Den foreliggende opfindelse angår en analogifremgangsmåde til fremstilling af naphthalenderivater, som er nyttige ved inhibering af visse dermatologiske tilstande. Den foreliggende opfindelse muliggør farmaceutiske præparater, der er nyttige ved lindring af virkningerne 5 af visse kroniske papillosquamøse dermatoser, fx psoriasis.
Psoriasis er en hudsygdom, der delvis er karakteriseret ved overdreven formering af celler i epidermis, der forbliver stærkt vedhængende og bygges op i et skællet udslet, der er typisk for sygdommen. De for tiden tilgængelige terapier, der ikke er helbredende, afhænger af 10 reguleringen af epidermal celleformering gennem anvendelse af hormonmidler såsom corticosteroider eller gennem anvendelse af forbindelser, der har relation til cancerkemoterapi såsom hydroxyurea, methotrexat og nitrogensennepsforbindelserne.
Selv om de ovennævnte midler til en vis grad er virksomme, forårsager 15 de talrige alvorlige uønskede bivirkninger, deriblandt renal irritation, levertoxicitet og erythema.
Forbindelsen 2,3-dimethoxy-l,4-diacetyloxynaphthalen er kendt og er beskrevet som værende et nyttigt syntetisk mellemprodukt, men forbindelsen har ikke været tilskrevet nogen nyttig biologisk virkning.

Jfr. fx US patentskrift nr. 4.229.478 og britisk patentskrift nr. 1.243.401. Disse forbindelser har imidlertid én eller flere ulemper såsom forårsagelse af hudirritation, farvning 25 af huden og sensitivisering af patienten. Det har overraskende vist sig, at forbindelserne fremstillet ved fremgangsmåden ifølge den foreliggende opfindelse er mindre irriterende, ikke farver huden og ikke sensitiviserer, når de anvendes ved behandling af psoriasis.
Endvidere er forbindelserne fremstillet ved fremgangsmåden ifølge 30 opfindelsen mere stabile i de normalt anvendte topiske formuleringer.

Inden for denne specifikke udførelsesform af opfindelsen er en fore-trukken gruppe forbindelser med den almene formel I sådanne, i hvilke R^ og R^ er lavere alkoxy med 1-3 carbonatomer, og W er lavere alkyl 15 med 1-5 carbonatomer.
I den foreliggende beskrivelse og krav betegner udtrykket "alkyl med 1-7 carbonatomer" ligekædede grupper eller forgrenede grupper. Eksempler på sådanne grupper er methyl, ethyl, n-propyl, i-propyl, n-hexyl, 2-methylpentyl og n-heptyl. Udtrykket "lavere alkylthio med 1-6 carbonatomer" betegner en lige-kædet eller forgrenet aliphatisk gruppe med 1-6 carbonatomer, på 5 hvilken der er bundet en svovldel. Eksempler på sådanne grupper er methylthio, ethylthio, n-propylthio, i-butylthio og n-hexylthio.
Udtrykket "halogen” betegner fluor, chlor og brom. Udtrykket "cyano" betegner gruppen -CN.
Med udtrykket "farmaceutisk acceptable syreadditionssalte" menes i 10 det foreliggende tilfælde de ikke-toxiske farmaceutisk acceptable syreadditionssalte, der ikke på ugunstig måde påvirker de farmaceutiske egenskaber af de oprindelige forbindelser. Med hensyn til sådanne additionssalte omfatter egnede uorganiske anioner fx chlorid, bromid, iodid, sulfat, phosphat, nitrat og lignende. Egnede organiske anioner 15 omfatter fx acetat, benzoat, lactat, picrat, propionat, butyrat, valerat, tartrat, maleat, fumarat, citrat, succinat, tosylat, ascor-bat, nicotinat, adipat, gluconat og lignende.
Forbindelserne fremstillet ved fremgangsmåden ifølge opfindelsen kan formuleres til administration via en hvilken som helst bekvem vej i 20 analogi med andre topiske præparater, der er tilpasset til anvendelse i pattedyr. Disse præparater kan være præsenteret til anvendelse på en hvilken som helst konventionel måde ved hjælp af en hvilken som helst af et bredt spektrum af farmaceutiske bærestoffer.
Naphthalenerne med den almene formel I kan være formuleret med egnede 25 farmaceutiske bærestoffer, der er kendte inden for teknikken til dannelse af særligt effektive topiske præparater. En effektiv mængde af naphthalenforbindelsen er fra ca. 0,001 vægtprocent til ca. 10 vægtprocent af det totale formulerede præparat. Resten af det formulerede præparat vil være fra ca. 90 vægtprocent til ca. 99,999 30 vægtprocent af en egnet excipiens, der kan omfatte et farmaceutisk acceptabelt opløsningsmiddel og andre farmaceutisk acceptable additiver til dannelse af en topisk virksom farmaceutisk formulering.

