Claims (42)
- 바이러스 게놈의 유전자내 영역(IGR)내로 삽입되는 이종 DNA 서열을 포함하는 재조합 변형 백시니아 안카라 바이러스(MVA).
- 제 1항에 있어서, 상기 이종 DNA 서열은 007R-008L, 018L-019L, 044L-045L, 064L-065L, 136L-137L, 148R-149L을 포함하는 그룹으로부터 선택된 두개의 인접한 오픈 리딩 프레임(ORF)간의 IGR내로 삽입되는 MVA.
- 제 1항 또는 제 2항에 있어서, 상기 이종 DNA 서열은 하나 이상의 코딩 서열, 바람직하게는 수두 바이러스 전사적 조절 인자의 전사적 조절하에서 하나 이상의 코딩 서열을 포함하는 MVA.
- 제 1항 내지 제 3항중 어느 한 항에 있어서, 상기 이종 DNA 서열은 하나 이상의 단백질, 폴리펩티드, 펩티드, 외래항원 또는 항원성 항원 결정기를 암호화하는 MVA.
- 제 1항 내지 제 4항중 어느 한 항에 있어서, 상기 이종 DNA 서열은 뎅기 바이러스, 일본 뇌염 바이러스, B형 간염 바이러스, C형 간염 바이러스 및/또는 면역 결핍 바이러스, 바람직하게는 인체 면역결핍 바이러스(HIV)로부터 유도된 MVA.
- 제 5항에 있어서, 뎅기 바이러스로부터 유도된 상기 이종 DNA 서열은 NS1 및 PrM을 포함하는 그룹으로부터 선택된 MVA.
- 제 6항에 있어서, 상기 NS1 유전자는 ORF 064L-065L간의 IGR내로 삽입되는 삽입되는 MVA.
- 제 6항 또는 제 7항에 있어서, 상기 PrM 유전자는 하나 이상의 4 뎅기 바이러스 혈청형으로부터 유도된 MVA.
- 제 6항 내지 제 8항중 어느 한 항에 있어서, 상기 PrM 유전자는 007R-008L, 044L-045L, 136L-137L, 148R-149L을 포함하는 그룹으로부터 선택된 ORF간의 IGR내로 삽입되는 MVA.
- 제 5항에 있어서, 면역결핍 바이러스 코드로부터 유도된 상기 이종 DNA 서열은 HIV env를 암호화하는 MVA.
- 제 10항에 있어서, 상기 HIV env 유전자는 ORF 007R-008L간의 IGR내로 삽입되는 MVA.
- 제 1항 내지 제 11항중 어느 한 항에 있어서, MVA는 ECACC에 기탁번호 제V00083008로 기탁된 MVA.
- 제 1항 내지 제 11항중 어느 한 항에 있어서, 의약 및/또는 백신으로 사용하기 위한 MVA.
- 바이러스 감염 및/또는 증식성 질환을 치료 및/또는 예방용 의약 및/또는 백시을 제조하기 위한 제 1항 내지 제 13항중 어느 한 항에 따른 MVA의 용도.
- 제 14항에 있어서, 뎅기 바이러스 또는 면역 결핍 바이러스 질환, 바람직하게는 HIV 감염의 치료 또는 예방을 위한 용도.

- DNA 서열을 사람을 비롯한 살아있는 동물의 몸에 도입하기 위한 방법에 있어서,상기 DNA 서열은 상기 세포의 게놈에 동종 및/또는 이종이며, 제 1항 내지 제 13항중 어느 한 항에 따른 MVA, 제 16항에 따른 백신 및/또는 제 17항에 따른 조성물을 사람을 비롯한 동물에게 투여하는 것을 특징으로 하는 방법.
- 제 1항 내지 제 13항중 어느 한 항에 따른 MVA를 포함하는 세포.
- MVA의 게놈으로부터 유도되거나 또는 MVA의 게놈에 동종인 DNA 서열을 포함하는 플라스미드 벡터에 있어서,상기 DNA 서열은 바이러스 게놈의 두개의 인접한 ORF간에 위치하며 IGR 서열내로 삽입되는 IGR 서열의 완전한 또는 부분적인 단편, MVA의 게놈에 이종인 DNA 서열 및 경우에 따라 리포터 및/또는 선별 유전자 카세트의 삽입용 클로닝 부위를 포함하는 것을 특징으로 하는 플라스미드 벡터.
- MVA의 게놈으로부터 유도되거나 또는 MVA의 게놈에 동종인 DNA 서열을 포함하는 플라스미드 벡터에 있어서,상기 DNA 서열은 IGR 서열내로 삽입되는, 두개의 인접한 ORF의 IGR 측면 서열을 포함하며, DNA 서열, MVA의 게놈에 이종인 DNA 서열 및 경우에 따라 리포터및/또는 선별 유전자 카세트의 삽입용 클로닝 부위를 포함하는 것을 특징으로 하는 플라스미드 벡터.
