Description
Verfahren zur Herstellung eines optisch reinen Enantiomeren von Formoterol
Gegenstand der Erfindung ist ein neues, vorteilhaftes Verfahren zur Herstellung des optisch reinen R,R-Enantiomeren von Formoterol. Formoterol: N-[2-Hydroxy-5-(l-hydroxy-2-((2-(4-methoxyphenyl)-l-methylethyl)-amino)-ethyl)-phenyl]-formamid so wie das Fumaratsalz dieser Verbindung, siehe Merck Index, Eleventh Edition Nr. 4159, gehort zur Gruppe der selektiven ß2-sympathomimetischen, bronchienerweiternden Wirkstoffe, welche als Antiasthmatika verwendbar sind. Besonders geeignete Darreichungsformen sind vorzugsweise Aerosole zur Inhalation. Ein bekanntes Handelsprodukt ist das Dosieraerosol ®Foradil (Ciba), welches in zahlreichen Ländem erhältlich ist.
In Chem. Pharm. Buil. 26, No.4, 1123*1129 (1978), ist das (R,R),(S,S)-Enantiomerenpaar von Formoterol sowie dessen Auftrennung in zwei optisch reine Enantiomere und zwei Diastereomere beschrieben. In Chirality 3:443-450 (1991) ist die verbesserte Wirkung des R,R-Enantiomeren gegenüber dem S,S-Enantiomeren beschrieben. Generell wird für chirale Beta-Sympathomimetika angenommen, dass nur ein optisch reiner Antipode eines Enantiomerenpaares wirksam ist. Der jeweils andere Antipode des Enantiomerenpaares ist entweder unwirksam oder kann sogar Nebenwirkungen verursachen, siehe hierzu auch TiPS, June 1992 [Vol. 13], Seiten 231-232. Die Herstellung des R,R-Enantiomeren mit hoher optischer Reinheit erfolgt mit geringer Ausbeute iiber mehrere aufwendig durchzuführende und daher kostspielige Syntheseschritte unter Verwendung von chiralen Ausgangsmaterialien und Zwischenprodukten.
Der vorliegenden Erfindung liegt die Aufgabe zugrunde, für das R,R-Enantiomere der freien Base von Formoterol ein vereinfachtes und verbessertes und daher kostengünstigeres Herstellungsverfahren mit erhöhter Ausbeute und besonders hoher Reinheit an optisch reinem Produkt bereitzustellen.
Diese Aufgabe wird durch die vorliegende Erfindung überraschenderweise gelost, welche ein Verfahren zur Herstellung von optisch reinem (R,R)-N-[2-Hydroxy-5-(l-hydroxy* 2-((2-(4-methoxyphenyl)-l-methylethyI)-amino)-ethyl)-phenyl]-formamid und dem
Fumaratsalz dieser Verbindung betrifft. Das Verfahren ist dadurch gekennzeichnet, daß man das racemische Gemisch oder Diastereomerengemisch der freien Verbindung in einer mobilen Phase enthaltend ein unpolares und gegebenenfalls ein weiteres polares, protisches oder aprotisches Lösungsmittel chromatographisch an einer chiralen, stationären Phase bestehend aus einem Polysaccharid, dessen freie Hydroxygruppe durch die 4-Methylbenzoyl-Gruppe derivatisiert sind, und gegebenenfalls einem inerten Trägermaterial, auftrennt und aus dem Eluat der mobilen Phase die optisch reine R,R-Verbindung isoliert und in das Fumaratsalz überführt.
Ein wesentlicher Vorteil dieses Verfahrens beruht in seiner Kombination mit bekannten, kostengünstigen Herstellungsverfahren von racemischem Formoterol. Man stellt racemisches Formoterol in an sich bekannter Weise her und unterwirft dieses anschliessend dem erfindungsgemässen chromatographischen Trennverfahren. In einem Verfahrensschritt wird das racemische Gemisch von Formoterol in die optisch reinen Enantiomeren aufgetrennt. Es ist keine aufwendige Synthese von optisch reinen Zwischenprodukten erforderlich.
Die in der Beschreibung der vorliegenden Erfindung verwendeten Begriffe und Bezeichnungen werden vorzugsweise wie folgt definiert:
Der Begriff "optisch rein" bezeichnet für einen definierten Stoff, der mindestens ein Chiralitätszentrum aufweist, einen Gehalt von mehr als 95 Gew.%, vorzugsweise mehr als 99 %, insbesondere mehr als 99,9 %, eines Antipoden mit definierter Konfiguration, z.B. nach den bekannten Sequenzregeln nach Kahn, Ingold, Prelog.