Et farmaceutisk acceptabelt opløsningsmiddel er et opløsningsmiddel, der i det væsentlige er ikke-toxisk og ikke-irriterende under anvendelsesbetingelserne, og som let kan formuleres til en hvilken som helst af de klassiske lægemiddelformuleringer såsom puddere, cremer, 5 salver, lotions, geler, skum, aerosoler, opløsninger og lignende.
Særligt egnede opløsningsmidler omfatter vand, ethanol, acetone, glycerin, propylencarbonat, dimethylsulfoxid (DMSO) og glycoler såsom 1,2-propylendiol, fx propylenglycol, 1,3-propylendiol, polyethylen-glycol med en molekylvægt på fra 100 til 10.000, dipr opy lenglycol, 10 etc. og blandinger af de ovennævnte opløsningsmidler med hinanden.
En topisk creme kan være fremstillet som en halvfast emulsion af olie i vand eller vand i olie. En cremebasisformulering er pr. definition en emulsion, som er et to-fasesystem med én væske (fx fedtstoffer eller olier) dispergeret som små globuler i en anden substans (fx en 15 glycol-vand-opløsningsmiddelfase), som kan anvendes som det primære opløsningsmiddel for naphthalenerne deri. Cremeformuleringen kan indeholde fedtalkoholer, overfladeaktive midler, mineralolie eller petrolatum og andre typiske farmaceutiske adjuvanser såsom antioxi-danter, antiseptika eller kompatible adjuvanser. En typisk cremeba-20 sisformulering er som følger:

Ved fremgangsmåden ifølge opfindelsen muliggøres en metode til afhjælpning af tilstanden psoriasis hos et pattedyr ved topisk admi-5 nistration af et præparat indeholdende en forbindelse med den almene formel I, hvor R^, n, og R^ er som ovenfor defineret. I almindelighed bekæmpes den psoriatiske tilstand i pattedyr, især i mennesker, ved at bringe de betændte områder i kontakt med en terapeutisk virksom mængde af de ved fremgangsmåden ifølge opfindelsen fremstil-10 lede naphthalenholdige præparater, dvs. en mængde, der resulterer i en formindskelse af den epidermale celleformering (en anti-psoriatisk virkning). Naphthalenerne formuleres fortrinsvis først til fremstilling af en egnet farmaceutisk formulering, sådan som det er diskuteret ovenfor, som derefter placeres i kontakt med det/de angrebne 15 område/områder. En effektiv mængde af naphthalenforbindelsen vil afhænge af den specifikke tilstand og det pattedyr, der modtager behandlingen, og vil variere mellem 0,001 og 10 vægtprocent af det farmaceutiske præparat og vil fortrinsvis være mellem 0,01 og 1 vægtprocent af formuleringen. Ved at anvende disse niveauer i formule-20 ringen påføres der til det/de ramte område/områder en terapeutisk virksom og ikke-bivirkningsgivende mængde, dvs. nok til at fremkalde et anti-psoriatisk respons, men ikke nok til på ugunstig måde at påvirke modtageren.

Reaktionen forløber på den måde, der er kendt fra polycycliske aromatiske forbindelser, og giver en blanding af 5- og 6-nitro-2,3-dichlor-l,4-naphthoquino-ner, forbindelser med den almene formel III. Reaktionen udføres 10 typisk med koncentreret salpetersyre i et opløsningsmiddelmedium med lav pH, fortrinsvis koncentreret svovlsyre, typisk ved fra 20°C til 100eC i et tidsrum, der er tilstrækkeligt til at fuldføre reaktionen.

