Abstract
【解決手段】 全身活性治療剤、局所活性治療剤、消毒及び衛生剤、清浄剤、芳香剤及び肥料から選択される活性剤を含有する基質を、有効量の可塑化された疎水性アクリル系ポリマーの水性分散液で被覆した後、該分散液のガラス転移点より高い温度で上記被覆を硬化させることから成る固体の放出制御型製剤の製法及びその放出制御型製剤。

Description
ヒドロモルホンビーズの製造
ヒドロモルホンビーズを、塩酸ヒドロモルホンを水に溶解し、オパドライ(Opadry〕Y-5-1442 ,ライトピンク(カラーオン、ウエストポイント、ペンシルバニア(Coloron, Westpoint, Pennsylvania)から市販されている製造物、ヒドロキシプロピルメチルセルロース(HPMC)、ヒドロキシプロピルセルロース、二酸化チタン、ポリエチレングリコール及びD&CレッドNo.30アルミニウムレーキを含む)を20%w/w で加え、約1時間混合し、次いで、ヌ・パリエル(nu pariel )18/20 メッシュのビーズ上にウルスター・インサート(Wurster insert)を用いてスプレーした。得られた製造物は、下記表1に示した配合であった。
遅延剤の被覆−硬化工程なし
実施例2においては、実施例1において製造したヒドロモルホンビーズを、第2表に示したように5%の付着重量となるようオイドラギットRS 30 Dでオーバーコートした。終点までの乾燥は行わなかった。
遅延剤被覆の保護
実施例2のヒドロモルホンビーズのゆっくりとした溶解が、ヒドロモルホンと遅延剤の間の安定性の問題によるものかどうかを検討するために、実施例3において、ヌ・パリエルヒドロモルホンビーズを実施例1と同様にして調製し、次いで、5%のHPMCでオーバーコートし、遅延剤層を形成せずに試験を行った。溶解試験を、最初と、そして、37℃乾燥及び37℃/80%RHの加速化された条件で貯蔵後に行った。
先行技術の硬化(文献で薦められているもの)
実施例4においては、実施例3によって製造したヒドロモルホンビーズを5%の付着重量となるようにオイドラギットRSで被覆した。被覆を塗布後、ビーズを流動床乾燥器中で、45℃において2時間乾燥(硬化)した。この温度は、クエン酸トリエチルで20%レベルの固形分で可塑化した、オイドラギットRS 30 DのTgを超える温度である。溶解試験を、最初と、そして、37℃乾燥及び37℃/80%RHで貯蔵後に行った。結果を以下の表5に示す。
硬化の最適化及び遅延被覆の成分
実施例2〜4から得られた結果は、遅延剤被膜でオーバーコートしたビーズの溶解は、ある点までは減速するが、それ以上は変わらないように思われる。しかしながら、達成された終点での溶解は遅すぎるものであった。
遅延剤被膜の膜厚の最適化
実施例8〜10においては、所望の放出パターンで使用するためのメタクリル酸エステルポリマーの最適重量を決定するため、並びに45℃の乾燥環境下48時間の硬化工程での再現性及び有効性を決定するために追加の試験を行った。メタクリル酸エステル量の異なるレベルで3バッチを製造し、45℃の乾燥オーブン中で硬化させた。
硫酸モルヒネ被覆ビーズ
実施例11においては、本発明の硬化工程を、硫酸モルヒネを薬剤として置換した製剤に適用した。
放出制御塩酸ヒドロモルホン8mg製剤−アクリル系ポリマー被覆実施例12は以下のように製造した。
実施例13においては、12人の患者について、単一の投与量で6種の方法を無作為化した交錯試験(1週間でウオッシュアウト)を行い、速放性製剤の等しい投与量で得られた結果と比較した。血液試料は血漿レベルを定量するために、投与後、初期、0.25、 0.5、 0.75 、1、1.5 、2、2.5 、3、3.5 、4、6、8、10、12、18、24、30、36及び48時間で採取した。比較例13Aは8mgのヒドロモルホン速放性製剤(ジラウジット(Dilaudid)4mg錠剤2錠、ノール(Knoll) 社から市販)である。実施例13は、8mg投与量の実施例12のカプセル封入ヒドロモルホンビーズである。
