Abstract
PURPOSE: To obtain the title compound useful as analgesics and antiphlogistics in a few steps and in high yield, by reacting 3-(2-fluoro-4-biphenylyl)-2-butanone with a hypohalogenite.
CONSTITUTION: 2-Fluoro-biphenylylacetic acid of formula IV is reacted with methyllithium in an anhydrous solvent, e.g. ethyl ether, and the reaction product is hydrolyzed to give a novel compound of formula III, whose methylene group at the α-position is reacted with a methyl halide in the presence of an alkali condensing agent, e.g. an alkali hydroxide or alkali metal alcoholate, in a solvent, e.g. methanol, to form a compound of formula II. The resulting compound is then reacted with an equivalent amount or slight excess of a hypohalogenite in a solvent, e.g. water or dioxane, at -5-W50°C for 2W24hr to give the compound of formula I.