Afhængig af reaktionstemperaturen og reaktionstiderne kan forhold i 5-nitroisomer:6-nitroisomer-blandingen ligge fra 10:1 til 1:10, typisk 8:1.
Forbindelser med den almene formel IV syntetiseres ud fra 5- og 6-5 nitro-2,3-dichlor-l,4-naphthoquinon, forbindelsen med den almene formel III, ved kondensering deraf med et alkalimetalalkoxid, i hvilket alkoxydelen er R^-R^. Reaktionen udføres fortrinsvis i et inert organisk opløsningsmiddel såsom tetrahydrofuran, dimethylsul-foxid, dimethylformamid og lignende ved temperaturer på fra ca. 20°C 10 til ca. 100°C i et tidsrum, der er tilstrækkeligt til at sikre fuldførelse af reaktionen, dvs. i fra ca. 2 timer til ca. 48 timer.
Forbindelser med den almene formel IV, hvor R^=R^ og er lavere al-kylthio, fremstilles ud fra forbindelser med den almene formel III på samme måde som for alkoxyforbindelserne med undtagelse af, at et 15 alkalimetalalkoxid erstattes med et alkalimetalsalt af en alkylmer-captan.
Forbindelserne med den almene formel V fremstilles ud fra forbindelser med den almene formel IV ved katalytiske eller ikke-katalytiske reduktionsprocesser, der er kendte inden for teknikken. Ved denne 20 reduktion kan der anvendes metal-syrereduktionsmiddelkompositioner såsom granuleret jern og saltsyre, tin og saltsyre og lignende, eller neutrale reduktionsmidcelkompositioner såsom zinkstøv og vandig alkohol eller aluminiumamalgam og vandig alkohol, såvel som organo-metalliske reduktionsmidler såsom lithiumaluminiumhydrid, natriurabor-25 hydrid og lignende. Reduktionen udføres fortrinsvis ved at behandle forbindelserne med den almene formel IV med overskud af hydrazin i nærværelse af en katalytisk tilstrækkelig mængde palladium, typisk på carbon. Reaktionen forløber let ved stuetemperatur, idet reaktionstiden styres af hastigheden af tilsætning af hydrazinen til reak-30 tionsblandingen og er typisk fra ca. 1 til ca. 10 timer.
Forbindelser med den almene formel VI, hvor R er hydrogen, kan fremstilles ved at omsætte forbindelser med den almene formel II med et alkalimetalalkoxid eller alkalimetalsalt af en mercaptan som beskrevet ovenfor.

Forbindelser med den almene formel V omdannes til forbindelser med den almene formel VI, hvor R-* er halogen, ved til forbindelsen med den almene formel V i en sur vandig opløsning at sætte en opløsning af et alkalimetalnitrit. Denne initiale reaktion danner diazoniumsal-5 tet i 5- eller 6-stillingen af naphthoquinonringen. Saltet dekompo-neres med en opløsning af kobber(I)halogenid dispergeret eller opløst i den tilsvarende halogensyre (Sandmeyer-reaktionen). 10 En modifikation af den ovennævnte Sandmeyer-reaktion er nyttig til fremstilling af forbindelser med den almene formel VI, hvor RJ er cyano, ved at diazoniumsaltet dekomponeres i nærværelse af et alka-limetalcyanid og et kobber(I)halogenid i stedet for at blive dekompo-neret i nærværelse af kobber(I)halogenid/halogensyre. Halogensubstitueret butadien omsættes med i,4-benzoquinon i 5 et opløsningsmiddel såsom eddikesyre ved en temperatur på fra -10°C til 30°C, fortrinsvis 25°C i 24-72 timer, fortrinsvis 40-48 timer.
5,7-Dihydroforbindelsen af den almene formel VIII udvindes og behandles med et oxideringsmiddel såsom natriumdichromat, natriumnitrit og lignende som beskrevet i de ovenstående artikler til dannelse af 10 forbindelser med den almene formel VIII. Forbindelser med den almene formel IX fremstilles ved at boble chlorgas gennem en opløsning af forbindelsen med den almene formel VIII opløst i et opløsningsmiddel såsom iseddike, nitrobenzen, carbontetrachlorid og lignende, fortrinsvis iseddike ved stuetemperatur. Denne forbindelse, der kan 15 isoleres på kendt måde, opløst i et opløsningsmiddel såsom eddikesyre, behandles med chlorgas og en passende katalysator såsom natriumacetat, iod, jern(III)chlorid, dimethylformamid eller alkoholer ved opvarmning under tilbagesvaling i fra 1/2 til 4 timer, fortrinsvis i 1-2,5 time til dannelse af forbindelser med den almene formel X.
20 Forbindelser med den almene formel VI, hvor R^ er halogen, fremstilles ved at omsætte forbindelser med den almene formel X med et alka-limetalalkoxid såsom natriumalkoxid, fx natriummethoxid, i et vandfrit opløsningsmiddel såsom methanol, dimethylformamid og lignende, idet opløsningsmidlet vælges efter længden af alkylkæden på alkoxy-25 gruppen. Reaktionsblandingen opvarmes under tilbagesvaling i fra 1/2 til 3 timer, fortrinsvis i fra 1/2 til 1 1/2 time. Forbindelserne med den almene formel VI udvindes på konventionel måde såsom ved krystallisation.
Forbindelserne med den almene formel VI, hvor R1 og R2 er lavere 30 alkylthio, kan fremstilles ved den for alkoxyforb inde Isen beskrevne metode med undtagelse af, at alkalimetalalkoxidet erstattes med alkalimetalsaltet af alkylmercaptanen, idet opløsningsmidlet er dimethylformamid og lignende.

