- 【特許請求の範囲】 1.植物性タンパク質源中のタンパク質を可溶化するための方法であって、次 の工程: (a)有効量の1種又はそれ以上のフィターゼ酵素を用いて植物性タンパク質源 を処理し;次いで (b)有効量の1種又はそれ以上のタンパク質分解酵素を用いて植物性タンパク 質源を処理することを含んでなる、前記方法。 2.工程(a)および工程(b)を同時に行なう、請求の範囲第1項記載の方 法。 3.更に(c)有効量の1種又はそれ以上の脂肪分解酵素(EC3.1.1.3 )および1種又はそれ以上のグリコシダーゼ酵素(EC3.2)を用いて植物性タ ンパク質源を処理する工程を含んでなる、請求の範囲第1又は2項記載の方法。 4.グルコシダーゼ酵素が、アミラーゼ特にα−アミラーゼもしくはβ−アミ ラーゼ、セルラーゼ、特にエンド−1,4−β−グルカナーゼ(EC3.2.1. 4)又はエンド−1,3−β−グルカナーゼ(3.2.1.6)、キシラナーゼ 、特にエンド−1,4−β−グルカナーゼ(EC3.2.1.8)又はキシラン− エンド−1,3−β−キシロシダーゼ(EC3.2.1.32)、α−ガラクトシダ ーゼ(EC3.2.1.22)、ポリガラクツロナーゼ(EC3.2.1.15)、セル ロース−1,4−β−セロビオシダーゼ(EC3.2.1.91)、エンドグルカナ ーゼ、特にエンド−1,6−β−グルカナーゼ(EC3.2.1.75)、エンド− 1,2−β−グルカナーゼ(EC3.2.1.71)、エンド−1,3−β−グルカ ナーゼ(EC3.2.1.39)又はエンド−1,3−α−グルカナーゼ(EC3.2 .1.59)である、請求の範囲第3項記載の方法。 5.グルコシダーゼ酵素が、ポリガラクツロナーゼ(EC3.2.1.15)、セ ルロース−1,4−β−セロビオシダーゼ(EC3.2.1.91)、又はエンドグ ルカナーゼ、好ましくはエンド−1,6−β−グルカナーゼ(EC3.2.1.75 )、エンド−1,2−β−グルカナーゼ(EC3.2.1.71)、エンド−1,3 −β−グルカナーゼ(EC3.2.1.39)又はエンド−1,3−α−グルカナー ゼ(EC3.2.1.59)である、請求の範囲第3項記載の方法。 6.工程(a),(b)および(c)を同時に行なう、請求の範囲第3〜5項 のいずれか1項に記載の方法。 7.工程(a)のフィターゼが、植物フィターゼ、菌類フィターゼ、細菌フィ ターゼ又は酵母により生産可能なフィターゼである、請求の範囲第1〜6項のい ずれか1項に記載の方法。 8.菌類フィターゼが、アスペルギルス(Aspergillus)菌株、好ましくはアス ペルギルス ニガー(Aspergillus niger)菌株、アスペルギルス フィクム(Asp ergillus ficuum)菌株、アスペルギルス アワモリ(Aspergillus awamori)菌株 、アスペルギルス オリゼ(Aspergillus oryzae)菌株、アスペルギルス テレ ウス(Aspergillus terreus)菌株、又はアスペルギルス ニドランス(Aspergill us nidulans)菌株から由来する、請求の範囲第7項記載の方法。 9.細菌フィターゼが、バシラス(Bacillus)菌株又はシュードモナス(Pseud omonas)菌株、好ましくはバシラス スブチリス(Bacillus subtilis)菌株から由 来する、請求の範囲第7項記載の方法。 10.フィターゼが属クルベロマイセス(Kluveromyces)又はサッカロマイセス (Saccharomyces)の酵母、好ましくはサッカロマイセス セレビシエ(Saccharom yces cerevisiae)菌株から由来する、 請求の範囲第7項記載の方法。 11.