- 【特許請求の範囲】 1.(a)キメラペントンベースタンパク質および(b)治療遺伝子を含有する組換え アデノウイルスであって、該キメラペントンベースタンパク質が、受容体との結 合に特異的な野生型ペントンベースアミノ酸配列に加えて、あるいはそれの代わ りに、受容体との結合に特異的であるか、受容体特異的抗体ドメインであるか、 またはエピトープである非ペントンベースアミノ酸配列を含み、また、該治療遺 伝子が、該組換えアデノウイルスが接着するか、あるいは内部移行された細胞内 で発現し得るものである当該アデノウイルス。 2.該野生型ペントンベースアミノ酸配列が、RGDアミノ酸配列である請求の 範囲1の組換えアデノウイルス。 3.該組換えアデノウイルスがファイバーを有しない請求の範囲2の組換えアデ ノウイルス。 4.該キメラペントンベースタンパク質に加えて、さらに、受容体との結合に特 異的であるか、受容体特異的抗体ドメインであるか、またはエピトープであるア ミノ酸配列を含むキメラコートタンパク質を含有する請求の範囲2の組換えアデ ノウイルス。 5.該キメラコートタンパク質中の、受容体との結合に特異的であるか、受容体 特異的抗体ドメインであるか、またはエピトープである該アミノ酸配列が、該キ メラペントンベースタンパク質中の非ペントンベースアミノ酸配列と同じ受容体 、抗体ドメイン、またはエピトープとの結合に特異的なものである請求の範囲4 の組換えアデノウイルス。 6.該キメラコートタンパク質が、キメラファイバーおよびキメラヘキソンから なる群より選択される請求の範囲4の組換えアデノウイルス。 7.該キメラコートタンパク質中の、受容体との結合に特異的であるか、受容体 特異的抗体ドメインであるか、またはエピトープである該アミノ酸配列が、該キ メラペントンベースタンパク質中の非ペントンベースアミノ酸配列と同じ受容体 、抗体ドメイン、またはエピトープとの結合に特異的なものである請求の範囲6 の組換えアデノウイルス。 8.キメラペントンベースタンパク質をコードするキメラアデノウイルスペント ンベース遺伝子配列を含有する組換えバキュロウイルスであって、キメラアデノ ウイルスペントンベース遺伝子配列が、(a)受容体との結合に特異的であるか、 受容体特異的抗体ドメインであるか、またはエピトープであるアミノ酸配列をコ ードする非ペントンベース遺伝子配列および(b)受容体との結合に特異的なアミ ノ酸配列をコードする野生型ペントンベース遺伝子配列を含む当該バキュロウイ ルス。 9.該野生型ペントンベース遺伝子配列が、RGDアミノ酸配列をコードする請 求の範囲8の組換えバキュロウイルス。 10.RGDアミノ酸配列をコードする遺伝子配列が変異した請求の範囲9の組 換えバキュロウイルス。 11.RGDアミノ酸配列をコードする遺伝子配列が、RGDアミノ酸配列をコ ードする遺伝子配列への非ペントンベース遺伝子配列の挿入によって変異した請 求の範囲10の組換えバキュロウイルス。 12.キメラペントンベースタンパク質中で、RGDアミノ酸配列が配座的に受 容体と結合できないように、RGDアミノ酸配列をコードする遺伝子配列がペン トンベース遺伝子配列内を移動させられた請求の範囲9の組換えバキュロウイル ス。 13.RGDアミノ酸配列をコードする遺伝子配列が、RGDアミノ酸配列をコ ードする遺伝子配列の近傍への非ペントンベース遺伝子配列の挿入によってペン トンベース遺伝子配列内を移動させられた請求の範囲12の組換えバキュロウイ ルス。 14.受容体との結合に特異的なアミノ酸配列をコードする野生型ペントンベー ス遺伝子配列の代わりに、受容体との結合に特異的であるか、受容体特異的抗体 ドメインであるか、またはエピトープであるアミノ酸配列をコードする非ペント ンベース遺伝子配列を含むキメラアデノウイルスペントンベース遺伝子配列を含 有する組換えバキュロウイルス。 15.該野生型ペントンベース遺伝子配列が、RGDアミノ酸配列をコードする 請求の範囲14の組換えバキュロウイルス。 16.受容体との結合に特異的な野生型ペントンベースアミノ酸配列に加えて、 あるいはそれの代わりに、受容体との結合に特異的であるか、受容体特異的抗体 ドメインであるか、またはエピトープである非ペントンベースアミノ酸配列を含 むキメラアデノウイルスペントンベースタンパク質。 17.該野生型ペントンベースアミノ酸配列が、RGDアミノ酸配列である請求 の範囲16のキメラアデノウイルスペントンベースタンパク質。 18.キメラペントンベースタンパク質をコードするキメラアデノウイルスペン トンベース遺伝子配列を含有するウイルス運搬ベクターであって、該キメラアデ ノウイルスペントンベース遺伝子配列が、受容体との結合に特異的なアミノ酸配 列をコードする野生型ペントンベース遺伝子配列に加えて、受容体との結合に特 異的であるか、受容体特異的抗体ドメインであるか、またはエピトープであるア ミノ酸配列をコードする非ペントンベース遺伝子配列を含むものである当該ベク ター。 19.該ウイルス運搬ベクターがアデノウイルス運搬ベクターである請求の範囲 18のウイルス運搬ベクター。 