Claims (1)
- 【特許請求の範囲】 1.(a)化合物を蛍光を発することができる信号伝達物質のPHドメインと インキュベートする; (b)PHドメインの蛍光のリン脂質誘発調節の測定(化合物の存在下の蛍光の 変化は化合物とPHドメインの間の機能的相互作用の指標である) の段階を含む、信号反応の調節剤と推定される化合物のスクリーニング法。 2.蛍光がPHドメインのN−末端に位置するTrp残基に関連するものである 、請求項1記載の方法。 3.信号伝達物質がプロテインキナーゼである、請求項1または2記載の方法 。 4.信号伝達物質がRACプロテインキナーゼであり、蛍光残基がTrp22であ る、請求項3記載の方法。 5.調節剤と推定される化合物を、精製PHドメインとインキュベートするこ とを含む、請求項1から4のいずれかに記載の方法。 6.PHドメインを、細菌宿主内でGST融合タンパク質としてコードする組 換え発現ベクターを発現させ、続いてスロンビンでGST融合タンパク質を開裂 させ、PHドメインを精製することにより製造する、請求項5記載の方法。 7.PHドメインを290nmで励起し、蛍光を345nmで検出する、請求項4 記載の方法。 8.ドメインをコードする核酸配列を発現させ、直接そのC−末端に少なくと も3個の親水性アミノ酸を結合させることを含む、細菌細胞内でRACプロテイ ンキナーゼのPHドメインを製造する方法。 9.親水性アミノ酸が同一または異なり、塩基性アミノ酸である、請求項1記 載の方法。 10.親水性アミノ酸がリジン残基である、請求項2記載の方法。 11.ドメインを発現を20から33℃の間で誘発させる、請求項3記載の方 法。
