Das proteolytische Enzym Renin, das ein Molekulargewicht von etwa 40 000 hat, wird in der Niere gebildet und von dieser ins Blut sekretiert. Es ist bekanntlich in vivo bei der Spaltung des natürlich-vorkommenden Plasma-Glycoproteins Angiotensinogen aktiv, im Fall von menschlichem Angiotensinogen an der Bindung zwischen den Leucin- (10.) und Valin- (11.) Aminosäureresten am N-endständigen Ende von Angiotensinogen:

Das zirkulierende Decapeptid mit endständigem N (Angiotensin I), das durch die obige Spaltwirkung von Renin gebildet wird, wird anschließend durch den Körper in ein als Angiotensin II bekanntes Octapeptid zerlegt. Angiotensin II ist bekanntlich eine potente Pressorsubstanz, d.h. eine Substanz, die in der Lage ist, eine signifikante Blutdruckerhöhung zu induzieren, und die vermutlich wirkt, indem sie eine Verengung der Blutgefäße und die Freisetzung des Natrium zurückhaltenden Hormons Aldosteron aus der Nebenniere verursacht. So ist das Renin-Angiotensinogen-System als verursachender Faktor in bestimmten Formen der Hypertension und des kongestiven Herzversagens impliziert worden.
Eine Maßnahme zur Linderung der nachteiligen Effekte der Wirkung des Renin-Angiotensinogen-Systems besteht in der Verabreichung einer Substanz, die die Angiotensinogen-Spaltwirkung von Renin inhibieren kann. Es sind zahlreiche derartige Substanzen bekannt,
einschließlich Antirenin-Antikörpern, Pepstatin und natürlichvorkoitimenden Phospholipidverbindungen. Die europäische Patentanmeldung 45 665 (veröffentlicht am 2. Feb. 1982) beschreibt eine Reihe von renin-inhibierenden Polypeptidderivaten der Formel

1 2 sein kann, wobei jedes R und R eine lipophile oder aromatische Seitenkette ist. Gemäß den in dieser veröffentlichten Patentanmeldung angegebenen Definitionen wird es nicht in Betracht gezogen, daß entweder A oder Z Statin sein könnte oder daß B Lysin sein könnte.
Die europäische Patentanmeldung 77 028A (veröffentlicht am 20.April 1983) beschreibt eine Reihe von renin-inhibierenden Polypeptidverbindungen mit einem nicht-endständigen Statin- oder Statinderivat-Rest. Zu dieser Reihe gehören Verbindungen, die eine Phenylalanin-Histidin-Statin-Sequenz aufweisen.
Die europäische Patentanmeldung 132 304A beschreibt ebenfalls die Verwendung statinhaltiger Polypeptide als renin-inhibierende, antihypertensive Mittel, und die europäische Patentanmeldung 114 993A beschreibt Cyclostatin enthaltende Polypeptide, die als renin-inhibierende Antihypertensiva verwendbar sind.

