Claims (11)

1. Mikrobiel organisme, omfattende en malonyl-CoA-uafhængig fedtsyresyntese-(FAS) omsætningsvej og en acyl-reduktionsomsætningsvej omfattende heterologe nukleinsyrer der koder for en malonyl-CoA-uafhængig FAS-omsætningsvej og heterologe nukleinsyrer der koder for en acyl-reduktionsomsætningsvej udtrykt i tilstrækkelige mængder til at producere en primær alkohol, hvor den primære alkohol er hexanol, heptanol, octanol, nonanol, decanol, dodecanol, tetradecanol, eller hexadecanol, hvor den malonyl-CoA-uafhængige FAS-omsætningsvej omfatter ketoacyl-CoA-acyltransferase eller ketoacyl-CoA-thiolase, 3-hydroxyacyl-CoA-dehydrogenase, enoyl-CoA-hydratase og enoyl-CoA-reductase, og acyl-reduktionsomsætningsvejen omfatter en acyl-Co A-reductase og en alkohol-dehydrogenase, hvor ketoacyl-CoA-acyltransferasen eller ketoacyl-CoA-thiolasen omdanner acyl-CoA til β-ketoacyl-CoA, hvor 3-hydroxyacyl-CoA-dehydrogenasen omdanner β-ketoacyl-CoA til β-hydroxyacyl-CoA, hvor enoyl-CoA-hydratasen omdanner β-hydroxyacyl-CoA til fra/7s-2-enoyl-CoA, hvor enoyl-CoA-reductasen omdanner fra/7S-2-enoyl-CoA til acyl-CoA, hvor acyl-CoA-reduetasen omdanner acyl-CoA til et aldehyd, hvor alkohol-dehydrogenasen omdanner et aldehyd til den primære alkohol.
2. Mikrobiel organisme ifølge krav 1, hvor den primære alkohol produceres i mængder til mindst 10% større niveauer sammenlignet med en mikrobiel organisme der mangler den heterologe nukleinsyre der koder for et malonyl-CoA-uafhængigt FAS-omsætningsvejsenzym.
3. Mikrobiel organisme ifølge krav 1, hvor de heterologe nukleinsyrer koder for ketoacyl-CoA-acyltransferase, 3-hydroxyacyl-CoA-dehydrogenase, enoyl-CoA-hydratase og enoyl-CoA-reductase.
4. Mikrobiel organisme ifølge krav 1, hvor de heterologe nukleinsyrer koder for ketoacyl-CoA-thiolase, 3-hydroxyacyl-CoA-dehydrogenase, enoyl-CoA-hydratase og enoyl-CoA-reductase.
5. Mikrobiel organisme ifølge krav 1, hvor de heterologe nukleinsyrer der koder for en acyl-reduktionsomsætningsvej endvidere omfatter et enzym med acyl-CoA-reductase og alkohol-dehydrogenase-aktivitet.
6. Mikrobiel organisme ifølge krav 5, hvor enzymet med acyl-CoA-reductase og alkohol-dehydrogenase-aktivitet omfatter fedtalkohol dannende acyl-CoA-reductase (FAR).
7. Dyrkningsmedium omfattende den mikrobielle organisme ifølge krav 1.
8. Mikrobiel organisme ifølge krav 1, hvor den malonyl-CoA-uafhængige FAS-omsætningsvej er udtrykt i cytosolen.
9. Fremgangsmåde til at producere en primær alkohol, omfattende dyrkning af den mikrobielle organisme ifølge et hvilket som helst af kravene 1-6 eller 8 under i det væsentlige anaerobe betingelser i et tilstrækkeligt tidsrum for at producere den primære alkohol, hvor den primære alkohol er hexanol, heptanol, octanol, nonanol, decanol, dodecanol, tetradecanol eller hexadecanol.
10. Fremgangsmåde ifølge krav 9, endvidere omfattende at isolere den primære alkohol fra den mikrobielle organisme.
11. Fremgangsmåde ifølge krav 10, hvor denne isolering omfatter oprensning, destillation eller krystallisering af den primære alkohol.