Mellemprodukterne med den almene formel VI, hvor er lavere alkyl 5 eller lavere alkoxy, kan fremstilles ved metoder, der er velkendte inden for teknikken, såsom ved omsætning af diazoniumsaltet af forbindelsen med den almene formel V med en passende forbindelse såsom en alkohol, fx methanol, ethanol og lignende. Disse mellemprodukter kan også fremstilles ved metoden vist i reakt ions sekvens II, hvor 2-10 halogen-1,3-butadienen erstattes med den passende 1,3-butadien såsom 2-methyl-l,3-butadien(isopren), 2-ethyl-1,3-butadien og lignende.

Følgende specifikke beskrivelse er givet for at gøre det muligt for fagmanden mere klart at forstå og udøve den foreliggende opfindelse.
FREMSTILLING 1 2.3- Dichlor-5- og 6-nitro-l,4-naphthoquinon 10 (Fremstilling af forbindelser med den almene formel III) 50 g fint pulveriseret 2,3-dichlor-l,4-naphthoquinon (0,22 mol) blev sat til en omrørt opløsning af 170 ml koncentreret svovlsyre og 102 ml 90% salpetersyre med en sådan hastighed, at den exoterme reaktion hævede temperaturen til 60°C. Den resulterende blanding blev omrørt 15 ved 60°C i yderligere 2 timer. Det gule krystallinske faste stof blev filtreret fra, vasket grundigt med vand og omkrystalliseret af chloroform, hvilket gav 22,9 g af 5-nitroisomeren, smeltepunkt 156-157°C. Det ovennævnte stærkt sure filtrat blev hældt ud i isvand. Det resulterende faste stof blev filtreret fra, vasket grundigt med vand 20 og tørret, hvilket gav 20,8 g af en blanding af 5- og 6-isomeren. Fraktioneret krystallisation af denne blanding af eddikesyre og chloroform:isopropanol gav 2,8 g af 6-nitroisomeren, smeltepunkt 184-187°C.
Yderligere mængder af både 5- og 6-isomeren blev opnået ud fra om-25 krystalliseringsmoderludene ved chromatografi på en silicagelkolonne elueret med chloroform/cyclohexan-blandinger.
FREMSTILLING 2 2.3- Dimethoxy-5-nitro-l,4-naphthoquinon (Fremstilling af forbindelser med den almene formel IV)

