Abstract
Un método para incrementar el rendimiento de una reacción de polimerasa sobre un ácido nucleico diana presente en una solución que contiene un inhibidor de la polimerasa, comprendiendo el método: (a) la puesta en contacto del ácido nucleico diana con una polimerasa, donde la polimerasa está unida covalentemente a un dominio de unión a ácido nucleico no específico de ninguna secuencia que: (i) se une a ácido nucleico bicatenario y (ii) incrementa la procesividad de la polimerasa en comparación con una polimerasa idéntica que no tenga fusionado a la misma el dominio de unión a ácido nucleico no específico de ninguna secuencia, y donde la solución tiene una composición que permite al dominio de unión unirse al ácido nucleico diana y al dominio polimerasa extender un cebador que esté hibridado a la secuencia de ácido nucleico diana; (b) la incubación de la solución en condiciones bajo las cuales el cebador sea extendido por la polimerasa, con la condición de que la sal no sea un inhibidor.
