Description
Verfahren zur Herstellung von Sulfamyl-3,4-dihydro-1,2,4-benzothiadiazin-1,1-dioxyden
Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung von 3,4-Dihydro-1, 2,4 benzothiadiazin-l, l-dioxyden der Formel

Darin stehen Ri für einen Alkylrest, besonders mit 1-5 Kohlenstoffatomen, R3, und R4 für Wasserstoff oder Alkylreste, besonders mit 1-5 Kohlenstoffatomen, und R2 für Wasserstoff, einen unsubstituierten oder substituierten Kohlenwasserstoff-oder heterocyclischen Rest. Als Kohlenwasserstoffreste R2 sollen besonders genannt werden Alkylreste, wie Methyl-, Athyl-, Propyl-, Isopropyl-, Butyl-oder Pentylreste, oder Alkenylreste, wie Vinyl-oder l-Propenylreste, oder Alkinylreste, z.
B. der Athinylrest, oder cycloaliphatische Reste, wie Cycloalkyloder Cycloalkenylreste, z. B. Cyclopentyl-oder Cyclohexenylreste, oder Cycloalkyl-alkyl-, z. B. Cyclohexyl- äthyl-, oder Cycloalkenyl-alkylreste, z. B. Cyclohexenyl äthylreste, oder Arylreste, besonders mono-oder bicyclische Arylreste, z. B. Phenyl-, Naphthyl- (1)- oder Naphthyl- (2)-reste, oder araliphatische Reste, z. B. Benzyl-, 2-Phenyl-äthyl-, Naphthyl- (l)- oder - (2)-methylreste. Als heterocyclische Reste Rg sind in erster Linie Furyl-, Thienyl-, Pyridyl-oder Chinolylreste zu nennen, die auch über einen Alkylrest an den Thiadiazinring gebunden sein können, wie der Furfuryl-oder Thenylrest.
Als Substituenten dieser Reste sollen z. B. genannt werden Alkylreste, freie, veresterte oder verätherte Hydroxyl-oder Mercaptogruppen, z. B. die Acetoxy-, Methoxy-, Phenoxy-oder Methylmercaptogruppe, die Methylendioxygruppe, Sulfamylreste, Aminogruppen, wie die Dimethylaminogruppe, oder Halogene.
Die neuen 3,4-Dihydro-1,2,4-benzothiadiazin 1, 1-dioxyde zeigen eine diuretische und natriuretische Wirksamkeit und können als Heilmittel Verwendung finden.
In erster Linie sind die Verbindungen der Formel

und deren Salze durch eine hervorragende diuretische Wirksamkeit ausgezeichnet, worin RI für einen Alkylrest mit 1-5 Kohlenstoffatomen, R3 für Wasserstoff oder einen Alkylrest mit 1-5 Kohlenstoffatomen und R2 für Wasserstoff, einen unsubstituierten oder durch Halogen, freie, veresterte oder verätherte Hydroxylgruppen oder durch Aminogruppen substituierten aliphatischen Kohlenwasserstoffrest mit 1-8 Kohlenstoffatomen, einen Cycloalkyl-oder Cycloalkenylrest, einen Phenyl-, Pyridyl-oder einen Phenylalkylrest stehen.
Aus dieser Gruppe ragen in bezug auf ihre Wirksamkeit noch die Verbindungen der Formel

oder deren Salze hervor, worin Rl und Rs die oben gezeigte Bedeutung besitzen und R2 für Wasserstoff, einen unsubstituierten oder durch Amino-oder Hydroxylgruppen oder Chlor substituierten Alkylrest mit 1-5 Kohlenstoffatomen, den Cyclohexenyl-, Benzyl-oder Phenyläthylrest steht.
Als besonders wirksame Verbindungen sollen z. B. genannt werden das 2-Methyl-6-chlor-7-sulfamyl3,4-dihydro-1,2,4-benzothiadiazin-l, 1-dioxyd und das 2-Methyl-6-chlor-7-methylsulfamyl-3, 4-dihydro-1, 2, 4- benzothiadiazin-l, 1-dioxyd und deren Salze.
