Description
- Die Erfindung betrifft cyclische Aminverbindungen und eine therapeutische Zusammensetzung zur Verwendung in der medizinischen Behandlung der senilen Demenz.
- Im Hinblick auf den raschen Anstieg des Bevölkerungsanteils älterer Menschen ist die Ermöglichung einer Therapie für die senile Demenz, wie beispielsweise die Alzheimer'sche senile Demenz, ein hoch erwünschtes Ziel.
- Verschiedene Versuche wurden angestellt, um die senile Demenz mit einem Arzneimittel zu behandeln. Bisher jedoch gibt es kein Arzneimittel, das zur Behandlung dieser Krankheiten sehr hilfreich wäre.
- Untersuchungen bezüglich der Entwicklung von therapeutischen Mitteln für diese Krankheiten wurden unter verschiedenen Aspekten angestellt. Da die Alzheimer'sche senile Demenz mit einer Verringerung der cholinergischen Hypofunktion einhergeht, wurde insbesondere die Entwicklung eines Arzneimittels unter dem Aspekt eines Acetylcholinvorläufers und eines Acetylcholinesteraseinhibitors vorgeschlagen und wird in der Tat versucht. Repräsentative Beispiele von Anti-Cholinesterase-Inhibitoren schliessen Physostigmin und Tetrahydroaminoacridin ein. Diese Arzneimittel haben jedoch Nachteile wie z. B. eine nicht ausreichende Wirkung und das Auftreten unerwünschter Nebenwirkungen. Gegenwärtig sind sie keine entscheidenden therapeutischen Mittel.
- Im Hinblick auf die obige Situation haben die Erfinder ausgedehnte und intensive Untersuchungen mit verschiedenen Verbindungen während vieler Jahre angestellt, um ein Arzneimittel zu entwickeln, das eine andauernde Wirksamkeit und hohe Sicherheit hat.
- Als Ergebnis entdeckten die Erfinder, dass ein Piperidinderivat mit der folgenden allgemeinen Formel (XXV) das erwünschte Ziel erreichen kann.
- Speziell hat die erfindungsgemässe Verbindung mit der folgenden allgemeinen Formel (XXV) grosse Vorteile einer starken und hochselektiven Antiacetylcholinesteraseaktivität, einer Erhöhung der Menge des im Hirn vorhandenen Acetylcholins, eines ausgezeichneten Effekts bei einem Modell für Gedächtnisstörungen, und einer andauernden Aktivität und hohen Sicherheit im Vergleich zu Physostigmin, das eine herkömmliche populäre Droge im Stand der Technik ist, was die erfindungsgemässe Verbindung sehr wertvoll macht.
- Die erfindungsgemässe Verbindung wurde auf Grundlage der inhibitorischen Wirkung auf die Acetylcholinesterase entdeckt und ist daher wirksam zur Behandlung und Verhinderung verschiedener Krankheiten, die man mit dem Fehlen von Acetylcholin als Neurotransmitter in vivo in Verbindung bringt.
- Beispiele solcher Krankheiten schliessen verschiedene Arten von Demenzen, einschliesslich der Alzheimer'schen senilen Demenz, ein und umfassen ausserdem die Chorea Huntington, die Pick'sche Krankheit und Ataxie.
- Daher sind die Ziele der vorliegenden Erfindung die Bereitstellung eines neuen Piperidinderivats, das wirksam ist als Arzneimittel, insbesondere zur Behandlung und Prävention von Krankheiten des Zentralnervensystems, die Bereitstellung eines Verfahrens zu ihrer Herstellung und die Bereitstellung eines Arzneimittels, das dieses Derivat als wirksamen Bestandteil umfasst.

- Vorzugsweise ist B -(CHR²²)r-; R²² ist ein Wasserstoffatom und r ist eine ganze Zahl von 1 bis 10.
- Bevorzugte erfindungsgemässe Verbindungen schliessen ein:
- 1-Benzyl-4-((5,6-dimethoxy-1-indanon)-2-yl)methylpiperidin,

- Unter den durch S wiedergegebenen Substituenten ist Methoxy der am meisten bevorzugte. t ist vorzugsweise eine ganze Zahl von 1 bis 4. Es ist am meisten bevorzugt, dass das Phenyl 1 bis 3 Methoxygruppen trägt.
- In der Definition B sind -(CHR²²)r- und =CH-(CH&sub2;)c- bevorzugt.
- Darüber hinaus stellt die Erfindung eine therapeutische Zusammensetzung zur Verfügung, die eine pharmakologisch wirksame Menge der cyclischen Aminverbindung mit der Formel (XXV) oder eines ihrer pharmakologisch annehmbaren Salze und einen pharmakologisch annehmbaren Träger umfasst.
- Gemäss einem weiteren Aspekt stellt die Erfindung ein Verfahren zur Herstellung einer cyclischen Aminverbindung der Formel (I) oder eines ihrer pharmakologisch akzeptablen Salze bereit:
- worin:
- S eine Niederalkylgruppe mit 1 bis 6 Kohlenstoffatomen, eine Niederalkoxygruppe mit 1 bis 6 Kohlenstoffatomen, ein Halogenatom, eine Hydroxylgruppe und t 0 bis 4 ist, oder (S)t eine Methylendioxygruppe oder eine Ethylendioxygruppe an zwei benachbarten Kohlenstoffatomen der Phenylgruppe, an die (S)t gebunden ist, bilden kann;