Die vorliegende Erfindung umfaßt auch ein Verfahren zur Behandlung von Hypertension in einem Sauger, das die Verabreichung einer antihypertensiv wirksamen Menge der erfindungsgemäßen Verbindungen und einer pharmazeutischen Zusammensetzung, die aus den erfindungsgemäßen Verbindungen und einem Träger besteht, an diesen Sauger umfaßt.
Wie oben angegeben, umfaßt die vorliegende Erfindung die pharmazeutisch annehmbaren Salze der biologisch aktiven Verbindungen. Diese Salze sind diejenigen, die bei den verabrechten Dosen
nicht-toxisch sind. Da die erfindungsgemäßen Verbindungen basische Gruppen enthalten können, sind Säureadditionssalze möglich. Pharmazeutisch annehmbare Säureadditionssalze umfassen z.B. das Hydrochlorid, Hydrobromid, Hydrojodid, Sulfat, Bisulfat, Phosphat, saures Phosphat, Acetat, Lactat, Maleat, Mesylat, Fumarat, Citrat, saures Citrat, Tartrat, Bitartrat, Succinat, Gluconat und Saccharat.
Die e"rf indungsgemäßen Verbindungen zeigen in vivo in Säugern, einschließlich dem Menschen, antihypertensive Aktivität. Mindestens ein wesentlicher Anteil dieser Aktivität stammt aus ihrer Fähigkeit, die Angiotensinogenspaltung durch Renin zu inhibieren. Ohne durch die folgende Theorie eines Mechanismus beschränkt werden zu wollen ist es wahrscheinlich, daß der Mechanismus der renin-inhibierenden Aktivität der erfindungsgemäßen Verbindungen in ihrer selektiven Bindung (im Vergleich zu Angiotensinogen) an Renin besteht. Die erfindungsgemäßen Verbindungen zeigen eine enzym-inhibierende Aktivität, die für Renin selektiv ist. Aufgrund ihrer niedrigen Molekulargewichte zeigen sie günstige Löslichkeitseigenschaften in wässrigen Medien und eine bessere Absorbierbarkeit, was eine orale Verabreichung ermöglicht; und sie können bei kommerziell realistischen Kosten hergestellt werden. Die erfindungsgemäßen Verbindungen sind auch gegen ein kongestives Herzversagen einsetzbar.
Die erfindungsgemäßen Verbindungen können nach den dem Fachmann geläufigen Methoden hergestellt werden. Die heterocyclischen Säuren werden mit dem erforderlichen Aminolacton unter Anwendung von in der Technik der Amidherstellung bekannten Kupplungsmethoden gekuppelt. Bei dieser besonderen Verbindungs-Reihe wird die Verwendung von Carbodiimiden und N-Hydroxybenzotriazol zur Bildung aktivierter Ester der heterocyclischen Säuren bevorzugt, obgleich irgendeine der vielen, zur Herstellung von aktivierten Estern bekannten Gruppen verwendet werden kann. Eine zweite bevorzugte Maßnahme des Kuppeins der erforderlichen Säure mit dem geeigneten Aminolacton ist die Verwendui g von Diethylcyanophosphat. Dieses Reagenz ist ebenso für seine Verwendung in der Synthese von Amiden bekannt.
Nach dem Kuppeln der entsprechenden heterocyclischen Säure und des Aminolactons wird das Lacton mit einem Amin behandelt, was zur Bildung des gewünschten Hydroxyamids führt, das die notwendige und gewünschte Stereochemie hat, um die antihypertensive Aktivität, die diese Verbindungen besitzen, zu verleihen.
Die Ausgangsreaktionsteilnehmer für die Synthese der erfindungsgemäßen Verbindungen werden hier beschrieben, oder sie können nach Literaturverfahren hergestellt werden. Die EPO-Anmeldung 86 305 995.2 (Veröffentlichung 0212903 (A2)) beschreibt die Herstellung von Polypeptiden unter Verwendung eines Aminolactons ähnlich dem hier beschriebenen. Weiterhin sind verschiedene der heterocyclischen Säuren im Handel verfügbar.
Die Fähigkeit der erfindungsgemäßen Verbindungen, durch Inhibierung von Renin als antihypertensive Mittel zu wirken, kann durch den folgenden in vitro-Versuch nachgewiesen werden:

Blutplasma wurde von gesundem Laboratoriumspersonal abgenommen, gepoolt und in gefrorenem Zustand bis zur Verwendung gelagert. Vor der Verwendung wurde eine gewisse Menge dieses Plasmas aufgetaut und zentrifugiert, und das überstehende Material wurde mit Protease-Inhibitoren gemischt und auf pH 7,4 gepuffert. Renin-Inhibitoren wurden unterschiedlichen Aliquoten des überstehenden Plasmamaterials in verschiedenen Konzentrationen zugefügt, und die erhaltenen Gemische (310 λ) wurden 3 h bei 37°C zusammen mit Renininhibitor-freien Kontrollgemischen inkubiert. Nach der Inkubation wurden die Gemische in Eiswasser abgeschreckt und jeweils unter Verwendung von Angiotensin I-Antikörper auf Angiotensin I untersucht. Die Bildung von Angiotensin I in Gegenwart eines Renin-Inhibitors wurde mit derjenigen verglichen, die in Abwesenheit des Inhibitors gebildet worden war, und es wurde eine prozentuale Inhibierung berechnet. Unter Verwendung von Daten, die aus duplizierten Inkubationen bei jeder der verschiedenen Inhibitorkonzentrationen
erhalten worden waren, wurde die Inhibitorkonzentration in dem Inkubationsgemisch, das zur Bildung einer 50-%igen Inhibierung der Angiotensinogen-Spaltaktivität von Renin notwendig war, d.h. die IC50 des Inhibitors, für die verschiedenen Inhibitoren berechnet.
Das Angiotensin I in den abgeschreckten Inkubationsgemischen wurde mittels Radioimmuno-Test unter Verwendung der Komponenten eines Renin-Radioimmunoassay-Kits, geliefert von Becton Dickinson & Co. (Orangeburg, N.Y., USA), bestimmt. Dieser Radioimmuno-Test beruht auf dem von Haber et al, J.Clin.Endocrinol., 2^, Seite 1349-1355 (1969) entwickelten Test.
Die erfindungsgemäßen Verbindungen können als antihypertensive Mittel entweder auf oralen oder parenteralem Verabreichungsweg verabreicht werden, wobei ersterer bevorzugt wird, weil er für den Patienten bequem und angenehm ist. Im allgemeinen werden die antihypertensiven Verbindungen normalerweise oral in Dosen im Bereich von etwa 0,1 bis etwa 20 mg pro kg Körpergewicht pro Tag und, bei parenteraler Verabreichung, von 0,1 bis etwa 5 mg pro kg Körpergewicht pro Tag verabreicht; Änderungen werden notwenigerweise in Abhängigkeit vom Zustand des behandelten Patienten und der besonderen verabreichten Verbindung auftreten. Typischerweise wird die Behandlung bei einer niedrigen Tagesdosis begonnen und vom Arz nur nach Bedarf erhöht. Es ist zu erwähnen, daß die erfindungsgemäßen Verbindungen in Kombination mit pharmazeutisch annehmbaren Trägern auf jedem der oben angegebenen Wege verabreicht werden können und daß eine derartige Verabreichung sowohl in Einzel- als auch in Mehrfachdosen erfolgen kann.
Die erfindungsgemäßen Verbindungen können oral in einer großen Vielzahl unterschiedlicher Dosierunysformen verabreicht werden, d.h. sie können mit verschiedenen pharmazeutisch annehmbaren, inerten Trägern in Form von Tabletten, Kapseln, Lutschtabletten, Pastillen, harten Bonbons, Pulvern, Sprays, wässrigen Suspensionen, Elixieren, Sirupen usw. formuliert werden. Die Träger umfassen feste Verdünnungs- oder Füllmittel, sterile, wässrige Medien und verschiedene nicht-toxische, organische Lösungsmittel

usw. Außerdem können orale pharmazeutische Formulierungen zweckmäßig mit verschiedenen Mitteln des für solche Zwecke üblicherweise verwendeten Typs gesüßt und/oder aromatisiert werden. Im allgemeinen sind die erfindungsgemäßen Verbindungen in derartigen oralen Dosierungsformen in Konzentrationen im Bereich von etwa 0,5 bis etwa 90 Gew.-% der gesamten Zusammensetzung in Mengen anwesend, die zur Bildung der gewünschten Einheitsdosierungen ausreichen.
Für die orale Verabreichung können Tabletten verwendet werden, die verschiedene Streckmittel, wie Natriumeitrat, Calciumcarbonat und Calciumphosphat| zusammen mit verschiedenen den Zerfall fördernden Mitteln, wie Stärke und vorzugsweise Kartoffel- oder Tapioka-Stärke, Alginsäure und bestimmte komplexe Silicate, zusammen mit Bindemitteln, wie Polyvinylpyrrolidon, Saccharose, Gelatine und Akaziengum, enthalten. Weiterhin sind für Tablettierzwecke sehr häufig Gleitmittel, wie Magnesiumstearat, Natriumlaurylsulfat und Talkum, geeignet. Feste Zusammensetzungen eines ähnlichen Typs können als Füllmittel in weichen und harten, gefüllten Gelatinekapseln verwendet werden; bevorzugte Materialien in diesem Zusammenhang umfassen auch Lactose oder Milchzucker sowie hochmolekulare Polyethylenglykole. Wenn wässrige Suspensionen und/oder Elixiere zur oralen Verabreichung gewünscht werden, kann der wesentliche aktive Bestandteil darin mit verschiedenen Süßungs- oder Aromatisierungsmi+'teln, färbender Materie oder Farbstoffen und, nach Wunsch, Emulgatoren und/oder Suspendierungsmitteln zusammen mit Verdünnungsmitteln, wie Wasser, Ethanol, Propylenglykol, Glycerin, und verschiedenen ähnlichen Kombinationen hiervon kombiniert werden.
Die erfindungsgemäßen Verbindungen sind auch in der Diagnose von Hypertension und kongestivem Herzversagen einsetzbar.

