittel, wie Dimethylformamid oder Dimethy!acetamid, 2 bis 3 Tage bei Raumtemperatur umsetzt.
Die Verbindung (XII) kann hergestellt werden, indem man die Verbindung (XI) mit Phosphoroxychlorid nach dem Verfahren von J. Org. Chem., Bd. 28, S. 945 (1963) umsetzt. Die Verbindung (XI) wird bei Raumtemperatur zu einer Lösung von Phosphoroxychlorid in Ν,Ν-Diethylanilin gegeben. Die Suspension wird 2 bis 30 Minuten, vorzugsweise bis 5 Minuten, unter Rückfluß erhitzt, worauf man überschüssiges Phosphoroxychlorid entfernt und das Produkt auf übliche Weise gewinnt, z.B. durch Extraktion mit einem organischen Lösungsmittel, gefolgt durch Chromatographieren.
Die Verbindung (XII) kann weiter chloriert werden, indem man das Diazosalz mit Natriumnitrit in Chlorwasserstoffsäure bildet; siehe J. Org. Chem., Bd. 31, S. 3258 (1966). Die Verbindung (XII) wird in wäßriger Salzsäure bei 0
bis 50C mit Natriumnitrit in Wasser versetzt. Die Lösung wird mit Wasser verdünnt und überschüssige Salzsäure wird mit wäßrigem Ammoniak neutralisiert. Die Verbindung (X III) kann nach üblichen Methoden gewonnen werden, 'z.B. durch Extrahieren mit einem organischen Lösungsmittel, gefolgt von Chromatographieren.
Die Verbindungen (XIV) können aus den Verbindungen (XII) und (XIII) nach bekannten Methoden hergestellt werden;

Die Verbindung (XIVb), in der A Wasserstoff ist, kann dadurch hergestellt werden, daß man eine Verbindung (XII)
1 bis 2 Stunden mit Thioharnstoff, gelöst in einem Lösungsmittel, wie Isopropanol,. unter Rückfluß umsetzt.
Das entstehende Thioharnstoffaddukt wird mit einer Alkalibase, z.B. wäßrigem Ammoniak, zersetzt, wobei die Thioverbindung entsteht. Die Reaktion führt auch zu einer Hydrolyse der Estergruppen.
Verbindungen (XIVa), bei denen A Wasserstoff und R Nieder-alkyl bedeutet, werden aus Verbindungen (XII) durch Behandeln mit einer Alkohollösung eines geeigneten Alkalimetallalkoxids, wie Natriummethoxid in Methanol, bei Raumtemperatur oder unter leichtem Erwärmen hergestellt.
Verschiedene Verbindungen der Formel (XIV) können aus der Verbindung (XIII) hergestellt werden. Beispielsweise wird die Verbindung (XIVc), bei der A Acetyl bedeutet, durch Behandeln der Verbindung (XIII) mit Natriumazid in einem Lösungsmittel, wie Ethanol/Wasser, Dimethylformamid oder Hexamethylphosphoramid, erhalten. Die Reaktanten werden
2 bis 24 Stunden auf 80 bis 2000C erhitzt. Die Verbindung (XIVd), bei der A Wasserstoff ist, wird durch Hydrieren der Verbindung (XIVc) z.B. mit Palladium-auf-Aktivkohle, erhalten.
Die Verbindung (XIII) wird in die Verbindung (XIVe),bei der A Wasserstoff ist, überführt, indem man sie mit

Ammoniak als Lösung in Methanol in einer Bombe 16 bis 24 Stunden auf 85 bis 110°C erhitzt. Die Verbindung kann noch weiter ammoniert werden, indem man die Verbindung (XIVe) gelöst in flüssigem Ammoniak in einer Bombe 24 bis 48 Stunden auf 110 bis 1500C erhitzt, wobei man die Verbindung (XIVd), in der A Wasserstoff ist, erhält. .
Ein weiteres Verfahren zur Herstellung der Verbindung (XIVd) besteht darin, die Verbindung (XIVe) mit Hydrazin und dann mit einer kalten Lösung von Natriumnitrit zu behandeln, wobei man die Verbindung (XIVf), in der A Wasserstoff ist, erhält, die man wiederum z.B. mit Palladiumauf-Aktivkohle zu der Verbindung (XIVd) hydriert.'
Die Aminogruppen der Verbindungen (XIV), bei denen R und/ oder R Amino sind, können durch Behandeln mit einem Alkylhalogenid, wie Methyljodid, zu den sekundären oder tertiären Aminen alkyliert werden. Ein bevorzugtes Verfahren zur Herstellung dieser alkylierten Aminoverbindungen besteht darin, eine Verbindung (XIII) oder (XIVe) mit einem Alkyl- oder Dialkylamin umzusetzen.
Verbindungen (IXa), bei denen R keine Methylgruppe ist, lassen sich dadurch herstellen, daß man die Hydroxylgruppen der Verbindung (IX)mit einem Trityl-Schutzmittel der nachstehend beschriebenen Art schützt, den Amino-Stickstoff desacetyliert, die .desacetylierte Verbindung mit einem geeigneten Säurechlorid umsetzt und die Schutzgruppen mit Eisessig/Wasser abspaltet.
Verbindungen (XIV), bei denen A Acetyl ist, können nach bekannten Methoden hydrolysiert werden, z.B. durch saure oder basische Hydrolyse.

