Abstract
Mejoras introducidas en un procedimiento para la síntesis de polinucleótidos de secuencia definida que comprende las siguientes etapas sucesivas: a) someter a reacción de copulación, como primer resto, un nucleótido que tiene un grupo diéster de 3''-fosfato enmascarado con, como segundo resto, un nucleósido o nucleótido que tiene un grupo 5''-hidroxilo libre disponibles, realizándose la reacción de copulación en presencia de un agente de copulación; b) opcionalmente, separar cualquier grupo protector presente en el compuesto procedente de la etapa anterior; c) si se desea, separar el dinucleótido procedente de la etapa b) de la mezcla de reacción y d) opcionalmente, repetir el proceso cuantas veces sea necesario para conseguir una cadena de polinucleótido tan larga como se desee. Cuya mejora comprende emplear un primer resto en un exceso molar considerable con relación al segundo resto y donde el segundo resto sustancialmente se consume completamente en la reacción de copulación.