Der Begriff "racemisches Gemisch" bezeichnet für einen definierten Stoff, der mindestens ein Chiralitätszentrum aufweist, ein ca. l:l-Gemisch von zwei Antipoden mit definierter Konfiguration.
Der Begriff "Diastereomerengemisch" bezeichnet für einen definierten Stoff, der mindestens zwei Chiralitütszentren aufweist, bezüglich eines Chiralitatszentrums eine definierte Konfiguration, wobei die Konfiguration bezüglich des weiteren Chiralitatszentrums racemisch ist.
Der Begriff "mobile Phase" definiert ein Lösungsmittel oder Lösungsmittelgemisch, worin das in die optisch reinen Antipoden aufzutrennende racemische Gemisch oder ein
Diastereomerengemisch von Formoterol gelöst ist. Als mobile Phase ist aber auch ein Inertgas, z.B. Argon, verwendbar, wenn man das Verfahren der präparativen Gaschromatographie anwendet.
Als Lösungsmittel für die mobile Phase eignen sich Mischungen eines unpolaren Lösungsmittels mit einem polaren, protischen oder aprotischen Lösungsmittel.
Geeignete unpolare Lösungsmittel sind z.B. n-Pentan, Isooctan, Petrolether, n-Hexan, n-Heptan, Cyclohexan, Cyclopentan, Diisopropylether, Cyclohexen, Dimethoxyethan oder Diethylether.
Geeignete polare, protische oder aprotische Lösungsmittel sind z.B. Amylalkohol, Acetonitril, i-Propanol, n-Propanol, n- oder tert-Butanol, Ethanol, Methanol, Ethylenglycol, Essigsäure oder Wasser.
Bevorzugt sind Mischungen eines unpolaren Lösungsmittels wie n-Pentan, Isooctan, Petrolether, n-Hexan, n-Heptan, Cyclohexan oder Cyclopentan mit einem polaren, protischen Lösungsmittel, wie i-Propanol oder Ethanol.
Besonders bevorzugt sind die Mischungen von n-Hexan oder n-Heptan mit Ethanol oder n-Hexan oder n-Heptan mit n-Isopropanol.
Der Begriff "chromatographische Trennung" definiert bekannte Trennverfahren von Stoffgemischen, welche in der mobilen Phase gelöst sind. Durch Absorption oder chemische Reaktion an einer stationâren Phase stellt sich für den betreffenden Stoff ein Gleichgewicht ein, welches charakteristische Retentionszeiten für den aufzutrennenden Stoff verursacht.
Geeignete chromatographische Trennverfahren sind unter Begriffen wie Adsorptionschromatographie, z.B. Säulenchromatographie oder Adsorptions-chromatographie an Adsorberharzen, Papierchromatographie, Dünnschicht-chromatographie oder präparative Gaschromatographie bekannt.
Besonders bevorzugt sind die unter der Bezeichnung HPLC (High Performance Liquid Chromatography) und SMBA (Simulated Moving Bed Adsorption) bekannten, chromatographischen Trennverfahren. Dabei verwendet man eine chirale stationäre Phase, welche aus einem Polysaccharid besteht, dessen freie Hydroxygruppen durch die 4-Methylbenzoyl-Gruppe derivatisiert sind. Mit diesem Polysaccharid kann ein inertes Trägermaterial beschichtet sein.
Für die chirale stationäre Phase geeignetes inertes Trägermaterial ist vorzugsweise makroporös, z.B. quervernetztes Polystyrol, Polyacrylamid, Polyacrylat, Quarz, Kieselgur, Aluminiumoxid, Aluminosilicat-Xerogele, saures Magnesiumsilicat, Magnesiumoxid, Titandioxid oder Kaolin. Bevorzugt ist Kieselgel.
Die Korngrösse des inerten Trägermaterials ist innerhalb weiter Grenzen variabel, beispielsweise von ca. 1 pm bis 1 mm, vorzugsweise ca. 1 bis 300 pm. Dieses ist vorzugsweise poros mit einer mittleren Porenweite von ca. 1,0 x 10'8 m bis 1,0 xlO"6 m.
Die Beschichtung des inerten Trâgermaterials mit dem derivatisierten Polysaccharid erfolgt in an sich bekannter Weise, indem man beispielsweise eine Lösung des derivatisierten Polysaccharids in einem organischen Lösungsmittel, z.B. Ethanol oder einem Methylenchlorid-Tetrahydrofuran-Gemisch, mit dem inerten Trägermaterial, z.B. makroporösem Silicagel, versetzt und das Lösungsmittel abdampft. Zahlreiche andere Verfahren sind bekannt, z.B. Behandeln im Wirbelschichtreaktor, Aufsprühen, Ausfällen etc.. Das makroporöse Silicagel kann man zuvor durch Umsetzung mit 3-Aminopropyltriethoxysilan, gelöst in Benzol, aktivieren.
