Description
Verfahren zur Herstellung von Phenanthrenderivaten Die vorliegende Erfindung bezieht sich auf ein Verfahren zur Herstellung neuartiger wertvoller Phen- anthrenverbindungen der allgemeinen Formel.

Die genannten Verbindungen sind wirksame und selektive Antiandrogenmittel.
In den oben erwähnten Phenanthrenverbindungen 1 kann für R ein Kohlenwasserstoffradikal beispielsweise angeführt werden, Methyl, Äthyl, Propyl, Isopropyl, Butyl, Pentyl, Hexyl, Cyclohexyl, Phenyl, Benzyl, Tolyl, Allyl, Vinyl, Äthynyl und dg., und für R' z.B. Äthyl, Propyl, Isopropyl, Butyl, Pentyl, Hexyl, Phenyl, Cyclo- hexyl, Allyl, Vinyl, Äthynyl, und dergl.
Das erfindungsgemässe Verfahren zur Herstellung oben genannter Phenanthrenverbindungen I ist dadurch gekennzeichnet, dass man ein 2-Tetralon der Formel:

Die durch die Formel<B>(1)</B> dargestellten und oben spezifisch illustrierten neuartigen Phenanthrenverbin- dungen weisen eine starke Antiandrogenwirkung und keine wesentlichen Nebenwirkungen auf (sie sind z.B. nicht toxisch in Blutbildanalysen und histologischen Untersuchungen und haben keine Wirkung auf die Sekretion der Geschlechtsdrüsen oder auf andere Hor mone). Z.
B. wirkt 4a-Äthyl-7-methoxy-2, 3, 4, 4a, 9, 10-hexahydrophenanthren-2-on (Verbindung 1 in nach folgender Tabelle), 4a-Propyl-7-methoxy-2, 3, 4, 4a, 9, 10-hexahydrophenanthren-2-on (Verbindung II), 4a-Al- lyl-7-methoxy-2, 3, 4, 4a, 9, 10-hexahydrophenanthren- 2-on (Verbindung III) und 4a-Äthyl-7-hydroxy-2, 3, 4, 4a, 9, 10-hexahydrophenanthren-2-on (Verbindung IV) der durch 1/10 der Menge Testosteronpropionat (TP)
hervorgerufenen Reaktion entgegen und entspricht der durch eine zweifache Menge Proge-steron verursachten Androgenicitätsreaktion bei Ratten im Falle von subku tanen Injektionen an<B>10</B> aufeinander folgenden Tagen <B>(10</B> mg/1 x im Tag), wie aus der folgenden Tabelle hervorgeht.

Als Katalysator kann beispielsweise ge nannt werden eine anorganische Base, wie ein Alkalime tallhydroxyd, z. B. Natriumhydroxyd, Kaliumhydroxyd, oder ähnliche, und Erdalkalimetallhydroxyd, z.B. Kal- ciumhydroxyd, ein Alkalimetallcarbonat z.B. Kaliumcar- bonat, ein Alkalimetallalkoxyd, z.B. Natriummethoxyd, Kalium-t-butoxyd, Natriumisopropoxyd oder ähnliche, ein Alkalimetallamid oder -hydrid, z.B. Natriumamid, Natriumhydrid; eine organische Base, wie ein Amin, Aminsalz oder Ammoniumverbindung, z.
B. Triäthyl- amin, Dimethylanilin, Pyridin, Pyridin-hydrochlorid, Pi- colin, Collidin, Trimethylbenzylammonium-hydroxyd oder ähnliche und Triphenylmethylnatrium oder ähnli che. Die Reaktion mit III, z.B.
Acrylonitril oder ähnl., kann in einem inerten Lösungsmittel, wie Methanol, Äthanol, Isopropanol, t-Butanol, Benzol, Äther, Tetra- hydrofuran, Dioxan, oder ähnl., oder Gemische dersel ben, oder in manchen Fällen ohne Lösungsmittel, in einem Temperaturbereich vom Schmelzpunkt bis zum Siedepunkt des Lösungsmittels, innerhalb einer Reak tionsdauer nicht mehr als zwei Tagen, durchgeführt werden; dabei entsteht ein Zwischenprodukt der For mel:

welche mit einem Alkali, z.B. Natriumhydroxyd oder ähnliche, hydrolysiert und dann einer Grignard-Reaktion mit einem Alkylmagnesiumhalogenid (B-CH2MgX, worin B die obige Bedeutung hat und X ein Halogen atom ist) und einem Ringschluss unterworfen wird.
Im oben angeführten erfindungsgemässen Verfahren brauchen die Reaktionszwischenprodukte nicht immer isoliert und gereinigt zu werden, nachdem durch die Isolierung in jeder Stufe nicht die besten Erträge erlangt werden. Für die Isolierung oder die Reinigung der Produkte können beispielsweise Destillationsverfahren, insbesondere Destillation unter vermindertem Druck, Chromatographie, oder chemische Umwandlung durch ein Carbonylderivat mit einem Carbonylreaktionsmittel, wie Semicarbazid, Girard T oder ähnliche, verwendet werden.
<I>Beispiel</I> 4a-Äthyl-7-methoxy-2, 3, 4, 4a, 9, 10-hexahydro- phenanthren -2-on Zu einer Lösung von 16,2 g Acrylonitril in einem Liter Dioxan werden 51 g 1-Äthyl-6-methoxy -1,2,3,4- tetrahydronaphthalen -2-on und 18 ml einer wässrigen 20 %igen Benzyltrimethyl-ammoniumhydroxyd-Lösung (Triton B) bei etwa<B>5-10</B> 'C zugesetzt, und die so erhaltene Lösung wird dann während 24 Stunden bei Zimmertemperatur unter einer Stickstoffatmosphäre ge rührt. Nach Entfernen des Dioxans unter vermindertem Druck wird der Rückstand mit Dichlormethan extra hiert.
Der Extrakt wird mit Wasser und dann mit einer Natriumchlorid-Lösung gewaschen, über wasserfreiem Natriumsulfat getrocknet und dann unter vermindertem Druck abgedampft; man erhält ein rötlich-orange gefärb tes Ölprodukt, das 51,4 g 1-Äthyl-2 -oxo-6- methoxy- 1,2,3,4-tetrahydronaphthalen-l-proprionitril, mit einem Ertrag von 79,9 % ergibt. (n24 D 1,5464, Kp 0,1-0,2 mm Hg 170-174 'C. Analyse berechnet für C17H19O2N C 74,68, H 7,44, N 5,44 Gefunden<B>C 74,81,</B> H<B>7,66, N 5,26</B> IR: v CCl 4 2227, 1714, 1613, 1578, 1503, 1266, max 1043 cm-') durch Destillation.
Zu einer Lösung von<B>30,5 g</B> oben erhaltener Verbin dung in<B>250</B> ml Äthanol werden<B>250</B> ml einer wässrigen 20 %igen Kaliumhydroxyd-Lösung zugegeben, und das erhaltene Gemisch wird während<B>6</B> Stunden unter einer Stickstoffatmosphäre bei Rückfluss behandelt, wobei sich Ammoniakgas entwickelt. Das Rekationsgemisch wird unter vermindertem Druck, zwecks Entfernung des Äthanols, eingeengt und der Rückstand wird mit Wasser verdünnt. Die so erhaltene alkalische Lösung wird mit Dichlormethan gewaschen, mit konzentrierter Chlorwas serstoffsäure angesäuert und mit Äther extrahiert.
Da nach wird der Extrakt mit Natriumchloridlösung gewa schen, über wasserfreiem Natriumsulfat getrocknet, und unter vermindertem Druck abgedampft; man erhält 31,6 g an roher Ketosäure, das ist der 1-Äthyl-2-oxo-6- methoxy-1, 2, 3, 4-tetrahydronaphthalen-1-propionsäure (IR: CCl 4 3200-2500 breit, 1710, 1610, 1575, max <B>1501, 1265,</B> 1045 cm-').
<B>1,38 g</B> der erhaltenen Ketosäure wird in einem Gemisch von 40 ml Essigsäure-Anhydrid und 0,02<B>g</B> wasserfreiem Natriumacetat gelöst. Die Lösung wird während drei Stunden unter einer Stickstoffatmosphäre bei Rückfluss erhitzt und dann, zwecks Entfernen des Essigsäureanhydrids, unter vermindertem Druck destil liert. Das so erhaltene Konzentrat wird mit Wasser verdünnt und mit Äther extrahiert.
Der Extrakt wird mit verdünnter wässeriger Natriumcarbonatlösung und dann mit gesättigter wässriger Natriumchloridlösung gewa schen, über wasserfreiem Natriumsulfatd getrocknet, und unter vermindertem Druck abgedampft; man erhält 1, 14 g des entsprechenden Enol-lactons, d. 1. das 4a- Äthyl-7-methoxy-3, 4, 4a, 9-tetrahydro-2H-1-oxaphen- anthren-2-on in rohem Zustand. Durch das Destillieren erhält man eine reine Substanz von Kp0,2-0,3 bei 200-220 'C.

