Description
Claims (7)
- 억제성 기질이 탄소원으로 사용되고, 상기 탄소원이 공급 단계동안 제한되고, 전체 배양 기간동안 연속적인 산소 제한이 없음을 특징으로하는, 형질전환되거나 비형질전환된 진핵 세포의 배양에 의한 내생성 또는 이종 단백질의 제조 방법.
- 제 1 항에 있어서,단백질이 효소, 특히 사료 효소, 예를 들면 피타제, 셀룰라제, 크실라나제, 또는 산업적 효소, 예를 들면 아밀라제, 프로티아제, 인버타제, 리파제, 카탈라제, 셀룰라제, 글루코즈 옥시다제, 알콜 옥시다제, 펙티나제, 나라기나제, 콜라게나제, 퍼옥시다제 또는 풀룰라나제인 방법.
- 제 1 항 또는 제 2 항에 있어서,세포가 메틸로트로픽(methylotropic)이고/이거나 메탄올 대사에 관련된 효소용 프로모터, 예를 들면 포르메이트 디하이드로게나제(FMD 또는 FMDH) 프로모터, 메탄올 옥시다제(MOX) 프로모터 또는 디하이드록시아세톤 신타제(DAS 또는 DHAS) 프로모터를 포함하는 DNA 서열로 형질전환된 방법.
- 제 1 항 내지 제 3 항중 어느 한 항에 있어서,세포가 메틸로트로픽 효모, 바람직하게는 한세뉼라(Hansenula), 예를 들면 한세뉼라 폴리모파(Hansenula polymorpha), 피치아(Pichia), 예를 들면 피치아 파스토리스(Pichia pastoris), 칸디다(Candida) 또는 토룰롭시스(Torulopsis)인 방법.
- 제 1 항 내지 제 4 항중 어느 한 항에 있어서,억제성 기질이 탄소원의 1 내지 100%, 바람직하게는 40 내지 100%, 보다 바람직하게는 90 내지 100%이거나, 또는 유일한 탄소원인 방법.
- 제 1 항 내지 제 5 항중 어느 한 항에 있어서,억제성 기질이 단당류, 이당류, 올리고당 또는 다당류, 예를 들면 글루코즈, 프락토즈, 슈크로즈, 말토즈, 전분, 글리코겐, 셀룰로즈 또는 덱스트로즈와 같은 당, 또는 당 함유 화합물, 예를 들면 당밀, 글루코즈 시럽 또는 프락토즈 시럽인 방법.
- 제 1 항 내지 제 6 항중 어느 한 항에 있어서,반복적인 공급-배치의 형태로 수행하는 방법.