Die Verbindungen (XIVg) und (XIVh) lassen sich dadurch herstellen, daß man Verbindungen (XII) bzw. (XIVe) mit einem Hydrierkatalysator, wie Palladium, in Gegenwart einer Base, wie Magnesiumoxid, hydriert.
Verbindungen (XIVi), bei denen A Wasserstoff ist, werden hergestellt durch Umsetzen einer Verbindung (XIVh) mit Natriumnitrit in Essigsäure unter Bildung der 6-Ketoverbindung, die wiederum mit einem Chlorierungsmittel, wie Phosphorylchlorid oder Phosphorpentachlorid, nach bekannten Methoden chloriert wird. Das 6-Chlor-Zwischenprodukt wird auf die oben beschriebene Weise mit Thioharnstoff umgesetzt.
Die Verbindungen (XIVj) können z.B. durch Umsetzen einer Verbindung (XIVe) mit methanolischem Schwefelwasserstoff/ Ammoniumcarbonat gemäß J. Org. Chem., Bd. 39 (9) S. 1256 (1974) oder mit Natriummercaptid in Dimethylformamid gemäß J. Med. Chem., Bd. 16 (1.2), S. 1381 (1973) hergestellt werden.
Verbindungen (XIV, bei denen R oder R Thio ist, können bei Raumtemperatur mit einem Alkylhalogenid, z.B. einem Alkyljodid, in einem Lösungsmittel, z.B. einem Alkanol, alkyliert werden.

sich Diester (I), bei denen R Nieder-alkyl ist, durch Umsetzen von Verbindungen (X) und (XIV), bei denen A Wasserstoff ist,, mit einem Überschuß eines geeigneten Säureanhydrids entweder als solches oder in einem Lösungsmittel, wie Dimethylformamid, Dimethylacetamid oder N-Methylpyrrolidin, herstellen. Die Reaktanten werden 1 bis 5 Tage, vorzugsweise 2 bis 3 Tage, bei 0 bis 500C, vorzugsweise Raumtemperatur, in Gegenwart eines Katalysators, z.B. 4-Dimethylaminopyridin, gerührt. Die Verbindungen (X) bzw. (XIV) und das Säureanhydrid liegen in einem Molverhältnis von etwa 1:10 bis 100 vor.
Die Säureanhydride sind leicht verfügbar oder falls dies nicht der Fall ist, lassen sie sich nach bekannten Methoden herstellen, z.B. durch Erhitzen der Säure in Gegenwart von Acetanhydrid oder Acetylchlorid.
Die Diester der Formel (I), bei denen R Wasserstoff ist, können nach dem oben beschriebenen Verfahren unter Verwendung des gemischten Anhydrids von Ameisensäure und Essigsäure hergestellt werden, das man durch Umsetzen von Ameisensäure mit Acetanhydrid bei 0 bis 400C erhält.
Ein weiteres Verfahren zur Herstellung der Diester der Formel (I) mit beliebigen Bedeutungen für R besteht darin zunächst eine Verbindung (XI) oder (XIV), bei der A Acetyl ist und R und/oder R Amino sind, mit einem Trityl-Schutzmittel, wie Triphenylmethylchlorid (Tritylchlorid)