Die neuen Verbindungen und deren Salze, besonders mit Alkalimetallen, können als Heilmittel in Form von pharmazeutischen Präparaten verwendet werden, welche diese Verbindungen zusammen mit pharmazeutischen organischen oder anorganischen, festen oder flüssigen Trägerstoffen, die für enterale, z. B. orale, oder parenterale Gabe geeignet sind, enthalten. Für die Bildung derselben kommen solche Stoffe in Frage, die mit den neuen Verbindungen nicht reagieren, wie z. B. Wasser, Gelatine, Milchzucker, Stärke, Magnesiumstearat, Talk, pflanzliche ) le, Benzylalkohole, Gummi, Polyalkylenglykole, Vaseline, Cholesterin oder andere bekannte Arzneimittelträger. Die pharmazeutischen Präparate können z. B. als Tabletten, Dragées, Kapseln oder in flüssiger Form als Lösungen, Suspensionen oder Emulsionen vorliegen.
Gegebenenfalls sind sie sterilisiert und bzw. oder enthalten Hilfsstoffe, wie Konservierungs-, Stabilisierungs-, Netz-oder Emulgiermittel, Salze zur Veränderung des osmotischen Druckes oder Puffer.
Sie können auch noch andere therapeutisch wertvolle Stoffe, z. B. hypotensive Mittel, enthalten, wie Rauwolfia-oder Veratrumalkaloide, beispielsweise Reserpin, Rescinnamin, Deserpidin, Germin oder Protoveratrin, synthetische hypotensive Mittel, z. B.
Hydralazin, oder Ganglienblocker, wie Chlorisondamin.
Die neuen Verbindungen werden erhalten, wenn man 3,4-Dihydro-1,2,4-benzothiadiazin-1,1-dioxyde der Formel

mit einen N-Alkylrest einführenden Mitteln behandelt.
So kann man Alkylreste, wie Methyl-oder Äthylreste, einführen durch Umsetzen einer Lösung der 6-Chlor7-sulfamyl-3,4-dihydro-1,2,4-benzothiadiazin-1,1dioxyde in wässriger Alkalimetallhydroxydlösung, wie Lithium-, Natrium-oder Kaliumhydroxydlösung, mit einem reaktionsfähigen Ester eines Alkohols, wie einem Dialkylsulfat, z. B. Dimethyl-oder Diäthylsulfat. Dabei kann auch die Sulfamylgruppe substituiert werden.
Die zu diesem Verfahren zu verwendenden Ausgangsstoffe lassen sich erhalten, wenn man ein 2-Sulf- amyl-anilin der Formel

oder eines seiner Salze mit einem Aldehyd der Formel R.,-CHO, umsetzt, wie dies z. B. in den Schweizer Patenten Nr. 337203 und Nr. 371454 beschrieben ist. Vorzugsweise nimmt man die Umsetzung mit dem Aldehyd in Gegenwart einer Säure, wie einer Mineralsäure, beispielsweise einer Halogenwasserstoffsäure, z. B. Salzsäure oder Bromwasserstoffsäure, oder Schwefelsäure, wenn erwünscht, in wasserfreier Form vor. Der Aldehyd der Formel R.,-CHO kann auch in Form eines seiner Polymeren oder reaktionsfähigen funktionellen Derivate, z. B. Paraformaldehyd, Trioxan, Hexamethylentetramin, oder eines Acetals, z. B.
Dimethoxymethan, Diäthoxy- methan, 1,1-Dimethoxy-äthan oder 1,1-Diäthoxy äthan, verwendet werden. Die Reaktion wird in erster Linie mit ungefähr äquivalenten Mengen der Reaktionskomponenten durchgeführt. Durch höhere Mengen an Aldehyd werden gegebenenfalls die Ausbeuten infolge einer Reaktion mit der

<tb> vermindert. Die Reaktion lässt sich in Abwesenheit oder vorzugsweise in Gegenwart von Lösungsmitteln, wie eines Athers, z. B. p-Dioxan oder Diäthylenglykol-dimethyläthern oder eines Formamids, z. B. Dimethylformamid, bei Raum-oder erhöhter Temperatur und bei normalem oder erhöhtem Druck oder in Gegenwart eines inerten Gases, wie Stickstoff, durchführen.
Als Salze der 2-Sulfamyl-aniline verwendet man bei dieser Reaktion besonders ein solches mit Alkalimetallen oder mit Säuren, in erster Linie mit anorganischen Säuren, wie Halogenwasser stoffsäure, z. B. Salzsäure oder Bromwasserstoffsäure.
Die zu dieser Reaktion zu verwendenden Ausgangsstoffe sind bekannt oder lassen sich nach an sich bekannten Methoden herstellen.