14 A. En opløsning af 2,72 g 2,3-dichlor-5-nitro-l,4-naphthoquinon (10 mmol) i 15 ml vandfrit tetrahydrofuran blev sat til 25 ml af en IN opløsning af natriummethoxid (25 mmol), og den resulterende opløsning henstod ved 22°C i 16 timer. 1 ml eddikesyre blev derefter sat til, 5 opløsningen blev koncentreret i vakuum, og remanensen blev delt mellem 50 ml vand og 100 ml chloroform. Den vandige fase blev yderligere ekstraheret med 2 x 50 ml chloroform. De forenede chlorofor-mekstrakter blev tørret med MgSO^ og koncentreret i vakuum. Remanensen blev omkrystalliseret af methanol, hvilket gav 1,99 g 2,3-dimet-10 hoxy-5-nitro-l,4-naphthoquinon, smeltepunkt 156-157°C.
B. På samme måde og ved at anvende den i afsnit A herover beskrevne fremgangsmåde, men, hvor det er passende, ved at erstatte 2,3-dich-lor-5-nitro-l,4-naphthoquinon med 2,3-dichlor-6-nitro-l,4- naphtho-quinon og natriummethoxid med det passende natriumalkoxid, fremstil- 15 les følgende forbindelse: 2.3- dimethoxy-6-nitro-l,4-naphthoquinon, smeltepunkt 113-114°C, FREMSTILLING 3 2.3- Dimethoxy-5-amino-l,4-naphthoquinon (Fremstilling af forbindelser med den almene formel V) 20 4,0 ml hydrazin (125 mmol, 97%) blev dråbevis i løbet af 2 timer sat til en omrørt blanding af 19,9 g af titelforbindelsen fra fremstilling 2 (75,6 mmol), 10 g 5% palladium på carbon og 750 ml ethanol under en nitrogenatmosfære. Katalysatoren blev filtreret fra gennem en Celite- kage, der blev vasket med 2 x 300 ml varm ethanol. Det 25 forenede filtrat og vaskefaser blev koncentreret til tørhed i vakuum, og remanensen blev omkrystalliseret af vand:ethanol (1,5:1), hvilket gav 14,6 g 2,3-dimethoxy-5-amino-l,4-naphthoquinon, smeltepunkt 116-117°C.

En opløsning af 0,69 g natriumnitrit (10 mmol) i 5 ml vand blev ved 0-5°C sat til en opløsning af 1,17 g 5-amino-2,3-dimethoxy-l,4-naph-10 thoquinon (5 mmol) i 25 ml 5:1 eddikesyre:vand indeholdende 1,7 ml koncentreret saltsyre. En yderligere mængde på 0,69 g natriumnitrit blev derefter sat til reaktionsblandingen efter afkøling til -5°C efterfulgt af en opløsning af 0,6 g kobber(I)chlorid i 5 ml koncentreret saltsyre. Blandingen fik lov at varme op til 22°C, og fast 15 kobber(I)chlorid blev sat til portionsvis, indtil blandingen antog en grøn farve. Der blev derefter sat vand til reaktionsblandingen, og det udfældede gule faste stof blev filtreret fra, vasket med vand og omkrystalliseret af methanol:vand (2:1), hvilket gav 1,01 g 5-chlor- 2.3- dimethoxy-l,4-naphthoquinon, smeltepunkt 120-121eC.
20 FREMSTILLING 5 5-Cyano-2,3-dimetoxy-l,4-naphthoquinon (Fremstilling af forbindelser med den almene formel VI), hvor R^ er cyano)
En opløsning af 2,21 g natriumnitrit (32 mmol) i 6 ml vand blev ved 25 0-5°C sat til en omrørt suspension af 3,73 g 5-amino-2,3-dimethoxy- 1.4- naphthoquinon (16 mmol) i 20 ml 3:1 vand:tetrahydrofuran indeholdende 6,7 ml koncentreret saltsyre, og den resulterende blanding blev omrørt ved 0-5°C i yderligere 1 1/4 time. Den næsten klare opløsning neutraliseres derefter med natriumcarbonat, filtreres og