oder 4-Methoxyphenyldiphenylmethylchlorid (Monomethoxytritylchlorid) umzusetzen. Die Trityl-Schutzmittel sind z.B. von der Aldrich Chemical Co. erhältlich. Die Reaktanten werden 8 bis 24 Stunden, vorzugsweise 12 bis 18 Stunden, in einem Lösungsmittel, wie Dimethylformamid oder Pyridin, mit einem Katalysator, z.B. 4-Dimethylpyridin, auf 40 bis 700C, vorzugsweise 45 bis 600C, erhitzt. Die Amino-geschützte Verbindung wird auf übliche Weise isoliert, z.B. durch Kristallisation, und die Acetatgruppen werden mit einer Base, z.B. einem Alkalimetallhydroxid, wie Natrium- oder Kaliumhydroxid, oder mit Ammoniumhydroxid hydrolysiert. Die Dihydroxyverbindung und ein Katalysator, wie 4-Dimethylaminopyridin, in Pyridin, werden zu dem entsprechenden Säurechlorid entweder als solchem oder in einem Lösungsmittel, wie Methylenchlorid oder Dichlorethan, getropft. Die Reaktanten werden 10 bis 24 Stunden, vorzugsweise 12 bis 18 Stunden, bei Raumtemperatur gerührt, worauf man das Produkt auf übliche Weise isoliert, z.B. durch Chromatographieren. Die Amino-geschützten Verbindungen werden durch Behandeln mit einer organischen Säure, wie Eisessig oder Trifluoressigsäure, unter Erwärmen auf 60 bis 1000C, vorzugsweise 60 bis 800C, 1 bis 12 Stunden, vorzugsweise 1 bis 5 Stunden, behandelt. Das Produkt wird durch Chromatographieren oder Kristallisieren isoliert.
Die Aminogruppe(n) der Verbindungen (XI) und (XIV) können auch dadurch geschützt werden, daß man die Verbindungen mit dem Acetal von Ν,Ν-Dimethylformamid umsetzt und dann auf die oben beschriebene Weise weiter verarbeitet.
Die Säurechloride'sind leicht verfügbar oder, falls dies nicht der Fall ist, lassen sich durch Umsetzen der entsprechenden Säure mit einem Chlorierungsmittel, wie Phosphor-

trichlorid, Phosphorpentachlorid oder Thionylchlorid, unter üblichen Reaktionsbedingungen herstallen.
Diester der Formel (I), bei denen R Amino ist, R zusammen mit R Keto bedeutet, R Wasserstoff ist und R Alkyl mit 7 bis 19 Kohlenstoffatomen, Alkenyl mit 7 bis 19 Kohlenstoffatomen, 2-Methyl-2-propyl, 1-Adamantyl oder Phenyl bedeutet, können auch durch Veresterung mit dem entsprechenden Säurechlorid hergestellt werden. Das Säurechlorid wird in einem Lösungsmittel, wie Methylenchlorid oder Dichlorethan, gelöst und mit einer Verbindung der Formel (IX), gelöst in einem Lösungsmittel, wie Pyridin oder Lutidinen, versetzt. Die Reaktanten werden 9 bis 24 Stunden, vorzugsweise 16 bis 24 Stunden, bei 15 bis 500C, vorzugsweise Raumtemperatur, gerührt. Die Aminogruppe wird durch Behandeln mit einer alkoholischen Lösung einer Base, z.B. einer Methanollösung von Ammoniumhydroxid, desacetyliert.
Ein anderes Verfahren zur Herstellung von Diestern der Formel (I), bei denen R Alkyl mit 7 bis 19 Kohlenstoffatomen ist, besteht darin, Verbindungen der Formeln (X) oder (XIV), bei denen A Wasserstoff ist, mit dem Kondensationsprodukt von Dicyclohexylcarbodiimid und der entsprechenden Carbonsäure umzusetzen. Die Carbonsäure in einem molaren Überschuß von Dicyclohexylcarbodiimid wird zu den genannten Verbindungen, gelöst in einem Lösungsmittel, wie Dimethylformamid oder Dimethylacetamid, gegeben. Die Lösung wird 16 bis 72 Stunden, vorzugsweise 24 bis 60 Stunden, unter Erhitzen auf 35 bis 75°C, vorzugsweise 40 bis 600C, gerührt. Die Diester der Formel (I) werden durch Ausfällen isoliert.

