Abstract
Description
学習能力および神経保護のためのモデルにおける化合物Iでの試験の目的は、該パラメーターの可能な影響を評価することである。というのは化合物Iは特にGABA作動性作用を展開するからである。てんかん患者は度重なる発作によりしばしば既に学習能力が欠乏しているので、健忘症ファクターを設定してある、つまり学習能力を低下させた動物でこの実験を行った。このために動物を繰り返し電気ショックで処置するか、またはアルコール遮断下においた。直接の神経保護作用を評価するために、供給血管の結紮により動物に慢性的な脳血行の減少を引き起こした。この損傷は全て学習能力の低下につながり、これを神経細胞損傷の指標として評価する。該モデルでGABAを強化する、抗てんかん作用を有する薬剤、例えばジアゼパムおよびナトリウムチャネル遮断薬、例えばフェニトインは有利な効果を示さず、しかも高い用量では学習能力に対して不利な効果が現れる場合がある。
試験モデル:
脳血行の減少による学習能力損傷
このモデルでは麻酔下でラットの頸動脈1本を結紮した。該動物は麻酔から目覚め、かつその後に学習能力が低下している。学習能力を棒跳び付きテスト(Stabsprungtest)で確認した。該試験の際に該動物は、音の信号により予め予告される、軽い電気的な足ショックを、床の上方に垂れ下がっている垂直な棒に飛び付くことにより回避することを学習しなくてはならない。該動物の学習能力を、条件づけられた反応(音の信号の段階での棒への跳躍)の数(パーセント)で測定する。