- wobei das Verfahren die folgenden Schritte umfasst:
- (i) Reduktion eines cyclischen Amins der Formel (11):
- worin S, t, r und K wie oben definiert sind; und
- (ii) gegebenenfalls Überführung der erhaltenen Verbindung der Formel (I) in ein pharmakologisch akzeptables Salz.
- Gemäss einem weiteren Aspekt stellt die vorliegende Erfindung ein Verfahren zur Herstellung einer cyclischen Aminverbindung der Formel (II) oder eines ihrer pharmakologisch akzeptablen Salze bereit:
- worin S, t, r und K wie oben im Zusammenhang mit Formel (I) definiert sind, welches die folgenden Schritte umfasst:
- (i) Umsetzung von
- worin S, t, r und K wie oben definiert sind, in einer Wittig- Reaktion; und
- (ii) gegebenenfalls Überführung der erhaltenen Verbindung der Formel (II) in ein pharmakologisch akzeptables Salz.
- Gemäss einem weiteren Aspekt stellt die vorliegende Erfindung ein Verfahren zur Herstellung einer cyclischen Aminverbindung der Formel (II) oder eines ihrer pharmakologisch akzeptablen Salze bereit:
- worin S, t, r und K wie oben im Zusammenhang mit Formel (I) definiert sind, welches die folgenden Schritte umfasst:
- (i) Umsetzung von
- worin S, t, r und K wie oben definiert sind, in Gegenwart von Lithiumdiisopropylamid; und
- (ii) gegebenenfalls Überführung der erhaltenen Verbindung der Formel (II) in ein pharmakologisch akzeptables Salz.
- Gemäss einem weiteren Aspekt stellt die vorliegende Erfindung ein Verfahren zur Herstellung einer cyclischen Aminverbindung der Formel (III) oder eines ihrer pharmakologisch akzeptablen Salze bereit:
- worin K wie oben im Zusammenhang mit Formel (I) definiert ist;
- ist, worin S und t wie oben im Zusammenhang mit Formel (I) definiert sind; und
- B die zweiwertige Gruppe -(CHR²²)r- ist, wobei r eine ganze Zahl von 1 bis 10 ist und R²² jeweils unabhängig entweder ein Wasserstoffatom oder eine Methylgruppe ist;
- wobei das Verfahren die folgenden Schritte umfasst:
- (i) Reduktion eines cyclischen Amins der Formel (IV):
- worin J und K wie oben definiert sind und B' B entspricht, wobei jedoch die terminale Gruppe, die ein Kohlenstoffatom enthält, weggelassen ist; und
- (ii) gegebenenfalls Überführung der erhaltenen Verbindung der Formel (III) in ein pharmakologisch akzeptables Salz.
- Gemäss einem weiteren Aspekt stellt die vorliegende Erfindung ein Verfahren zur Herstellung einer cyclischen Aminverbindung der Formel (V) oder eines ihrer pharmakologisch akzeptablen Salze bereit:
- worin S, t und K wie oben im Zusammenhang mit Formel (I) definiert sind;
- B eine der zweiwertigen Gruppen =(CH-CH=CH)b-, in der b eine ganze Zahl von 1 bis 3 ist; =CH-(CH&sub2;)c-, in der c eine ganze Zahl von 0 bis 9 ist; oder =(CH-CH)d= , in der d eine ganze Zahl von 0 bis 5 ist, bedeutet und
- eine Einfach- oder eine Doppelbindung bezeichnet, wobei das Verfahren die folgenden Schritte umfasst:
- (i) Umsetzung von
- worin S, t und K wie oben definiert sind und B' B entspricht, wobei jedoch die terminale Gruppe, die ein Kohlenstoffatom enthält, weggelassen ist, in einer Wittig-Reaktion; und
- (ii) gegebenenfalls Überführung der erhaltenen Verbindung der Formel (V) in ein pharmakologisch akzeptables Salz.
- Gemäss einem weiteren Aspekt stellt die vorliegende Erfindung ein Verfahren zur Herstellung einer cyclischen Aminverbindung der Formel (V) oder eines ihrer pharmakologisch akzeptablen Salze bereit:
- worin S, t und K wie oben im Zusammenhang mit Formel (I) definiert sind;
- B eine der zweiwertigen Gruppen =(CH-CH=CH)b-, in der b eine ganze Zahl von 1 bis 3 ist; =CH-(CH&sub2;)c-, in der c eine ganze Zahl von 0 bis 9 ist; oder =(CH-CH)d = , in der d eine ganze Zahl von 0 bis 5 ist, bedeutet und
- eine Einfach- oder eine Doppelbindung bezeichnet, wobei das Verfahren die folgenden Schritte umfasst:
- (i) Umsetzung von
- worin S, t und K wie oben definiert sind und B' B entspricht, wobei jedoch die terminale Gruppe, die ein Kohlenstoffatom enthält, weggelassen ist; und
- (ii) gegebenenfalls Überführung der erhaltenen Verbindung der Formel (V) in ein pharmakologisch akzeptables Salz.
- Gemäss einem weiteren Aspekt stellt die vorliegende Erfindung ein Verfahren zur Herstellung einer cyclischen Aminverbindung der Formel (VI) oder eines ihrer pharmakologisch akzeptablen Salze bereit:
- worin S, t und K wie oben im Zusammenhang mit Formel (I) definiert sind; und
- r eine ganze Zahl von 0 bis 10 ist und R²² jeweils unabhängig ein Wasserstoffatom oder eine Methylgruppe ist;
- wobei das Verfahren die folgenden Schritte umfasst:
- (i) Dehydratation einer Indanolverbindung der Formel:
- worin S, t, r und K wie oben definiert sind; und
- (ii) gegebenenfalls Überführung der erhaltenen Verbindung der Formel (VI) in ein pharmakologisch akzeptables Salz.
- In der vorliegenden Erfindung schliesst der Begriff "pharmakologisch annehmbare Salze" solche mit anorganischen Säuren ein, wie z. B. Hydrochloride, Sulfite, Hydrobromide und Phosphate, und solche mit organischen Säuren, wie z. B. Formiate, Acetate, Trifluoracetate, Methansulfonate, Benzolsulfonate und Toluolsulfonate. Wenn ausserdem eine bestimmte Art von Substituenten gewählt wird, kann die erfindungsgemässe Verbindung z. B. Alkalimetallsalze, z. B. ein Natrium- oder Kaliumsalz, Erdalkalimetallsalze, z. B. ein Calcium- oder Magnesiumsalz, organische Aminsalze, z. B. ein Salz mit Trimethylamin, Triethylamin, Pyridin, Picolin, Dicyclohexylamin oder N,N- Dibenzylethylendiamin, bilden.
- Ausserdem können die erfindungsgemässen Verbindungen in Abhängigkeit von der Art des Substituenten ein asymmetrisches Kohlenstoffatom enthalten und daher Stereoisomere bilden. Diese liegen selbstverständlich im Bereich der vorliegenden Erfindung.
- Ein spezifisches Beispiel dafür wird nun beschrieben. Wenn J ein Indanonskelett hat, hat die erfindungsgemässe Verbindung ein asymmetrisches Kohlenstoffatom und kann daher Stereoisomere, optische Isomere, Diastereomere, etc., aufweisen. Alle diese Isomere liegen im Bereich der vorliegenden Erfindung.
- Die erfindungsgemässe Verbindung kann durch verschiedene Verfahren hergestellt werden. Repräsentative Verfahren zur Herstellung der erfindungsgemässen Verbindungen werden nun angegeben.
- Wenn J in der allgemeinen Formel (XXV) eine einwertige oder zweiwertige Gruppe ist, die sich von einem Indanon mit einer unsubstituierten oder substituierten Phenylgruppe ableitet, und B eine Gruppe mit der Formel -(CH&sub2;)n- ist, wobei n eine ganze Zahl von 1 bis 6 ist, kann die erfindungsgemässe Verbindung nach dem folgenden Verfahren hergestellt werden:
- Insbesondere kann eine Verbindung (X), die eine der Zielverbindungen ist, hergestellt werden, indem man ein substituiertes 1-Indanon-2-ylphosphonat mit der allgemeinen Formel (VII) mit einer Aldehydverbindung der Formel (VIII) (d. h. durch eine Wittig-Reaktion) umsetzt und so eine Verbindung (IX) herstellt, die eine der Zielverbindungen ist, und dann diese Verbindung (IX) katalytisch reduziert.
- Beispiele des in der Wittig-Reaktion verwendeten Katalysators schliessen Natriummethylat (MeONa), Natriumethylat (EtONa), tert-BuOK und NaH ein. Beispiele des bei dieser Reaktion verwendeten Lösungsmittels schliessen Tetrahydrofuran (THF), Dimethylformamid (DMF), Ether, Nitromethan und Dimethylsulfoxid (DMSO) ein. Eine Reaktionstemperatur im Bereich von Raumtemperatur bis etwa 100ºC liefert vorteilhafte Ergebnisse.
