Description
- Die vorliegende Erfindung betrifft neue und bekannte alpha-Aminoamidverbindungen, Verfahren zu ihrer Herstellung, sie enthaltende pharmazeutische Zubereitungen und ihre Verwendung als therapeutische Mittel.
- Insbesondere besitzen die erfindungsgemäßen Verbindungen analgetische Eigenschaften und sind besonders nützlich für die Behandlung und Linderung chronischer und neuropathischer Schmerzen.
- Chronischer und neuropathischer Schmerz ist assoziiert mit einer längeren Gewebeschädigung oder Verletzungen des peripheren oder zentralen Nervensystems und beruht auf einer Reihe komplexer Änderungen in den Schmerzleitungswegen.
- Klinische Manifestationen des chronischen Schmerzes umfassen eine Empfindung des Brennens oder des elektrischen Schocks, das Gefühl einer körperlichen Verzerrung, Allodynie und Hyperpathie.
- Trotz einer großen Zahl verfügbarer Analgetika ist ihre Verwendung durch ernste Nebenwirkungen und mäßige Aktivität bei einigen Schmerzzuständen beschränkt. Es besteht daher noch ein eindeutiger Bedarf für die Entwicklung neuer Verbindungen.
- In den internationalen Anmeldungen WO 90/14334, WO 94/22808, WO 97/05102 und WO 97/05111 werden substituierte Benzylaminopropionamidverbindungen beschrieben, die gegenüber dem zentralen Nervensystem Aktivität zeigen und die als antiepileptische, Anti- Parkinson-Mittel, Neuroprotektiva, Antidepressiva, antispastische Mittel und hypnotische Mittel nützlich sind.
- Die vorliegende Erfindung beruht auf der Erkenntnis, dass Verbindungen, die aus den oben erwähnten internationalen Anmeldungen bekannt sind, und neue, die sehr eng mit diesen verwandt sind, analgetische Eigenschaften in Säugetieren einschließlich Menschen zeigen.
- Dementsprechend ist es Aufgabe der vorliegenden Erfindung, die Verwendung einer Verbindung der Formel (I)
- worin:
- A eine -(CH&sub2;)m-, -(CH&sub2;)n-X- oder -(CH&sub2;)v- O-Gruppe bedeutet, worin m eine ganze Zahl von 1 bis 4 bedeutet, n Null oder eine ganze Zahl von 1 bis 4 bedeutet, X -S- oder -NH- bedeutet und v Null oder eine ganze Zahl von 1 bis 5 bedeutet;
- s 1 oder 2 bedeutet;
- R einen Furyl-, Thienyl- oder Pyridylring oder einen Phenylring, gegebenenfalls substituiert durch ein oder zwei Substituenten, unabhängig ausgewählt aus Halogen, Cyano, C&sub1;-C&sub4;-Alkyl, C&sub1;-C&sub4;-Alkoxy und Trifluormethyl, bedeutet;
- R&sub1; Wasserstoff oder C&sub1;-C&sub4;-Alkyl bedeutet;
- wobei einer von R&sub2; und R&sub3; Wasserstoff und der andere Wasserstoff oder C&sub1;-C&sub4;-Alkyl, gegebenenfalls substituiert durch Hydroxy oder Phenyl, bedeutet; oder worin R&sub2; und R&sub3; zusammen mit dem Kohlenstoffatom, an das sie gebunden sind, einen C&sub3;-C&sub6;- Cycloalkylring bilden; oder worin R&sub2; und R&sub3; beide Methyl bedeuten;
- R&sub4; Wasserstoff oder C&sub1;-C&sub4;-Alkyl bedeutet;
- oder der pharmazeutisch annehmbaren Salze davon;
- und wobei, wenn
- A eine -(CH&sub2;)&sub5;-O-Gruppe bedeutet, dann s 1 bedeutet, R eine Phenylgruppe, gegebenenfalls substituiert durch einen oder zwei Substituenten, ausgewählt unabhängig aus Halogen, Trifluormethyl und C&sub1;-C&sub4;-Alkoxy, bedeutet, R&sub1; Wasserstoff bedeutet und einer von R&sub2; und R&sub3; Wasserstoff und der andere Wasserstoff oder C&sub1;-C&sub4;-Alkyl, gegebenenfalls substituiert mit Hydroxy, bedeutet;
- und wobei,
- wenn R&sub2; und R&sub3; beide Methyl bedeuten, dann R eine andere Bedeutung als ein Furyl-, Thienyl- oder Pyridylring besitzt, zur Herstellung eines Arzneimittels für die Verwendung als Analgetikum, insbesondere für die Behandlung und Linderung von chronischem und neuropathischem Schmerz zur Verfügung zu stellen.