- 제 25항에 있어서, 상기 DNA 서열은 IGR 서열의 완전한 또는 부분적인 단편을 포함하며, 상기 IGR 서열은 이종 DNA 서열 및 경우에 따라 리포터 및/또는 선별 유전자 카세트의 삽입용 클로닝 부위을 포함하는 것을 특징으로 하는 플라스미드 벡터.
- 제 24항 내지 제 26항중 어느 한 항에 있어서, 상기 IGR 서열은 007R-008L, 018L-019L, 044L-045L, 064L-065L, 136L-137L, 148R-149L을 포함하는 그룹으로부터 선택된 ORF간의 IGR로부터 유도된 플라스미드 벡터.
- 제 24항 내지 제 27항중 어느 한 항에 있어서, 상기 IGR-유도된 서열은 뉴클레오티드 서열-서열번호 32의 527-608;-서열번호 33의 299-883;-서열번호 34의 339-852;-서열번호 35의 376-647;-서열번호 36의 597-855;-서열번호 37의 400-607을 포함하는 그룹으로부터 선택된 서열을 포함하는플라스미드 벡터.
- 제 24항 내지 제 28항중 어느 한 항에 있어서, 두개의 인접한 ORF의 IGR 측면 서열은 007R-008L, 018L-019L, 044L-045L, 064L-065L, 136L-137L, 148R-149L을 포함하는 그룹으로부터 선택된 플라스미드 백터.
- 제 24항 내지 제 29항중 어느 한 항에 있어서, 두개의 인접한 ORF의 IGR 측면 서열은 뉴클레오티드 서열-서열번호 32의 1-526 및 609-1190;-서열번호 33의 101-298 및 884-1198;-서열번호 34의 1-338 및 853-1200;-서열번호 35의 1-375 및 648-1200;-서열번호 36의 1-596 및 856-1200;-서열번호 37의 1-399 및 608-1081을 포함하는 그룹으로부터 선택된 서열을 포함하는 플라스미드 벡터.
- 제 24항 내지 제 30항중 어느 한 항에 있어서, 상기 DNA 서열은 ECACC에 기탁번호 제V00083008로 기탁된 MVA의 게놈으로부터 유도되거나 또는 MVA의 게놈에 동종인 플라스미드 벡터.
- - 제 24항 내지 제 31항중 어느 한 항에 따른 플라스미드 벡터에 의해 세포를 형질감염시키는 단계;- 형질감염된 세포를 MVA에 의해 감염시키는 단계;- 제 1항 내지 제 13항중 어느 한 항에 따른 MVA를 확인, 분리 및 경우에 따라 정제시키는 단계를 포함하는 제 1항 내지 제 13항중 어느 한 항에 따른 MVA의 제조방법.
- 제 32항에 있어서, 상기 세포는 ECACC에 기탁번호 제V00083008로 기탁된 MVA에 의해 감염된 방법.
- MVA의 게놈으로부터 유도되거나 또는 MVA의 게놈에 동종인 DNA 서열에 있어서,상기 DNA 서열은 바이러스 게놈의 두개의 인접한 ORF간에 위치한 IGR 서열의 완전한 또는 부분적인 단편을 포함하는 DNA 서열.
- MVA의 게놈으로부터 유도되거나 또는 MVA의 게놈에 동종인 DNA 서열에 있어서, 상기 DNA 서열은 두개의 인접한 ORF의 IGR 측면 서열을 포함하는 DNA 서열.
- 제 35항에 있어서, 상기 DNA 서열은 IGR 서열의 완전한 또는 부분적인 단편을 포함하는 DNA 서열.
- 제 34항 내지 제 36항중 어느 한 항에 있어서, MVA 게놈에 이종인 외인성 DNA 서열이 IGR내로 삽입되는 DNA 서열.
- 제 34항 내지 제 37항중 어느 한 항에 있어서, 상기 IGR 서열은 007R-008L, 018L-019L, 044L-045L, 064L-065L, 136L-137L, 148R-149L을 포함하는 그룹으로부터 선택된 ORF간의 IGR로부터 유도된 DNA 서열.
- 제 34항 내지 제 38항중 어느 한 항에 있어서, 상기 IGR-유도된 서열은 뉴클레오티드 서열-서열번호 32의 527-608;-서열번호 33의 299-883;-서열번호 34의 339-852;-서열번호 35의 376-647;-서열번호 36의 597-855;-서열번호 37의 400-607을 포함하는 그룹으로부터 선택된 서열을 포함하는 DNA 서열.