Diester und Monoester der Formel (I), bei denen R Hydroxyalkyl ist, werden dadurch hergestellt, daß man zunächst die Hydroxylgruppe der Hydroxysäure mit einer Benzylgruppe schützt, die geschützte Säure mit einer Trityl-geschützten Verbindung der Formel (X) oder (XIV), bei der A Wasserstoff ist, umsetzt und die Benzylgruppe durch katalytische Hydrierung abspaltet, worauf man die Tritylgruppe(n) abspaltet.
Die Monoester der Formel (I) können durch Umsetzen von Verbindungen der Formel (X) oder (XIV), bei denen A Wasserstoff ist, mit einer der oben genannten Trityl-Schutzgruppen hergestellt werden. Die Schutzgruppe reagiert mit einer Hydroxylgruppe und dem gegebenenfalls vorhandenen Amino-Stickstoff. Die nicht-geschützte Hydroxylgruppe wird mit einem Säurechlorid nach dem oben beschriebenen Verfahren verestert, worauf man die Schutzgruppen wie oben erläutert, abspaltet.
100 g (2,08 Mol) Natriumhydrid (5.0 % Dispersion in Mineralöl) werden zweimal mit 1 Liter Hexan gewaschen und dann unter Stickstoff getrocknet. Hierauf gibt man 1,5 Liter trockenes Dimethylformamid und dann innerhalb 2 Stunden 400 ml Benzylalkohol derart zu, daß die Temperatur unter 5O0C bleibt. Anschließend tropft man 92,5 g (1 Mol) Epichiorhydrin innerhaib 30 Minuten unter Eiskühlung zu, um die Temperatur unter 400C zu halten. Die Lösung wird dann 16 Stunden bei 210C und hierauf 2,5 Stunden bei 500C ge-

rührt. Der durch Abdampfen von Dimethylformamid unter vermindertem Druck erhaltene ölige Rückstand wird in 2,5 Liter Diethylether gelöst, worauf man die organische Lösung mit 2 Liter Wasser, 2 Liter 2 % Salzsäure, 2 Liter 1 % Natriumbicarbonat und 1 Liter Salzlösung wäscht, über Natriumsulfat trocknet und zu einem braunen Öl konzentriert, Durch Destillation erhält man 147,8 g 1,3-Di-O-benzylglycerin (Kp. 170 bis 180°C/1 Torr).
Herstellung von 1,S-Di-O-benzyl^-O-chlormethylglycerin Trockenes Chlorwasserstoffgas wird 1,5 Stunden in eine Lösung von 15 g (55 mMol) 1 ,3-Di-O-benzylglycerin aus Herstellungsbeispiel I und 3,3 g (110 mMol) Paraformaldehyd in 175 ml 1,2-Dichlorethan von 00C geleitet. Die Lösung wird dann in einem verschlossenen Kolben 24 Stunden bei 40C gelagert, hierauf unter Erwärmen auf 210C über Magnesiumsulfat getrocknet und dann filtriert und konzentriert, wobei 17,5 g 1,3-Di-0-benzyl-2-0-chlormethylglycerin erhalten werden.
Herstellungsbeispiel III
Herstellung von 2-0-Acetoxymethyl-1,3-di-O-benzylglycerin Eine Lösung von 17,5 g (55 mMol) 1,3-Di-O-benzyl-2-O-chlormethylglycerin aus Herstellungsbeispiel II in 400 ml Dimethylformamid von 00C, die sich unter einem Trockenrohr befindet, wird mit 6 g Natriumacetat versetzt, worauf man die Lösung auf 210C erwärmt und 15 Stunden magnetisch rührt. Der durch Abdampfen des Lösungsmittels unter vermindertem Druck erhaltene ölige Rückstand wird in 500 g Diethylether gelöst. Die Etherlösung wird einmal mit 750 ml

Wasser, zweimal mit 250 ml Wasser und einmal mit 250 ml Salzlösung gewaschen, über Natriumsulfat getrocknet und konzentriert, wobei 19 g 2-0-Acetoxymethyl-1,3-di-O-benzylglycerin als Öl erhalten werden.
Herstellungsbeispiel IV
20 g (0,132 Mol) Guanin werden mit 300 ml Acetanhydrid vereinigt und 16 Stunden unter Rückfluß erhitzt. Hierauf kühlt man das Gemisch, dampft überschüssiges Acetanhydrid unter vermindertem Druck ab und erhält durch Umkristalli-
sieren des Rückstands aus Dimethylsulfoxid 25,6 N ,9-Diacetylguanin.
A. Herstellung von N -Acetyl-0-(1,3-dibenzyloxy-2-propoxy-
methyl)-guanin 15,61 g (66 mMol) N ,9-Diacetylguanin, 19g (55 mMol) 2-0-Acetoxymethyl-1,3-di-O-benzylglycerin aus Herstellungsbeispiel III und 0,5 g Bis-(p-nitrophenyl)-phosphat werden zusammen mit 150 ml Diethylether gerührt. Der durch Abdampfen des Lösungsmittels erhaltene Rückstand wird 1,5 Stunden unter einem Stickstoffstrom auf 175°C erhitzt.
Durch Säulenchromatographie und Eluieren mit Methanol/ Methylenchlorid (1 : 9), gefolgt von Umkristallisieren aus
Ethylacetat, erhält man 4,7 6 g N -Acetyl-9-(1,3-dibenzyloxy-2-propoxymethyl)-guanin, F. 145 bis 146°C.·

