g)200~400mgのラジカル捕捉剤
を含有することができる。
a)1ml当たり200~400単位のボツリヌス神経毒素、好ましくはA型ボツリヌス神経毒素
b)0.25~0.9% w/vのヒアルロン酸
c)0.36~2.0% w/vの塩、好ましくは塩化ナトリウム
d)0~1.5% w/vまたは0.2~1.5% w/vの神経毒素安定剤、好ましくはヒト血清アルブミン
e)0.4~0.94% w/vの結晶化阻害剤
f)0.03~0.06% w/vの緩衝液、好ましくはリン酸緩衝液
g)20~40% w/vのラジカル捕捉剤
・限局性ジストニア(1箇所の身体部位のみに影響を及ぼす)
・分節性ジストニア(2箇所以上の身体部位に影響を及ぼす)
・全身性ジストニア(全身に影響を及ぼす)
(i)本発明の組成物を投与するステップと、
(ii)4~9ヵ月後に投与を繰り返すステップと、
を含む方法である。
本発明は、以下の実施例によって示すことができる。
実施例1
組成物の2つのサブユニットを調製した。
A型ボツリヌス神経毒素 25単位
非架橋ヒアルロン酸 7mg
ヒト血清アルブミン(HAS) 0.125mg
ショ糖 0.59mg
マンニトール 17.0mg
グリセリン 1.5mg
リン酸緩衝液 0.3mg
塩化ナトリウム 4.5mg
参照組成物を以下のようにして取得する。50単位のA型ボツリヌス神経毒素、25mgのヒト血清アルブミン(HAS)、及び1.18mgのショ糖を含有する凍結乾燥物を、1mlの0.9%食塩水で再構成した。
A型ボツリヌス神経毒素 50単位
ヒト血清アルブミン(HAS) 0.25mg
ショ糖 1.18mg
塩化ナトリウム 0.9mg
2.1 10匹のマウスの群2群に対し、各々の右腓腹筋に、参照実施例の組成物0.8mlまたは実施例1の組成物1.6ml(両実施例とも40単位のA型ボツリヌス神経毒素を含有)を注射した。いずれの群においても死亡は観察されなかった。軽微な全身毒性の徴候が、2~10日目に同じ程度で両群に生じた。
Claims (14)
- a)ボツリヌス神経毒素、及び
b)平均分子量が2.5MDa~4.5MDaである非架橋ヒアルロン酸
を含み、25℃において1Hzの発振周波数を用いて決定される場合の動的粘度が1.5~4Pa・sである、組成物。 - 前記ボツリヌス神経毒素がA型ボツリヌス神経毒素である、請求項1に記載の組成物。
- 前記非架橋ヒアルロン酸の多分散指数が1.1~4.0である、請求項1または2に記載の組成物。
- 1ミリグラムの非架橋ヒアルロン酸当たり2~4単位のボツリヌス神経毒素または20~40単位のボツリヌス神経毒素を含有する、請求項1~3のいずれか1項に記載の組成物。
- c)塩、
d)任意選択により神経毒素安定剤、
e)結晶化阻害剤、
f)緩衝液、及び/または
g)ラジカル捕捉剤
をさらに含む、請求項1~4のいずれか1項に記載の組成物。 - 200~500単位のボツリヌス神経毒素が投与される、ジストニアの処置または防止における使用のための請求項1~5のいずれか1項に記載の組成物。
- 500~1000単位のボツリヌス神経毒素が投与される、痙縮の処置または防止における使用のための請求項1~5のいずれか1項に記載の組成物。
- 40~50単位のボツリヌス神経毒素が投与される、皺の防止または処置における使用のための請求項1~5のいずれか1項に記載の組成物。
- 適用してから最大24時間投与部位に残存する、請求項1~8のいずれか1項に記載の組成物。
- 前記ボツリヌス神経毒素が12時間以内に放出される、請求項1~9のいずれか1項に記載の組成物。
- 4~9ヵ月ごとに投与される、請求項1~10のいずれか1項に記載の組成物。
- 哺乳類の皺を、美容的に滑らかにするまたは防止するために、4~9ヵ月ごとに投与される、請求項1~5のいずれか1項に記載の組成物。
- 40~50単位のボツリヌス神経毒素を含む、請求項12に記載の組成物。
- 皺を滑らかにするまたは防止するために、40~50単位のボツリヌス神経毒素を含む、請求項1~5のいずれか1項に記載の組成物。