16 sættes ved 5°C til en kraftigt omrørt opløsning af 4,75 g kobber(I)-chlorid og 5,88 g natriumcyanid i 80 ml vand. 100 ml ethylacetat blev sat til, og blandingen blev opvarmet til 45eC i 0,5 time, filtreret gennem en Celite-kage og delt i de to faser. Den vandige fase blev 5 ekstraheret med 2 x 100 ml ethylacetat. De forenede organiske faser blev ekstraheret med 150 ml saltvand, tørret over MgS04 og koncen-, treret til tørhed i vakuum. Remanensen blev omkrystalliseret af iso-propanol, hvilket gav 2,96 g 5-cyano-2,3-dimethoxy-l,4-naphthoquinon, smeltepunkt 171-172eC.
10 FREMSTILLING'6 2,3-Dimethoxy-5-phenylthio-l,4-naphthoquinon (Fremstilling af forbindelser med den almene formel VI, hvor R^ er phenylthio)
Metode A. 0,18 g natriumnitrit blev ved 0-5°C sat til en omrørt sus-15 pension af 233 mg 5-amino-2,3-dimetoxy-l,4-naphthoquinon (1 mmol) i 10 ml 0,6N saltsyre og 1 ml tetrahydrofuran. Blandingen blev omrørt ved 5eC i 10 minutter, indtil der blev opnået en klar opløsning og blev derefter neutraliseret ved tilsætning af natriumcarbonat. Den iskolde opløsning blev derefter under en nitrogenatmosfære og ved 20 50°C langsomt sat til en kraftigt omrørt 2-faseblanding af 0,16 g kaliumhydroxid, 10 ml vand, 0,32 ml thiophenol og 35 ml ethylacetat.
Efter en total reaktionstid på 20 minutter blev blandingen delt mellem 40 ml ethylacetat og 100 ml saltvand. Ethylacetatfasen blev tørret over MgS04 og koncentreret i vakuum. Remanensen blev oprenses 25 ved chromatografi på en tyktlagssilicagelplade under anvendelse af acetone:toluen:chloroform (1:20:20), hvilket gav et fast stof, der, efter omkrystallisering af isopropanol, gav 70 mg 2,3-dimethoxy-5-phenylthio-1,4-naphthoquinon, smeltepunkt 76-77eC.
Metode B. 6,0 ml thiophenol (59 mmol) blev ved -30°C sat til en om-30 rørt blanding af 1,4 g 100%'s natriumhydrid (59 mmol) og 200 ml dime thyl formamid, og den resulterende blanding blev omrørt ved 22°C i 16 timer. Denne blanding blev derefter afkølet til -50eC, og en opløsning af 13,0 g di(2,3-dimethoxy-l,4-naphthoquinon-5)-disulfid (49 17 mmol) i 100 ml dimethylformamid blev sat til i løbet af 30 minutter. Den resulterende blanding fik lov at varme op til 22°C i løbet af 1 time før den blev afkølet til -50°C og neutraliseret med 5,4 ml eddikesyre (90 mmol). Reaktionsblandingen blev derefter hældt ud i en 5 blanding af 1,8 liter vand og 700 ml methanol. Det udfældede materiale blev filtreret fra og omkrystalliseret af methanol, hvilket gav 10,3 g 2,3-dimethoxy-5-phenylthio-l,4-naphthoquinon, smeltepunkt 76-79°C. En yderligere mængde på 2,2 g med smeltepunkt 76-77eC blev opnået i anden omgang fra omkrystalliseringen.
10 FREMSTILLING-7 (Fremstilling af forbindelser med den almene formel X, hvor er 6-chlor) A. Gennem en opløsning af 193 g 6-chlor-l,4-naphthoquinon i 1600 ml iseddikesyre blev boblet chlorgas, indtil TLC af en portion viste 15 fuldkommen forsvinden af 6-chlor-l,4-naphthoquinon. Det resulterende bundfald blev opsamlet ved filtrering, vasket med 200 ml eddikesyre og 2 x 300 ml hexan og lufttørret, hvilket gav 157 g 2,3,6-trichlor- 2.3- dihydro-l,4-naphthoquinon. Det faste stof blev overført til en kolbe, der var udstyret med en mekanisk omrører og tilbagesvaler.
20 98,4 g natriumacetat og 1,5 liter eddikesyre blev sat til, og chlor gas blev boblet gennem suspensionen. Blandingen blev bragt til tilbagesvaling og holdt der i 2 timer. Den afkølede blanding blev hældt over 3,5 liter vand, og det resulterende bundfald blev opsamlet ved filtrering, vasket med 2 x 500 ml vand, lufttørret og derefter va-25 kuumtørret over phosphorpentoxid, hvilket gav 139 g 2,3,6-trichlor- 1.4- naphthoquinon, smeltepunkt 147,5-148,5°C.
B. Gennem en suspension af 100 g 6-chlor-l,4-naphthoquinon (520 mmol) i 800 ml eddikesyre, der var opvarmet til 70eC, blev der under mekanisk omrøring boblet chlorgas. Opvarmningen blev forøget for at 30 bringe blandingen til tilbagesvaling, på hvilket tidspunkt 13,2 g fast iod (104 mmol, 0,2 ækvivalenter) blev tilsat. Chlortilsætning ved tilbagesvaling blev fortsat, indtil TLC viste fuldstændig omdannelse til produkt, i alt 7-10 timer. Reaktionen blev afkølet,

18 hvilket gav et tykt bundfald af produkt, der blev opsamlet ved filtrering. Endnu en portion blev vundet ved koncentrering og/eller fortynding med vand. Det totale udbytte af tørret produkt af 2,3,6-trichlor-l,4-naphthoquinon er 121,2 g (464 mmol, 89%), smeltepunkt 5 147,5-148,5°C.