- Die -(CH&sub2;)m-, -(CH&sub2;)n-X- oder -(CH&sub2;)v-Kette kann eine verzweigte oder gerade Kette sein.
- Die Alkyl- und Alkoxygruppen können verzweigte oder geradkettige Gruppen sein. Repräsentative Beispiele von C&sub1;-C&sub4;-Alkylgruppen umfassen Methyl, Ethyl, n- und Isopropyl, n-, Iso-, sek.- und tert.-Butyl.
- Repräsentative Beispiele von C&sub1;-C&sub4;-Alkoxygruppen umfassen Methoxy und Ethoxy.
- Eine C&sub3;-C&sub6;-Cycloalkylgruppe ist beispielsweise Cyclopropyl, Cyclopentyl oder Cyclohexyl, insbesondere Cyclopentyl oder Cyclohexyl.
- Ein Halogenatom ist ein Fluor-, Brom-, Chlor- oder Iodatom, insbesondere ein Chlor- oder Fluoratom.
- Die pharmazeutisch annehmbaren Salze der erfindungsgemäßen Verbindungen umfassen Säureadditionssalze mit anorganischen Säuren, beispielsweise Salpetersäure, Chlorwasserstoffsäure, Bromwasserstoffsäure, Schwefelsäure, Perchlorsäure und Phosphorsäure oder mit organischen Säuren, beispielsweise Essigsäure, Trifluoressigsäure, Propionsäure, Glykolsäure, Milchsäure, Oxalsäure, Malonsäure, Äpfelsäure, Maleinsäure, Weinsäure, Citronensäure, Benzoesäure, Zimtsäure, Mandelsäure und Salicylsäure.
- Die Verbindungen der Formel (I) besitzen asymmetrische Kohlenstoffatome und sie können daher entweder als racemische Gemische oder als individuelle optische Isomere (Enantiomere) vorliegen.
- Gegenstand der Erfindung sind ebenfalls alle möglichen Isomere und ihre Gemische, und beides, die Metaboliten und die pharmazeutisch annehmbaren Biovorstufen (anderweitig als Prodrugs bekannt) der Verbindungen der Formel (I).
- Bevorzugte Verbindungen der Formel (I) sind solche, worin
- A eine Gruppe bedeutet, ausgewählt aus -CH&sub2;-, -CH&sub2;-CH&sub2;-, -CH&sub2;-S-, -CH&sub2;-CH&sub2;-S- und -(CH&sub2;)v-O-, worin v eine ganze Zahl von 1 bis 5 bedeutet;
- s 1 oder 2 bedeutet;
- R einen Phenylring, der gegebenenfalls substituiert ist mit ein oder zwei Substituenten, unabhängig ausgewählt aus Halogen, Cyano, oder einen Thienylring bedeutet;
- R&sub1; Wasserstoff oder C&sub1;-C&sub4;-Alkyl bedeutet;
- einer von R&sub2; und R&sub3; Wasserstoff und der andere C&sub1;-C&sub4;-Alkyl bedeutet, gegebenenfalls substituiert durch Hydroxy oder Phenyl; oder worin R&sub2; und R&sub3; beide Methyl bedeuten; und
- R&sub4; Wasserstoff oder C&sub1;-C&sub4;-Alkyl bedeutet; und die pharmazeutisch annehmbaren Salze davon.