Eine Lösung von 4,62 g (9,67 mMol) N -Ac&tyl-9-(1,3-dibenzyloxy-2-propoxymethyl)-guanin in 15o ml Methanol + 4 0 ml Wasser wird mit 20 % Palladiumhydroxid-auf-Aktivkohle, aufgeschlämmt in 10 ml Wasser, versetzt. Das Gemisch wird in einer Parr-Bombe 38 Stunden bei 4,14 bar Wasserstoff hydriert, dann durch Celite filtriert und zu einem weißen Feststoff konzentriert. Durch Umkristallisieren aus Methanol/Ethylacetat erhält man 1,4 g N2-Acetyl-9-(1,3-dihydroxy-2-propoxymethyl)-guanin, F. 205 bis 2080C.
Die Mutterlauge wird weiter mit 1 g 10 % Palladium-auf-Kohle in 150 ml Methanol + 50 ml Wasser 47 Stunden bei
2 - -— 4,14 bar hydriert. Die Gesamtausbeute an N -Acetyl-9-( dihydroxy-2-propoxymethyl)-guanin beträgt 2,11 g.
C. Herstellung von 9-1,3-Dihydroxy-2-propoxymethyl)-guanin
2 721,9 mg (2,4 mMol) N -Acetyl-9-(1,3-dihydroxy-2-propoxymethyl) ^-guanin werden 17 Stunden bei 210C mit 50 ml methanolischer Ammoniaklösung (Methanol bei 00C mit Ammoniak gesättigt) gerührt. Durch Konzentrieren der Lösung zu einem weißen Feststoff und Umkristallisieren des Rückstands aus Wasser oder Methanol erhält man 582,3 mg 9-(1,3-Dihydroxy-2-propoxymethyl)-guanin, F. 25O0C.

ε Ein Gemisch von 3,00 g 9-(1,3-Dihydroxy-2-propoxymethyl)-guanin, 300 mg 4-Dimethylaminopyridin und 100 ml Acetanhydrid wird 3 Tage kräftig bei Raumtemperatur gerührt. Durch Abdampfen des Acetanhydrids unter vermindertem Druck und Umkristallisieren des Rückstands aus Methanol erhält man 3,62 g. der gewünschten Verbindung, F. 237 bis 239°C.
Beispiel 2
9-(1,3-(2,2-Dimethylpropanoyloxy)-2-propoxymethyl)-guanin
12 (Verbindung, bei der R und R 2,2-Dimethylpropanoyl, R Amino, R Wasserstoff und R und R Keto bedeuten)
Ein Gemisch von 1,306 g 9-(1,3-Dihydroxy-2-propoxymethyl)-guanin, 150 mg 4-Dimethylaminopyridin und 50 ml 2,2-Dimethylpropansäureanhydrid, sowie 75 ml trockenes Dimethylformamid werden 2 Tage kräftig gerührt. Hierauf dampft man das Dimethylformamid unter vermindertem Druck ab und versetzt den Rückstand mit 200 ml Ethylether. Nach dem Kühlen auf 00C wird der Niederschlag abfiltriert und aus Methanol umkristallisiert. Man erhält 1,83 g der gewünschten Verbindung, F. 230 bis 232°C.

Beispiel 3
9-.(1 ,3-Di-n-hexadecanoyloxy-2-propoxymethyl) -guanin 50 mg 9-(1,3-Dihydroxy-2-propoxymethyl)-guanin in 5 ml Ν,Ν-Dimethylformamid werden mit 290 mg Dicyclohexylcarbodiimid und 300 mg n-Hexadecansäure versetzt und 50 Stunden bei 50°C gerührt. Die Verbindung wird dann in Eiswasser ausgefällt, worauf man das Wasser dreimal mit Methylenchlorid extrahiert, die vereinigten Methylenchloridextrakte konzentriert und dann auf präparativen Silikagelplatten unter Entwicklung mit Methanol/Methylenchlorid (1:9) reinigt. Es werden 140 mg der gewünschten Verbindung erhalten, F. 150 - 154°C.

