Description

【発明の詳細な説明】 〔産業上の利用分野〕 本発明は局所投与による炎症の治療そしてそのための製
薬組成物に関する。
〔従来の技術〕
英国特許第1474377号は式（Ｉ） 〔式中Ｘは塩素又は臭素原子又はメトキシル、メチルチ
オ又は１〜４個の炭素原子のアルキル基であり;Yは−CH
R1−CH2−又は−CR1＝CH−（式中R1は水素原子又はメチ
ル基である）でありそしてＡはCHOH又はCO基である〕の
化合物及び製薬上許容しうる担体よりなる製薬組成物と
炎症の治療におけるその用途を開示している。その特許
は式（Ｉ）の化合物の経口投与のみを教示しているのに
過ぎない。
〔発明の構成〕
本発明の要旨は、式（Ｉ）の好ましい化合物である式
（Ia） 〔式中Ｘはメトキシル基であり； Ｙは−CH2−CH2−基でありそしてＡはCO基である〕 の化合物及び製薬上許容しうる局所に有効な担体よりな
る局所投与用抗炎症剤である。
式（Ｉ）の化合物が予想されないことであつたが局限さ
れた局所の抗炎症活性を有しそして局所的に投与された
とき前身性の抗炎症活性を有することが見い出された。
従つて本発明はヒトを含む哺乳動物への局所投与による
炎症の治療又は予防のための薬剤を製造する式（Ｉ）の
化合物の用途を提供する。
本発明は又抗炎症の局所に有効な量の式（Ｉ）の化合物
の局所投与よりなるヒトを含む哺乳動物の炎症を治療又
は予防する方法も提供する。
式（Ｉ）の化合物は局所投与用の製薬組成物例えばクリ
ーム、軟膏、ローシヨン又はゲルとして投与されよう。
式（Ｉ）の化合物はアトピー性及び接触性皮膚炎、乾せ
ん、湿疹及び他の炎症性皮膚炎の局所的治療そして目、
耳、鼻及びのどの炎症に用いられよう。それらは又変形
性関節症及び慢性関節リウマチ及び捻座、挫傷及び滑液
のう炎の局所投与による治療に用いられよう。
用いられる式（Ｉ）の化合物の局所的に有効な量は多数
のフアクター例えば治療される病気の性質及び程度そし
て用いられる特定の化合物に依存することは理解されよ
う。しかし説明のため有効な治療はヒドロコーチゾンの
化合物で用いられているのとほとんど同様な量の式
（Ｉ）の化合物を用いて達成されうることが考えられて
いる。成人を治療するのに適当な代表的な処方は適当に
は式（Ｉ）の化合物の0.1〜10％さらに適当には0.5〜５
％を含むだろう。
本発明の治療法用の局所製薬組成物は１日１〜６回そし
てさらに普通には１日２〜４回投与されよう。
本発明は又局所用の製薬上許容しうる担体とともに式
（Ｉ）の化合物よりなる局所的に有効な抗炎症製薬組成
物を提供する。
式（Ｉ）の化合物について用いられるクリーム、軟膏、
ローシヨン又はゲルは当業者に周知の従来用いられてい
る処方であり例えば製剤学及び化粧学の標準的は教科書
例えばレオナード・ヒル・ブツクス（Leonard Hill Boo
ks）により刊行された「ハリーズ・コスメチコロジイ
（Harry′s Cosmeticology）」及び英国薬局方に記載さ
れている。標準的な乳化用の油中水又は水中油用媒体又
は無水ポリエチレングリコール基剤はこのような適当な
処方の例である。
標準的な乳化軟膏基剤に含まれるのに適した油の例は鉱
油、植物油、合成脂肪酸エステル、脂肪族アルコール、
ラノリン及びその誘導体を含む。
本発明の組成物は普通適当な乳化剤を含む。組成物は存
在するであろう濃厚化剤の量及びエマルジヨンの型に従
つて液体から半液体を経てゲル型に及ぶ。乳化剤の例は
多価アルコールエステル例えばソルビタンモノステアレ
ート、脂肪酸エステル例えばグリセリルモノステアレー
ト及び脂肪酸又は脂肪族アルコールのポリエステル誘導
体を含む。
組成物は又抗酸化剤及び他の従来用いられている成分例
えば保存剤、香料及びアルコールを含むだろう。有利に
は浸透剤例えばアゾン（Azone）も含まれよう。
本発明の組成物はさらに他の治療剤例えば抗感染症剤及
び／又は抗ウイルス剤も含みうる。適当な抗感染症剤は
既に局所の抗炎症剤において用いられている局所的に適
用されうる抗菌剤、抗イースト剤及び抗かび剤を含む。
抗ウイルス剤として特に抗ヘルペス剤が挙げられる。
式（Ｉ）の化合物においてＸは好ましくはメトキシであ
りＹは好ましくはCHR1−CH2（式中R1は水素原子であ
る）でありそしてＡは好ましくはCO基であつて本発明の
組成物及び治療に用いられる式（Ｉ）の好ましい化合物
は式（II） のそれである。
式（Ｉ）の他の化合物は又英国特許第1474377号に開示
されている。
式（Ｉ）及び（II）の化合物は英国特許第1474377号に
記載されているようにして製造されよう。式（II）の化
合物は又ヨーロッパ特許第0003374又は0003643号に記載
されているようにして製造されよう。
〔実施例〕
下記の薬理学上のデータは炎症の局所的治療又は予防に
おける式（Ｉ）の化合物の有効性を示す。薬理学上のデ
ータ 式（II）の化合物をジー・ビー・フレグナン（G.B.Freg
nan）及びエー・エル・トルセロ（A.L.Torsello）（197
5）により「カレント・テラピューチカル・リサーチ（C
urrent Therapeutical Research）」17,4号,375〜381ペ
ージに記載されたのに基く動物モデルの局所的抗炎症活
性についてテストした。大略方法は以下の通りである。
動物： ラツト，チヤールス・リバー・ウイスター種，メス,1郡
９匹，体重200〜240g。
刺戟剤： 適用される刺戟溶液はテトラヒドロフラン中１％クロト
ン油よりなる。
方法： 0.05mlの刺戟溶液を各耳につけ、テスト化合物を片方の
耳の刺戟溶液に含ませる。６時間後耳をヘアラインに沿
つて切断しそして重量を測る。
−Veコントロール；刺戟溶液なし。
＋Veコントロール；両方の耳に刺戟溶液あり。
結果を表に示すが式（II）の化合物は（II）で示す。
毒性 上述のテストにおいて毒性は認められなかつた。

    【特許請求の範囲】
  1. 【請求項１】式（Ia） 〔式中Ｘはメトキシル基であり； Ｙは−CH2−CH2−基でありそしてＡはCO基である〕 の化合物及び製薬上許容しうる局所に有効な担体よりな
    る局所投与用抗炎症剤。
  2. 【請求項２】式（Ia）の化合物が４−（６′−メトキシ
    −２′−ナフチル）ブタン−２−オンである特許請求の
    範囲第（１）項記載の局所投与用抗炎症剤。