- Die Erfindung betrifft pharmazeutische Zubereitungen mit analgetischer Aktivität, insbesondere gegen chronischen und neuropathischen Schmerz, umfassend eine Verbindung der Formel (I), wie zuvor definiert, als aktives Mittel und ein pharmazeutisch annehmbares Salz davon.
- Die Erfindung betrifft weiterhin ein Verfahren zur Behandlung eines Säugetiers einschließlich von Menschen, die ein analgetisches Mittel benötigen, wobei das Verfahren umfasst die Verabreichung einer wirksamen Menge einer Verbindung der Formel (I) oder eines pharmazeutisch annehmbaren Salzes davon.
- Neuropathische und chronische Schmerzzustände bei Säugetieren können somit erleichtert und behandelt werden. Beispiele von Schmerzzuständen, die mittels einer Verbindung der Formel (I) behandelt werden können, umfassen:
- - periphere Neuropathien, wie trigeminale Neuralgie, posttherapeutische Neuralgie, diabetische Neuropathie, glossopharyngeale Neuralgie, Radiculopathie und Neuropathie, sekundär zu metastatischer Infiltration, Adiposis dolorosa und Verbrennungsschmerz; und
- - zentrale Schmerzzustände nach Schlaganfall, thalamische Läsionen und mutiple Sklerose.
- Der Ausdruck "Behandlung", wie er hier verwendet wird, beinhaltet irgendeine Behandlung eines Krankheitsbildes bei einem Säugetier, insbesondere bei Menschen, und umfasst:
- (i) die Prophylaxe einer Krankheit, die in einem Subjekt auftritt, das Veranlagung für die Krankheit besitzt, bei dem diese jedoch noch nicht diagnostiziert wurde;
- (ii) die Inhibierung eines Krankheitsbildes, beispielsweise Aufhalten seiner Entwicklung; oder
- (iii) die Linderung des Krankheitsbildes, beispielsweise die Verursachung der Regression der Krankheit.
- Die Erfindung betrifft weiter neue Verbindungen der Formel (IA):
- worin:
- A eine -(CH&sub2;)m- oder -(CH&sub2;)n-E-Gruppe bedeutet, worin m eine ganze Zahl von 1 bis 4 bedeutet, n Null oder eine ganze Zahl von 1 bis 4 bedeutet und E -O-, -S- oder -NH- bedeutet;
- s 1 oder 2 bedeutet;
- einer von R&sub1;&sub0; und R&sub1;&sub1; Cyano bedeutet und der andere unabhängig ausgewählt ist aus Wasserstoff, Halogen, Cyano, C&sub1;-C&sub4;-Alkyl, C&sub1;-C&sub4;-Alkoxy und Trifluormethyl;
- R&sub1; Wasserstoff oder C&sub1;-C&sub4;-Alkyl bedeutet;

- Die Verbindungen der Formel (IA) fallen unter den Rahmen der Verbindung der Formel (I), wie zuvor definiert. Daher gelten alle Definitionen und biologischen Eigenschaften, die oben für die Verbindung der Formel (I) angegeben wurden, ebenfalls für Verbindungen der Formel (IA).
- Insbesondere sind bevorzugte Verbindungen der Formel (IA) solche, worin
- A die Gruppe -CH&sub2;-O- oder -CH&sub2;-CH&sub2;-O- bedeutet,
- s 1 bedeutet;
- einer von R&sub1;&sub0; und R&sub1;&sub1; Cyano und der andere Wasserstoff, Cyano oder Halogen bedeutet; und einer von R&sub2; und R&sub3; Wasserstoff und der andere C&sub1;-C&sub4;-Alkyl, gegebenenfalls substituiert durch
- Hydroxy, bedeutet; oder worin R&sub2; und R&sub3; beide Methyl bedeuten, und die pharmazeutisch annehmbaren Salze davon.
- Spezifische Beispiele von Verbindungen der Formel (IA) sind:
- 2-[4-(3-Cyanobenzyloxy)benzylamino]-3-hydroxy-N-methylpropanamid;
- 2-[N-(4-(3-Cyanobenzyloxy)benzyl)-N-methylamino]-3-hydroxy-N-methylpropanamid, entweder als einfaches Isomeres oder als Gemisch davon und die pharmazeutisch annehmbaren Salze davon.
