Abstract

"métodos para a produção de 3-o-4<39>monofosforil desativado lipídeo a (3d-mla)". a invenção refere-se um método para produção de lipopolissacarídeo (lps), que compreende: (a) desenvolver uma cultura de uma cepa bacteriana mutante extremamente rudimentar em um meio; (b) manter a cultura em fase estacionária por pelo menos cerca de 5 horas; (c) colher células da cultura; e (d) extrair lps das células. o método permite para a produção de um lps que pode ser usado para produzir um 3-o-monofosforil desacilado lipídeo a (3d-mla) que apresenta pelo menos cerca de 20% em mol do grupo de congêneres de hexacila. é também apresentado nesta invenção um método de extrair lipopolissacarídeo (lps) de uma cultura de células de cepa bacteriana mutante extremamente rudimentar, que compreende: (a) extrair as células com uma solução que consiste essencialmente pelo menos de cerca de 75% em peso de um álcool alifático que apresenta de 1 a 4 átomos de carbono e o equilíbrio água, produzindo desse modo células com teor de fosfolipídeo reduzido; e (b) extrair as células com teor de fosfolipídeo reduzido com uma solução que compreende clorofórmio e metanol, produzindo desse modo uma solução de lps em clorofórmio e metanol (cm). esse método proporciona soluções de lps em cm que apresentam teor de fosfolípideo reduzido e são produzidas através de etapas de processo relativamente simples e econômico.
