Abstract

Un método para caracterizar moléculas de ácido nucleico, que comprende las etapas de: I) introducir una base modificada que es un sustrato de una DNA-glicosilasa en una molécula de DNA; II) cortar la base modificada por medio de dicha DNA-glicosilasa de manera que se genera un sitio abásico; III) escindir el DNA en el sitio abásico de manera que se genera un fragmento de DNA aguas arriba que puede ser extendido; y IV) incubar el fragmento aguas arriba y extensible en presencia de una enzima que permite su extensión y de un ácido nucleico molde, y analizar el fragmento(s) resultante.