Man kann aber auch die zu verwendenden Ausgangsstoffe erhalten, wenn man in Benzothiadiazin 1, 1-dioxyden der Formel

die C=N-Doppelbindung reduziert, wie dies z. B. in den Schweizer Patenten Nrn. 337205 und 376514 beschrieben ist. Vorzugsweise führt man diese Reduktion mit Alkalimetallborhydriden, wie Lithium-, Kalium-oder besonders Natriumborhydrid, durch.
Diese Metallhydride verwendet man zweckmässig in Gegenwart eines Lösungsmittels, wie wässrigen Lösungen von Alkalimetallhydroxyden, z. B. Lithium-, Natrium-oder Kaliumhydroxyden, eines Äthers, wie Diäthylenglykol-dimethyläther, oder eines flüssi- gen Carbonsäureamids, wie eines Formamids, z. B.
Formamid selbst oder Dimethylformamid. Die Reduktion lässt sich bei Raumtemperatur oder bei erhöh- ter Temperatur, wenn erwünscht, in Gegenwart eines inerten Gases, wie Stickstoff, durchführen. Die C=N Doppelbindung kann aber auch elektrolytisch nach an sich bekannten Verfahrensmethoden reduziert werden.
Die zu dieser Verfahrensmethode verwendeten Ausgangsmaterialien sind bekannt oder lassen sich auf an sich bekannte Weise erhalten.
Je nach den Reaktionsbedingungen erhält man die neuen Verbindungen in freier Form oder in Form ihrer Salze. Erhaltene Metallsalze können z. B. durch Reaktion mit wässrigen sauren Mitteln, wie Mineralsäure, z. B. Halogenwasserstoffsäure, beispielsweise Salzsäure, oder Schwefelsäure, in die freien Verbindungen übergeführt werden. Diese wiederum lassen sich in die Metallsalze, wie Alkalimetallsalze, überführen durch Behandeln z. B. mit einem Metallhydroxyd, wie Natrium-oder Kaliumhydroxyd, in einem Lösungsmittel, wie einem Alkanol, z. B.
Methanol oder Athanol, oder in Wasser, und anschliessendes Abdampfen des Lösungsmittels oder durch Reagierenlassen der freien Verbindung in einem Ather, wie p-Dioxan oder Diäthylenglykol- dimethylätherlösung, mit einem Alkalimetallhydrid oder-amid, z. B. Natrium-oder Kaliumhydrid oder -amid. Dabei lassen sich Mono-oder Polysalze erhalten.
Die neuen Verbindungen können auch in Form ihrer optischen Antipoden vorliegen. Erhaltene Racemate lassen sich in üblicher Weise in ihre Antipoden auftrennen.
Die Erfindung betrifft auch Ausführungsformen des Verfahrens, bei denen ein Ausgangsstoff unter den Reaktionsbedingungen gebildet wird.
Die Temperaturen sind in den nachfolgenden Beispielen in Celsiusgraden angegeben.
Beispiel 1
2,6 g 6-Chlor-7-sulfamyl-3, 4-dihydro-1, 2,4-benzo thiadiazin-1, 1-dioxyd werden in 11 cm3 einer 1-n. wässrigen Natriumhydroxydlösung und 40 cm3 Wasser gelöst. Man kühlt auf 10 ab, gibt 1,4 g Dimethylsulfat zu und rührt 1 Stunde bei 10 und eine weitere Stunde bei Raumtemperatur. Dann filtriert man den Niederschlag ab, kristallisiert ihn 4mal aus wässrigem Athanol um und erhält so das 2-Methyl-6-chlor-7sulfamyl-3,4-dihydro-1,2,4-benzothiadiazin-1,1dioxyd von F. 240-245 .
Aus der Mutterlauge lässt sich das 2-Methyl-6- chlor-7- (N-methyl-sulfamyl)-3,4-dihydro-1,2,4 benzothiadiazin-l, l-dioxyd vom F. 203-206 isolieren.
Beispiel 2
9,4 g 3-Methyl-6-chlor-7-sulfamyl-3, 4-dihydro 1, 2,4-benzothiadiazin-1,1-dioxyd (F. 258-260 ) werden in einer Mischung von 33 cm3 einer 1-n. wässri- gen Natriumhydroxydlösung und 120 cm3 Wasser gelöst. Vom Unlöslichen wird abfiltriert, dann auf 10 gekühlt, 2,4 g Dimethylsulfat zugegeben und bei dieser Temperatur 1 Stunde stehengelassen. Man hält eine weitere Stunde bei Raumtemperatur, filtriert die Reaktionsmischung und kristallisiert den Rück- stand 2mal aus einer 1 : l-Mischung aus Athanol und Wasser um. Man erhält so das 2,3-Dimethyl-6-chlor7-sulfamyl-3,4-dihydro-1,2,4-benzothiadiazin-1,1dioxyd vom F. 274-276 .
