- compounds of the invention could be prepared according to Scheme 3 by taking ester 2 and deprotonating N5 with a base such as NaH or K 2 CO 3 , and then reacting the subsequent anion with an alkyl halide such as 2-picolyl halide in a suitable solvent such as DMF or DMSO to give esters of type 9 . These are then treated with a base such as NaOH (3N in ethanol) with heating to give enaminones of type 10 . Reaction of compounds of type 10 with isocyanates typically at room temperature in a solvent such as 1,2-dichloroethane affords target compounds of type 1 .

- the compounds of this invention were tested for affinity for the benzodiazepine sites of the GABA-A receptor. Since compounds which bind to this receptor can be useful in treating central nervous system disorders, the compounds were also tested in appropriate screens to evaluate specific activities. The results of the various screens are shown in Table 1.
- IC 50 value for all tested compounds is listed in Table 1. An IC 50 value of over 10,000 for a particular compound would indicate that the compound was not active in this screen. This is a general screen and compounds active here are have potential utility for the treatment of one or more disorders of the central nervous system.
- mice Male CD 1 mice, were fasted at least 16 hours, were divided into equal groups and test compounds or vehicle were administered parenterally. Water was not withheld except during the period of observations.
- anti-pentylenetetrazol (anti-metrazol) activity was evaluated by the subcutaneous administration of the CD 90 dose of metrazol (the dose of metrazol was determined from the dose-response curve producing clonic convulsions in 90% of animals that received the corresponding vehicle for this experiment).
- Metrazol was dissolved in 0.9% sodium chloride solution, and its dose volume was 10 mL/kg. Animals were housed individually for observation of clonic convulsions, tonic convulsions and death for a period of 30 min. Test compounds that blocked the clonic seizure component of the convulsion in at least 50% of the animals were considered active. The biological assay was considered to be valid if the effects of a known anticonvulsant (positive control) were detected, within the same experiment. Activity was reported as percent reduction of clonic convulsions from the vehicle group. The ED 50 values of active compounds were calculated by the method of probits (Finney, D.J. 1971. Probit Analysis. London: Cambridge University Press) and are listed in Table 1. An ED 50 value of greater than 30 indicates that an active dose for the compound being tested had not been determined. Compounds active in this screen are considered active anticonvulsion/antiepileptic agents.
- the anxiolytic activity of selected compounds of the invention was assessed by determining their ability to release (disinhibit) behavior that had been suppressed by punishment (Vogel, J.R. et al. Psychopharmacology 1971 , 21 , 1).
- Male rats were deprived of water for 48 hrs and were deprived of food for 24 hrs prior to testing. After the first 24 hours of water deprivation, they were placed in the conflict chamber for a training period; wherein, they were allowed 200 unpunished licks from a bottle containing tap water. The experiment was run the next day. At the expected time of peak activity, the animals were placed in the chamber and allowed access to tap water. If they failed to drink, the experiment was terminated in 5 min, and animals were evaluated for signs of CNS depression.
- a compound was considered active if there is a significant difference in the median number of shocks tolerated between the drug-treated group and the control group.
- the minimum effective doses (MED) for the active compounds of the invention are listed in Tables 1 to 5.