20.該野生型ペントンベース遺伝子配列がRGDアミノ酸配列をコードする請 求の範囲19のウイルス運搬ベクター。 21.RGDアミノ酸配列をコードする遺伝子配列が変異した請求の範囲20の ウイルス運搬ベクター。 22.RGDアミノ酸配列をコードする遺伝子配列が、RGDアミノ酸配列をコ ードする遺伝子配列中への非ペントンベース遺伝子配列の挿入によって変異した 請求の範囲21のウイルス運搬ベクター。 23.キメラペントンベースタンパク質中で、RGDアミノ酸配列が配座的に受 容体と結合できないように、RGDアミノ酸配列をコードする遺伝子配列がペン トンベース遺伝子配列内を移動させられた請求の範囲20のウイルス運搬ベクタ ー。 24.RGDアミノ酸配列をコードする遺伝子配列が、RGDアミノ酸配列をコ ードする遺伝子配列の近傍への非ペントンベース遺伝子配列の挿入によってペン トンベース遺伝子配列内を移動させられた請求の範囲23のウイルス運搬ベクタ ー。 25.キメラペントンベースタンパク質をコードするキメラアデノウイルスペン トンベース遺伝子配列を含有するウイルス運搬ベクターであって、該キメラアデ ノウイルスペントンベース遺伝子配列が、受容体との結合に特異的なアミノ酸配 列をコードする野生型ペントンベース遺伝子配列の代わりに、受容体との結合に 特異的であるか、受容体特異的抗体ドメインであるか、またはエピトープである アミノ酸配列をコードする非ペントンベース遺伝子配列を含むものである当該ベ クター。 26.該ウイルス運搬ベクターがアデノウイルス運搬ベクターである請求の範囲 25のウイルス運搬ベクター。 27.該野生型ペントンベース遺伝子配列がRGDアミノ酸配列をコードする請 求の範囲26のウイルス運搬ベクター。 28.キメラペントンベースタンパク質をコードするキメラアデノウイルスペン トンベース遺伝子配列を含有する原核または真核生物の発現ベクターであって、 該キメラアデノウイルスペントンベース遺伝子配列が、受容体との結合に特異的 なアミノ酸配列をコードする野生型ペントンベース遺伝子配列の代わりに、受容 体との結合に特異的であるか、受容体特異的抗体ドメインであるか、またはエピ トープであるアミノ酸配列をコードする非ペントンベース遺伝子配列を含むもの である当該ベクター。 29.該野生型ペントンベース遺伝子配列がRGDアミノ酸配列をコードする請 求の範囲28の原核または真核生物の発現ベクター。 30.キメラペントンベースタンパク質をコードするキメラアデノウイルスペン トンベース遺伝子配列を含有する原核または真核生物の発現ベクターであって、 該キメラアデノウイルスペントンベース遺伝子配列が、受容体との結合に特異的 なアミノ酸配列をコードする野生型ペントンベース遺伝子配列に加えて、受容体 との結合に特異的であるか、受容体特異的抗体ドメインであるか、またはエピト ープであるアミノ酸配列をコードする非ペントンベース遺伝子配列を含むもので ある当該ベクター。 31.該野生型ペントンベース遺伝子配列がRGDアミノ酸配列をコードする請 求の範囲30の原核または真核生物の発現ベクター。 32.RGDアミノ酸配列をコードする遺伝子配列が変異した請求の範囲31の 原核または真核生物の発現ベクター。 33.RGDアミノ酸配列をコードする遺伝子配列が、RGDアミノ酸配列をコ ードする遺伝子配列中への非ペントンベース遺伝子配列の挿入によって変異した 請求の範囲32の原核または真核生物の発現ベクター。 34.キメラペントンベースタンパク質内で、RGDアミノ酸配列が配座的に受 容体と結合できないように、RGDアミノ酸配列をコードする遺伝子配列がペン トンベース遺伝子配列内を移動させられた請求の範囲31の原核または真核生物 の発現ベクター。 35.RGDアミノ酸配列をコードする遺伝子配列が、RGDアミノ酸配列をコ ードする遺伝子配列の近傍への非ペントンベース遺伝子配列の挿入によってペン トンベース遺伝子配列内を移動させられた請求の範囲34の原核または真核生物 の発現ベクター。 36.遺伝子治療を必要とする患者に治療上有効な量の請求の範囲1の組換えア デノウイルスを投与することを含む遺伝子治療方法。 37.遺伝子治療を必要とする患者に、請求の範囲2の組換えアデノウイルスを 治療上有効な量投与することを含む遺伝子治療方法。 38.遺伝子治療を必要とする患者に、請求の範囲3の組換えアデノウイルスを 治療上有効な量投与することを含む遺伝子治療方法。 39.遺伝子治療を必要とする患者に、請求の範囲4の組換えアデノウイルスを 治療上有効な量投与することを含む遺伝子治療方法。 40.遺伝子治療を必要とする患者に、請求の範囲5の組換えアデノウイルスを 治療上有効な量投与することを含む遺伝子治療方法。 41.遺伝子治療を必要とする患者に、請求の範囲6の組換えアデノウイルスを 治療上有効な量投与することを含む遺伝子治療方法。 42.遺伝子治療を必要とする患者に、請求の範囲7の組換えアデノウイルスを 治療上有効な量投与することを含む遺伝子治療方法。