Die Umkristallisationsmutterlaugen werden eingeengt, wobei der Rückstand fest wird und sich aus Methanol kristallisieren lässt. Man kristallisiert aus wässrigem Athanol um. Das 2,3-Dimethyl-6-chlor-7 (N-methyl-sulfamyl)-3,4-dihydro-1,2,4-benzothia diazin-1, l-dioxyd schmilzt bei 248-251 .
Das Ausgangsmaterial lässt sich wie folgt erhalten :
Eine Mischung von 2,9 g 5-Chlor-2,4-disulfamylanilin, 20 cm3 wasserfreiem Diäthylenglykol-dimethyl- äther, 0,44 g Acetaldehyd und 0,5 cm3 einer Salzsäurelösung in Essigsäureäthylester (109,5 g Chlorwasserstoffsäure pro Liter) wird auf 80-90a erhitzt und 1 Stunde bei dieser Temperatur gehalten. Die Reaktionsmischung wird unter vermindertem Druck eingeengt, dann Wasser zugefügt und das auskristallisierte Produkt abfiltriert. Das so erhaltene 3-Methyl6-chlor-7-sulfamyl-3,4-dihydro-1,2,4-benzothiadiazin 1, 1-dioxyd vom F. 258-260 lässt sich aus wässrigem Athanol umkristallisieren.
Beispiel 3
Zu einer Lösung von 12,2 g 6-Chlor-7-sulfamyl- 3,4-dihydro-1,2,4-benzothiadiazin-1,1-dioxyd in einer Mischung von 55 cm3 1-n. wässrigem Natriumhydroxyd und 200 cm3 Wasser gibt man 6,9 cm3 Diäthyl- sulfat. Die Reaktionsmischung wird bei 10-20 5 1/2 Stunden gerührt und dann über Nacht bei Raumtemperatur stehengelassen. Der viskose Niederschlag wird abgetrennt, in wenig Athanol gelöst und vom nicht reagierten Ausgangsmaterial durch Filtrieren abgetrennt. Nach dem Stehenlassen des Filtrates bei Raumtemperatur kristallisiert das 2-Athyl-6-chlor7-sulfamyl-3, 4-dihydro-1,2,4-benzothiadiazin-1,1-dioxyd aus, das aus wässrigem Athanol umkristallisiert wird. F. 195-198 .
Beispiel 4
Eine Lösung von 10,6 g 3-n-Butyl-6-chlor-7-sulfamyl-3,4-dihydro-1,2,4-benzothiadiazin-1, 1-dioxyd (F. 176-179 ) in 66 cm3 einer 1-n. wässrigen Na triumhydroxydlösung und 120 cm3 Wasser wird auf 20 abgekühlt und 4,2 g Dimethylsulfat langsam zugegeben. Dann rührt man die Reaktionsmischung
1 Stunde bei dieser Temperatur, dann eine weitere Stunde bei Raumtemperatur und extrahiert 3mal mit Essigsäureäthylester.
Die organische Schicht wird über Natriumsulfat getrocknet, das Lösungsmittel eingedampft, worauf man eine amorphe Mischung von 2-Methyl-3-n-butyl-6-chlor-7-sulfamyl-3, 4-dihydro- 1, 2,4-benzothiadiazin-1,1-dioxyd und 2-Methyl-3-n butyl-6-chlor-7- (N-methyl-sulfamyl)-3, 4-dihydro 1, 2,4-benzothiadiazin-1,1-dioxyd erhält. Sie schmilzt bei 90-95 (unter Zersetzung und Schäumen).
Das Ausgangsmaterial lässt sich wie folgt erhalten :
Eine Mischung von 2,9 g 5-Chlor-2,4-disulfamylanilin, 15 cm3 Diäthylenglykol-dimethyläther, 1, 1 g n-Valeraldehyd und 0,5 cm3 einer gesättigten Lösung von Salzsäure in Essigsäureäthylester wird 2 Stunden auf 80-90 erhitzt. Man dampft die Reaktionsmischung unter vermindertem Druck ein, fügt Wasser zum Rückstand und dekantiert vom ausgefallenen Öl ab. Durch Aufnehmen in Athanol und Stehenlassen bei Raumtemperatur fällt einiges Ausgangsmaterial aus, das abfiltriert wird.