Claims (10)
- Composé de formule 1 suivante: dans laquelle X est indépendamment choisi parmi l'un quelconque d'un atome d'hydrogène, un groupe alkyle en C1 à C8, un atome d'halogène, un groupe perfluoro(alkyle en C1 à C8), hydroxy, alcoxy en C1 à C8, di(alkyl en C1 à C8)amino, alcoxycarbonyle en C1 à C8 ou alkylthio en C1 à C8;
dans laquelle R est choisi parmi l'un quelconque d'un (CH2)n-hétérocycle où n = 1 à 4 et dans laquelle l'hétérocycle est choisi parmi l'un quelconque de la morpholine, la pyridine, le pyridine-N-oxyde, le thiazole, le thiophène, le furane, l'indole, le benzothiophène, le pyridazine, la pyrimidine, l'indoline, la quinoléine, l'indazole, l'imidazole, le benzofurane, la triazine, la pyrazine, l'isoquinoléine, l'isoxazole, le thiadiazole, le benzothiazole, le triazole, le benzotriazole ou une pipérazine substituée où le substituant est un atome d'hydrogène ou un groupe alkyle en C1 à C8, un hétérocycle substitué où il y a un ou plusieurs substituants qui sont indépendamment choisis parmi l'un quelconque d'un atome d'halogène, un perfluoroalklyle (en C1 à C8), nitro, alkylthio en C1 à C8, alcoxy en C1 à C8, alkyle en C1 à C8, di(alkyl en C1 à C8)amino, carboxy ou alcoxycarbonyle en C1 à C8;
à condition que R ne soit pas (CH2)n-NR2R3, où R2 et R3 sont pris ensemble avec l'atome d'azote pour former une pipérazine substituée dans laquelle la pipérazine substituée est substituée par un groupe alkyle en C1 à C4 ou aralkyle (en C1 à C4);
dans laquelle Ar est choisie parmi l'un quelconque d'un groupe phényle et d'un groupe phényle substitué, dans lequel les substituants du phényle sont un groupe alkyle en C1 à C8, un atome d'halogène, un groupe perfluoro(alkyle en C1 à C8), hydroxy, alcoxy en C1 à C8, di(alkyl en C1 à C8)amino, alcoxycarbonyle à C1 à C8 ou alkylthio en C1 à C8; un hétérocycle dans lequel l'hétérocycle est choisi parmi l'un quelconque de la pyridine, le pyridine-N-oxyde, le thiazole, le thiophène, le furane, l'indole, le benzothiophène, la pyridazine, la pyrimidine, l'indole, l'indoline, la quinoléine, l'indazole, l'imidazole, le benzofurane, la triazine, la pyrazine, l'isoquinoléine, l'isoxazole, le thiadiazole, le benzothiazole, le triazole ou le benzotriazole; un hétérocycle substitué où il y a un ou plusieurs substituants qui sont indépendamment choisis parmi l'un quelconque d'un atome d'halogène, d'un groupe perfluoro(alkyle en C1 à C8), nitro, alkylthio en C1 à C8, alcoxy en C1 à C8, alkyle en C1 à C8, di(alkyl en C1 à C8)amino, carboxy, alcoxycarbonyle en C1 à C8;
ou un sel, solvate, hydrate, tautomère ou rotomère pharmaceutiquement acceptable de celui-ci;

- Composé selon l'une quelconque des revendications 1 à 3, dans lequel R est choisi parmi l'un quelconque du (2-furfuryl)méthyle; du (4-imidazo)méthyle; du (3-pyridyl-N-oxyde)méthyle; du (4-pyridyl-N-oxyde)méthyle; du 2-(2-pyridyl)éthyle; du (4-pyridyl)méthyle; du (3-pyridyl)méthyle; du 2(2-thiényl)éthyle; du (3-thiényl)méthyle; du 2(3-thiényl)éthyle; du (2-thiényl)méthyle; du (2-pyridyl)méthyle; du 2-(3-thiényl)éthyle et du (3-furfuryl)méthyle.
- Composé selon la revendication 4, dans lequel R est le 4-(imidazo)méthyle.
- Composé selon l'une quelconque des revendications 1 à 5, dans lequel Ar est le 2-fluorophényle ou le 2,6-difluorophényle.
- Composé selon la revendication 1, choisi parmi l'un quelconquedu 7-fluoro-1,2-dihydro-5-[2-(3-thiényl)éthyl]-3-oxo-N-(2-fluorophényl)pyrido-[2,2-a]benzimidazole-4-carboxamide,du 7-fluoro-1,2-dihydro-5-[2-(2-thiényl)éthyl]-3-oxo-N-(2-fluorophényl)pyrido-[1,2-a]benzimidazole-4-carboxamide,du 7-fluoro-1,2-dihydro-5-[(2-thiényl)méthyl]-3-oxo-N-(2-fluorophényl)pyrido-[1,2-a]benzimidazole-4-carboxamide,du 7-fluoro-1,2-dihydro-5-[(3-thiényl)méthyl]-3-oxo-N-(2-fluorophényl)pyrido-[1,2-a]benzimidazole-4-carboxamide, etdu 7-fluoro-1,2-dihydro-5-[(4-imidazo)méthyl]-3-oxo-N-(2-fluorophényl)pyrido-[1,2-a]benzimidazole-4-carboxamide.
- Composition pharmaceutique comprenant un composé de formule 1 tel que défini selon l'une quelconque des revendications 1 à 7 ou un sel, solvate, hydrate, tautomère ou rotomère pharmaceutiquement acceptable de celui-ci en une quantité efficace pour traiter des troubles du système nerveux central et un véhicule ou diluant pharmaceutiquement acceptable.
- Composé selon l'une quelconque des revendications 1 à 7 ou composition selon la revendication 8 destiné à traiter des troubles du système nerveux central, tels que l'anxiété, les convulsions, l'insomnie, le spasme musculaire ou la surdose de médicament benzodiazépine.
- Composé ou composition selon la revendication 9, pour utilisation en une quantité efficace de 0,2 à 25 mg/kg par jour.