実施例14〜15において、10人の対象に対し、単一の投与量で4種の無作為化した交錯試験を行った。実施例14は、実施例13のヒドロモルホンビーズ8mg−絶食で試験し、実施例15は、実施例13のヒドロモルホンビーズ8mg−食餌で試験した。比較例14Aにおいて、8mgの速放性ヒドロモルホン(ジラウジッド(Dilaudid)4mg錠剤を2錠)を絶食した者に投与した。比較例15Aにおいて、8mgの速放性ヒドロモルホン(ジラウジッド(Dilaudid)4mg錠剤を2錠)を食事を与えた者に投与した。
実施例16〜17において、定常状態で3種の交錯試験を4日間行った。比較例16Aにおいては、対象に6時間毎に速放性ヒドロモルホン(ジラウジッド4mg錠剤を2錠)8mgを投与した。実施例16では、実施例15のヒドロモルホンビーズ8mgを12時間毎に投与した。実施例17では、実施例13のヒドロモルホンビーズ8mgを24時間毎に投与した。4日目に血液試料を採取した。
放出制御硫酸モルヒネ30mg製剤−アクリル系ポリマー被覆実施例18は上記実施例と同様の方法で調製した。実施例18のビーズの完全な配合は、下記表21に記載する。
実施例19〜20においては、硫酸モルヒネの配合量が多いように製造された高配合基ビーズであり、それ故、多い投与量が1日1回容易に投与できる。高配合ビーズは、グラット・ローター処理器(Glatt Rotor Processor) 中で粉末を層化することにより製造される。実施例19〜20の製剤は、下記表24に記載した。
放出制御アセトアミノフェン(APAP)錠剤を本発明に従い、以下のようにして製造した。
実施例22においては、放出制御アセトアミノフェン(APAP)錠剤を製造した。迅速な溶解を与えることが要求される場合には、オイドラギットRL-30 Dの量を増加し、オイドラギットRS-30 Dの量を減少する。かくして、オイドラギットRL-30 Dのみを含有し、オイドラギットRS-30 Dを含有しない放出制御型APAP錠剤を製造した。APAP核は実施例4と同様にして製造した。次に、上記のように製造したAPAP錠剤核を、以下のようにして本発明の放出制型御被膜で被覆した:精製水をオイドラギットRL-30 Dに加えた。精製水の量は、最終被覆懸濁液の濃度が、固体ポリマー、可塑剤及びタルクで約20%となるように計算した。次いで、クエン酸トリエチルを添加し、15分間攪拌した。次いで、タルクを添加し、15分間攪拌した。適当量のAPAP錠剤核をアクセラ・コータ(Accela Cota) 塗装槽に添加した。被覆懸濁液を、適当なスプレーガンから、ポリマーが錠剤当たり4%の付着重量が得られるまで噴霧した。
Claims (21)
- 有効量の治療活性剤を含む固体基材を含む放出制御型製剤であって、該固体基材は、この基材が被覆された後に胃液にさらされたときに該治療活性剤を制御して放出するのに有効な量であって、かつ、1日1回の投与に適した製剤を提供する量の、可塑化されたアクリル系ポリマーおよび透過性を増大させる化合物を含む水性分散液で被覆されており、該被覆された基材は、硬化後40℃、75%の相対湿度で少なくとも1カ月の加速化された貯蔵条件下にさらした後に生体外溶解を行ったとき、所定の溶解時間のいかなる時点においても、貯蔵前に行った生体外での溶解と比較して、放出される治療活性剤の総量の約20%より大きく変わることのない量の該治療活性剤を放出するという終点に達するまで、前記水性分散液のガラス転移温度より高い温度で硬化させられており、該被覆が少なくとも24時間の間、治療に有効な血中レベルを与えることを特徴とする、放出制御型製剤。
- 経口投与されたときに所望の治療効果を提供するのに充分な量の全身的に活性な治療剤を含有する基材を含む固体の放出制御型経口投与製剤であって、該基材が約2%〜約25%の付着重量で、可塑化されたアクリル系ポリマーおよび透過性を増大させる化合物を含む水性分散液で被覆されており、該被覆が、USPのバドル法で37℃において900mlの水性緩衝液(pHが1.6 〜7.2)により100rpmで測定したとき、1時間後に12.5〜42.5%(重量)の活性剤を放出し、2時間後に25%〜55%(重量)の活性剤を放出し、4時間後に45%〜75%(重量)の活性剤を放出し、8時間後に55%〜85%(重量)の活性剤を放出するという該活性剤の制御された放出を得るのに充分であり、該被覆された基材が、硬化後40℃、75%の相対湿度で少なくとも1カ月の加速された貯蔵条件にさらしたとき、生体外溶解の際いかなる時点においても、貯蔵前に行った生体外での溶解と比較して、放出される治療活性剤の総量の約20%より大きく変わることのない量の該治療活性剤を放出するという終点に達するまで、前記水性分散液のガラス転移温度より高い温度で硬化させられており、該被覆が少なくとも24時間の間、治療に有効な血中レベルを与えることを特徴とする、固体の放出制御型経口投与製剤。