IA. 2R,4S,5S-6-Cyclohexyl-5-( 5 ' -chlorindol-2'-ylrcarbonylaniino) 2-(2'-chlor-2'-propenyl)-f-hexanolacton
Zu 25 ml Methylenchlorid wurden bei 00C 165,5 mg (0,5 mMol) 2R, 4S, 5S-6-Cyclohexyl-5-amino-2-( 2 ' -chlor-2 ' --propenyl) -fhexanolacton-hydrochlorid, 50,6 mg (0,5 mMol) N-Methylmorphclin, 97,8 mg (0,05 mMol) 5-Chlorindol-2-carbonsäure, 67,5 mg (0,5 mMol) N-Hydroxybenzotriazol und 103 mg (0,5 mMol) Dicyclohexylcarbodiimid zugefügt, und das anfallende Reaktionsgemisch wurde bei Raumtemperatur über Nacht gerührt. Das Reaktionsgemisch wurde filtriert und das Filtrat zur Trockne konzentriert. Der Rückstand wurde erneut in Ethylacetat gelöst, nacheinander mit Wasser, einer gesättigten Natriumbicarbonatlösung und Kochsalzlösung gewaschen und über Magnesiumsulfat getrocknet. Das Lösungsmittel wurde entfernt, und der Rückstand, 280 mg, wurde an Siliciumdioxidgel unter Verwendung von Chloroform als Eluens chromatographiert, um 226 mg des gewünschten Produktes zu ergeben.
IB. 2R,4S,5S-6-Cyclohexyl-5-(5'-chlorindol-2'-yl-carbonylamino)-4-hydroxy-2-(2'-chlor-2'-propenyl)-N-methylhexanamid
Eine Lösung aus 226 mg des Produktes von Beispiel IA in 10 ml Methanol wurde mit gasförmigem Methylamin gesättigt, und das Reaktionsgemisch wurde 1,5 h bei Raumtemperatur stehen gelassen. Das Reaktionsmaterial wurde zur Trockne konzentriert und der Rückstand mit Ether verrieben, um 155 irg des gewünschten Produktes zu liefern.
Das NMR-Spektrum (CD OD) zeige Absorption bei 2,7 (3H, s), 5,15 (2H, m) und 7,0-8,0 (4H, m) ppm.
Unter Verwendung der Arbeitsweise von Beispiel 1A-1B und ausgehend von den entsprechenden Ausgangsreagenzien wurden die folgenden Verbindungen hergestellt:

Zu 10 ml Methylenchlorid wurden bei 00C 20 mg (0,1 mMol) 5-Chlorindol—3-carbonsäure, 30 mg (0,1 mMol) 2R,4S,5S-6-Cyclohexyl-5-amino-2-(2'-methylpropyl) -f-hexanolacton-hydrochlorid, 15 μΐ (0,1 mMol) Diethylcyanophosphat und 42 μΐ (0,3 mMol) Triethylamin zugefügt. Dimethylformamid (1 ml) wurde zugegeben, und das Reaktionsgemisch wurde bei Raumtemperatur über Nacht gerührt. Das Reaktionsmaterial wurde zur Trockne konzentriert und erneut in Ethylacetat gelöst, das nacheinander mit Wasser (2 x), einer gesättigten Natriumbicarbonatlösung (2 x) und Kochsalzlösung gewaschen wurde. 3B. 2R,4S,5S-6-Cyclohexyl-5-(5'-chlorindol-3'-yl-carbonylamino)-4-hydroxy-2-(2'-methylpropyl)-N-methylhexanamid
Eine Lösung des Produktes von Beispiel 3A in 10 ml Methanol wurde bei Raumtemperatur mit Methylamin gesättigt, und das anfallende Reaktionsgemisch wurde bei Raumtemperatur 2 h

gerührt. Das Reaktionsmaterial wurde zur Trockne eingeengt und ergab 61 mg eines Schaumes, der an Siliciumdioxidgel unter Verwendung von Methanol-Ch1.^ oform als Eluens chromatographiert wurde. Die das Produkt enthaltenden Fraktionen wurden vereinigt und konzentriert und ergaben 14 mg des gewünschten Produktes.
Das NMR-Spektrum (CD OD) zeigte Absorption bei 0,95 (6H, d, J=5 Hz), 2,7 (3H, s) und 7,0-8,0 (4H, m) ppm.
Unter Anwendung der Arbeitsweise von Beispiel 3A-3B und ausgehend von den erforderlichen Ausgangsreaktionsteilnehmern wurden die folgenden Verbindungen hergestellt:

5A. 2R,4S,5S-6-Cyclohexyl-5-(51-bromindol-3'-yl-
acetylamino)-2-(2'-methylpropyl)-f-hexanolacton
Zu 10 ml Methylenchlorid und 0,3 ml Dimethylformamid wurden bei 00C 30 mg (0,1 mMol) 2R,4S,5S-6-Cyclohexyl-5-amino-2-(2'-methylpropyl)-jf-hexanolacton-hydrochlorid, 25 mg (0,1 mMol) 5-Bromindol-3-essigsäure, 21 mg (0,1 mMol) Dicyclohexylcarbodiimid, 14 mg (0,1 mMol) N-Hydroxybenzotriazol und 11 μΐ (0,1 mMol) N-Methylmorpholin zugefügt, und das Reaktionsgemisch wurde über Nacht bei Raumtemperatur gerührt. Das Reaktionsmaterial wurde filtriert und das Filtrat zur Trockne eingeengt. Der Rückstand wurde erneut in Ethylacetat gelöst, das nacheinander mit Wasser, einer gesättigten Natriumbicarbonatlösung und Kochsalzlösung gewaschen und über Natriumsulfat getrocknet wurde. Das Lösungsmittel wurde unter Vakuum entfernt und der Rückstand, 54 mg, an Siliciumdioxidgel unter Verwendung von

3 % Aceton als Eluens chromatographiert, 28 mg.
5B. 2R,4S,5S-6-Cyclohexyl-5-(5'-bromindol-3'-ylacetylamino)-4-hydroxy-2-(2'-methylpropyl)-N-methylhexanamid
Eine Methanol-(10 ml) -lösung, enthaltend 28 mg des Produktes von Beispiel 5A, wurde bei Raumtemperatur mit Methylamin gesättigt und 2 h gerührt. Das Reaktionsmaterial wurde zur Trockne eingeengt und der Rückstand mit Hexan verrieben, um 30 mg des gewünschten Produktes zu ergeben.
Das NMR-Spektrum (CD OD) zeigte Absorption bei 0,95 (6H, d, J=5 Hz), 2,7 (3H, s) und 7,0-8,0 (4H, m) ppm.
Unter Anwendung der Arbeitsweise von Beispiel 5Ä-5B und mit den entsprechenden Ausgangsmaterialien wurden die folgenden Verbindungen hergestellt:

7A. 2R,4S,5S-6-Cyclohexyl-5-(6'-azaindol-2'-ylcarbonylamino) -2-( 2 ' -methylpropyl) -f^-hexanolacton
Zu 10 ml Methylenchlorid und 0,5 ml Dimethylformamid wurden bei 00C 37 mg (0,1 mMol) 2R,4S,5S-6-Cyclohexyl-5-amino-2-(2'-methylpropyl)-^hexanolacton-hydrochlorid, 33 mg (0,2 mMol) 6-Azaindol-2-carbonsäure, 25 mg (0,1 mMol) Dicyclohexylcarbodiimid, 17 mg (0,1 mMol) N-Hydroxybenzotriazol und 20 μΐ (0,1 mMol) N-Methylmorpholin zugefügt, und das Reaktionsgemisch wurde bei Raumtemperatur 48 h gerührt. Das Reaktionsmaterial wurde filtriert, mit Wasser und gesättigter Natriumbicarbonatlösung gewaschen und über Natriumsulfat getrocknet. Abdampfen des Lösungsmittel ergab 55 mg des gewünschten Produktes.
7B. 2R,4S,5S-6-Cyclohexyl-5-(6'-azaindol-2'-y1-carbonylamino)-4-hydroxy-2-(2'-methylpropyl)-N-methylhexanamid
Eine Lösung aus 55 mg des Produktes von Beispiel 7A in 10 ml Methanol wurde bei Raumtemperatur mit Methylamin gesättigt, und das Reaktionsgemisch wurde 2 h stehen gelassen. Das Lösungsmittel wurde unter Vakuum entfernt und der Rückstand an SiIiciumdioxidgel unter Verwendung von Methanol-Chloroform als

