Claims (8)

  1. Méthode de purification du facteur stimulant les colonies de granulocytes-macrophages (GM-CSF) d'un milieu de culture d'un micro-organisme obtenu par une technique d'ADN recombinant et capable de produire GM-CSF, comprenant les étapes séquentielles d'enlèvement des protéases par chromatographie par échange d'anions, d'enlèvement des impuretés hydrophobes par chromatographie par affinité ligand-colorant, de séparation des impuretés de fort et faible poids moléculaire par chromatographie par filtration sur gel et séparation des protéines en rapport par chromatographie en phase inverse.
  2. Méthode selon la revendicaiton 1, où GM-CSF est GM-CSF humain.
  3. Méthode selon la revendication 2, où GM-CSF est GM-CSF humain recombinant.
  4. Méthode selon l'une quelconque des revendications 1 à 3, où la résine d'échange d'anions est une résine d'aminoéthyle quaternaire.
  5. Méthode selon l'une quelconque des revendications 1 à 4, où la colonne par affinité ligand-colorant comprend un colorant de triazinyle sur un support d'agarose.
  6. Méthode selon l'une quelconque des revendications 1 à 5, où la colonne de filtration sur gel est un gel à base de dextrane réticulé ayant une plage de coupure comprise entre environ 1.000 et environ 200.000 pour les protéines.
  7. Méthode selon l'une quelconque des revendications 1 à 6, où la chromatographie en phase inverse est une chromatographie liquide rapide des protéines en utilisant une colonne C4 ou C8.
  8. Méthode selon la revendication 1, comprenant l'élimination de la protéase avec une résine d' aminoéthyle quaternaire, l'élimination des impuretés hydrophobes avec une colonne comprenant un colorant de triazinyle sur un support d'agarose, l'éliminaiton des impuretés de faible et fort poids moléculaire avec un gel à base de dextrane réticulé ayant une plage de coupure d'environ 1.000 à environ 200.000 pour les protéines et l'élimination des protéines en rapport par chromatographie liquide rapide des protéines en utilisant une colonne C4 ou C8.
