Abstract
Die Erfindung betrifft ein Verfahren zur Herstellung von Cysteinderivaten, die fuer die Verwendung in der Humanmedizin und Veterinaermedizin fuer therapeutische Zwecke zu Praeparaten verarbeitet werden. Durch das erfindungsgemaesse Verfahren werden Cysteinderivate der allgemeinen Formel I, worin R CH(CH3)2 oder C(CH3)3 bedeutet, hergestellt. Formel I

worin R -CH(CH)I2 oder -C(CHj)] bedeutet, oder physiologisch verträgliche Salze derselben oder optische Isomere hiervon mit besserer biologischer Verfügbarkeit als N-Acetyl-L-cystein nach oraler Aufnahme. So erreichen nach oraler Aufnahme die neuen Verbindungen Konzentrationen in der systematischen Zirkulation, die Größenordnungen höher als die maximalen Konzentrationen an N-Acetyl-L-cystein sind. Da die Verbindungen der Formel I ähnliche oder identische Fähigkeiten wie N-Acetyl-L-cvsteln 1. zum Aufbrechen von Disulfidbrücken, 2. zur Wirkung als Antioxidantien und 3. zur Wirkung als Wurzelspülmittel haben, sollte eine orale Behandlung mit den neuen Substanzen viel wirksamer als die mit n-Acetyl-L-cystein gegen Lungenerkrankungen sein, vorausgesetzt, daß die Erkrankung durch irgendeine Art von oxidativer Beanspruchung verursacht oder aufrechterhalten wird.
Es muß auch daraufhlngowieson werden, daß sine andere Folge der biologischen Stabilität der Verbindungen der Formel I darin besteht, daß sehr wenig L-Cysteln, wenn überhaupt, freigesetzt wird. Dies bedeutet, daß dlose Verbindungen einen Anstieg nur bis zu sohr geringen Gehalten an Glutathfonvorläufern ergeben. Daher sind die nachfolgend beschriebenen Wirkungen in dem Sauorstofftoxlzitätssystem wahrscheinlich abhängig von den synthetischen Thiolon selber und nicht von der Glutathlonbiosynthese.
Die Erfindung liofert somit Verbindungen und physlolgoslch verträgliche Salzo und Isomere derselben, die In der thorapoutlschon Behandlung entzündlicher Lungonerkrankungen, wie chronischer Bronchitis, und anderer Erkrankungen, wie der folgernden, brauchbar sind:
1. Andere Lungenorkrankungon, die durch vlskoson Schleim kompliziort werden, wie zystischeί Voao, Asthma und Emphysem,
2. Bindegewebeerkrankungen, wie rheumatoide Arthritis,
3. Lungenschädigungsorkrankun^on, wie septischer Schock, ARDS und bronchlopulmonare Dysplasie,
4. durch Strahlung hervorgerufene Erkrankungen, wie durch Gammastrahlen induzierte Pneumonitis und Fibröse,
6. Erkrankungen im Lungenparenchym, wie Sarcoidose, Fibröse, Granulomatose, Collagenose, und
8. mit Diabetes verbundene Krankheiten, wie Abbau von ß-Zellen und Rolinopathie.