- Die katalytische Reaktion in Gegenwart eines Katalysators aus Palladium-Kohle etc., liefert günstige Ergebnisse.
- Das folgende Schema zeigt spezifisch ein Verfahren zur Herstellung der erfindungsgemässen Verbindung, in der J eine Gruppe mit der Formel
- ist, wobei R&sup6; und R&sup7; gleich oder verschieden sein können und jeweils ein Wasserstoffatom, eine Niederalkylgruppe, eine Niederalkylalkoxygruppe oder ein Halogenatom unter den durch A definierten Gruppen sind, B eine Gruppe mit der Formel -(CH&sub2;)n- ist, wobei n eine ganze Zahl von 1 bis 6 ist, K eine Gruppe mit der Formel
- ist, wobei R&sup8; und R&sup9; jeweils die selbe Bedeutung wie R&sup6; und R&sup7; haben:
- Wenn J in der allgemeinen Formel (XXV) eine einwertige oder zweiwertige Gruppe ist, die sich von einem Indanon mit einer unsubstituierten oder substituierten Phenylgruppe ableitet, und B eine Gruppe mit der Formel -(CH&sub2;)n- ist, wobei n eine ganze Zahl von 1 bis 6 ist, kann die erfindungsgemässe Verbindung auch nach dem folgenden Verfahren hergestellt werden:
- Spezifisch werden beispielsweise Diisopropylamin und n-Butyllithium/Hexan zu einem Lösungsmittel, wie Tetrahydrofuran, gegeben. Ein substituiertes 1-Indanon mit der allgemeinen Formel (XI) und Hexamethylphosphorsäureamid werden dazu bei einer Temperatur von vorzugsweise etwa -80ºC gegeben. Dann wird eine Aldehydverbindung mit der allgemeinen Formel (VIII) zugegeben und anschliessend eine Reaktion nach einer gewöhnlichen Methode ausgeführt. Die Reaktionsmischung wird dehydriert und dadurch die Verbindung (IX) hergestellt. Diese Verbindung kann katalytisch auf die selbe Weise wie in Verfahren B zur Herstellung der Verbindung (X) reduziert werden.
- Ein spezifisches Beispiel des Verfahrens C wird nun auf die selbe Weise wie zuvor bei Verfahren B beschrieben.
- Die cyclische Aminverbindung mit der Formel (XXV), in der J (2) Indanonyl, (5) Indandionyl ist und B -(CHR²²)r-, =(CH-CH=CH)b, =CH-(CH&sub2;)c- oder =(CH-CH)d = ist, kann nach der folgenden Prozedur hergestellt werden. B' ist eine Gruppe, bei der die Endgruppe von B, die ein Kohlenstoffatom enthält, fehlt.
- Bei dieser Prozedur wird das Phosphat mit einer Aldehydverbindung durch eine Wittig-Reaktion umgesetzt und das Produkt katalytisch reduziert. Der in der Wittig-Reaktion verwendete Katalysator schliesst Natriummethylat, Natriumethylat, Kalium-t-butyrat oder Natriumhydrid ein. Die Reaktion kann in einem Lösungsmittel, wie Tetrahydrofuran, Dimethylformamid, Ether, Nitromethan und Dimethylsulfoxid bei einer Temperatur von Raumtemperatur bis 100ºC ausgeführt werden. Bei der katalytischen Reduktion wird vorzugsweise ein Katalysator verwendet, wie beispielsweise ein Katalysator aus Palladium und Kohle, Raney-Nickel und ein Katalysator aus Rhodium und Kohle.
- In der oben gezeigten Prozedur verläuft ein Beispiel, in dem J Indanonyl ist, wie folgt:
- Die in Schritt 1 definierte Verbindung kann auch auf die folgende Weise erhalten werden:
- Die Verbindung J-H, wie beispielsweise Indanon, wird mit einem Aldehyd durch eine herkömmliche Aldolkondensation unter Erhalt einer Zielverbindung umgesetzt. Die Reaktion kann in einem Lösungsmittel, wie Tetrahydrofuran, ausgeführt werden, indem man zuerst Lithiumdiisopropylamid aus Diisopropylamin und einer n-Butylhexanlösung von Lithium herstellt, dazu eine Verbindung J-H bei einer Temperatur von vorzugsweise etwa -80ºC gibt, dann den Aldehyd zugibt, eine Reaktion auf herkömmliche Weise ausführt, dann die Produktionsmischung auf Raumtemperatur erwärmt, um eine Dehydrierung auszuführen, und schliesslich die Zielverbindung in Enonform erhält. In einer weiteren Ausführungsform werden die zwei Reaktanten in einem Lösungsmittel, wie Tetrahydrofuran, gelöst, eine Base, wie z. B. Natriummethylat, wird zur Lösung bei etwa 0ºC gegeben, und die Reaktion bei Raumtemperatur ausgeführt.
- Die auf diese Weise erhaltene Enonform kann zum Erhalt der Zielverbindung reduziert werden.
- Ein Beispiel, in dem J Indanonyl ist und B -(CH&sub2;)r- ist, verläuft wie folgt:
- Die Verbindung mit einer Indenylgruppe wird nach der folgenden Prozedur hergestellt. Diese Prozedur gilt für eine Verbindung, die Indenylgruppen mit einem oder mehreren Substituenten am Phenyl aufweist.
- Die Dehydratation wird in herkömmlicher Weise ausgeführt, beispielsweise mit Salzsäure.
- Die Indanonverbindung, die in den oben gezeigten Verfahren I und K verwendet wurde, ist im Handel erhältlich und wird durch die folgenden Schritte hergestellt:
- Die oben verwendete Aldehydverbindung wird nach dem folgenden Verfahren hergestellt:
- Die oben gezeigte Ausgangsverbindung wird zum Aldehyd umgewandelt und der Aldehyd zur Wittig-Reaktion zur Erhöhung der Kohlenstoffzahl verwendet. Die Wittig-Reaktion wird wiederholt oder in Kombination mit einer anderen Art einer Wittig-Reaktion ausgeführt. Dies ist für den Fachmann offensichtlich. Das Wittig-Reagens schliesst Methoxymethylentriphenylphosphoran zur Hinzufügung eines Kohlenstoffatoms und Formylmethylentriphenylphosphoran zur Hinzufügung von zwei Kohlenstoffatomen ein. Methoxymethylentriphenylphosphoran wird durch Reaktion zwischen Methoxymethylentriphenylphosphoniumchlorid und n-Butyllithium in Ether oder Tetrahydrofuran erhalten. Dann wird eine Keton- oder Aldehydverbindung zur Produktmischung gegeben, um deren Methoxyvinylverbindung zu erhalten, und die resultierende Mischung mit einer Säure zum Erhalt des entsprechenden Aldehyds behandelt. Ein Beispiel verläuft wie folgt:
- Wenn Formylmethylentriphenylphosphoran verwendet wird, wird eine Lösung des Ausgangsketons oder -aldehyds in Ether, Tetrahydrofuran oder Benzol mit diesem Wittig-Reagens gemischt und die Mischung zum Rückfluss erhitzt, um die Zielverbindung zu erhalten.
- Die erhaltene ungesättigte Aldehydverbindung kann in die entsprechende gesättigte Verbindung durch katalytische Reduktion unter Verwendung eines Katalysators aus Palladium und Kohle, Raney-Nickel oder einem Rhodium/Kohle-Katalysator umgewandelt werden. Ein Beispiel verläuft wie folgt:
- Die so hergestellten Verbindungen und deren Säureadditionssalze mit der allgemeinen Formel (XXV) sind verwendbar zur Behandlung verschiedener Arten der senilen Demenz, insbesondere der senilen Demenz vom Alzheimer'schen Typ.
- Die Erfindung wird hinsichtlich ihrer therapeutischen Nützlichkeit zusammen mit pharmakologischen Experimentaldaten beschrieben.
- Ein Mäusehirnhomogenat wurde als Acetylcholinesterasequelle verwendet und seine Esteraseaktivität nach der Methode von Ellman et al bestimmt (Ellman, G.L., Courtney, K.D., Andres, V., und Featherstone, R.M. (1961) Biochem. Pharmacol., 7, 88-95).
- Acetylthiocholin als Substrat, eine zu detektierende Probe und DTNB wurden zum Mäusehirnhomogenat gegeben, gefolgt von Inkubation. Die Menge einer durch Reaktion zwischen dem Thiocholin und DTNB gebildeten gelben Substanz wurde durch die Absorption von 412 nm bestimmt und auf die Acetylcholinesteraseaktivität bezogen.
- Die Acetylcholinesterase-inhibierende Aktivität der Probe wurde als die inhibitorische Konzentration 50% (IC&sub5;&sub0;) ausgedrückt.