Das Polysaccharid lässt sich z.B. durch Umsetzung mit 4-Methylbenzoylchlorid derivatisieren. Besonders geeignet ist Cellulose vom Typ ® Avicel (Merck).
Als derivatisierbare Polysaccharide sind optisch aktive natlirliche Oder chemisch modifizierte Polysaccharide geeignet, z.B. mikrokristalline oder native Cellulose, Baumwolle-Linters oder Cellulose aus Pflanzenfasem wie Baumwolle, Flachs, Hanf, Jute oder Ramiefaser.
Das Polysaccharid, insbesondere die Cellulose, ist an 1-3, vorzugsweise 3, freien Hydroxygruppen durch die 4-Methylbenzoyl-Gruppe derivatisiert und ist als Beschichtungsmittel für ein inertes Trägermaterial Oder selbst in Form von "Beads" verwendbar, siehe publizierte Europäische Patentanmeldung Nr. 186 133.
Geeignete Materialien für die chirale, stationäre Phase sind bekannt und kommerziell erhältlich, insbesondere die unter der Bezeichnung ®ChiraIcel (Daicel) OJ bekannte Handelsware, welche aus Silicagel besteht, das mit veresterter Cellulose beschichtet ist. Die Estergruppe ist die 4-Methylbenzoyl-Gruppe.
Für die chromatographische Trennung mittels HPLC eignen sich insbesondere Säulen für Trennungen im semipräparativen oder präparativen Masstab, z.B. mit 1-10 cm Durchmesser und 20-60 cm Länge. Besonders geeignete durchschnittliche Partikelgrössen des Trägermaterials sind z.B. 10-20 μπι für HPLC und 10-60 pm für SMBA.
Die Überführung der freien Verbindung in ihr Fumaratsalz erfolgt, falls erwünscht, in an sich bekannter Weise durch übliche Umsetzung der freien Verbindung mit Fumarsäure oder Umsetzung eines Natrium- oder Kaliumsalzes mit Fumarsäure Oder dem Säurechlorid davon.
Die folgenden Beispiele illustrieren die Erfindung:
Beispiel 1 2 g einer 2,5 %-igen Lösung von racemischem Formoterol in Hexan/Ethanol (85:15 Vol-%) werden auf eine chirale CHIRALCEL OJ (Daicel Chem.Ind., Japan) HPLC-Säule (10 X 50 cm, Komgrösse 20 pm) aufgetragen. Das Trägermaterial besteht aus einem mit p-Methylbenzoylcellulose beschichteten Kieselgel. Bei einem Durchfluss von ca. 150 ml/min. und Hexan/Ethanol (85:15 Vol-%) als Fliessmittel werden die Enantiomeren mit einem Trennfaktor von α = 1,54 wie folgt getrennt:
Aus 2 g Racemat werden optisch reine Fraktionen erhalten und eingeengt. Sie ergeben 0,9 g des ersten eluierten Enantiomeren mit einer optischen Reinheit > 99,9 % und 1,15 g des zweiten eluierten Enantiomeren mit einer optischen Reinheit > 98 %. Die mit dem zweiten eluierten Enantiomeren angereicherten Fraktionen werden weiter bis zum Erhalt einer optischen Reinheit von von mindestens 99,5 % chromatographiert. Beide Fraktionen werden noch weiter durch Flash- Chromatographie gereinigt. Die Reinigung erfolgt an Kieselgel (34 g, Komgrösse 40-63 pm, Glassäule 2,5 x 30 cm) nacheinander mit Gemischen aus a) 250 ml Hexan/Ethanol (2:1 Volumenteile), b) 250 ml Hexan/Ethanol (1:3 Volumenteile) und c) 250 ml Hexan/Ethanol (1:6 Volumenteile) als Fliessmitteln bei einem Druck von ca. 0,2 bar. Die gereinigten Fraktionen des betreffenden Enantiomeren werden gesammelt und anschliessend eingeengt. Anschliessend wird der jeweilige
Rückstand in Ether aufgeschlämmt, wieder eingeengt und getrockneL Das erste (0,730 g) und zweite (0,780 g) Enantiomère werden jeweils als weisses Pulver isolierL
Zur Prüfung der biologischen Wirksamkeit werden beide Enantiomeren in ihr Fumaratsalz umgewandelL Dazu lost man 1,72 g des jeweiligen optisch reinen Enantiomeren in 10 ml Methanol und gibt dazu eine äquimolare (0,29 g) Menge Fumarsäure. Nach einer Reaktionszeit von einer Stunde bei Raumtemperatur wird die betreffende klare Lösung bei 40°C am Rotationsverdampfer eingeengt und anschliessend bei 40°C sechs Stunden lang im Hochvakuum getrocknet. Die erhaltenen Fumarat-Salze werden als Hemifumarat Monohydrate isoliert und charakterisiert: a) Erstes eluiertes optisch reines Enantiomères: (-)-(R,R)-Formoterol: [a]D= -44,7 ±2.3°; b) Zweites eluiertes optisch reines Enantiomères: (+)-(S,S)-Formoterol: [a]D= +47,0 ±0,2°.