erhält man mit Umsetzungen von Cystein oder eines optischen Isomeren hiervon mit dem geeigneten Aldehyd, um das entsprechende Thiazolidin zu bekommen, welches nach den im Verfahren A beschriebenen Methoden acyliert wird.
Eine Suspension von 35,2g (0,20 Mol) L-Cysteinhydrochloridmonohydrat in 10OmI eines Gemisches von 80% Tetrahydrofuran (THF) und 20% Wasser wurde unter Stickstoff bei Raumtemperatur gerührt und mit 44,0g (0,40 Mol) Natriumisobulyrat behandelt. Das Reaktionsgemisch (weißer Schlamm) wurde auf 0 bis 5°C gekühlt, und unter Stickstoff wurdon 35ml (0,21 Mol) Isobutyrylanhydrid tropfenweise zugegeben. Die resultierende bewegliche Suspension wurde 6h bei Raumtemperatur gerührt, dann ließ man über Nacht stehen, und schließlich wurde 4 h unter Rückfluß erhitzt. Das Reaktionsgemisch wurde auf einem Eisbad gekühlt, und 16ml konzentrierter Salzsäure wurden zugegoben. Die organische Phase wurde eingedampft und ergab ein farbloses öl. Das öl wurde mit lexan gewaschen und danach mit Ether behandelt und ergab 12,9g der in der Überschrift angegebenen Verbindung als weißen Feststoff. F. = 103 bis 1040C (umkristallisiert aus Butylacetat). DIo organische Phase wurdo eingodampft und das Rohprodukt mehrere Male mit Hexan gewaschen und danach in11SO ml Chloroform gelöst. Nach dom Filtrieren und Vordampfen des Lösungsmittels wurde das Rohprodukt aus Ethylacetat

Aus den obigen Zusammensetzungen der Rezepturen A und B wurden 2000 Tabletten hergestellt, die 50mg bzw. 100mg Wirkstoff enthielten. Gegebenenfalls können die erhaltenen Tabletten mit beispielsweise Methylcellulose in einem organischen Lösungsmittel filmbeschichtet werden.

Ein Ziel der vorliegenden Erfindung Ist es, Moleküle zu liefern, die nach oraler Aufnahme hohe Gehalte an freien Thiolan Im Plasma und letztlich in der Lunge ergeben. Daher wurdon Experimente durchgeführt, um die biologische Beständigkeit und biologische Verfügbarkeit dor Vorbindungen nach Formol I zu untersuchen und cJioso mit N-Acetyl-L-cystein zu vergleichen.