- Die Verbindungen der Formel (I) und (IA) und die pharmazeutisch annehmbaren Salze davon können nach gut bekannten Verfahren, wie in den oben erwähten internationalen Anmeldungen beschrieben, hergestellt werden. Insbesondere kann eine Verbindung der Formel (IA) und die Salze davon gemäß einem Verfahren erhalten werden, das umfasst:
- a) Umsetzung einer Verbindung der Formel (II):
- worin R&sub1;&sub0;, R&sub1;&sub1;, A und s die oben gegebenen Definitionen besitzen, mit einer Verbindung der Formel (III)
- worin R&sub2;, R&sub3; und R&sub4; die oben gegebenen Definitionen besitzen, in Anwesenheit eines Reduktionsmittels, wobei eine Verbindung der Formel (IA) erhalten wird, worin R&sub1; Wasserstoff bedeutet; oder
- b) Umsetzung einer Verbindung der Formel (IV):
- worin R&sub2;, R&sub3;, R&sub4;, R&sub1;&sub0;, R&sub1;&sub1;, A und s die oben gegebenen Definitionen besitzen, mit einer Verbindung der Formel (V) oder (VI), im letzteren Fall in Anwesenheit eines Reduktionsmittels,
- R'&sub5;W (V) R"&sub5;CHO (VI)
- worin W ein Halogenatom bedeutet, R'&sub5; C&sub1;-C&sub4;-Alkyl bedeutet und R"&sub5; Wasserstoff oder C&sub1;-C&sub3;- Alkyl bedeutet, wobei eine Verbindung der Formel (IA) erhalten wird, worin R&sub1; C&sub1;-C&sub4;-Alkyl bedeutet; und gewünschtenfalls Umwandlung einer Verbindung der Formel (IA) in eine andere Verbindung der Formel (IA) und/oder gewünschtenfalls Umwandlung einer Verbindung der Formel (IA) in ein pharmazeutisch annehmbares Salz und/oder gewünschtenfalls Umwandlung des Salzes in eine freie Verbindung.
- Alle die oben beschriebenen Verfahren sind Analogieverfahren und können nach an sich gut bekannten Verfahren der organischen Chemie durchgeführt werden.
- Eine Verbindung der Formel (IV) ist eine Verbindung der Formel (JA), worin R&sub1; Wasserstoff bedeutet.
- Die Umsetzung einer Verbindung der Formel (II) mit einer Verbindung der Formel (III) unter Bildung einer Verbindung der Formel (IA) oder (IV) ist eine reduktive Aminierungsreaktion, die nach gut bekannten Verfahren durchgeführt werden kann. Gemäß einer bevorzugten erfindungsgemäßen Ausführungsform kann sie unter Stickstoffatmosphäre in einem geeigneten organischen Lösungsmittel, wie einem Alkohol, beispielsweise einem niedrigen Alkohol, insbesondere Methanol, oder in Acetonitril bei einer Temperatur im Bereich von 0ºC bis etwa 40ºC in Anwesenheit eines Reduktionsmittels durchgeführt werden, wobei das geeignetste Natriumcyanoborhydrid ist.
- Gelegentlich können Molekularsiebe zu dem Reaktionsgemisch zur Erleichterung der Reaktion gegeben werden.
- In einer Verbindung der Formel (IV) ist das Halogenatom W bevorzugt Iod. Die Alkylierungsreaktion einer Verbindung der Formel (IV) mit einer Verbindung der Formel (V) kann in einem geeigneten organischen Lösungsmittel, wie einem Alkohol, beispielsweise Methanol, Ethanol oder Isopropanol, insbesondere in Ethanol, bei einer Temperatur im Bereich von 0ºC bis etwa 50ºC durchgeführt werden.
