Description

【発明の詳細な説明】 本発明はにきび用組成物に関する。更に詳しく
は本発明はにきび用外用組成物に関する。
サリチル酸はにきび治療に有用なにきび分解活
性をもつものとして報告されている（J.
Investigative Dermatology73，434（1979））。ア
スピリン（アセチルサリチル酸）およびその他の
サリチル酸塩をにきび治療用の代表的調剤に使用
することは米国特許第4126881号、英国特許第
842404号、英国特許第1292503号、カナダ特許第
938555号、仏国特許第2077798号および日本特許
73／03364号に示唆されている。サリチル酸塩の
有効性は恐らくその角質分解、剥離および／また
は角質溶解の活性によるものである。
にきび治療用の過酸化ベンゾイル組成物が米国
特許第3535422号に報告されている。過酸化ベン
ゾイルは恐らくそのP.にきび生育阻止活性のため
に有用である。
過酸化ベンゾイルとサリチル酸との両方を含む
にきび治療用組成物が西独特許公開公報第
2418386号に報告されている。
アセチルサリチロイルパーオキサイドは周知の
化合物であるPharm.Zeiting47，847（1902）に記
載されている。
次式のアシルサリチロイルパーオキサイドがサ
リチル酸塩と過酸化ベンゾイルとの所望の効果な
らびに有力な抗菌活性の両者を兼ね備えるにきび
に有用な化合物であることが今や発見された。
上記の化合物は好ましくは溶液、クリームまた
はローシヨンの形体の賦形剤中で皮膚のにきびに
侵された区域に塗布される。この組成物は活性パ
ーオキサイド化合物を約１〜約10重量％含む。皮
膚に塗布するのに好適な任意の溶媒、ローシヨン
またはクリームが賦形剤として使用しうる。
本発明を以下の実施例によつて更に具体的に説
明する。これらの実施例は説明のためのものであ
つて本発明を限定するものと考えるべきではな
い。
実施例 １ Ｏ−アセチルサリチロイルパーオキサイド Ｏ−アセチルサリチロイルクロライド（12ｇ）
を氷冷アセトン（20ml）中にとかし、次いで30％
過酸化水素（3.4ｇ）および無水硫酸ナトリウム
（３ｇ）をこの溶液に加えた。次いでこの反応混
合物に重炭酸ナトリウム（5.1ｇ）を０℃におい
て１時間にわたつて少しつづ加えた。この反応混
合物を更に１時間かくはんした。
過後、少量のメタノールを液に加え、放置
して白色針状物を沈澱させた。過して1.8ｇの
生成物をえた。融点105〜106℃。
実施例 ２ Ａ ローシヨン賦形剤の調製 相 重量％ Promulgen Ｄ 2.5 （高分子量天然脂肪アルコール類のポリエチレ
ングリコールエーテル錯体） Schercomol D1D 1.5 （ジイソプロピルダイマーレート ポリエチレングリコール400 5.0 蒸留水 88.2 相 Avicel CL−611（微結晶セルロース） 2.8 製造操作 １ プロペラ型ミキサを使用して適当なステンレ
ス鋼容器中で相の全材料を一縮に混合し、75
℃に加熱する。
２ 高せん断下で混合しながら相に相
（Avicel CL−611）を迅速に加える。60℃で５
分間高せん断かくはんした後、おだやかにかく
はんしながら冷却を始め、捕促された空気を逃
散させる。このローシヨンを30℃に冷却する。
３ 水の損失を補なつて処方どおりとする。
Ｂ 活性成分含有生成物の調製 乳ばちと乳棒を使用して、実施例１により製造
した化合物7.4ｇを実施例2Aにより製造したロー
シヨン30ｇ中にまぜてすりつぶし、十分な粉砕と
混合を行なう。十分に混合しながら追加のローシ
ヨンを加えてＯ−アセチルサリチロイルパーオキ
サイドの最終濃度を約7.4重量％とする。
実施例 ３ 実施例２の方法により、約1.48重量％のＯ−ア
セチルサリチロイルパーオキサイドを含むローシ
ヨンを調製した。このローシヨンをほほ、ひたい
および皮膚の中程度のにきびの状態の上に使用し
た。このローシヨンを一日２回（午前と午後）、
30秒にわたつて塗布し一夜保持した。この治療を
４週間にわたつて続けた。にきびの数の75％減少
が認められた。また、炎症吹き出物の数の90％減
少が約10〜18日間で達成された。
本発明の組成物はそうよう症かさぶた皮膚炎、
感染うろこ状皮膚炎、皮脂腺炎症およびその他の
類似皮膚炎にも有用である。

  1. 【特許請求の範囲】 １ 次式の化合物を有効成分として含むにきび用
    組成物。 ２ 溶液、ローシヨンまたはクリームの形態の特
    許請求の範囲第１項記載の組成物。 ３ 該化合物を１〜10重量％含む特許請求の範囲
    第１項記載の組成物。
