Claims (1)
- 【特許請求の範囲】 1.細菌宿主内におけるタンパク質の発現に有用な組換えDNA構築体において、 タンパク質のコード領域がそのタンパク質の宿主内での発現を可能にするプロモ ーターに操作可能に連結してなり、プロモーター領域は 5′-TTGACAACATAAAAAACTTTGTGTTATACT-3′ および 5′-TTGACACTTTATGCTTCCGGCTCGTATACT-3′ から選択されるDNA配列からなる組換えDNA構築体。 2.プロモーター領域はDNA配列 5′-TTGACAACATAAAAAACTTTGTGTTATACT-3′ からなる請求項1記載の組換えDNA構築体。 3.細菌宿主は大腸菌である請求項1記載の組換えDNA構築体。 4.制御領域はさらにタンパク質の発現を調節するオペレーターからなる請求項 3記載の組換えDNA構築体。 5.オペレーターはlacオペレーターである請求項4記載の組換えDNA構築体。 6.タンパク質はヒトPTHである請求項5記載の組換えDNA構築体。 7.タンパク質の宿主ペリプラスムへの分泌を可能にする、コード領域とリーデ ィングフレームの合致したOmpAシグナルペプチドをコードするDNAからさらに構 成される請求項6記載の組換えDNA構築体。 8.制御領域はDNA配列:ATTAAAGAGGAGAAATTAAGCをもつT5リボソーム結合部位 からなる請求項3記載の組換えDNA構築体。 9.図2に定義された配列を有する請求項3記載の組換えDNA構築体。 10.図3に定義された配列を有する請求項3記載の組換えDNA構築体。 11.請求項1〜10のいずれかに記載の組換えDNA構築体からなる、細菌宿主細胞 トランスフォーマントの産生に有用なベクター。
