Description

【発明の詳細な説明】 〔産業上の利用分野〕 本発明はイブプロフエンを含有する薬剤学的
製剤およびその製造法に関する。
〔従来技術〕
口腔及び咽頭腔内で強く粘膜を刺激する酸か
ら成る作用物質、又はこの部分を強く刺激する
酸基を含む作用物質が存在することは公知であ
り、この種の更に難溶性又は不溶性の作用物質
にイブプロフエンがある。
イブプロフエンはリウマチ及び関節炎治療用
の作用物質として益々重要視されるようになつ
た。この物質は水に不溶であるが極めて不快な
味を有し、又おそらく化合物２−（４−イソブ
チルフエニル）プロピオン酸の存在により特に
食道の粘膜を刺激する。同様に文献で公知のよ
うに、イブプロフエンは、アセチルサリチル酸
に似て、この物質が錠剤の形であれカプセルの
形であれ高濃度で胃壁に達した場合は特に、胃
腸の出血を引き起すおそれがある。従つて胃腸
内で局部的な高濃度を生ずることのないように
この物質を水に懸濁させてから服用させるのが
治療上有利である。しかしこの水に懸濁させた
液は、その作用物質の一部が口や食道に残留
し、そこでひりひり刺激するような感覚を与え
るので、何か適当な対策をとらない限り味覚の
点から服用不能である。
〔発明が解決しようとする問題点〕
本発明の目的は、前述した味覚上のマイナス
の効果を除去し、イブプロフエンを含有する薬
剤の経口服用を問題なく実施できるようにす
る、冒頭に述べた種類の薬剤学的製剤およびそ
の製造法を提供することにある。
〔問題点を解決するための手段〕
本発明によれば、前述の種類の薬剤学的製剤
に於ける上記の目的は、イブプロフエン粒子を
少くとも一種のプソイドコロイド及びフマル酸
から成る外被で被覆することにより達成され
る。
ここでいうプソイドコロイドとは、水に溶解
してコロイド様の粘性溶液を生成する有機高分
子物質を意味し、とくにキサンタンガム及び／
又はマルトデキスリトンを含有するものとする
ことができる。
更に本発明による薬剤学的製剤は場合によつ
ては発泡性混合物を含有することを特徴とす
る。
この発泡性混合物としては、クエン酸及び炭
酸カルシウムを含有するものとすることがで
き、その際炭酸カルシウムは、炭酸カルシウム
とクエン酸の結晶の表面に近い層との反応によ
り生成した接着層の接着作用により、クエン酸
を被覆する。
本発明による種類の薬剤学的製剤の製造法
は、本発明によればイブプロフエン粒子を真空
混合機の中で真空下で少くとも一種のプソイド
コロイドとフマル酸とから成る外被で被覆し、
次いで真空乾燥することを特徴とする。
この場合特に、先ずイブプロフエンとプソイ
ドコロイド又はプソイドコロイドと水とを真空
混合機の中で約0.1バールの真空下で混合し、
次に約0.2バールで乾燥した後でフマル酸を添
加し、次いで完全に乾燥するようにすることが
できる。
本発明は、従来のイブプロフエン製剤の味覚
上のマイナス効果をイブプロフエンをプソイド
コロイドとフマル酸で被覆することにより除去
できるという知見に基づくもので、イブプロフ
エン結晶をプソイドコロイドで、それより低い
pH−値を有するフマル酸の存在下で被覆すれ
ば、この味覚上のマイナス効果を防止できる。
ここでプソイドコロイドは水に不溶性のイブプ
ロフエンと難溶性のフマル酸とを固定する機能
を果すだけである。本発明による薬剤学的製剤
の製造乃至本発明による方法の実験を、真空混
合機を用い、西独公開特許公報P3434774.7号の
主題をなす方法によつて行うのが、本目的のみ
ならずその他の点でも有利である。この点に関
する本発明の思想の補足的説明には前記西独公
開特許公報が全面的に引用される。
次に、実施例により本発明を詳細に説明す
る。
実施例 １ イブプロフエン200部を、キサンタンガム10
部とマルトデキストリン10部とを50部の水に溶
かした溶液で処理し、真空中で完全に乾燥する
前に、約200ミリバールでフマル酸20部を加え
る。完全に乾燥すると粒状物が得られ、これを
粒径約0.3mmに粉砕する。
この粒子を瞬間技法（instantaneous
technology）により砂糖の結晶に付着させる
ことができ、この場合好ましくは10〜15倍の量
の砂糖を使用し、又２倍量のクエン酸を加えて
もよい。
一回量20mgのイブプロフエンのこの種の混合
物を作れば、適当に香料を添加することにより
飲み易い飲料が得られ、これは粘膜に何等の刺
激を与えることがない。
実施例 ２ 前述と同じシステムは、これにクエン酸と炭
酸カルシウムとの発泡性混合物を追加すること
により更に改良できる。
クエン酸50部と炭酸カルシウム50部とに50％
エタノール５部を加えて真空下で反応させる。
この発泡性混合物を乾燥し、実施例１の方法で
処理したイブプロフエンに５倍量加える。
こうして作つた調合物は緩やかに泡を発生す
る混合物による自己懸濁効果を示し、この再存
在するカルシウムイオンは明らかにイブプロフ
エンの味覚を更に改善させる効果がある。

Claims (1)

  1. 【特許請求の範囲】 １ イブプロフエン粒子を少くとも一種のプソ
    イドコロイドおよびフマル酸から成る外被で被
    覆することを特徴とする、イブプロフエンを含
    有する薬剤学的製剤。 ２ プソイドコロイドがキサンテンを含有する
    ことを特徴とする特許請求の範囲第１項記載の
    製剤。 ３ プソイドコロイドがマルトデキストリンを
    含有することを特徴とする特許請求の範囲第１
    項又は第２項記載の製剤。 ４ 一種の発泡性混合物を含有することを特徴
    とする特許請求の範囲第１乃至３項の何れかに
    記載の製剤。 ５ 発泡性混合物がクエン酸と炭酸カルシウム
    を含有し、その際炭酸カルシウムは、炭酸カル
    シウムとクエン酸の結晶の表面に近い層との反
    応により生成した接着層の接着作用によりクエ
    ン酸を被覆することを特徴とする特許請求の範
    囲第４項記載の製剤。 ６ イブプロフエン粒子を真空混合機の中で真
    空下で、少くとも一種のプソイドコロイドとフ
    マル酸とから成る外被で被覆し、次いで真空乾
    燥することを特徴とする薬剤学的製剤の製造
    法。 ７ 先ずイブプロフエンとプソイドコロイド類
    と水とを真空混合機の中で約0.1バールの真空
    下で混合し、次に約0.2バールで乾燥した後で
    フマル酸を添加し、次いで完全に乾燥すること
    を特徴とする特許請求の範囲第６項記載の方
    法。