A. N -(4-Methoxyphenyldipheny!methyl·)-9-(T,3-diacetoxy- 2-propoxymethyl·) -guanin
Eine Lösung von 3,3 g 9-(1,3-Diacetoxy-2-propoxymethyl)-guanin, 7,0 g 4-Methoxyphenyldiphenylmethylchlorid, 7,0 mi Triethylamin und 300 mg 4-Dimethyiaminopyridin in 50 ml trockenem Dimethylformamid wird 15 Stunden unter magneti-
schem Rühren auf 500C erwärmt. Hierauf gibt man 5 ml Methanol zu, und konzentriert die Lösung unter vermindertem Druc zu einem braunen Öl, das an Silikagel mit Methanol/Methylenchlorid (1:12) als Eluiermittel chromatographiert wird. Durch Kristallisation des Öls aus Ethylacetat/Hexan erhält man 5,42 g der gewünschten Verbindung, F. 139-141 C.
Bei Ersatz des 4-Methoxyphenyldiphenylmethylchlorids in Teil A durch Triphenylmethylchlorid erhält man die ent-

Zu einer magnetisch gerührten Lösung von 1,05 g N -(4-Met! oxyphenyldiphenylmethyl)-9-(1,3-dihydroxy-2-propoxymethyl guanin und 25 mg 4-Dimethylaminopyridin in 30 ml trockene: Pyridin wird eine Lösung von 1,2g N-Octanoylchlorid in 10 ml Methylenchlorid getropft. Nach 15 Stunden gibt man

A. N -(4-Methoxyphenyldiphenylmethyl)-9-[1-hydroxy-3-
(4-methoxyphenyldiphenylmethoxy)-2-propoxymethyl]-guanin 2 g 9-(1,3-Dihydroxy-2-propoxymethyl)-guanin in 50 ml Ν,Ν-Dimethylformamid werden mit 6,33 g 4-Methoxyphenyldiphenylmethylchlorid, 20 mg N,N-Dimethylaminopyridin und 10 ml Triethylamin versetzt. Das Gemisch wird 60 Stunden bei 50 bis 60 C gerührt und dann in 900 ml Eiswasser ausgefällt. Der Niederschlag wird abfiltriert und das Produkt aus Ethanol umkristallisiert. Durch weitere Reinigung des etwas verunreinigten Produkts durch Säulenchromatographie an Silikagel mit Methanol/Methylenchlorid (1:9) als Eluiermittel und Umkristallisieren aus Methanol erhält man 3,5 g der gewünschten Verbindung, F. 159 -
161 °C.

B. 9-(1-n-Hexadecanoyloxy-S-hydroxy^-propoxymethyl)-guanin
Ein gerührtes Gemisch von 3 g n-Hexadecanoylchlorid -in
2 50 ml Pyridin wird mit 1,5 g N -(4-Methoxyphenyldiphenylmethyl)-9-[hydroxy-3-(4-methoxyphenyldiphenylmethoxy-2-propoxymethyl]-guanin, versetzt. Das Gemisch
wird 48 Stunden bei 50 bis 60°C gerührt und dann in 500 ml Eiswasser ausgefällt. Der erhaltene Niederschlag wird mit 80prozentiger wäßriger Essigsäure 1 Stunde bei 80 C behandelt, worauf man die Essigsäure unter vermindertem Druck abdampft und den Rückstand an Silikagel mit Methanol/ Methylenchlorid (1:9) als Eluiermittel chromatographiert. Auf ähnliche Weise werden durch katalytische Abspaltung der Benzylgruppe und anschließende Weiterverarbeitung wie oben die folgenden Verbindungen hergestellt:
9- (1 -Hydroxyacetyloxy-S-hydroxy^-propoxymethyl] -guanin; 9-[1 -(3-Hydroxypropanoyloxy)-3-hydroxy-2-propoxymethyl]-guanin; und
9-[1-(4-Hydroxybutanoyloxy)-3-hydroxy-2-propoxymethyl]-guanin.
Ein zweifacher stöchiometrischer Überschuß von 3 % Chlorwasserstoff in Methanol wird zu einer Lösung von 1,0g 9-(1,3-di-n-Octanoyloxy-2-propoxymethyl)-guanin in 20 ml Methanol gegeben. Diethylether wird zugegeben, bis die Ausfällung vollständig ist. Das Produkt wird filtriert, mit Ether gewaschen, an der Luft getrocknet und zu 9-(1 ,3-Di-n-octanoyloxy-2-propoxymethyl)-guanin-hydrochlorid umkristallisiert.
Auf ähnliche Weise können alle Verbindungen der Formel (I), die als freie Basen vorliegen, in die Säureadditionssalze überführt werden, indem man sie mit der entsprechenden Sau-