植物性タンパク質源を、1kgのタンパク質源当たり約2〜約50000フィタ ーゼ単位(FYT)、好ましくは1kgのタンパク質源当たり約50〜約30000FYT、最も 好ましくは1kgのタンパク質源当たり約100〜約10000FYTの量で工程(a)のフ ィターゼを用いて処理する、請求の範囲第1〜10項のいずれか1項に記載の方法 。 12.工程(b)のタンパク質分解酵素が、微生物プロテアーゼ、菌類プロテア ーゼ、又はペプシンもしくはトリプシンである、請求の範囲第1〜11項のいずれ か1項に記載の方法。 13.微生物プロテアーゼが、バシラス(Bacillus)菌株、好ましくはバシラス リケニホルミス(Bacillus licheniformis)菌株、および/又はバシラス ス ブチリス(Bacillus subtilis)菌株から由来する、請求の範囲第12項記載の方法 。 14.菌類プロテアーゼが、アスペルギルス菌株、好ましくはアスペルギルス アクレタス(Aspergillus aculeatus)菌株、アスペルギルス ニガー(Aspergillu s niger)菌株、アスペルギルス オリゼ(Aspergillus oryzae)菌株から由来す る、請求の範囲第12項に記載の方法。 15.1kgのタンパク質源当たり約0.0001〜約0.5AU、好ましくは1kgのタンパ ク質源当たり約0.001〜約0.05AU、より好ましくは1kgのタンパク質当たり約0.0 05〜約0.03AUの量で工程(b)の1種又はそれ以上のタンパク質分解酵素を用い て植物性タンパク質源を処理する、請求の範囲第1〜14項のいずれか1項に記載 の方法。 16.植物性タンパク質源が、マメ科植物、キク科植物、十字花科植物、又は穀 物植物又はそれらの混合物である、請求の範囲第1〜15項のいずれか1項に記載 の方法。 17.植物性タンパク質源が大豆である、請求の範囲第1〜15項のい ずれか1項に記載の方法。 18.酵素処理を、3〜9のpH、好ましくは4〜7のpHで行なう、請求の範囲第 1〜17項のいずれか1項に記載の方法。 19.酵素処理を、約10〜約70℃の温度、好ましくは約35℃〜約65℃の温度で行 なう、請求の範囲第1〜18項のいずれか1項に記載の方法。 20.i)植物性タンパク質源を水中に懸濁し; ii)工程1)の水性懸濁液を、フィターゼおよびタンパク質分解酵素を用いて 酵素処理し; (iii)工程ii)の酵素を失活させ;次いで所望により (iv)可溶化タンパク質を水性懸濁液から分離する 工程を含んでなる、請求の範囲第1〜19項のいずれか1項に記載の方法。 21.フィターゼおよびタンパク質分解酵素を含んでなる、動物飼料添加剤。 22.脂肪分解酵素(EC3.1.1.3)およびグリコシダーゼ酵素(EC3.2 )から成る群から選ばれる1種又はそれ以上の追加の酵素を含んでなる、請求の 範囲第22項記載の動物飼料添加剤。 23.グルコシダーゼ酵素が、アミラーゼ特にα−アミラーゼもしくはβ−アミ ラーゼ、セルラーゼ、特にエンド−1,4−β−グルカナーゼ(EC3.2.1. 4)又はエンド−1,3−β−グルカナーゼ(3.2.1.6)、キシラナーゼ 、特にエンド−1,4−β−グルカナーゼ(EC3.2.1.8)又はキシラン− エンド−1,3−β−キシロシダーゼ(EC3.2.1.32)、α−ガラクトシダ ーゼ(EC3.2.1.22)、ポリガラクツロナーゼ(EC3.2.1.15)、セル ロース−1,4−β−セロビオシダーゼ(EC3.2.1.91)、エンドグルカナ ーゼ、特にエンド−1,6−β−グルカ ナーゼ(EC3.2.1.75)、エンド−1,2−β−グルカナーゼ(EC3.2. 1.71)、エンド−1,3−β−グルカナーゼ(EC3.2.1.39)又はエンド −1,3−α−グルカナーゼ(EC3.2.1.59)である、請求の範囲第22項記 載の動物飼料添加剤。 