Claims (1)
- 【特許請求の範囲】 1.細胞内又は適当な無細胞系において: (a)リコンビナーゼ活性が、リガンド結合性ドメインに結合するリガンドの 非存在中で阻害され、そして (b)リコンビナーゼ活性が、リガンド結合性ドメインへのリガンドの結合に より誘導され又は変更されるように、核レセプタのリガンドに結合するアミノ酸 が含まれるように核レセプタの一部又は全部に融合された、リコンビナーゼ・タ ンパク質又はリコンビナーゼ複合体の成分を含んで成る、融合タンパク質。 2.核レセプタが、ホルモン・レセプタである、請求項1に記載の融合タンパ ク質。 3.核レセプタが、ステロイド・ホルモン・レセプタである、請求項1又は2 に記載の融合タンパク質。 4.核レセプタが、それが請求項1に記載の融合タンパク質の特徴を保持する ような核レセプタの突然変異された誘導体である、請求項1〜3のいずれか1項 に記載の融合タンパク質。 5.核レセプタが、脊椎動物のグルココルチコイド、エストロゲン、プロゲス テロン又はアンドロゲン・レセプタである、請求項1〜4のいずれかに記載の融 合タンパク質。 6.リコンビナーゼ・タンパク質又はリコンビナーゼ複合体の成分が:FLPリ コンビナーゼ、Creリコンビナーゼ、ザイゴサッカロミセス・ロウキシイ(Zygos accharomyces rouxii)プラスミドpSR1由来のRリコンビナーゼ、クルイベロ ミセス・ドロソフィラリウム(Kluyveromyces drosophilarium)プラスミドpKD 1由来のAリコンビナーゼ、クルイベロミセス・ワルチイ(Kluyveromyces wal tii )プラスミドpKW1由来のAリコンビナーゼ、λInt組換 え系のいずれかの成分、Gin組換え系のいずれかの成分、又はその変異体から成 る群から選ばれる、請求項1〜5のいずれか1項の融合タンパク質。 7.リコンビナーゼ・タンパク質又はリコンビナーゼ複合体の成分が、遺伝子 融合により核レセプタ又はそのリガンド結合性ドメインに融合される、請求項1 〜6のいずれかに記載の融合タンパク質。 8.核レセプタ又はそのリガンド結合性ドメインが、リコンビナーゼ・タンパ ク質又はリコンビナーゼ複合体の成分のN−又はC−末端に融合されている、請 求項1〜7のいずれかに記載の融合タンパク質。 9.核レセプタが、ペプチド・リンカーを介してリコンビナーゼ・タンパク質 又はリコンビナーゼ複合体の成分に融合されている、請求項1〜8のいずれかに 記載の融合タンパク質。 10.FLPリコンビナーゼとエストロゲン・レセプタのリガンド結合性ドメイン を含んで成る、請求項1〜9のいずれかに記載の融合タンパク質。 11.請求項1〜10の中のいずれか1項に記載の融合タンパク質をコードする核 酸。 12.DNA又はRNAである、請求項11に記載の核酸。 13.請求項11又は12の核酸の中の少なくとも1のコピーを含んで成る組換えベ クター。 14.プラスミドである、請求項13に記載のベクター。 15.プラスミドpHFE1(DSM9265)。 16.請求項11又は12に記載の核酸あるいは請求項13〜15の中のいずれか1項に 記載の組換えベクターを含む細胞。 17.哺乳動物細胞である、請求項16に記載の細胞。 18.核レセプタのリガンド結合性ドメインへのリガンドの結合を測定するため の方法であって、以下の段階: (a)組換えのためのDNA標的部位を含む、細胞又は適当な無細胞系内に、請 求項1〜9のいずれかに記載の融合タンパク質、又は請求項10又は11に記載の核 酸を導入し、 (b)場合により、評価されるべき、上記リガンド、又は、リガンド(単数又 は複数)を含むと疑われる混合物を投与し、 (c)組換え又は上記DNA標的部位間のその組換え速度における変化を検出す ることにより、それが存在する場合に、その融合タンパク質のリコンビナーゼ活 性を検出する、 を含んで成る方法。 19.DNA標的部位間の組換えが、それらの標的部位物理的編成の直接的な測定 により検出される、請求項18に記載の方法。 20.DNA標的部位間の組換えが、組換えの前後において標的部位を担持するDNA 領域によりコードされる特性を評価することにより検出される、請求項18に記載 の方法。 21.リガンドに結合するその能力に対する核レセプタのリガンド結合性ドメイ ン内の突然変異の効果を測定するための方法であって、以下の段階: (a)請求項1〜10のいずれかに記載の融合タンパク質のリガンド結合性ドメ イン内に突然変異を導入し、 (b)請求項18に記載の段階(a)〜(c)に従う を含んで成る方法。 22.DNA標的部位の組換えの調節方法であって、以下の段階: (a)部位特異的リコンビナーゼのためのDNA標的部位を含む細胞又は適当な 無細胞系を提供し、 (b)請求項1〜10のいずれか1項に記載の融合タンパク質とDN A標的部位を接触させ、そして (c)上記融合タンパク質の核レセプタ成分についてのリガンドとその融合タ ンパク質を接触させることにより上記DNA標的部位の組換えを行う、 を含んで成る方法。