Die Thiole wurden in die Därme anästhesierter Ratten injiziert. Zu verschiedenen Zeiten nach den Injektionen wurden Blutproben abgenommen und die Plasmathlole analysiert. Die Konzentrationen der Thiole im Plasma waren folgende: N-Isobutyryl-L-cystein 13,0 + 2,9 μΜ (η = 9), N-Pivaloyl-L-cystein 11,8 + 2,9 μ M (η - 9) und N-Acetyl-L-cystein 0,7 μΜ (nurein Experiment durchgeführt). Die angegebenen Werte stellen das Mittel ± Standardabweichung dar, wenn nichts anderes angegeben ist. Die Ergebnisse zeigen, daß der Plasmathiolgehalt, den man in vivo bekommt, in Beziehung zu der in vitro bestimmten biologischen Beständigkeit steht. So war eine leicht hydrolysierbare Verbindung, wie N-Acetyl-L-cystein, nach intratesetinaler Injektion kaum feststellbar, während N-lsobutyryl-L-cystein, das in vitro nicht hydrolysiert wurde, beachtliche Konzentrationen erreichte.
Die Plasmakonzentrationen und Harnausscheidung bei gesunden menschlichen Patienten wurden nach oraler Aufnahme von N-Acetyl-L-cystein und N-lsobutyryl-L-cystein, jeweils 1,23mMol gemessen. Die Spitzenplasmakonzentrationen der freien Thile waren < 1 μΜ bzw. 10μΜ. Die Prozentsäue der im Urin unverändert ausgeschiedenen Dosen waren 2 % und 74%. Zusammenfassung der Stoffwechselexperimente
Die oben beschriebenen Experimente zeigten, daß N-Acetyl-L-cystein biologisch instabil ist und daß dies zu einer schlechten biologischen Verfügbarkeit nach oraler Aufnahme führt. Eine Modifikation des Acylrestas verbessert die biologische Beständigkeit und auch die biologische Verfügbarkeit drastisch. So erreichte N-lsobutyryl-L-cystein im Plasma hohe Konzentrationen und wird voraussichtlich die Lunge in viel größerem Umfang als N-Acetyl-L-cystein erreichen. Wirkungsmessungen
Einwirkung von reinem Sauerstoff führt zu Lungenödem. Der Mechanismus ist der, daß der erhöhte Sauerstoffdruck zu unvollständiger Sauerstoffreduktion und folglich zur Bildung von aktiviertem Sauerstoff, wie Superoxid, Peroxiden und Hydroxylradikalen, führt. Einige dieser aktivierten Sauerstoffarten sind identisch mit jenen, die während Entzündungen auftreten.
Andere Forscher zeigten, daß N-Acetyl-L-cystein intravenös infundiert teilweise gegen sauorstoffinduziertes Ödem bei Ratten schützt. Diese Erkenntnis wurde bestätigt, und es wurde demonstriert, daß N-lsobutyryl-L-cystein einen Schutz ergibt, der etwas besser ist, wenn es auf dem gleichen Weg verabreicht wird. Wie oben aufgezeigt, führte die Konzentration von N-lsobutyryl-L-cystein im menschlichen Plasma nach oraler Verabreichung zu einer Spitze bei 10μΜ. Daher scheint es durchaus möglich, einen Schutz gegen oxidative Beanspruchung und vielleicht gegen Entzündung in der Lunge durch orale Aufnahme der vorliegenden neue Thiole zu erhalten
Schlußfolgerungen
Die metabolischen Studien demonstrierten, daß N-lsobutyryl-L-cystein und N-pivaloyl-L-cystein biologisch viel beständiger als das therapeutisch verwendete N-Acetyl-l.-cystein waren. Die Plasmagehalte der neuen Verbindungen nach oraler Aufnahme waren viel höher als die Gehalte von N-Acetyl-L-cystein. Das Sauerstofftoxizitätsmodell zeigten, daß die neuen Verbindungen gegen Lungenschädigung wenigstens so wirksam wie N-Acetyl-L-cystein nach intravenöser Verabreichung schützten. Die Resultate zeigen, daß die neuen Verbindungen viel wirksamer als N-Acetyl-L-cystein gegen oxidative Beanspruchung in der Lunge sind, wenn sie oral verabreicht werden.
Claims (3)
- Erfindungsanspruch:1. Verfahren zur Herstellung von Cysteinderivaten der allgemeinen Formel HSHN COOH COworin R-CH(CH3I2 oder -C(CHa)3 bedeutet, oder eines physiologisch verträglichen Salzes derselben oder eines optischen Isomers hiervon, gekennzeichnet dadurch, daß man a) eine Verbindung der FormelHSH2N<COOHoder ein optisches Isomeres hiervon mit einem Acylierungsmittel der allgemeinen Formel RCOX umsetzt, worin R die obige Bedeutung hat und -COX eine reaktive Gruppe ist, die mit einer Aminogruppe unter Bildung eines Amidrestes reagieren kann, oder b) eine Verbindung der allgemeinen FormelHN COOH_j2 COoder ein optisches Isomeres hiervon, worin R die obige Bedeutung hat, unter Verwendung eines Reduktionsmittels oder elektrochemisch reduziert, oder c) von einer Verbindung der allgemeinen FormelR1S.HN' COOH CO Roder einem optischen Isomeren hiervon, worin R die obige Bedeutung hat und R1 eine Schutzgruppe bedeutet, unter Verwendung eines Reduktionsmittels oder durch Acidolyse die Schutzgruppe R1 abspaltet, oder d) eine Verbindung der allgemeinen FormelCOCOOHoder ein optisches Isomeres hiervon, worin R die obige Bedeutung hat und R" eine Schutzgruppe bedeutet, hydrolysiertund danach gegebenenfalls die nach einer der Methoden a) bis d) erhaltene Verbindung in an sich bekannter Weise in ein optisches Isomeres oder ein physiologisch verträgliches Salz derselben umwandelt.
- 2. Verfahren nach Anspruch 1, gekennzeichnet dadurch, daß man Ausgangsverbindungen verwendet, in denen R-CH(CH3J2 ist.
- 3. Verfahren nach Anspruch 1 oder 2, gekennzeichnet dadurch, daß man als optisches Isomeres das D-Isomere verwendet.