re, z.B. Chlorwasserstoff-, Bromwasserstoff-, Schwefel-, Phosphor-, Essig-, Propion-, Glykol-, Bernstein-, Oxal-, Halon-, Brenztrauben-, Äpfel-, Malein-, Fumar-, Wein-, Citronen-, Benzoe-, Zimt-, Mandel-, Methansulfon-, Ethansulfon- oder p-Toluolsulfonsäure umsetzt.
1,0 g 9- (1 , B-Di-n-octanoyloxy^-propoxymethyl) -guanin . HCl, die in 50 ml Ether suspendiert sind, wird mit dem zweifachen stöchiometrischen Überschuß verdünnter wäßriger
Kaliumcarbonatlösung gerührt, bis sich das Salz vollständig löst. Hierauf trennt man die-organische Schicht ab, wäscht zweimal mit Wasser, trocknet über Magnesiumsulfat und erhält durch Eindampfen 9-(1,3-Di-n-octanoyloxy-2-
propoxymethyl)-guanin als freie Base.
Alkalimetallsalze von Mono- und Diestern der Formel (I), insbesondere solche, bei denen R 1-Adamantyl oder 2-Methyl-2-propyl bedeutet, können gemäß dem folgenden Beispiel hergestellt werden.
Beispiel 9
Das folgende Beispiel erläutert die Herstellung repräsentativer pharmazeutischer Formulierungen, die einen Wirkstoff der Formel (I) enthalten,- z.B. 9-[1,3-Di-(2,2-dimethylpropanoyloxy-2-propoxymethyl]-guanin.

A. Topische Formulierung

-Alle-Bestandteile mit Ausnahme von Wasser werden vereinigt und unter Rühren auf 60 C erhitzt. Eine ausreichende Menge Wasser von 600C wird dann unter kräftigem Rühren zugegeben um 100 g der Cremeformulierung zu erhalten, die dann auf Raumtemperatur abgekühlt wird.
Die folgende Formulierung eignet sich für die intraperitoneale und intramuskuläre Injektion.

Der Wirkstoff wird in Propylenglykol, Polyethylenglykol 400 und Tween 80 gelöst. Dann wird unter Rühren eine ausreichende Menge 0,9 % Kochsalzlösung zugegeben, um 100 ml der Injektionslösung zu erhalten, die man durch ein 0,2 μΐη-Membranf ilter filtriert und unter sterilen Bedin-
10 gungen abfüllt.
Die folgende Formulierung eignet sich für die intravenöse Injektion.

Der Wirkstoff wird zu einer Lösung von Polysorbat 80 und Propylenglykol oder Polyethylenglykol 400 in 20 ml Wasser gegeben und vermischt. Die erhaltene Lösung wird mit Wasser auf 100 ml verdünnt und durch ein 0,2 μτη-Membranfilter filtriert.

Die Bestandteile werden vereinigt und unter Verwendung von Methanol als Lösungsmittel granuliert. Die Formulierung wird dann getrocknet und mit einer Tablettenpresse zu Tabletten verarbeitet, die 200 mg Wirkstoff enthalten.
Die antivirale Aktivität der erfindungsgemäßen Verbindungen ist in den folgenden Versuchen erläutert:
Herpes-Simplex-Virus 2-Stamm G wird für die Infektion in ΗΞρ-2-Zellkultüren gezüchtet. Das Virus wird 1 Stunde adsorbiert, worauf man frisches Medium auf die Zellen aufbringt und diese bei 35°C inkubiert, bis alle Zellen infiziert sind. Die Zellsuspension wird bei -7 0 C eingefroren, dann aufgetaut und zentrifugiert, um Zellbruchstücke zu entfernen. Die überstehende Flüssigkeit wird in gleiche Teile aufgeteilt und bis zur Verwendung bei -70 C gefroren gelagert. Eine 10 ' -Verdünnung des überstands übergibt eine 50 % Zellkultur-Infektionsdosis (CCID _) in HEp-2-
-3 7 Zellen und eine 10 ' -Verdünnung ergibt eine 50 % lethale Reaktion (LC ») bei Mäusen.
Gruppen von 20 weiblichen Swiss Webster-Mäusen (15 bis 17 g) werden intraperitoneal mit 0,2 ml EMEM infiziert, das 10 LC--./Maus Virus enthält. Mäuse, die mit 10 ' mehr oder we-
-2 niger Virus als die 10 LC1-Q-DoSiS infiziert werden, dienen als Virulenzkontrolle, um die Funktionstüchtigkeit des Modells zu kontrollieren.
Die Behandlung mit den Testverbindungen beginnt 6 Stunden nach der Infektion. Den in Gruppen von je 20 unterteilten Mäusen werden die Verbindungen in Salzlösung subcutan mit