- Die Alkylierungsreaktion einer Verbindung der Formel (IV) mit einem Aldehyd der Formel (VI) kann in einem geeigneten organischen Lösungsmittel, wie einem Alkohol, beispielsweise Methanol, Ethanol oder Acetonitril, in Anwesenheit eines geeigneten Reduktionsmittels, wie Natriumcyanoborhydrid, bei einer Temperatur im Bereich von etwa 0ºC bis etwa 30ºC durchgeführt werden.
- Eine Verbindung der Formel (IA) kann, wie oben angegeben, in eine andere Verbindung der Formel (IA) nach gekannten Verfahren überführt werden. Die obige Verfahrensvariante b) kann als Beispiel einer möglichen Umwandlung einer Verbindung der Formel (IA) in eine andere Verbindung der Formel (IA) angesehen werden.
- Die mögliche Salzbildung einer Verbindung der Formel (IA), wie auch die Umwandlung eines Salzes in die freie Verbindung kann nach bekannten Verfahren erfolgen.
- Die Verbindungen der Formeln (II) und (III), (V) und (VI) sind bekannte Verbindungen oder können nach bekannten Verfahren erhalten werden.
- Wenn in den erfindungsgemäßen Verbindungen und in den Zwischenprodukten davon Gruppen vorhanden sind, die geschützt werden müssen, bevor sie den hier oben erläuterten Reaktionen unterworfen werden, können sie vor der Umsetzung geschützt werden und dann kann die Schutzgruppe nach an sich bekannten Verfahren in der organischen Chemie wieder abgespalten werden.
- Die Verbindungen der Formel (I), (IA) und die pharmazeutisch annehmbaren Salze davon werden im Folgenden als "erfindungsgemäße Verbindungen" oder als "aktive erfindungsgemäße Mittel" bezeichnet.
- Wie oben angegeben, sind die erfindungsgemäßen Verbindungen als analgetische Mittel wirksam, was beispielsweise durch die Tatsache bewiesen wird, dass sie bei dem Formalin-Test aktiv sind.
- Der Formalin-Test ist ein nützliches Werkzeug der Erzeugung einer neurogenen Entzündung und kontinuierlicher Schmerzen (Shibata et al., Pain, 38: 347-352, 1989).
- Formalin erzeugt eine genaue zweiphasische Antwort. Die frühe Phase scheint überwiegend durch C-Faser-Aktivierung, bedingt durch peripheren Stimulus, verursacht zu werden, während die letzte Phase von der Kombination einer inflammatorischen Reaktion in dem peripheren Gewebe und funktionellen Änderungen im Hinterhorn des Rückenmarks abhängt. Diese funktionellen Änderungen scheinen durch die C-Faser-Sperre während der frühen Phase initiiert zu werden (Tjolsen et al., Pain 51, 5-17, 1992). Die Substanz P und das Bradykinin sind in der frühen Phase, während Histamin, Serotonin, Prostaglandine und Bradykinin in der späten Phase involviert sind.
- Männlichen NMRI-Mäusen (22 bis 25 g) wurden 20 ml einer 2,7%igen Lösung von Formalin in die rechte Hinterpfote injiziert und dann wurden sie umgehend in Beobachtungskammern gesetzt. Die kumulativen Leck-Zahlen der injizierten Pfote wurden in der akuten Phase (0 bis 5 min) und in der chronischen Phase (30 bis 40 min) der Schmerzreaktion auf Formalin aufgezeichnet.
- Zwei repräsentative erfindungsgemäße Verbindungen (S)-2-[4-(3-Fluorbenzyloxy)- benzylamino]propanamid, Methanolsulfonat (innere Bezeichnung PNU 151774E) und (S)-2-[4- (3-Fluorbenzyloxy)benzylamino]-2-methylpropanamid (innere Bezeichnung PNU 156654E) wurden 60 min vor der Formalin-Injektion in Dosismengen von 7,5, 15,0, 30,0 und 60,0 mg/kg verabreicht; po. Morphin (5 mg/kg; sc) wurde als positiver Standard verwendet. Die Aktivitätswerte wurden gemäß dem Dunnett's-t-Test analysiert.
