Description
Claims (1)
- 【特許請求の範囲】 1.標的結合性ポリペプチドであって、 a)安定なコアポリペプチド領域(SCR)と、 b)少なくとも一つの標的結合領域(TBR) とを具備し、 前記標的結合領域は、前記SCRに共有結合体に結合しており、また標的に対 する特異性、親和性または結合活性を改変するために、任意に成熟プロセスを受 けているポリペプチド。 2.請求項1に記載のポリペプチドであって、自己会合して安定な二量体、凝 集体またはアレイを形成することができるポリペプチド。 3.請求項1または2に記載のポリペプチドであって、前記SCRおよびTB Rがリンカー成分によって結合されているポリペプチド。 4.請求項1〜3の何れか1項に記載のポリペプチドであって、二官能性また は多官能性であるポリペプチド。 5.請求項1〜4の何れか1項に記載のポリペプチドであって、前記SCRが ヒト起源であり、また血清中に存在するか若しくは細胞表面に表現されるタイプ のタンパクであるポリペプチド。 6.請求項1〜5の何れか1項に記載のポリペプチドであって、前記TBRが 、グリコホリンもしくは他の赤血球細胞表面タンパク、インフルエンザウイルス ・ノイラミニダーゼ、 ウイルス抗原、抗体もしくは他のIgGファミリーのメンバー、腫瘍増殖因子α (TGF−α)、腫瘍マーカー、細胞表面タンパクおよび白血病阻害因子(LI F)からなる群から選択される標的に結合することができるポリペプチド。 7.請求項1〜6の何れか1項に記載の標的結合性ポリペプチドであって、、 修飾抗体もしくは抗体フラグメント、scFvフラグメント、修飾CD8分子、 および抗体分子もしくはそのフラグメントとCD8との組合せを含む、免疫グロ ブリンスーパーファミリーに対して相同性を有するポリペプチド。 8.請求項1〜7の何れか1項に記載の標的結合性ポリペプチドをコードする DNA構築物。 9.請求項1〜7の何れか1項に記載の標的結合性ポリペプチドをコードする DNA構築物を製造する方法であって、標的結合性ポリペプチドコードするDN Aを、突然変異および選別の一以上のサイクルに供して、改変された親和性、特 異性または結合活性の特性をもった標的結合性ポリペプチドをコードするDNA のサブポピュレーションを得ることを具備した方法。 10.請求項9に記載の方法であって、標的結合性ポリペプチドをコードする 前記DNAが、複製の能力のある要素または表現ベクター内に存在する方法。 11.請求項10に記載の方法であって、前記表現ベクターが、バクテリオフ ァージ、繊維状バクテリオファージ、ウイルス、バクテリア、酵母、粘菌、また は哺乳類細胞からな る群から選択される方法。 12.請求項10に記載の方法であて、前記表現ベクターがpHFAである方 法。 13.請求項12に記載の方法であって、突然変異が大腸菌の突然変異株を用 いて行なわれる方法。 14.請求項1〜7の何れか1項に記載の標的結合性ポリペプチドを製造する 方法であって、 a)一以上の所望の標的結合性ポリペプチドのフレームワーク構造をコードす るDNAを、ポリメラーゼチェイン反応によって単離する工程と、 b)該DNAを一以上の表現ベクターに挿入する工程と、 c)所望の特異性、結合活性または親和性の標的結合性ポリペプチドを表現す る表現ベクターのサブポピュレーションを選別することと、 d)改変された親和性、特異性または結合活性の特性を有する標的結合性ポリ ペプチドを表現する表現ベクターのサブポピュレーションを得るために、前記該 選別されたサブポピュレーションを、突然変異誘発および選別の一以上のサイク ルに供する工程と、 e)改変された標的結合性ポリペプチドをコードするDNAを、高レベル発現 ベクターに挿入する工程と、 f)上記の高レベル発現ベクターを適切な発現ホスト中に導入する工程と、 g)前記標的結合性タンパクを発現させる工程と、 h)こうして産生されたタンパクを単離する工程とを具備 した方法。 14.請求項1〜7の何れか1項に記載の標的結合性ポリペプチドと、薬学的 に許容され得る担体とを含有する薬剤組成物。 15.請求項1〜7の何れか1項に記載の標的結合性ポリペプチドと、稀釈剤 とを含有する診断試薬。 16.請求項1〜7の何れか1項に記載の標的結合性ポリペプチドの診断にお ける使用。 17.請求項1〜7の何れか1項に記載の標的結合性ポリペプチドの医薬とし ての使用。
