Claims (1)
- 【特許請求の範囲】 1.(a)水性媒体中に実質的の不溶性の生体適合性で熱可塑性のポリマー; (b)水性媒体に混和または分散しうる生体適合性の有機溶媒;および (c)活性剤を含む制御放出成分 を含む、制御放出インプラントを形成するのに適した液体デリバリー組成物。 2.(a)水性媒体に実質的に不溶性の第一の生体適合性で熱可塑性のポリマーに 活性剤が共有結合している、活性剤/ポリマーコンジュゲート;および (b)水性媒体に混和または分散しうる生体適合性の有機溶媒 を含む、制御放出インプラントを形成するのに適した液体デリバリー組成物。 3.水性媒体に実質的に不溶性の第二の生体適合性で熱可塑性のポリマーをさら に含む、請求項2に記載の液体デリバリー組成物。 4.水性媒体と接触したときにスキンによって取り囲まれたコアを有する固体の 微多孔性マトリックスを形成することができ、該スキンが該コアの孔よりも実質 的に小さな直径を有することを特徴とする、請求項1または2に記載の液体デリ バリー組成物。 5.スキンが約5%〜約10%の多孔性を有し、コアが約40%〜約60%の多 孔性を有する、請求項4に記載の液体デリバリー組成物。 6.組成物のエーロゾル化を有効に可能にする粘度を有する、請求項1または2 に記載の液体デリバリー組成物。 7.(a)水性媒体に実質的に不溶性の生体適合性のポリマーの固体微多孔性マト リックス;および (b)活性剤を含み、微多孔性マトリックス内に埋設された制御放出成分 を含み、 その際、該マトリックスは水性媒体と請求項1または2に記載の液体デリバリー 組成物との接触により調製されることを特徴とする、制御放出インプラントとし て使用するのに適したポリマーシステム。 8.請求項1または2に記載の液体デリバリー組成物から形成された生分解性の 微多孔性フィルム包帯。 9.請求項1または2に記載の液体デリバリー組成物から形成され、外側の袋お よび液体充填物からなる構造を有する、患者に植え込むための制御放出インプラ ント前駆体。 10.熱可塑性ポリマーが、ポリラクチド、ポリグリコリド、ポリカプロラクト ン、ポリ無水物、ポリアミド、ポリウレタン、ポリエステルアミド、ポリオルト エステル、ポリジオキサノン、ポリアセタール、ポリケタール、ポリカーボネー ト、ポリオルトエステル、ポリホスファゼン、ポリヒドロキシブチレート、ポリ ヒドロキシバレレート、ポリアルキレンオキサレート、ポリアルキレンスクシネ ート、ポリ(リンゴ酸)、ポリ(アミノ酸)、ポリ(メチルビニルエーテル)、ポリ( 無水マレイン酸)、キチン、キトサン、およびこれらのコポリマー、ターポリマ ー、および混合物よりなる群から選ばれたものである、請求項1、2、7、8ま たは9のいずれか。 11.熱可塑性ポリマーが、ポリグリコリド、ポリ(D,L−ラクチド)、ポリカ プロラクトン、ポリオルトエステル、ポリカーボネート、ポリアミド、ポリ無水 物、ポリウレタン、ポリエステルアミド、ポリホスファゼン、ポリヒドロキシブ チレート、ポリヒドロキシバレレート、ポリアルキレンオキサレート、およびこ れらのコポリマー、ターポリマー、および混合物よりなる群から選ばれたもので ある、請求項1、2、7、8または9のいずれか。 12.熱可塑性ポリマーが、グリコリド、カプロラクトンまたはラクチドのコポ リマー、またはD,L−ラクチドおよびマロラクトン酸のコポリマーである、請 求項1、2、7、8または9のいずれか。 13.熱可塑性ポリマーが生分解性、生腐食性またはその両者である、請求項1 、2、7、8または9のいずれか。 14.