<tb> max 1,14 g des erhaltenen rohen Enol-lactons werden in einem Gemisch von<B>100</B> ml trockenem Äther und<B>30</B> ml trockenem Benzol gelöst und die so erhaltene Lösung wird während<B>7,5</B> Stunden bei<B>-50</B> 'C gerührt, während welcher Zeitspanne eine Lösung von Methylmagnesium- jodid hergestellt aus 1,2 g Magnesiummetall und 7,0 g Methyljodid in 30 ml trockenem Äther, tropfenweise zugesetzt.
Zu diesem Reaktionsgemisch werden dann 12 ml Aceton und 50 ml wässriger 20 %iger Essigsäure- Lösung unter Kühlung zugegeben. Die organische Schicht wird gesammelt und mit dem ätherischen Ex trakt aus der wässrigen Schicht vereinigt. Die vereinigten organischen Schichten werden dann mit wässerigem verdünntem Natriumcarbonat und dann mit Wasser gewaschen, über wasserfreiem Natriumsulfat getrocknet und unter vermindertem Druck abgedampft; man erhält 1,4 g eines öligen Rückstandes, der hauptsächlich aus 1- Äthyl-1-(3-oxobutyl)-6-methoxy-1, 2, 3, 4 tetrahydro- naphthalen -2-on besteht.
1,4<B>g</B> des erhaltenen öligen Rückstandes werden in<B>60</B> ml Methanol gelöst und zu einer Lösung 2,0 g Kalium hydroxyd in 6,0 ml Wasser gefügt. Das Gemisch wird während<B>1,5</B> Stunden unter einer Stickstoffatmosphäre bei Rückfluss erhitzt und dann abgedampft. Der Rück stand wird mit einem Gemisch von Wasser und Äther behandelt und die Ätherschicht wird gesammelt. Die Ätherschicht wird dann mit verdünnter wässriger Na triumchloridlösung gewaschen, über Natriumsulfat ge trocknet und abgedampft; man erhält 0,8 g an rohem 4a- Äthyl-7-methoxy-2, 3, 4, 4a, 9, 10-hexahydrophenan- thren-2-on als gelblich- orange gefärbtes Öl.
Dieses so erhaltene rohe Produkt wird in das entsprechende Semicarbazon mit Hilfe des üblichen Verfahrens unter Verwendung von<B>13</B> ml Methanol, 434 mg Semicarbazid-hydrochlorid, 319 mg Natriumacetat und<B>2,5</B> ml Wasser, umgewandelt; man erhält 0,4<B>g</B> des Semicarbazons mit F von 2119-220 'C. Man gewinnt das freie Keton auf übliche Weise unter Verwendung von Essigsäure, Pyrotraubensäure und Natriumacetat in Wasser und man erhält, nach, Umkristallisieren aus Äther-penthan, 264 mg reiner Kristalle von 4a-Äthyl-7 - methoxy-2, 3, 4, 4a 9, 10-hexahydrophenanthren-2-on, F von<B>75-76</B> 'C.
Claims (1)
- <B>PATENTANSPRUCH</B> Verfahren zur Herstellung von Verbindungen der Formel EMI0004.0021 worin R Wasserstoff oder ein Kohlenwasserstoffrest, R' ein 2-6 Kohlenstoffatome aufweisender Kohlenwasser stoffrest ist, und A und B für Wasserstoffatome oder Niederalkyle, stehen, dadurch gekennzeichnet, dass man ein 2-Tetralon der Formel EMI0004.0022 mit einer Verbindung der Formel EMI0004.0023 in Gegenwart eines basischen Katalysators zur Reaktion bringt, das erhaltene Zwischenprodukt mittels Alkalibe handlung hydrolysiert, mit einer Grignard-Verbindung der Formel B-CH2MgX, worin X ein Halogen be deutet<B>'</B> umsetzt und das erhaltene Produkt einem Ringschluss unterwirft.<I>Anmerkung des</I> Eidg. <I>Amtes für geistiges Eigentum:</I> Sollten Teile der Beschreibung mit der im Patentansprach gegebenen Definition der Erfindung nicht in Einklang ste hen, so sei daran erinnert, dass gemäss Art.<B>51</B> des Patentgesetzes der Patentanspruch für den sachlichen Geltungsbe reich des Patentes niassgebend ist.