- Die Wirkungen dieser Verbindungen auf die lokomotorische Aktivität und Rotarod (ein Test zur Bewertung der motorischen Koordination) wurden untersucht um Änderungen in diesen Parametern als verwechselnde Faktoren bei der Bewertung des Formalin-Ansprechens auszuschließen. Der lokomotorische Aktivitätstest dauerte 15 min. Fünf Minuten nach dem Testen der lokomotorischen Aktivität wurden die Mäuse auf den Rotarod während 2 min gegeben und die Zahl der Mäuse, die innerhalb dieser Zeit abfielen, wurde gezählt.
- Die Verbindungen PNU 151774E und PNU 156654E wurden in Dosismengen von 7,5, 15,0, 30,0 und 60,0 mg/kg getestet; po. Die Verbindungen wurden 60 min vor dem lokomotorischen Aktivitätstest verabreicht.
- Die Verbindungen PNU 151774E und PNU 156654E verringern dosisabhängig die kumulative Zeit in beiden Phasen des Formalin-Tests (Tabelle 1) und demonstrieren die analgetische Aktivität ohne Wirkung auf die lokomotorische oder Rotarod-Aktivität (Tabelle 2). Tabelle 1

- Aufgrund ihrer biologischen Aktivität sind die erfindungsgemäßen Verbindungen bei Säugetieren einschließlich Menschen als analgetische Mittel nützlich. Insbesondere sind sie nützlich zur Behandlung von Schmerz, der mit Schäden oder permanenten Änderungen des peripheren oder zentralen Nervensystems einhergeht, beispielsweise periphere Neuropathien, wie trigeminale Neuralgie, posttherapeutische Neuralgie, diabetische Neuropathie, Radikulopathie, glossopharyngeale Neuralgie und Neuropathie, sekundär für metastatische Infiltration, Adiposis dolorosa und Verbrennungsschmerz; und zentrale Schmerzbedingungen, folgend auf Schlaganfall, thalmische Läsionen und multiple Sklerose.
- Der Zustand eines Patienten, der eines analgetischen Mittels bedarf, können so verbessert werden.
- Die erfindungsgemäßen Verbindungen können in einer Vielzahl von Dosisformen, beispielsweise oral, in Form von Tabletten, Kapseln, mit Zucker oder mit Film beschichteten Tabletten, flüssigen Lösungen oder Suspensionen; rektal in Form von Suppositorien; parenteral, beispielsweise intramuskulär oder durch intravenöse Injektion oder Infusion verabreicht werden.
- Die Dosis hängt vorn Alter, Gewicht, dem Zustand des Patienten und von dem Verabreichungsweg ab; beispielsweise kann die Dosis, die zur oralen Verabreichung bei erwachsenen Menschen angepasst ist, beispielsweise für repräsentative erfindungsgemäße Verbindungen
- (S)-2-[4-(3-(Fluorbenzyloxy)benzylamino]propanamin,
- Methansulfonat,
- (S)-2-[4-(3-(Fluorbenzyloxy)benzylamino]-2-methylpropanamid,
- (S)-[2-[4-(3-Cyanobenzyloxy)benzylamino]-3-hydroxy-N-methylpropanamid, im Bereich von etwa 1 bis etwa 500 mg pro Dosis, 1- bis 5mal täglich liegen.
- Die Erfindung betrifft pharmazeutische Zubereitungen, die eine Verbindung der Formel (IA) als Wirkstoff in Kombination mit einem pharmazeutisch annehmbaren Exzipiens, welches ein Träger oder ein Verdünnungsmittel sein kann, enthalten. Die pharmazeutischen Zubereitungen, die die erfindungsgemäßen Verbindungen enthalten, werden üblicherweise nach bekannten Verfahren hergestellt und werden in einer pharmazeutisch geeigneten Form verabreicht.