FREMSTILLING'8 10 (Fremstilling af forbindelser med den almene formel VI, hvor er 6-chlor).
Til en mekanisk omrørt opløsning af 55,5 g natriummethoxid i 1,5 liter vandfrit methanol under en nitrogenatmosfære blev så hurtigt som muligt sat 130 g 2,3,6-trichlor-l,4-naphthoquinon fra fremstil-15 ling 10. Temperaturen steg til 50°C under tilsætningen, og reaktionen blev derefter opvarmet til tilbagesvaling i 1 time. Blandingen blev afkølet og gjort sur med 6M saltsyre, hvilket gav en klar gul farve.
Efter tilsætning af 300 ml vand blev reaktionsblandingen filtreret, og bundfaldet blev vasket med vandig methanol (4:1 vand:methanol), 20 indtil filtratet var gul-orange. Bundfaldet blev lufttørret, hvilket gav 102 g 6-chlor-2,3-dimethoxy-l,4-naphthoquinon, smeltepunkt 125-126°C.
FREMSTILLING 9 A. Til en opløsning af 25,8 g natrium-n-butoxid i 125 ml tørt dimeth-25 ylformamid blev på én gang sat 28 g 2,3,6-trichlor-l,4-naphthoquinin. Blandingen blev tilbagesvalet i 2 timer, afkølet, gjort sur med 6M saltsyre og inddampet. Remanensen blev chromatograferet på silicagel ved eluering med dichlormethan, hvilket gav 12,3 g 2,3-di-n-butyloxy-6-chlor-l,4-naphthoquinon som et rødt olieagtigt fast stof.
19 FREMSTILLING 10 (Forbindelser med den almene formel VI, hvor er hydrogen).
Til en mekanisk omrørt opløsning af 11,1 g natriummethoxid i 200 ml vandfrit methanol under nitrogenatmosfære blev så hurtigt som muligt 5 22,7 g 2,3-dichlor-l,4-naphthoquinon tilsat. Temperaturen steg til 50°C under tilsætningen, og reaktionen blev derefter opvarmet til tilbagesvaling i 1 time. Blandingen blev afkølet og gjort sur med 6M saltsyre, hvilket gav en klar gul farve. Efter tilsætningen af 800 ml vand, blev reaktionsblandingen filtreret, og bundfaldet blev vasket 10 med vandig methanol (4:1 vand:methanol) , indtil filtratet var gulorange. Bundfaldet blev lufttørret, hvilket gav 21,2 g 2,3-dimethoxy- 1,4-naphthoquinon, smeltepunkt 116-117°C.
EKSEMPEL 1 A. En opløsning af 50,5 g 6-chlor-2,3-dimethoxy-l,4-naphthoquinon 15 (200 mmol) i 500 ml tetrahydrofuran blev hydrogeneret ved atmosfærisk tryk over 5,0 g palladium-på-trækul (10%), indtil opløsningen var farveløs, ca. 4 timer.
B. Stadig under en hydrogenatmosfære blev en opløsning af 47 ml eddikesyreanhydrid, 40 ml pyridin og 1,22 g DMAP i 50 ml tetrahy- 20 drofuran sat til blandingen. Efter omrøring i 1 time blev blandingen inddampet. Remanensen blev opløst i 500 ml ether og blev vasket med 3 x 250 ml 1M HC1 og med 2 x 250 ml saltvand. Den organiske fase blev tørret over natriumsulfat, filtreret og inddampet, hvilket gav en olie, der krysalliserede i højvakuum. Omkrystallisering af ether/pe-25 troleumsether gav 6-chlor-l ,4-diacetyloxy-2,3-dimethoxynaphthalen, smeltepunkt 93-94°C.
På samme måde og ved at anvende den ovenstående fremgangsmåde, men, hvor det er passende, erstatte 6-chlor-2,3-dimetoxy-l,4-naphthoquinon med 6-chlor-2,3-dimethylthio-l,4-naphthoquinon fremstilles følgende 30 forbindelse:

EKSEMPEL 2
En opløsning af 20,0 g 2,3-dimethoxy-l,4-naphthoquinon i 150 ml te-5 trahydrofuran blev hydrogeneret ved atmosfærisk tryk over 0,5 g Pd-C (10%), indtil opløsningen var farveløs, ca. 4 timer. Stadig under en hydrogenatmosfære, blev en opløsning af 20 ml eddikesyreanhydrid og 18 ml pyridin i 50 ml tetrahydrofuran sat til blandingen. Efter omrøring i ltime blev blandingen inddampet. Remanensen blev opløst i 10 100 ml ether og blev vasket med 3 x 50 ml 1M saltsyre og med 2 x 50 ml saltvand. Den organiske fase blev tørret over natriumsulfat, filtreret og inddampet. Omkrystallisering af ether gav 22,5 g 2,3-dimethoxy-l,4-diacetyloxynaphthalen, smeltepunkt 138-139°C.

EKSEMPEL 4 Toxicitet LD^q af forbindelsen 6-chlor-l,4-diacetyloxy-2,3-dimethoxynaphthalen 10 blev undersøgt på følgende måde:
Akut toxicitet ved subcutan dosering
Mus blev givet enkelte subcutane doser på op til 640 mg/kg af forbindelsen og blev observeret i 3 uger. Der forekom ingen dødsfald ved denne undersøgelse. Den subcutane LD,.q af forbindelsen i mus blev 15 anslået til at være større end 640 mg/kg.
Akut toxicitet ved oral dosering
Mus blev givet enkelte orale doser på op til 1280 mg/kg af forbindelsen og blev observeret i 3 uger. Dosisrelaterede dødsfald forekom hos 1/12 af musene, der var doseret med 640 mg/kg og hos 4/12 af 20 musene, der var doseret med 1280 mg/kg. Den orale LDj.q af forbindelsen i mus blev anslået til at være større end 1280 mg/kg, EKSEMPEL 5 Biologiske data

Test for topisk anti-inflammatorisk virkning ved inhibering af ara-chidonsyreinduceret museøreødem
MATERIALER OG METODER
Swiss Webster han- og hunmus, der vejede 18-27 g, blev tilfældigt 5 fordelt i behandlingsgrupper på 8 eller 10 dyr, sat i bur sammen og blev givet føde og vand ad libitum.
Testmaterialerne blev fremstillet som opløsninger eller suspensioner i kemisk ren acetone i et dosisniveau på 100 mg/ml, og arachidonsyre blev fremstillet som en opløsning i acetone, også på 100 mg/ml. Test-10 materialerne blev påført højre øre på mus ved hjælp af en automatisk mikroliterpipette, således at 10 μΐ opløsning blev påført hver af de indre og ydre overflader. Hvert øre fik derfor i alt 20 μΐ opløsning indeholdende 2 mg testmateriale. 1 time efter påføring af testmaterialet blev arachidonsyreopløsningen påført på samme måde. 1 time efter 15 arachidonsyrepåføringen blev dyrene dræbt ved cervikal dislokation, og det højre øre blev fjernet. En 8 mm skive blev stanset ud fra hvert øre ved hjælp af en biopsistanser, og skiverne blev vejet til det nærmeste 0,1 mg. Med det formål at bestemme den anti-inflammatoriske virkning af testmaterialerne fik i hvert eksperiment en 20 negativ kontrolgruppe to påføringer kun af acetone, og en positiv kontrolgruppe fik kun acetone efterfulgt af arachidonsyre. Den procentuelle inhibering, der fremkom ved behandling med testmaterialet, blev beregnet på følgende måde:

1. Analogifremgangsmåde til fremstilling af en forbindelse med den almene formel 1 DK 164105 B 0C(0)W R3-rYiI 0C(0)K hvor 1 9
2. Fremgangsmåde ifølge krav i, kendetegnet ved, at der fremstilles en forbindelse med den almene formel 1' OC(0) W ftV1 0C(O)W 10 hvor R^, R^, R3 og W er som defineret i krav 1.
3. Fremgangsmåde ifølge krav 2, kendetegnet ved, at der fremstilles en forbindelse, hvor R^- og R^ er lavere alkoxy med 1-6 carbonatomer.
4. Fremgangsmåde ifølge krav 3, kendetegnet ved, at der fremstilles en forbindelse, hvor R3 er valgt fra klassen bestående af fluor, chlor og brom.
5. Fremgangsmåde ifølge krav 3, kendetegnet ved, at der fremstilles en forbindelse, hvor 20 R3 er cyano.