有機溶媒が、N−メチル−2−ピロリドン、2−ピロリドン、2〜8の炭 素原子を有する脂肪族アルコール、プロピレングリコール、グリセリン、テトラ グリコール、グリセリンホルマール、ソルケタール、酢酸エチル、乳酸エチル、 酪酸エチル、マロン酸ジブチル、クエン酸三ブチル、アセチルクエン酸三n−ヘ キシル、コハク酸ジエチル、グルタール酸ジエチル、マロン酸ジエチル、クエン 酸三エチル、トリアセチン、トリブチリン、炭酸ジエチル、炭酸プロピレン、ア セトン、メチルエチルケトン、ジメチルアセトアミド、ジメチルホルムアミド、 カプロラクタム、ジメチルスルホキシド、ジメチルスルホン、テトラヒドロフラ ン、カプロラクタム、デシルメチルスルホキシド、オレイン酸、N,N−ジエチ ル−m−トルアミド、およびドデシルアザシクロヘプタン−2−オン、1,3− ジメチル−3,4,5,6−テトラヒドロ−2(1H)−ピリミジノン、およびこれ らの混合物よりなる群から選ばれたものである、請求項1、2、7、8または9 のいずれか。 15.少なくとも一つの重合可能なエチレン性不飽和基を有する液体の生体適合 性プレポリマー、および活性剤を含む制御放出成分を含む、制御放出インプラン トを形成するのに適した液体デリバリー組成物。 16.硬化剤をさらに含む請求項15に記載の液体プレポリマー組成物。 17.重合可能なエチレン性不飽和基がα,β−不飽和カルボニル基である、請 求項15に記載の液体プレポリマー組成物。 18.液体の生体適合性プレポリマーがアクリル酸エステル末端プレポリマーで ある、請求項15に記載の液体プレポリマー組成物。 19.水性媒体に混和または分散しうる生体適合性の有機溶媒をさらに含む、請 求項15に記載の液体プレポリマー組成物。 20.制御放出成分が、マイクロ構造、マクロ構造、活性剤の低水溶性塩、担体 分子に共有結合した活性剤のコンジュゲート、活性剤と担体分子との錯体、また は活性剤、担体分子および金属カチオンの錯体である、請求項1、7、8、9ま たは15のいずれか。 21.制御放出成分が、マイクロカプセル、マイクロ粒子、ナノ粒子、シクロデ キストリン、リポソーム、ミセル、繊維、フィルム、棒、円盤、および円筒より なる群から選ばれたものである、請求項1、2、7、8、9または15のいずれ か。 22.活性剤が生物活性剤または診断剤である請求項1、2、7、8、9または 15のいずれか。 23.活性剤が抗菌剤、抗真菌剤、抗ウイルス剤、抗炎症剤、抗寄生虫剤、抗新 生物剤、鎮痛剤、麻酔薬、抗精神病薬、ワクチン、中枢神経系剤、成長因子、ホ ルモン、抗ヒスタミン剤、骨形成剤、心血管剤、抗腫瘍剤、気管支拡張剤、血管 拡張剤、産児制限剤、抗高血圧症剤、抗凝血剤、鎮痙剤、および排卵促進剤より なる群から選ばれたものである、請求項1、2、7、8、9または15のいずれ か。 24.液体デリバリー組成物が生理学的に許容しうる、放出速度調節剤、孔形成 剤、またはその両者をさらに含む、請求項1、2、7、8、9または15のいず れか。 25.患者に活性剤を送達するための制御放出インプラントを形成するための医 薬の製造のための、請求項1、2または15のいずれかに記載の液体デリバリー 組成物の使用。 26.請求項1、2または15のいずれかに記載の液体デリバリー組成物を患者 のインプラント部位に投与し、ついで該組成物から、制御放出成分がその中に埋 設された固体のインプラントを形成させることを特徴とする、患者のインプラン ト部位中またはインプラント部位上にインシトゥで微多孔性の除放インプラント を形成する方法。 27.請求項1、2または15のいずれかに記載の液体デリバリー組成物を患者 のインプラント部位に投与し、その際、制御放出成分がその中に埋設された固体 のインプラントが形成されることを特徴とする、活性剤を患者に送達する方法。 28.液体デリバリー組成物を組織上に分配してフィルムを形成させ、該組成物 は該組織上で固化してフィルム包帯を形成する、請求項26または27に記載の 方法。 29.液体デリバリー組成物をインプラント部位に噴霧、塗布または噴射するこ とにより分配する請求項26、27または28に記載の方法。 30.請求項1または2に記載の液体デリバリー組成物を水性媒体と接触させる ことにより患者の体外で形成した微多孔性の除放インプラントを患者のインプラ ント部位に挿入することを特徴とする、活性剤を患者に送達する方法。