- Beispielsweise können die festen oralen Formen zusammen mit der aktiven Verbindung Verdünnungsmittel, beispielsweise Lactose, Dextrose, Saccharose, Cellulose, Maisstärke oder Kartoffelstärke; Gleitmittel, beispielsweise Silica, Talk, Stearinsäure, Magnesium- oder Calciumstearat, und/oder Polyethylenglykole; Bindemittel, beispielsweise Stärken, Gummi arabicum, Gelatine, Methylcellulose, Carboxymethylcellulose oder Polyvinylpyrrolidon; Zerfallsmittel, beispielsweise Stärke, Alginsäure, Alginate oder Natriumstärkeglycolat; Brausegemische; Farbstoffe; Süßstoffe; Benetzungsmittel, wie Lecithin, Polysorbate, Laurylsulfate; und im Allgemeinen nichttoxische und pharmakologisch inaktive Substanzen, die in pharmazeutischen Zubereitungen verwendet werden, enthalten. Die pharmazeutischen Präparate können in an sich bekannter Weise, beispielsweise durch Misch-, Granulier-, Tablettierungs-, Zuckerbeschichtungs- oder Filmbeschichtungs-Verfahren hergestellt werden.
- Die flüssigen Dispersionen für die orale Verabreichung können beispielsweise in Form von Sirupen, Emulsionen und Suspensionen vorliegen.
- Die Sirupe können als Träger beispielsweise Saccharose oder Saccharose mit Glycerin und/oder Mannit und/oder Sorbit enthalten.
- Die Suspension und die Emulsion kann als Träger beispielsweise Naturgummi, Agar, Natriumalginat, Pectin, Methylcellulose, Carboxymethylcellulose oder Polyvinylalkohol enthalten.
- Die Suspension oder Lösung für intramuskulare Injektionen kann zusammen mit der aktiven Verbindung einen pharmazeutisch annehmbaren Träger, beispielsweise steriles Wasser, Olivenöl, Ethyloleat, Glykole, beispielsweise Propylenglykol, und gegebenenfalls eine geeignete Menge an Lidocainhydrochlorid enthalten. Die Lösungen für intravenöse Injektionen oder Infusion kann als Träger beispielsweise steriles Wasser enthalten oder bevorzugt können sie in Form steriler, wässrigen, isotoner Salzlösungen vorliegen.
- Die Suppositorien können zusammen mit der aktiven Verbindung einen pharmazeutisch annehmbaren Träger, beispielsweise Kakaobutter, Polyethylenglykol, ein Polyoxyethylensorbitanfettsäureester-grenzflächenaktives Mittel oder Lecithin enthalten.
- Die folgenden Beispiele erläutern die Erfindung, ohne sie zu beschränken.
- Zu einer Lösung von N-Methylserinamidhydrochlorid (2 g; 0,0129 mol) in Methanol (40 ml) werden 2 g gepulverte 3A-Molekularsiebe zugegeben; nach dem Rühren während 15 Minuten bei Raumtemperatur werden 0,65 g (0,0102 mol) Natriumcyanoborhydrid in einer einzigen Portion, gefolgt von 2,85 g (0,012 mol) 4-(3-Cyanobenzyloxy)benzaldehyd gegeben. Das Gemisch wird 2 Stunden bei Raumtemperatur gerührt, dann filtriert, und der Rückstand wird nach Verdampfen durch Flashchromatographie an Silicagel (Eluierungsmittel: Chloroform 98 : Methanol 2 : 30% NH&sub4;OH 0,2) getrennt. Es werden 2,6 g (63%) reine Titelverbindung erhalten (Fp. 130 bis 134ºC).