Das Filtrat wird zur Trockne eingedampft, der Rückstand mit Benzol behandelt, dann mit wässrigem Athanol und ergibt das 3-n-Butyl-6-chlor-7-sulfamyl-3, 4-dihydro-1, 2,4 benzothiadiazin-1, 1-dioxyd, das nach dem Umkristallisieren aus wässrigem Athanol bei 176-179 schmilzt.
In analoger Weise zu den vorstehenden Beispielen erhält man, ausgehend von dem entsprechenden Ausgangsmaterial, die folgenden Verbindungen :
2-Methyl-3-benzyl-6-chlor-7-sulfamyl-3, 4-dihydro 1, 2,4-benzothiadiazin-l, 1-dioxyd und 2-Methyl-3-benzyl-6-chlor-7- (N-methyl-sulfamyl)- 3,4-dihydro-1,2,4-benzothiadiazin-1, 1-dioxyd.
2-Athyl-6-chlor-7- (N-äthyl-sulfamyl)-3, 4-dihydro 1, 2,4-benzothiadiazin-1,1-dioxyd vom F. 163-166 (aus wässrigem Athanol).
2-n-Butyl-6-chlor-7- (N-n-butyl-sulfamyl)-3, 4-dihydro-1, 2,4-benzothiadiazin-1, 1-dioxyd vom F. 170 171 .
Claims (1)
- PATENTANSPRUCH Verfahren zur Herstellung von 3,4-Dihydro-1,2,4 benzothiadiazin-1, 1-dioxyden der Formel EMI4.1 worin Ri für einen Alkylrest, R und R4 für Wasserstoff oder Alkylreste und R für Wasserstoff, einen unsubstituierten oder substituierten Kohlenwasserstoff-oder heterocyclischen Rest stehen, dadurch gekennzeichnet, dass man 3,4-Dihydro-1,2,4-benzo thiadiazin-1, 1-dioxyde der Formel EMI4.2 mit einen N-Alkylrest einführenden Mitteln behandelt.UNTERANSPRUCHE 1. Verfahren nach Patentanspruch, dadurch gekennzeichnet, dass man mit einem reaktionsfähigen Ester eines Alkanols umsetzt.2. Verfahren nach Patentanspruch und Unteranspruch 1, dadurch gekennzeichnet, dass man mit Dialkylsulfat umsetzt.3. Verfahren nach Patentanspruch und den Unteransprüchen 1 und 2, dadurch gekennzeichnet, dass man als Ausgangsstoffe Verbindungen der Formel EMI4.3 verwendet, worin R2 für Wasserstoff, einen unsubstituierten oder durch Halogen, freie, veresterte oder verätherte Hydroxylgruppen oder durch Aminogruppen substituierten aliphatischen Kohlenwasserstoffrest mit 1-8 Kohlenstoffatomen, einen Cycloalkyloder Cycloalkenylrest, einen Phenyl-, Pyridyl-oder einen Phenylalkylrest steht.4. Verfahren nach Patentanspruch und den Un teransprüchen 1 und 2, dadurch gekennzeichnet, dass man als Ausgangsstoffe Verbindungen der Formel EMI5.1 verwendet, worin R2 für Wasserstoff, einen unsubstituierten oder durch Amino-oder Hydroxylgruppen oder Chlor substituierten Alkylrest mit 1-5 Kohlenstoffatomen, den Cyclohexenyl-, Benzyl-oder Phenyl äthylrest steht.5. Verfahren nach Patentanspruch und den Unteransprüchen 1 und 2, dadurch gekennzeichnet, dass man das 6-Chlor-7-sulfamyl-3,4-dihydro-1,2,4-benzo thiadiazin-1, 1-dioxyd mit Dimethylsulfat umsetzt.6. Verfahren nach Patentanspruch und den Unteransprüchen 1 und 2, dadurch gekennzeichnet, dass man das 6-Chlor-7-sulfamyl-3, 4-dihydro-1, 2,4-benzo thiadiazin-1, 1-dioxyd mit Diäthylsulfat umsetzt.7. Verfahren nach Patentanspruch und den Unteransprüchen 1 und 2, dadurch gekennzeichnet, dass man das 3-Methyl-6-chlor-7-sulfamyl-3, 4-dihydro 1, 2,4-benzothiadiazin-1,1-dioxyd mit Dimethylsulfat umsetzt.8. Verfahren nach Patentanspruch, dadurch gekennzeichnet, dass man erhaltene Benzothiadiazine in ihre Metall-oder Säureadditionssalze überführt.