- Eine zu untersuchende Probe wurde oral an Ratten verabreicht. Eine Stunde nach Verabreichung wurden die Hirnhälften seziert und homogenisiert und anschliessend die Acetylcholinesteraseaktivität bestimmt. Eine Gruppe von Ratten, die mit physiologischer Kochsalzlösung behandelt worden war, wurde als Kontrolle verwendet. Die Inhibierung von AChE durch Proben ex vivo wurde ausgedrückt als Prozentsatz der Inhibierung gegenüber dem Kontrollwert. Ergebnisse werden in Tabelle 2 gezeigt.

- Männliche Wistar-Ratten wurden als Testtiere verwendet und eine Lichtschranke und ein dunkler Behälter als Vorrichtung verwendet. Eine zu untersuchende Probe wurde oral 1 Stunde vor dem Training verabreicht und die Ratten mit 0,5 mg/kg (i.p.) Scopolamin 30 Minuten vor dem Training behandelt. In einem Trainingsexperiment wurde das Tier in einen hellen Raum gestellt und gleich nach dem Eintreten des Tieres in einen dunklen Raum eine Falltür geschlossen und anschliessend ein elektrischer Schock vom Bodengitter aus verabreicht. Nach 6 Stunden wurde das Tier wieder in einen hellen Raum für ein Retentionsexperiment gestellt und die Zeit, die das Tier dort verbrachte, bis es wieder in den dunklen Raum eintrat, als Bewertung der Wirkung der Probe gemessen.
- Der Unterschied in der Responsezeit zwischen der Gruppe, die physiologische Kochsalzlösung verabreicht erhielt, und der Gruppe, die Scopolamin verabreicht erhielt, wurde als 100% angesetzt und die Wirkung der Probe als Prozentsatz Antagonismus durch die Probe ausgedrückt (Reverse %).
- Die Ergebnisse werden in Tabelle 3 gezeigt. TABELLE 2 TABELLE 3
- Die Anzahl der Tiere pro Dosis betrug 10 bis 17. NE: nicht wirksam.
- Die oben beschriebenen pharmakologischen Experimente ergaben, dass die erfindungsgemässe Verbindung eine potente Acetylcholinesteraseinhibierende Wirkung hatte.
- Unter den Verbindungen (I) der vorliegenden Erfindung ist eine Verbindung, in der J eine Indanonyl- oder eine Indanolidenylgruppe ist, die sich von einem Indanon mit einem unsubstituierten oder substituierten Phenylring ableitet, bevorzugt, und eine Verbindung, in der J eine Indanonylgruppe ist, ist besonders bevorzugt. Insbesondere eine Verbindung, in der J eine Gruppe ist, die sich von einem Indanon mit einem unsubstituierten oder substituierten Phenylring ableitet, hat Charakteristika, wie einen deutlichen Unterschied zu herkömmlichen Acetylcholinesteraseinhibitoren in der Struktur, Vorteile hinsichtlich der Herstellung der pharmazeutischen Präparate aufgrund seiner potenten Acetylcholinesterase-inhibierenden Wirkung, einen grossen Abstand zwischen den Haupt- und Nebenwirkungen, eine persistente Aktivität, hohe Wasserlöslichkeit, ausgezeichnete Stabilität, Vorteile bei der Formulierung zu Präparaten, hohe Bioverfügbarkeit und ausgezeichnete Penetration in das Gehirn.
- Daher sind die Ziele der vorliegenden Erfindung die Bereitstellung einer neuen Verbindung, die wirksam ist für verschiedene Arten der Demenz und von Folgeerscheinungen von cerebrovaskulären Krankheiten, die Bereitstellung eines Verfahrens zur Herstellung und die Bereitstellung eines neuen Arzneimittels mit denselben als Wirkstoff.
- Die repräsentative erfindungsgemässe Verbindung Nr. 4 in der obigen Tabelle 3 wurde für Toxizitätstests bei Ratten verwendet. Als Ergebnis wies die Verbindung eine Toxizität von 100 mg/kg oder mehr auf, d. h. keine ernsthafte Toxizität.
- Die erfindungsgemässe Verbindung ist wirksam zur Behandlung, Vorbeugung, Linderung, Verbesserung, etc., verschiedener Arten der senilen Demenz, insbesondere der Alzheimer'schen senilen Demenz; von cerebrovaskulären Krankheiten im Gefolge eines Hirnschlags, z. B. Cerebralhämorrhagie oder Cerebralinfarkten, Cerebralarteriosklerose, Kopfverletzungen, etc.; und Aprosexie, Sprechstörungen, Hyperbulie, emotionale Veränderungen, Kurzzeitgedächtnisstörungen, halluzinatorische paranoide Syndrome, Verhaltensveränderungen, einer begleitenden Encephalitis, Cerebrallähmung, etc..
- Ausserdem hat die erfindungsgemässe Verbindung eine starke und hochselektive Anticholinesterasewirkung, was die erfindungsgemässe Verbindung auch als Arzneimittel auf Grundlage dieser Wirkung verwendbar macht.
- Speziell ist die erfindungsgemässe Verbindung beispielsweise wirksam für die Chorea Huntington, die Pick'sche Krankheit und verzögerte Ataxie oder spät eintretende motorische Fehlentwicklungen, die von der Alzheimer'schen senilen Demenz verschieden sind.
- Wenn die erfindungsgemässe Verbindung als Arzneimittel für diese Krankheiten verwendet wird, kann sie oral oder parenteral verabreicht werden. Im allgemeinen wird sie parenteral in Form von Injektionen, z. B. intravenös, subkutan, und intramuskulär, als Suppositorien oder Sublingualtabletten verabreicht. Die Dosis wird insbesondere in Abhängigkeit von den Symptomen; Alter, Geschlecht, Gewicht und der Empfindlichkeit der Patienten; der Verabreichungsweise; der Zeit und den Verabreichungsintervallen und Eigenschaften, der Darreichung und der Art der pharmazeutischen Präparate; der Art der Wirkstoffe, etc., abhängen, so dass hinsichtlich der Dosis keine besondere Beschränkung besteht. Normalerweise kann die Verbindung in einer Dosis von etwa 0,1 bis 300 mg, vorzugsweise 1 bis 100 mg pro Tag pro Erwachsener, gewöhnlich in 1 bis 4 Portionen, verabreicht werden.
- Pharmazeutische Präparate in der Dosierungsform von z. B. Injektionen, Suppositorien, Sublingualtabletten, Tabletten und Kapseln, werden nach Verfahren hergestellt, die im Stand der Technik gewöhnlich angewandt werden.
- Bei der Herstellung von Injektionen wird der Wirkstoff, falls erforderlich, mit einem pH-Modifikator, einem Puffer, einem Suspendiermittel, einem Solubilisiermittel, einem Stabilisator, einem tonizitätsherstellenden Mittel, einem Konservierungsmittel, etc., gemischt und anschliessend nach einem gewöhnlichen Verfahren eine intravenöse subkutane oder intramuskuläre Injektion hergestellt. In diesem Fall ist es, falls erforderlich, möglich, diese Präparate nach einem gewöhnlichen Verfahren zu lyophilisieren.
- Beispiele von Suspendiermitteln schliessen Methylcellulose, Polysorbat 80, Hydroxyethylcellulose, Acacia, gepulverten Tragacanth, Natriumcarboxymethylcellulose und Polyoxyethylensorbitonmonolaurat ein.
- Beispiele von Solubilisierungsmitteln schliessen polyoxyethylenhydriertes Castoröl, Polysorbat 80, Nikotinamid, Polyoxyethylensorbitanmonolaurat, Macrogol und einen Ethylester einer Castoröl-Fettsäure ein.
- Beispiele von Stabilisatoren schliessen Natriumsulfit, Natriummetasulfit und Ether ein und Beispiele von Konservierungsmitteln schliessen Methyl-p-hydroxybenzoat, Ethyl-p-hydroxybenzoat, Sorbinsäure, Phenol, Cresol und Chlorcresol ein.
- Die vorliegende Erfindung wird nun in grösserer Ausführlichkeit unter Bezug auf die folgenden Beispiele beschrieben. Es erübrigt sich zu sagen, dass die technische Lehre der vorliegenden Erfindung nicht nur auf diese Beispiele beschränkt ist.
- In den folgenden Beispielen wurden alle NMR-werte mit Verbindungen in freier Form gemessen.
- 0,37 g 1-Benzyl-4-[2-[(1-indanon)-2-yl]]-ethylpiperidin wurden in 10 ml Methanol gelöst und anschliessend 0,1 g 5% Rhodium-Kohle zugegeben. Die Mischung wurde bei Raumtemperatur unter Atmosphärendruck 24 Stunden hydriert. Der Katalysator wurde abfiltriert und das Filtrat im Vakuum konzentriert. Der Rückstand wurde durch Silicagel-Säulenchromatografie (Methylenchlorid/Methanol = 200 : 1) gereinigt. Das Eluat wurde im Vakuum konzentriert und der Rückstand in Methylenchlorid gelöst. Eine 10%-ige Chlorwasserstofflösung in Ethylacetat wurde zur resultierenden Lösung zugegeben, gefolgt von Konzentrierung im Vakuum unter Erhalt von Kristallen, die aus Methanol/IPE umkristallisiert wurden, so dass man 0,33 g (Ausbeute 80%) der Titelverbindung mit den folgenden Eigenschaften erhielt:

- Gefunden (%): 74,66 7,65 3,77 BEISPIEL 2 1-Benzyl-4-[2-[(1-indanon)-2-ylidenyl]]ethylpiperidin-hydrochlorid:
- 0,32 g 60%-iges Natriumhydrid wurden mit Hexan gewaschen und 10 ml THF zugegeben. Eine Lösung von 2,12 g Diethyl-1-indanon-2-ylphosphonat in 30 ml THF wurde bei 0ºC zugetropft. Die Mischung wurde bei Raumtemperatur 30 Minuten gerührt und wieder auf 0ºC gekühlt, gefolgt von Zugabe einer Lösung von 3,43 g 1-Benzyl-4-piperidinacetaldehyd in 10 ml DMF. Die Mischung wurde bei Raumtemperatur 2 Stunden und bei 50ºC 2 Stunden gerührt und dann 2 Stunden unter Erwärmen der Mischung am Rückfluss gehalten. Methanol und 20%-ige Schwefelsäure wurden bei 0ºC zur Reaktionsmischung zugegeben. 10 Minuten nach der Zugabe wurde die Reaktionsmischung mit Natronlauge basisch gemacht und mit Ethylacetat extrahiert. Die organische Phase wurde mit einer gesättigten Kochsalzlösung gewaschen, über Magnesiumsulfat getrocknet und im Vakuum konzentriert. Der resultierende Rückstand wurde mittels Silicagel-Säulenchromatografie (Methylenchlorid/Methanol = 500 : 1) gereinigt. Das Eluat wurde im Vakuum konzentriert und der Rückstand in Methylenchlorid gelöst. Eine 10%-ige Chlorwasserstofflösung in Ethylacetat wurde zur resultierenden Lösung zugegeben, gefolgt von Konzentrierung im Vakuum unter Erhalt von 0,78 g (Ausbeute 27%) der Titelverbindung. 1,37 g Diethyl-1-indanon-2-ylphosphorat wurden ebenfalls gewonnen.
- Molekülformel: C&sub2;&sub3;H&sub2;&sub5;NO·HCl
- ¹H-NMR (CDCl&sub3;) δ: 1,10-2,13 (7H, m), 2,26 (2H, t), 2,88 (2H, bd), 3,48 (2H, s), 6,72-7,07 (2H, m), 7,30 (5H, s), 7,10-8,00 (5H, m). BEISPIEL 3 1-Benzyl-4-piperidin-carboaldehyd mit der Formel
- wurde auf die folgende Weise hergestellt:
- 26 g Methoxymethylentriphenylphosphoniumchlorid wurden in 200 ml wasserfreiem Ether suspendiert. Eine 1,6 M-Lösung von n-Butyllithium in Hexan wurde zur Suspension bei Raumtemperatur zugetropft. Die Mischung wurde bei Raumtemperatur 30 Minuten gerührt und auf 0ºC gekühlt. Dann wurden 30 ml einer Lösung von 14,35 g 1-Benzyl-4-piperidon in wasserfreiem Ether zur Mischung zugegeben. Diese wurde bei Raumtemperatur 3 Stunden gerührt und vom Unlöslichen abfiltriert. Die filtrierte Flüssigkeit wurde bei reduziertem Druck konzentriert. Das erhaltene Konzentrat wurde in Ether gelöst und mit 1 N Salzsäure extrahiert. Eine wässrige Natronlauge wurde zum Extrakt zugegeben und so der pH-Wert auf 12 eingestellt. Die resultierende Lösung wurde mit Methylenchlorid extrahiert. Der Extrakt wurde über Magnesiumsulfat getrocknet und bei reduziertem Druck konzentriert. Der Rückstand wurde mit einer Silicagelsäule unter Erhalt von 5,50 g eines Öls mit einer Ausbeute von 33% gereinigt.
- Das Öl wurde in 40 ml Methanol gegeben und 40 ml 1 N Salzsäure zur Lösung gegeben. Die Mischung wurde auf Rückfluss 3 Stunden erhitzt und dann unter reduziertem Druck konzentriert. Der Rückstand wurde in Wasser gelöst. Eine wässrige Natronlauge wurde zur Lösung zugegeben, so dass ein pK von 12 eingestellt wurde, und die Lösung mit Methylenchlorid extrahiert. Der Extrakt wurde mit gesättigter Salzlösung gewaschen und über Magnesiumsulfat getrocknet. Er wurde weiter unter reduziertem Druck konzentriert und der Rückstand in einer Silicagelsäule gereinigt. 2,77 g Zielverbindung wurden mit einer Ausbeute von 54% erhalten. In der Analyse wurde die Summenformel zu C&sub1;&sub3;H&sub1;&sub7;NO festgestellt und die ¹H-NMR-Daten (CDCl&sub3;) sind δ: 1,40-2,40 (7H, m), 2,78 (2H, dt), 3,45 (2H, s), 7,20 (5H, s), 9,51 (1H, d).
- Die Verbindung kann nach den Methoden hergestellt werden, die angegeben sind in (1) Arm. Kim. Zh., 36 (9), 614-17 (1983) von R.A. Kuroyan, A.I. Markosyan, G.M. Snkhchyan und S.A. Vartangan und (2) Ind. Chim. Beige, 32, 64-5 (1967) von B. Hermans und P. Van Daele. 1-Benzyl-4-[(5,6-dimethoxy-1-indanon)-2-ylidenyl]methylpiperidin-hydrochlorid:
- Diese Reaktion wurde in einer Argonatmosphäre ausgeführt.
- 2,05 ml Diisopropylamin wurden zu 10 ml wasserfreiem THF gegeben, gefolgt von Zugabe von 9,12 ml einer 1,6 M Lösung von n-Butyllithium in Hexan bei 0ºC. Die Mischung wurde bei 0ºC 10 Minuten gerührt und dann auf -78ºC gekühlt und eine Lösung von 2,55 g 5,6-Dimethoxy-1-indanon in 30 ml wasserfreiem THF und 2,31 ml Hexamethylphosphorsäureamid zugegeben. Die Mischung wurde bei -78ºC 15 Minuten gerührt und eine Lösung von 2,70 g 1-Benzyl-4- piperidin-carboaldehyd in 30 ml wasserfreiem THF zugegeben. Die Temperatur der Mischung wurde allmählich auf Raumtemperatur erhöht und anschliessend 2 Stunden gerührt. Eine wässrige 1%-ige Ammoniumchloridlösung wurde zugegeben und die organische Phase abgetrennt. Die Wasserphase wurde mit Ethylacetat extrahiert und die organischen Phasen miteinander vereinigt. Die vereinigten organischen Phasen wurden mit gesättigter Kochsalzlösung gewaschen, über Magnesiumsulfat getrocknet und im Vakuum konzentriert. Der resultierende Rückstand wurde mittels einer Silicagelsäule (Methylenchlorid/- Methanol = 500 : 1 bis 100 : 1) gereinigt. Das Eluat wurde im Vakuum konzentriert und der Rückstand in Methylenchlorid gelöst. Eine 10%-ige Chlorwasserstofflösung in Ethylacetat wurde zur resultierenden Lösung gegeben und anschliessend im Vakuum unter Erhalt von Kristallen konzentriert, welche aus Methanol/IPE umkristallisiert wurden, so dass man 3,40 g (Ausbeute 62%) Titelverbindung mit den folgenden Eigenschaften erhielt:

- 0,4 g 1-Benzyl-4-[(5,6-dimethoxy-1-indanon)-2-ylidenyl]methylpiperidin wurden in 16 ml THF gelöst, gefolgt von Zugabe von 0,04 g 10% Palladium-Kohle. Die Mischung wurde bei Raumtemperatur unter Atmosphärendruck 6 Stunden hydriert. Der Katalysator wurde abfiltriert und das Filtrat im Vakuum konzentriert. Der Rückstand wurde unter Verwendung einer Silicagelsäule (Methylenchlorid/Methanol = 50 : 1) gereinigt. Das Eluat wurde im Vakuum konzentriert und der Rückstand mit Methylenchlorid gelöst. Eine 10%-ige Chlorwasserstofflösung in Ethylacetat wurde zur resultierenden Lösung gegeben und anschliessend im Vakuum unter Erhalt von Kristallen konzentriert, welche aus Methanol/IPE umkristallisiert wurden, so dass man 0,36 g (Ausbeute 82%) Titelverbindung mit den folgenden Eigenschaften erhielt:

- 0,85 g 5,6-Dimethoxy-1-indanon und 1,38 g 1-Benzoyl-4-piperidincarbaldehyd wurden in 20 ml wasserfreiem THF unter Erhalt einer Lösung gelöst. 1,02 g 28%-iges Natriummethylat wurden zur Lösung bei 0ºC gegeben. Die erhaltene Mischung wurde bei Raumtemperatur 2 Stunden gerührt, mit Ethylacetat verdünnt, mit einer gesättigten wässrigen Kochsalzlösung gewaschen, über Magnesiumsulfat getrocknet und im Vakuum konzentriert. Der erhaltene Rückstand wurde durch Silicagel-Säulenchromatografie gereinigt, so dass 1,23 g 1-Benzoyl-4-[(5,6-dimethoxy-1-indanon)-2-ylidenyl]methylpiperidin (Ausbeute 71%) erhalten wurden.
- 1,23 g dieser Verbindung wurden in 20 ml THF gelöst, gefolgt von Zugabe von 0,3 g 10% Palladium-Kohle. Nach Ausführung der Hydrierung bei Raumtemperatur unter Normaldruck während eines Tages wurde der Katalysator abfiltriert und das Filtrat im Vakuum konzentriert. Der Rückstand wurde aus Methylenchlorid/Hexan umkristallisiert, wodurch man 1,10 g Titelverbindung (Ausbeute 89%) erhielt. Seine Charakteristika sind wie folgt:

- 9,00 g 1-Benzoyl-4-[(5,6-dimethoxy-1-indanon)-2-yl]methylpiperidin wurden in 90 ml Dioxan gelöst und anschliessend 90 ml 6 N Salzsäure zugegeben. Die erhaltene Mischung wurde unter Rückfluss 10 Stunden erhitzt und im Vakuum konzentriert. Der Rückstand wurde mit Wasser verdünnt und mit Ethylacetat extrahiert. Der pH der wässrigen Schicht wurde mit einer 50%-igen Natronlauge auf 12 eingestellt und mit Methylenchlorid extrahiert. Die organische Schicht wurde mit einer gesättigten wässrigen Kochsalzlösung gewaschen, über Magnesiumsulfat getrocknet und im Vakuum konzentriert. Der erhaltene Rückstand wurde in sein Hydrochlorid nach einem gewöhnlichen Verfahren umgewandelt. Das erhaltene Produkt wurde aus Methanol/Ethanol unter Erhalt von 6,30 g Titelverbindung (Ausbeute 85%) umkristallisiert. Seine Charakteristika sind wie folgt:

- 0,25 g 4-[(5,6-Dimethoxy-1-indanon)-2-yl]-methylpiperidin wurden in 6 ml THF gelöst und anschliessend 0,29 ml Triethylamin und 0,13 ml 3-Fluorbenzylbromid zugegeben. Die erhaltene Mischung wurde unter Rückfluss 2 Stunden erhitzt und im Vakuum konzentriert. Der Rückstand wurde mit Ethylacetat verdünnt, mit einer 10%-igen Natriumcarbonatlösung und einer gesättigten Kochsalzlösung nacheinander gewaschen, über Magnesiumsulfat getrocknet und im Vakuum konzentriert. Der erhaltene Rückstand wurde durch Silicagel-Säulenchromatografie gereinigt und in sein Hydrochlorid nach einem gewöhnlichen Verfahren umgewandelt. Das erhaltene Produkt wurde aus Methylenchlorid/IPE unter Erhalt von 0,27 g Titelverbindung (Ausbeute 72%) umkristallisiert. Seine Charakteristika sind wie folgt:

- 0,50 g 1-Benzyl-4-[(5,6-dimethoxy-1-indanon)-2-yl]methylpiperidin wurden in 8 ml Benzol gelöst und anschliessend 0,15 ml Ethylchloroformiat zugegeben. Die erhaltene Mischung wurde unter Rückfluss 3 Stunden erhitzt, mit Ethylacetat verdünnt, mit einer gesättigten Natriumhydrogencarbonatlösung und einer gesättigten wässrigen Kochsalzlösung nacheinander gewaschen, über Magnesiumsulfat getrocknet und im Vakuum konzentriert. Der erhaltene Rückstand wurde aus Ethylacetat/Hexan umkristallisiert und so 0,45 g Titelverbindung (Ausbeute 94%) erhalten. Seine Charakteristika sind wie folgt:

- 0,24 g 1-Benzyl-4-[(5,6-dimethoxy-1-indanol)-2-yl]methylpiperidin wurden in 5 ml Methylenchlorid gelöst, gefolgt von Zugabe einer 10%-igen Chlorwasserstofflösung in Ethylacetat. Die erhaltene Mischung wurde im Vakuum konzentriert. Der erhaltene Rückstand wurde aus Methylenchlorid/IPE umkristallisiert und so 0,24 g Titelverbindung (Ausbeute 95%) erhalten.

- 0,31 ml Diisopropylamin wurden zu 5 ml wasserfreiem THF gegeben. 1,39 ml einer 1,6 M Lösung von n-Butyllithium in Hexan wurden weiter zur erhaltenen Mischung bei 0ºC zugesetzt. Die erhaltene Mischung wurde bei 0ºC 10 Minuten gerührt und auf -78ºC gekühlt und anschliessend eine Lösung von 0,39 g 5,6-Dimethoxy-1-indanon in 5 ml wasserfreiem THF und 0,35 ml Hexamethylphosphorsäureamid zugegeben. Die erhaltene Mischung wurde bei -78ºC 15 Minuten gerührt, gefolgt von Zugabe einer Lösung von 0,50 g 3-(1-Benzyl-4- piperidin)propionaldehyd in 5 ml wasserfreiem THF. Die erhaltene Mischung wurde allmählich auf Raumtemperatur erwärmt, bei dieser Temperatur 3 Stunden gerührt, mit Ethylacetat verdünnt, mit einer gesättigten wässrigen Kochsalzlösung gewaschen, über Magnesiumsulfat getrocknet und im Vakuum konzentriert. Der erhaltene Rückstand wurde durch Silicagel-Säulenchromatografie gereinigt und in sein Hydrochlorid nach einem gewöhnlichen Verfahren umgewandelt, so dass man 0,55 g Titelverbindung als Öl (Ausbeute 61%) erhielt.

- ¹H-NMR (CDCl&sub3;) δ: 1,10-3,00 (13H, m), 3,45 (2H, s), 3,50 (2H, s), 3,90 (3H, s), 3,95 (3H, s), 6,58-7,20 (3H, m), 7,27 (5H, s) BEISPIEL 24 1-Benzyl-4-[3-[(5,6-dimethoxy-1-indanon)-2-yl]]propylpiperidin-hydrochlorid:
- 0,40 g 1-Benzyl-4-[3-[(5,6-dimethoxy-1-indanon)-2-ylidenyl]]propylpiperidin wurden in 15 ml THF gelöst und anschliessend 0,1 g 10% Palladium-Kohle zugegeben. Nach Ausführung einer Hydrierung bei Raumtemperatur unter Normaldruck während 2 Stunden wurde der Katalysator abfiltriert und das Filtrat im Vakuum konzentriert. Der Rückstand wurde durch Silicagel-Säulenchromatografie gereinigt und in sein Hydrochlorid nach einem gewöhnlichen Verfahren umgewandelt, so dass 0,37 g Titelverbindung als Öl erhalten wurden (Ausbeute 84%).
- Summenformel: C&sub2;&sub6;H&sub3;&sub3;NO&sub3;·HCl
- ¹H-NMR (CDCl&sub3;) δ: 1,00-3,3 (18H, m), 3,38, 3,43 (insgesamt 2H, jeweils s), 3,85 (3H, s), 3,90 (3H, s), 6,77, 6,83 (insgesamt 1H, jeweils s), 7,05, 7,10 (insgesamt 1H, jeweils s), 7,18, 7,20 (insgesamt 5H, jeweils s).

