Claims (6)
- 1.HCVポリヌクレオチドを有すると思われる分析物ストランドにおいてHC V配列を検出する方法であって、該HCVポリヌクレオチドが、選択された標的 領域を含み、(a)分析物ポリヌクレオチドストランドにおけるHCV配列にハ イブリダイズし得るオリゴマーを提供する工程であって、該オリゴマーが、図1 に示すHCVゲノムの以下の領域:16−45、49−78、82−111、1 15−144、148−177、211−240、242−271、275−3 04、332−361、365−394、398−427、および457−48 6、に相補的なオリゴヌクレオチドからなる群から選択されるポリヌクレオチド 配列を含む、工程; (b)該分析物ストランドを、標的する配列と標的配列との間に特異的なハイブ リッド二重鎖を形成し得る、該(a)のオリゴマーと共にインキュベートする工 程;および(d)標的領域が存在する場合には、該標的領域と、該オリゴマーと の間に形成されるハイブリッドを検出する工程、を包含する方法。
- 2.以下の工程: (a)オリゴマーのセットを提供する工程であって、該オリゴマーが、ポリメラ ーゼ鎖反応法のプライマーであり、前記標的領域に隣接して存在する、工程;お よび(b)該標的領域をポリメラーゼ領反応法により増幅する工程、 をさらに包含する、請求項1に記載の方法。
- 3.適切な容器にパッケージされた請求項1に記載のオリゴマーを含む、分析物 ストランド中のHCV標的配列を検出するためのキット。
- 4.HCVを含まない血液を調製する方法であって、(a)HCV標的配列を含 むと思われる血液試料からの分析物核酸を提供する工程; (b)分析物ポリヌクレオチドストランド中にHCV配列が存在する場合には、 該HCV配列にハイブリダイズし得るオリゴマーを提供する工程であって、該オ リゴマーが、図1に示すHCVゲノムの以下の領域:16−45、49−78、 82−111、115−144、148−177、211−240、242−2 71、275−304、332−361、365−394、398−427、お よび457−486に相補的なオリゴヌクレオチドからなる群から選択されるポ リヌクレオチド配列を含む、工程;(c)標的する配列と、標的配列が存在する 場合には該配列との間にポリヌクレオチド二本鎖を形成し得る条件下で、(a) と(b)とを反応させる工程; (d)(c)で形成される二本鎖が存在する場合には、該二本鎖を検出する工程 ;および (e)(d)において複合体が検出されなかった血液を保存する工程、 を包含する方法。
- 5.HCVの検出に有用な試薬であって、図1に示す塩基16−45、49−7 8、82−111、115−144、148−177、211−240、242 −271、275−304、332−361、365−394、398−427 、および457−486からなる群から選択されるHCVゲノムの領域に相補的 な配列を有するオリゴヌクレオチドを含む試薬。
- 6.前記オリゴヌクレオチドがさらに検出可能な標識を含む、請求項5に記載の 試薬。