- [α]²&sup0;D: +12,8º (c = 1,25 AcOH)
- (S) -2-[4-(3-Cyanobenzyloxy)benzylamino]-3-hydroxy-N-methylpropanamid (2 g; 0,0059 mol) werden in Methanol (30 ml) gelöst, und 1,8 g (0,013 mol) wasserfreies Kaliumcarbonat werden zu der Lösung gegeben. Methyiodid (1,5 ml; 0,025 mol) wird in das Gemisch, welches 2 Stunden bei Raumtemperatur gerührt wird, eingetropft und dann wird zur Trockene eingedampft. Der rohe Rückstand wird an Silicagel (Eluierungsmittel : Chloroform/Methanol; 95/5) chromatographiert. Es werden 1,88 g (90%) (S)-2-[N-(4-(3-Cyanobenzyloxy)benzyl)-N- methylamino]-3-hydroxy-N-methylpropanamid erhalten. Elementaranalyse:
- Unter Verwendung üblicher Verfahren zur Herstellung pharmazeutischer Zubereitungen können Kapseln der folgenden Zusammensetzung hergestellt werden:

1. Verwendung zur Herstellung eines Arzneimittels für
die Verwendung als Analgetikum von Verbindungen, die
alpha-Aminoamide der Formel (I)
sind, worin:
A eine -(CH&sub2;)m-, -(CH&sub2;)n-X- oder -(CH&sub2;)v-O-Gruppe
bedeutet, worin m eine ganze Zahl von 1 bis 4
bedeutet, n Null oder eine ganze Zahl von 1 bis 4
bedeutet, X -SO- oder -NH- bedeutet und v Null oder eine
ganze Zahl von 1 bis 5 bedeutet;
s 1 oder 2 bedeutet;
R einen Furyl-, Thienyl- oder Pyridylring oder einen
Phenylring, der gegebenenfalls substituiert ist
durch ein oder zwei Substituenten, unabhängig
ausgewählt aus Halogen, Cyano, C&sub1;-C&sub4;-Alkyl, C&sub1;-C&sub4;-Alkoxy
und Trifluormethyl, bedeutet;

2. Verwendung nach Anspruch 1, wobei das Arzneimittel
für die Behandlung oder Linderung von chronischen oder
neuropathischen Schmerzen verwendet wird.
3. Verwendung nach Ansprüch 1, wobei in der Formel (I)
A eine Gruppe, ausgewählt aus -CH&sub2;-, -CH&sub2;-CH&sub2;-,
-CH&sub2;-S-, -CH&sub2;-CH&sub2;-S- und -(CH&sub2;)v-O- bedeutet, worin v
eine ganze Zahl von 1 bis 5 bedeutet;
s 1 oder 2 bedeutet;
R einen Phenylring bedeutet, gegebenenfalls
substituiert durch einen oder zwei Substituenten,
unabhängig ausgewählt aus Halogen, Cyano oder einem
Thienylring;
R&sub1; Wasserstoff oder C&sub1;-C&sub4;-Alkyl bedeutet;
einer von R&sub2; und R&sub3; Wasserstoff und der andere C&sub1;-C&sub4;-Alkyl

oder ein pharmazeutisch annehmbares Salz davon.
6. Verbindung nach Anspruch 5, worin
A die Gruppe -CH&sub2;-O- oder -CH&sub2;-CH&sub2;-O- bedeutet,
s 1 bedeutet;
einer von R&sub1;&sub0; und R&sub1;&sub1; Cyano und der andere Wasserstoff,
Cyano oder Halogen bedeutet; und
einer von R&sub2; und R&sub3; Wasserstoff bedeutet und der andere C&sub1;-
C&sub4;-Alkyl, gegebenenfalls substituiert durch Hydroxy,
bedeutet; oder worin R&sub2; und R&sub3; beide Methyl
bedeuten.
7. Verbindung, ausgewählt aus:
2-[4-(3-Cyanobenzyloxy)benzylamino]-3-hydroxy-N-
methylpropanamid; und
2-[N-(4-(3-Cyanobenzyloxy)benzyl)-N-methylamino]-3-
hydroxy-N-methylpropanamid
als einfaches Isomeres oder als
Gemisch davon und die pharmazeutisch annehmbaren Salze.
8. Pharmazeutische Zubereitung, umfassend ein
pharmazeutisch annehmbares Exzipiens und als ein aktives Mittel
eine Verbindung nach Anspruch 5.
9. Verbindung nach Anspruch 5 für die Verwendung in
einem Verfahren zur Behandlung des menschlichen oder
tierischen Körpers durch Therapie.
10. Verbindung nach Anspruch 9 für die Verwendung als
Analgetikum.