Beispiel 2
Trennung der Enantiomeren von Formoterol durch "Simulated Moving Bed Adsorption". Konfiguration des Systems:
Anlage: Prep-SMB-System L von der Firma UOP (Universal Oil Products, Des Plaines Illinois 60017-5017, USA).
Säule: 16-Säule(Bed)-System mit karusselförmiger Anordnung. Jede Säule hat einen Innendurchmesser von 16 mm und eine Längevon 60 mm. Die Säulen (Vol. 0.193 ml) werden mit der Slurry-Methode gefüllt. Chirales Trägermaterial: Chiralcel® OJ, 20 pm; Fliessmittel: Heptan-Ethanol 70:30.
Durchführung der Trennung:
Bei einer Konzentration der Racematlösung von 0.25% in Heptan-Ethanol 70:30; einer Fliessgeschwindigkeit der Racematlösung von 0,52 ml/min; einer Fliessgeschwindigkeit der mobilen Phase von 6,69 ml/min; einer Zykluszeit von 90 min; einer "Extract rate" von 3,59 ml/min; einem "Raffinate rate" von 3,62 ml/min erreicht man für jedes Enantiomer eine Produktivität von 0,44 g pro Stunde und pro kg chirale stationäre Phase. Die optische Reinheit beträgt 100 % für das Raffinât (RR Enantiomer) und 97,4% für den Extrakt (SS Enantiomer).
Claims (6)
1. Verfahren zur Herstellung von optisch reinem (R,R)-N-[2-Hydroxy-5-(l-hydroxy-2-((2-(4-methoxyphenyl)-l-methylethyl)-amino)-ethyl)-phenyl]-formamid Oder dem Fumaratsalz dieser Verbindung, dadurch gekennzeichnet, daß man das racemische Gemisch Oder Diastereomerengemisch der freien Verbindung in einer mobilen Phase enthaltend ein unpolares und gegebenenfalls ein weiteres polares, protisches Oder aprotisches Lösungsmittel chromatographisch an einer chiralen stationären Phase bestehend aus einem Polysaccharid, dessen freie Hydroxygruppen durch die 4-Methylbenzoyl-Gruppe derivatisiert sind, und gegebenenfalls einem inerten Trägermaterial, auftrennt und aus dem Eluat der mobilen Phase die optisch reine R,R-Verbindung isoliert und in das Fumaratsalz überführt.
2. Verfahren gemäss Anspruch 1, dadurch gekennzeichnet, dass man ein racemisches Gemisch des Fumaratsalzes von N-[2-Hydroxy-5-(l-hydroxy-2-((2-(4-methoxyphenyl)-l-methylethyl)-amino)-ethyl)-phenyl]-formamid chromatographisch auftrennt.
3. Verfahren gemäss Anspruch 2, dadurch gekennzeichnet, dass man ein racemisches Gemisch des Fumaratsalzes von N-[2-Hydroxy-5-(l-hydroxy-2-((2-(4-methoxyphenyl)-l-methylethyl)-amino)-ethyl)-phenyl]-formamid mittels HPLC auftrennt.
4. Verfahren gemäss Anspruch 2, dadurch gekennzeichnet, dass man ein racemisches Gemisch des Fumaratsalzes von N-[2-Hydroxy-5-(l-hydroxy-2-((2-(4-methoxyphenyl)-l-methylethyl)-amino)-ethyl)-phenyl]-formamid mittels SMBA auftrennt.
5. Verfahren gemäss Anspruch 3 Oder 4, dadurch gekennzeichnet, dass man das racemische Gemisch in einer mobilen Phase enthaltend Hexan Oder n-Heptan als unpolares und Ethanol als polares protisches Lösungsmittel chromatographisch auftrennt.
6. Verfahren gemäss Anspruch 3, dadurch gekennzeichnet, dass man das racemische Gemisch chromatographisch an einer stationären Phase bestehend aus dem inerten Träger Kieselgel versehen mit dem Polysaccharid Cellulose, welche durch die 4-Methylbenzoyl--Gruppe derivatisiert ist, auftrennt. .