Eluens chromatographiert, um 9,7 mg Produkt zu ergeben.
Das NMR-Spektrum (CD3OD) zeigte Absorption bei 0,95 (6H, d, J = 5 Hz) und 2,7 (3H, s) ppm.
Unter Verwendung der Arbeitsweise von Beispiel 7A-7B und ausgehend von den entsprechenden Ausgangsmaterialien wurden die folgenden Verbindungen hergestellt:

2R,4S,5S-6-Cyclohexyl-5-(indol-2'-yl-carbonylamino)-4-hydroxy-2-(4'-aminobutyl)-N-methylhexanamid (HET = Indol-2-yl; R, = Cyclohexyl, R2 = (CH2J4NH2; R3 = CH3)
Eine Mischung aus 75 mg 2R,4S,SS-ö-Cyclohexyl-S-(5'-chlorindol-2'-yl-carbonylamino)-4-hydroxy-2-(4'-azido-2'-butenyl)-N-methylhexanamid, 35 mg Palladium-auf-Tierkohle und 5 ml Essigsäure in 5·ml Methanol wurden in einer Wasserstoffatmosphäre von 3,45 bar (50 psi) 6hgeschüttelt. Weitere 35 mg Katalysator wurden zugefügt, und die Reduktion wurde über Nacht fortgesetzt. Der Katalysator wurde abfiltriert und das FiItrat unter Vakuum zu 66 mg Produkt konzentriert. Der Rückstand wurde mit Ether verrieben und filtriert, 50 mg.
Das NMR-Spektrum (CD3OD) zeigte Absorption bei 2,7 (3H, 2) und 7,0-8,0 (5H, m) ppm.
HERSTELLUNG A 2R,4S,5S-6-Cyclohexyl-5-(t-butoxycarbonylamino)-2-(2'-chlor-
Zu einer Tetrahydrofuranlösung, enthaltend 37,5 mMol Lithiumdiethylamid bei -700C (hergestellt aus 23,4 ml 1,6 m Butyllithiumhexan und 4,26 g Diethylamin in 50 ml trockenem Tetrahydrofuran) wurde eine Lösung aus 4,67 g (15 mMol) 4S,5S-6-Cyclohexyl-5-( t-butoxycarbonylamino)-γ^-hexanolacton in 25 ml Tetrahydrofuran zugetropft. Nach 30 min bei -78°C wurde eine Lösung aus 3,64 g (16 mMol) 2-Chlor-3-jodpropen in 25 ml Tetrahydrofuran bei -7O0C zugetropft. Nach 2 h wurde das Reaktionsgemisch mit 10 ml einer gesättigten Ammoniumchloridlösung, die bei -780C zugetropft wurde, abgeschreckt, und das erhaltene Gemisch konnte sich auf Raumtemperatur erwärmen. Das Lösungsmittel wurde unter Vakuum entfernt und der Rückstand mit Diethylether extrahiert. Die Etherlösung wurde mit einer 10-%igen Zitronensäurelösung, einer gesättigten Natriumbicarbonatlösung und Kochsalzlösung gewaschen. Dann wurde die Etherlösung über Magnesiumsulfat getrocknet und konzentriert, um 6,83 g eines

Öles zu ergeben, das an Siliciumdioxidgel unter Verwendung von Ethylacetat-Hexan als Eluens chromatographiert wurde. Die das Produkt enthaltenden Fraktionen wurden vereinigt und konzentriert und ergaben 2,38 g des gewünschten Produktes.
Das NMR-Spektrum (CDCl,) zeigte Absorption bei 1,4 (9H, s) ppm.
Unter"Verwendung der Arbeitsweise von Herstellung A und der entsprechenden Ausgangsreaktionsteilnehmer wurden die folgenden Zwischenstufen synthetisiert:

Eine Lösung aus 710 mg (1,6 mMol) 2R,4S,SS-ö-Cyclohexyl-S-;t· butoxycarbonylamino)-2-(4'-brom-2'-buteny])- f-hexanolacton
und 986 mg (15,2 mMol) Natriumazid in 75 ml Dimethylsulfoxid-Wasser (2:1, Vol.:Vol.) wurde über Nacht bei Raumtemperatur gerührt. Das Reaktionsmaterial wurde in 500 ml Wasser gegossen und das Produkt mit Ethylacetat extrahiert. Die Extrakte wurden vereinigt und nacheinander mit Wasser und Kochsalzlösung gewaschen und über Magnesiumsulfat getrocknet. Das Lösungsmittel wurde unter Vakuum entfernt, um 73 mg des gewünschten Produktes zu er.geben.
2R,4S,5S-6-Cyclohexyl-5-(t-butoxycarbonylami no)-2-(2'-
Eine Lösung aus 100 mg (0,27 mMol) 2R,4S,SS-fi (t-butoxycarbonylamino)-2-(2'-methyl-2'-propenyl)-^hexanolacton in 15 ml Methylenchlorid wurde bei -78°C mit Ozon gesättigt, bis eine Blaufärbung bestehen blieb. Das Reaktionsgemisch wurde 30 mi/, bei -78°C stehen gelassen und dann von überschüssigem Ozon bei -78°C mit eil.em schnellen Stickstoffstrom befreit. Tetrabutylammoniumborhydrid (140 mg, 0,54 mMol) wurde bei -78°C zugefügt, und das Reaktionsgemisch wurde 2 d bei 00C stehen gelassen. Weitere 140 mg Borhydrid wurden zugefügt, und das Reaktionsgemisch wurde über Nacht bei Raumtemperatur stehen gelassen. Die Lösung wurde mit Wasser und Kochsalzlösung gewaschen, über Natriumsulfat getrocknet und eingeengt, um 250 mg Rohprodukt zu ergeben. Blitzchromatographie unter Verwendung von Chloroform-Methanol (99:1; Vol.:Vol.) ergab 45 mg der gewünschten Zwischenstufe.
Das NMR-Spektrum (CDCl,) zeigte Absorption bei 1,4 (9H, s) ppm.
2R, 4S, 5S-6-Cyclohexyl-5-amino-2- ( 2 ' -chi ->r-2 ' -propenyl) -^f-
Eine Lösung aus 385 mg (1 mMol) 2R,4S,5S-6-Cyclohexyl-5-(tbutoxycarbonylamino) -2-( 2 ' -chlor-2 ' -propenyl) -y'-hexanolacton in 10 ml 4,7 η Chlorwasserstoff in Dioxan wurde 2 h bei Raum-

temperatur gerührt. Das Lösungsmittel wurde unter Vakuum entfernt, um 331 mg des gewünschten Aminhydrochlorids zu ergeben,

Claims (1)
- PATENTANSPRÜCHE1.- Verfahren zur Herstellung einer Verbindung der Formel•R,HET-CONHCONHR.OHoder eines pharmazeutisch annehmbaren Salzes derselben, worin HEToder eine Gruppe der Formel(CH2)n- ist,worin X Wasserstoff, (C1-C3)AlKyI, (C1-C3)AIkOXy, Fluor, Chlor, Brom oder Cyano ist; Y Wasserstoff,(C,-C,)Alkyl, (C1-C3)AIkOXy, Fluor oder Chlor ist; R4 Wasserstoff oder (C^C-jAlkyl ist; η eine ganze Zahl von 0 bis 2 ist; R1 (C,-C0)Cycloalkyl oder i-Propyl ist; R3 (C3-C5)Alkyl, Phenyl, Methylvinyl, Dimethyl-vinyl, Halogenvinyl, Hydroxy(C,-C,)alkyl oder Amino(C,-C.)alkyl ist; und R^ (C,-Cfi)Alkyl oder Morpholinoethyl ist, gekennzeichnet durch die Umsetzung einer Verbindung der Formelmit einem Amin der Formel R,NH» in einem Niederalkanol bei Raumtenperatur bis praktisch zur Beendigung und, gegebenenfalls, Reduzieren der Verbindung, worin R_ W-N^(C9-C4)Alkylen oder N,CH„- ist, mit Wasserstoff und Palladium-auf-Tierkohle, um die Verbindung zu ergeben, worin R- Amino(C,-C.)alkyl ist.