20 mg/kg injiziert. Eine Gruppe von 20 Mäusen wird als Kontrollgruppe verwendet und erhält Kochsalzlösung subkutan. Die Behandlung wird 24, 48, 72 und 96 Stunden nach der Infektion wiederholt.
Die erfindungsgemaßen Verbindungen zeigen antivirale Wirkung, wie aus der folgenden Tabelle ersichtlich ist.

Akute Toxizität bei der Maus Gruppen von 5 männlichen Swiss-Webster-Mäusen (Simonsen) mit einem Körpergewicht etwa 25 g werden die in der folgenden Tabelle genannten Verbindungen subcutan verabfolgt. Die Testverbindungen werden nur an Tag 1 gegeben und die Mäuse werden 21 Tage lang täglich bezüglich der Mortalität beobachtet.

- Erfindungsanspruch1. Verfahren zur Herstellung von Purinderivaten der allgemeinen Formel I(DCH0OCHCH OR1Δ ! Z"und deren Säureadditionssalzen, vorzugsweise den pharmazeutisch verträglichen Säureadditionssalzen,1 7 7in der R Wasserstoff oder -C(O)R bedeutet, wobei R Wasserstoff, Alkyl mit 1 bis 19 Kohlenstoffatomen, Hydroxyalkyl mit 1 bis 8 Kohlenstoffatomen, Alkoxyalkyl mit 2 bis 9 Kohlenstoffatomen, Alkenyl mit 2 bis Kohlenstoffatomen, Phenyl, 1-Adamantyl, 2-Carboxyethyl oder Carboxymethyl oder ein pharmazeutisch ver- trägliches Alkalimetallsalz davon ist;2 7 7R -C(O)R darstellt, wobei R wie oben definiert ist, R Wasserstoff, Halogen, Thio, Nieder-alkylthio mit 1η if! 9-(T -n-Hexadecanoyloxy-S-hydroxy^-propoxymethyl) guanin;9- [ 1 - (1 -Adamantylcarboxyl) -S-hydroxy-^-propoxymethyl] guanin;9- [ 1 - (3-Carboxypropanoyloxy) -S-hydroxy^-propoxymethyl]-guanin und die entsprechenden pharmazeutisch verträglichen Alkalimetallsalze. - 8. Verfahren nach einem der vorangehenden Punkte, gekennzeichnet dadurch, daß man eine Verbindung der FormelR6CH2OCHCH2OHCH2OH3
herstellt, in der R Wasserstoff, Halogen, Thio, Nieder-alkylthio mit 1 bis 6 Kohlenstoffatomen,9 10 9 10Azido oder -NR R , wobei R und R unabhängig voneinander Wasserstoff oder Nieder-alkyl mit 1 bis 6 Kohlenstoffatomen sind, bedeutet und R Wasserstoff, Halogen, Nieder-alkoxy mit 1 bis 6 Kohlenstoffatomen, Azido, Thio, Nieder-alkylthio mit 1 bis 6 Kohlenstoff-9 10 9 10 atomen oder -NR R , wobei R und R wie oben defi-247593niert sind, darstellt, mit der Maßgabe, daß wenn R Wasserstoff ist, R nicht -NH oder Chlor bedeutet. - 9. Verfahren nach einem der vorangehenden Punkte, gekennzeichnet dadurch, daß man eine Verbindung der FormelCH2OCHCH2OH CH2OH20 herstellt, in der R Wasserstoff, Alkyl mit 1 bis Kohlenstoffatomen oder 1-Adamantyl ist.
