Claims (15)
1. Postupak procjene neurotoksina iz bakterije Clostridium botulinum (BoNT) na aktivnost, naznačen time što se sastoji u:
a) stavljanju u kontakt živčane stanice nastale iz ljudske inducirane pluripotentne matične stanice (hiPS) s pripravkom koji sadrži BoNT; i
b) procjeni navedenog BoNT na biološku aktivnost.
2. Postupak u skladu s patentnim zahtjevom 1, naznačen time što dodatno uključuje korak stavljanja u kontakt navedenog BoNT s ispitivanim spojem prije stavljanja u kontakt navedenih živčanih stanica nastalih iz hiPS.
3. Postupak u skladu s patentnim zahtjevom 2, naznačen time što je navedeni ispitivani spoj protutijelo.
4. Postupak u skladu s patentnim zahtjevom 3, naznačen time što je navedeno protutijelo neutralizirajuće protutijelo.
5. Postupak u skladu s patentnim zahtjevom 4, naznačen time što je navedeno neutralizirajuće protutijelo prisutno u uzorku koji se bira iz skupine koju čine pročišćena protutijela, serum i antitoksini.
6. Postupak procjene neurotoksina iz bakterije Clostridium botulinum (BoNT) na aktivnost, naznačen time što se sastoji u:
a) stavljanju u kontakt živčane stanice nastale iz ljudske inducirane pluripotentne matične (hiPS) stanice s pripravkom koji sadrži i) BoNT; i ii) neutralizirajuće protutijelo; i
b) procjeni navedenog BoNT na biološku aktivnost.
7. Postupak u skladu s bilo kojim od patentnih zahtjeva 1 do 6, naznačen time što navedeni BoNT ima serotip koji se bira iz skupine koju čine A, B, C, E i modificirane varijante navedenih BoNT.
8. Postupak u skladu s bilo kojim od patentnih zahtjeva 1 do 7, naznačen time što se navedenu biološku aktivnost bira iz skupine koju čine cijepanje SNAP-25, cijepanje VAMP2 i otpuštanje neurotransmitera.
9. Postupak u skladu s bilo kojim od patentnih zahtjeva 1 do 8, naznačen time što je navedeno ispitivanje kvalitativno.
10. Postupak u skladu s bilo kojim od patentnih zahtjeva 1 do 9, naznačen time što je navedeno ispitivanje kvantitativno.
11. Postupak u skladu s bilo kojim od patentnih zahtjeva 1 do 10, naznačen time što je navedeni BoNT pročišćen.
12. Postupak u skladu s bilo kojim od patentnih zahtjeva 1 do 10, naznačen time što je navedeni BoNT u kompleksu.
13. Postupak u skladu s bilo kojim od patentnih zahtjeva 6 do 12, naznačen time što je navedeno neutralizirajuće protutijelo prisutno u uzorku koji se bira iz skupine koju čine pročišćena protutijela, serum i antitoksini.
14. Postupak određivanja količine biološki aktivnog BoNT u pripravku koji sadrži biološki aktivni BoNT, naznačen time što se navedeni postupak sastoji u koracima:
(a) stavljanja u kontakt živčane stanice nastale iz stanice hiPS s uzorkom pripravka koji sadrži biološki aktivni BoNT; i
(b) određivanja količine biološki aktivnog BoNT prisutnog u pripravku procjenom navedenog uzorka na biološku aktivnost BoNT.
15. Upotreba živčane stanice nastale iz ljudske inducirane pluripotentne matične (hiPS) stanice, naznačena time što je navedena živčana stanica namijenjena procjeni BoNT na aktivnost.