- 使用環境下において所望の効果を与えるのに充分な量の活性剤を含有する基材を含む放出制御型製剤であって、該活性剤は、消毒剤、清浄剤、芳香剤、肥料、消臭剤、染料、動物忌避剤、昆虫忌避剤、殺虫剤、除草剤、殺菌剤、及び植物成長刺激剤から成る群より選択され、該基材は、該製剤が周囲の液体にさらされたときに、該活性剤の制御された放出を得るのに充分な量で、可塑化されたアクリル系ポリマーおよび透過性を増大させる化合物を含む水性分散液で被覆されており、該被覆された基材は、加速された貯蔵条件にさらした後にも変化しない該活性剤の安定した溶解性を与えるという硬化の終点に到達するまで、少なくとも24時間の間、前記水性分散液のガラス転移温度よりも高い温度で硬化させられており、該終点は、硬化直後の製剤の溶解パターンと、37℃の温度、80%の相対湿度で少なくとも1カ月の加速された貯蔵条件にさらした後の製剤の溶解パターンとを比較することにより決定され、該被覆が少なくとも24時間の間、治療に有効な血中レベルを与えることを特徴とする、放出制御型製剤。
- 該被覆された基材が、生体外溶解試験の際に、40℃の温度及び75%の相対湿度で少なくとも1カ月の加速された貯蔵条件にさらされた後の溶解パターンを該加速された条件にさらされる前の溶解パターンと比較した場合に、いかなる時点においても放出される総活性剤の約15%より広くないバンド領域を与えることを特徴とする、請求項1または2に記載の製剤。
- 該被覆された基材が、該バンド領域が得られるまで、該被覆中の個々のアクリル系ポリマー粒子を合体させ、周囲の液体にさらされたとき該活性剤の放出を徐々に遅くするように、前記水性分散液のガラス転移温度より高い温度で硬化させられていることを特徴とする、請求項3または4に記載の製剤。
- 該基材が約2%〜約25%の付着重量で被覆されていることを特徴とする、請求項1または3に記載の製剤。
- 該治療活性剤が、抗ヒスタミン剤、鎮痛剤、非ステロイド抗炎症剤、胃腸剤、制吐剤、抗てんかん剤、血管拡張剤、鎮咳剤、去啖剤、抗喘息剤、ホルモン剤、利尿剤、抗低血圧薬、抗高血圧薬、気管支拡張剤、抗生物質、抗ウィルス剤、抗痔疾剤、ステロイド剤、睡眠剤、向精神剤、下痢止め剤、粘液溶解剤、鎮静剤、うっ血除去剤、緩下剤、ビタミン剤及び興奮剤から成る群より選ばれたものであることを特徴とする、請求項1〜6のいずれか1項に記載の製剤。
- 該基材が医薬的に許容可能なビーズであり、複数の該被覆され硬化したビーズが、水性溶液に接触したときに、放出が制御された有効薬量を与えるのに充分な量でカプセル中に入れられていることを特徴とする、請求項1〜7のいずれか1項に記載の製剤。
- 該基材が錠剤の核であることを特徴とする、請求項1〜8のいずれか1項に記載の製剤。
- 該治療活性剤が、ヒドロモルホン、オキシコドン、モルヒネ、レボルファノール、メタドン、メペリジン、ヘロイン、ジヒドロコデイン、コデイン、ジヒドロモルヒネ、ブプレノルフィン、それらのいずれかの塩、及びこれらの混合物から成る群より選ばれたオピオイド鎮痛剤であることを特徴とする、請求項1〜8のいずれか1項に記載の製剤。
- 該被覆が、約24〜約48時間の間、硬化させられていることを特徴とする、請求項1〜3のいずれか1項に記載の製剤。
- 該透過性を増大させる化合物が、該硬化された被覆基質からの該治療活性剤の放出速度を調節するのに有効な量で含有されることを特徴とする、請求項1〜11のいずれか1項に記載の製剤。
- 該透過性を増大させる化合物が、遊離ラジカル重合が可能なモノエチレン性不飽和第4級アンモニウム化合物であることを特徴とする、請求項1〜3のいずれか1項に記載の製剤。