24.グルコシダーゼ酵素が、ポリガラクツロナーゼ(EC3.2.1.15)、セ ルロース−1,4−β−セロビオシダーゼ(EC3.2.1.91)、又はエンドグ ルカナーゼ、好ましくはエンド−1,6−β−グルカナーゼ(EC3.2.1.75 )、エンド−1,2−β−グルカナーゼ(EC3.2.1.71)、エンド−1,3 −β−グルカナーゼ(EC3.2.1.39)又はエンド−1,3−α−グルカナー ゼ(EC3.2.1.59)である、請求の範囲第22項記載の動物飼料添加剤。 25.フィターゼが、植物フィターゼ、菌類フィターゼ、細菌フィターゼ又は酵 母により生産可能なフィターゼである、請求の範囲第21〜24項のいずれか1項に 記載の動物飼料添加剤。 26.菌類フィターゼが、アスペルギルス(Aspergillus)菌株、好ましくはアス ペルギルス ニガー(Aspergillus niger)菌株、アスペルギルス フィクム(Asp ergillus ficuum)菌株、アスペルギルス アワモリ(Aspergillus awamori)菌株 、アスペルギルス オリゼ(Aspergillus oryzae)菌株、アスペルギルス テレ ウス(Aspergillus terreus)菌株、又はアスペルギルス ニドランス(Aspergil lus nidulans)菌株から由来する、請求の範囲第25項記載の動物飼料添加剤。 27.細菌フィターゼが、バシラス(Bacillus)菌株又はシュードモナス(Pseud omonas)菌株、好ましくはバシラス スブチリス(Bacillus subtilis)菌株から由 来する、請求の範囲第25項記載の動物飼料添加剤。 28.フィターゼが、属クルベロマイセス(Kluveromyces)又はサッカロマイセ ス(Saccharomyces)の酵母、好ましくはサッカロマイセス セレビシエ(Sacchar omyces cerevisiae)菌株から由来する、請求の範囲第25項記載の動物飼料添加 剤。 29.組成物1g当たり約200〜約50000フィターゼ単位(FYT)、好ましくは、組 成物1g当たり約500〜約10000FYT、より好ましくは組成物1g当たり約2000〜 約6000FYTに相当する量の1種又はそれ以上のフィターゼ酵素を含んでなる、請 求の範囲第21〜28項のいずれか1項に記載の動物飼料添加剤。 30.タンパク質分解酵素が、微生物プロテアーゼ、菌類プロテアーゼ、又はペ プシンもしくはトリプシンである、請求の範囲第21〜29項のいずれか1項に記載 の動物飼料添加剤。 31.微生物プロテアーゼが、バシラス(Bacillus)菌株、好ましくはバシラス リケニホルミス(Bacillus licheniformis)菌株、および/又はバシラス ス ブチリス(Bacillus subtilis)菌株から由来する、請求の範囲第30項記載の動物 飼料添加剤。 32.菌類プロテアーゼが、アスペルギルス菌株、好ましくはアスペルギルス アクレタス(Aspergillus aculeatus)菌株、アスペルギルス ニガー(Aspergillu s niger)菌株、アスペルギルス オリゼ(Aspergillus oryzae)菌株から由来す る、請求の範囲第30項に記載の動物飼料添加剤。 33.組成物1g当たり約0.01〜約20AU(アンソン単位)、好ましくは組成物1 g当たり約0.1〜約10AU、より好ましくは組成物1g当たり約0.2〜約5AUに相当 する量のタンパク質分解酵素を含んでなる、請求の範囲第21〜32項のいずれか1 項に記載の動物飼料添加剤。 34.1種又はそれ以上の酵素が、粉末、無粉塵性粒質物、安定化 液体、スラリー又は保護された酵素の形で提供される、請求の範囲第21〜23項の いずれか1項に記載の動物飼料添加剤。