Claims (9)
1. Verfahren zur Herstellung einer cyclischen Aminverbindung der
Formel (I) oder eines pharmakologisch akzeptablen Salzes davon:
worin:
S eine Niederalkylgruppe mit 1 bis 6 Kohlenstoffatomen, eine
Niederalkoxygruppe mit 1 bis 6 Kohlenstoffatomen, ein Halogenatom, eine
Hydroxylgruppe und t 0 bis 4 ist, oder (S)t eine Methylendioxygruppe oder
eine Ethylendioxygruppe an zwei benachbarten Kohlenstoffatomen der
Phenylgruppe, an die (S)t gebunden ist, bilden kann; r eine ganze Zahl von
1 bis 6 ist; und K eine Phenylalkylgruppe, wobei das Phenyl gegebenenfalls
mit einer gegebenenfalls halogenierten C&sub1;&submin;&sub6;-Alkylgruppe substituiert ist,
eine C&sub1;&submin;&sub6;-Alkoxygruppe, eine Nitrogruppe, ein Halogenatom, eine
Carboxylgruppe, eine Benzyloxygruppe, eine C&sub1;&submin;&sub6;-Alkoxycarbonylgruppe, eine
Aminogruppe, eine C&sub1;&submin;&sub6;-Monoalkylaminogruppe, eine C&sub1;&submin;&sub6;-Dialkylaminogruppe,
eine Carbamoylgruppe, eine C&sub1;&submin;&sub6;-Acylaminogruppe, eine
Cyclohexyloxycarbonylgruppe, eine C&sub1;&submin;&sub6;-Alkylaminocarbonylgruppe, eine C&sub1;&submin;&sub6;-
Alkylcarbonyloxygruppe, eine Hydroxylgruppe, eine Formylgruppe oder eine
C&sub1;&submin;&sub6;-Alkoxy-C&sub1;&submin;&sub6;-alkylgruppe ist;
wobei das Verfahren die folgenden Schritte umfasst:
(i) Reduktion eines cyclischen Amins der Formel (II):
worin S, t, r und K wie oben definiert sind; und
(ii) gegebenenfalls Überführung der erhaltenen Verbindung der Formel
(I) in ein pharmakologisch akzeptables Salz.
2. Verfahren gemäss Anspruch 1, wobei die Verbindung der Formel (I):
oder ein pharmakologisch akzeptables Salz davon ist.
3. Verfahren zur Herstellung einer cyclischen Aminverbindung der
Formel (II) oder eines pharmakologisch akzeptablen Salzes davon:
worin S, t, r und K wie in Anspruch 1 definiert sind, das die
folgenden Schritte umfasst:
(i) Umsetzung von
worin S, t, r und K wie oben definiert sind, in einer Wittig-
Reaktion; und
(ii) gegebenenfalls Überführung der erhaltenen Verbindung der Formel
(II) in ein pharmakologisch akzeptables Salz.
4. Verfahren zur Herstellung einer cyclischen Aminverbindung der
Formel (II) oder eines pharmakologisch akzeptablen Salzes davon:
worin S, t, r und K wie in Anspruch 1 definiert sind, das die
folgenden Schritte umfasst:
(i) Umsetzung von
worin S, t, r und K wie oben definiert sind, in Gegenwart von
Lithiumdiisopropylamid; und
(ii) gegebenenfalls Überführung der erhaltenen Verbindung der Formel
(II) in ein pharmakologisch akzeptables Salz.
5. Verfahren zur Herstellung einer cyclischen Aminverbindung der
Formel (III) oder eines pharmakologisch akzeptablen Salzes davon:
worin K wie in Anspruch 1 definiert ist;
ist, worin S und t wie in Anspruch 1 definiert sind; und
B die zweiwertige Gruppe -(CHR²²)r- ist, wobei r eine ganze Zahl von
1 bis 10 ist und R²² jeweils unabhängig entweder ein Wasserstoffatom oder
eine Methylgruppe ist;
wobei das Verfahren die folgenden Schritte umfasst:
(i) Reduktion eines cyclischen Amins der Formel (IV):
worin J und K wie oben definiert sind und B' B entspricht, wobei
jedoch die terminale Gruppe, die ein Kohlenstoffatom enthält, weggelassen
ist; und
(ii) gegebenenfalls Überführung der erhaltenen Verbindung der Formel
(III) in ein pharmakologisch akzeptables Salz.
6. Verfahren gemäss Anspruch 1 oder Anspruch 5, wobei die Reduktion
in Schritt (i) katalytisch durchgeführt wird.
7. Verfahren zur Herstellung einer cyclischen Aminverbindung der
Formel (V) oder eines pharmakologisch akzeptablen Salzes davon:
worin S, t und K wie in Anspruch 1 definiert sind;
B eine der zweiwertigen Gruppen =(CH-CH=CH)b-, in der b eine ganze
Zahl von 1 bis 3 ist; =CH-(CH&sub2;)c-, in der c eine ganze Zahl von 0 bis 9
ist; oder =(CH-CH)d = , in der d eine ganze Zahl von 0 bis 5 ist, bedeutet
und
eine Einfach- oder eine Doppelbindung bezeichnet, wobei das
Verfahren die folgenden Schritte umfasst:
(i) Umsetzung von
worin S, t und K wie oben definiert sind und B' B entspricht, wobei
jedoch die terminale Gruppe, die ein Kohlenstoffatom enthält, weggelassen
ist, in einer Wittig-Reaktion; und
(ii) gegebenenfalls Überführung der erhaltenen Verbindung der Formel
(V) in ein pharmakologisch akzeptables Salz.
8. Verfahren zur Herstellung einer cyclischen Aminverbindung der
Formel (V) oder eines pharmakologisch akzeptablen Salzes davon:
worin S, t und K wie in Anspruch 1 definiert sind;
B eine der zweiwertigen Gruppen =(CH-CH=CH)b-, in der b eine ganze
Zahl von 1 bis 3 ist; =CH-(CH&sub2;)c-, in der c eine ganze Zahl von 0 bis 9
ist; oder =(CH-CH)d= , in der d eine ganze Zahl von 0 bis 5 ist, bedeutet
und
eine Einfach- oder eine Doppelbindung bezeichnet, wobei das
Verfahren die folgenden Schritte umfasst:
(i) Umsetzung von
worin S, t und K wie oben definiert sind und B' B entspricht, wobei
jedoch die terminale Gruppe, die ein Kohlenstoffatom enthält, weggelassen
ist; und
(ii) gegebenenfalls Überführung der erhaltenen Verbindung der Formel
(V) in ein pharmakologisch akzeptables Salz.
9. Verfahren zur Herstellung einer cyclischen Aminverbindung der
Formel (VI) oder eines pharmakologisch akzeptablen Salzes davon:
worin S, t und K wie in Anspruch 1 definiert sind; und r eine ganze
Zahl von 0 bis 10 ist und R²² jeweils unabhängig ein Wasserstoffatom oder
eine Methylgruppe ist;
wobei das Verfahren die folgenden Schritte umfasst:
(i) Dehydratation einer Indanolverbindung der Formel:
worin S, t, r und K wie oben definiert sind; und
(ii) gegebenenfalls Überführung der erhaltenen Verbindung der Formel
(VI) in ein pharmakologisch akzeptables Salz.
