Abstract
설로덱사이드 또는 그의 구성성분은 심혈관계질환, 당뇨에 의해 야기된 심혈관계질환, 정맥류, 만성정맥부전 (chronic venous insufficiency, CVI), 위장 궤양, 폐질환, 및 종양성 병리 (neoplastic pathology)와 같은, MMP가 연관된 병리의 치료에 유용하다.

Claims (8)
- AT III로 관능화된 정지상을 이용한 친화성 크로마토그래피 방법을 의해 술로덱사이드로부터 얻어지는 데르마탄 술페이트로서, 상기 데르마탄 술페이트는 비흡수된 통과액 (flow-through)에 수집되고, 평균 분자량이 25 kDa 이고, 아가로스 겔에서 전기영동 상대적 이동도 (Rf)가 0.84인 것으로 규명된 것인 데르마탄 술페이트, 또는 그의 약제학적으로 허용가능한 염, 용매화물, 포접화합물 (clathrate).
- 청구항 1에 있어서, 감염 또는 종양 질환 (neoplastic disease), 높은 수준의 MMPs가 특징인 혈관 질환, 위장 궤양 (gastrointestinal ulcer), 종양의 증가 및 진행, 원발성 종양 (primary tumor)의 성장, 혈관 형성 (angiogenesis), 종양 세포의 혈관 외 유출 및 혈관 내 주입, 및 이들의 조합으로 이루어진 군으로부터 선택되는 질환의 치료에 사용하기 위한 것인 데르마탄 술페이트.
- 청구항 2에 있어서, 상기 혈관 질환은 만성 정맥 부전 (chronic venous insufficiency, CVI), 정맥류 (varicose vein), 심근 경색 후 심정지 (cardiac break), 및 복부 대동맥류 (abdominal aortic aneurysm)로 이루어진 군으로부터 선택되는 것인 데르마탄 술페이트.
- 청구항 1에 있어서, 5 ㎍/ml의 농도에서 50 %보다 높은 양으로 MMPs의 억제 또는 감소에 사용하기 위한 것인 데르마탄 술페이트.
- 청구항 1에 있어서, 5 ㎍/ml의 농도에서 50 %보다 높은 양으로 MMP-9의 억제 또는 감소에 사용하기 위한 것인 데르마탄 술페이트.
- 청구항 1에 따른 데르마탄 술페이트를 약제학적으로 허용되는 부형제와 함께 포함하는, 정제, 캡슐, 현탁액, 용액, 또는 에어로졸 형태인 것인 조성물.
- 청구항 6에 있어서, 상기 부형제는 하나 이상의 리간트 (ligants), 활택제 (glidant), 붕해제 (disintegrants), 천연 물질(natural agents) 중에서 선택되는 것인 조성물.
- 하기의 단계를 포함하는, 청구항 1의 데르마탄 술페이트를 제조하는 방법:
a) 5 내지 10 mg/ml 겔 농도로 ATIII을 포함하는 친화성 크로마토그래피 컬럼을 준비하는 단계;
b) pH 7.4에서, TRIS 50 mM, NaCl 50 mM를 포함하는 버퍼 중 0.1 내지 5 mg/ml 겔에 속하는 농도로, 상기 단계 a)의 컬럼에 설로덱사이드를 로딩하는 단계;
c) 흡수되지 않은 분획 (non absorbed fraction) 내의 용출된 데르마탄 술페이트 분획을 통합 (pool)하고, 선택적으로 통합된 데르마탄 술페이트를 동결건조하는 단계;
d) 선택적으로 상기 데르마탄 술페이트를 그의 약제학적으로 허용가능한 염, 용매화물, 수화물, 포접화합물로 변형하는 단계.
