Description
Verfahren zur Herstellung von neuen SulfonylharnstofFen
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von neuen Sulfonylharnstoffen.
Verbindungen der allgemeinen Formel I,

in welcher Ri eine Alkyl-oder Alkenylgruppe von höchstens 10 Kohlenstoffatomen, eine Cycloalkyl-oder Cycloalkenylgruppe von höchstens 12 Kohlenstoff- atomen, oder eine Polycycloalkylgruppe von höchstens
11 Kohlenstoffatomen bedeutet, sowie ihre Salze mit anorganischen oder organischen Basen sind bisher nicht bekanntgeworden.
Wie nun gefunden wurde, besitzen diese Verbindungen sowie ihre pharmazeutisch annehmbaren Salze wertvolle pharmakologische Eigenschaften. Sie zeigen überraschenderweise bei peroraler oder parenteraler Verabreichung lang anhaltende hypoglykämische Wirkung, die sie als geeignet zur Behandlung der Zuckerkrankheit charakterisiert.
In den Verbindungen der allgemeinen Formel I kann Rl die folgenden Gruppen sein : als Alkylgruppe z. B. die Propyl-, Isopropyl-, Butyl-, Isobutyl-, sek. Butyl-, tert. Butyl-, Pentyl-, Isopentyl-, tert. Pentyl-, 1 Methyl-butyl-, 2-Methyl-butyl-, 1,2-Dimethyl-propyl-, 2,2-Dimethyl-propyl-, Hexyl-, 2-Methyl-pentyl-, Heptyl-, 4-Methyl-hexyl-, Octyl-, Nonyl-oder die Decylgruppe ; als Alkenylgruppe z. B. die Allyl-, Crotyl-oder Methallylgruppe ; als Cycloalkylgruppe z.
B. die Cyclobutyl-, Cyclopentyl-, Cyclohexyl-, Cycloheptyl-, Cyclooctyl-, Cyclononyl-, Cyclodecyl-, Cyclododecyl-, Cyclopentylmethyl-, Cyclohexylmethyl-, Cycloheptylmethyl-oder die Cyclooctylmethylgruppe sowie Derivate von solchen Resten, die gegebenenfalls durch Methyl-, Athyl-oder tert. Butylgruppen substituiert sind ; ferner als Cycloalkenylgruppe, z. B. die 2-Cyclopenten-1-yl-oder die 3-Cyclohexen-1-yl-gruppe und als Polycycloalkylgruppe z. B. die Bornyl-, Norbornyl-, Norbornylmethyl-, Adamantyl-oder die Octahydro-1, 2,4-methenopentalenylgruppe.
Die Verbindungen der allgemeinen Formel I werden erfindungsgemäss hergestellt, indem man eine Verbindung der allgemeinen Formel II,

in welchen Ri die unter Formel I angegebene Bedeutung hat, umsetzt, nötigenfalls das Reaktionsprodukt zur Abspaltung eines an den Ringstickstoff gebundenen Acylrestes partiell hydrolysiert und gewünschtenfalls eine erhaltene Verbindung der allgemeinen Formel I mit einer anorganischen oder organischen Base in ein Salz überführt.
Ra kann als Acylrest von Verbindungen der allgemeinen Formel II z. B. eine niedere Alkanoylgruppe, insbesondere die Acetylgruppe, oder eine niedere Alkoxycarbonylgruppe, wie z. B. die Methoxycarbonyl-oder Athoxycarbonylgruppe, sein. Als reaktionsfähige Ester von Carbaminsäuren der allgemeinen Formel IV kommen niedere Alkylester, insbesondere der Methyl-oder Äthylester, ferner auch Phenylester, in Betracht.
Die erfindungsgemässe Umsetzung erfolgt beispielsweise in der Kälte oder durch Erwärmen, vorzugsweise in einem inerten organischen Lösungsmittel und in Gegenwart eines Kondensationsmittels. Geeignete inerte organische Lösungsmittel sind beispielsweise Kohlen- wasserstoffe, wie Benzol, Toluol oder Xylol, ätherartige Flüssigkeiten, wie Ather, Dioxan oder Tetrahydrofuran, chlorierte Kohlenwasserstoffe, wie Methylenchlorid, oder niedere Ketone, wie Aceton oder Methyläthylketon. Als Kondensationsmittel kann ein Alkalimetallhydroxid, wie Natrium-oder Kaliumhydroxid, oder ein Alkalimetallalkanolat, wie Natriumoder Kaliummethylat oder ein entsprechendes Äthylat, eingesetzt werden.
Als weitere Kondensationsmittel kön- nen bei der Umsetzung eines Isocyanderivates tertiäre organische Basen Verwendung finden.
Verfahrensgemäss erhaltene Reaktionsprodukte, die am Ringstickstoff durch eine Acylgruppe substituiert sind und vorzugsweise der allgemeinen Formel V

entsprechen, in welcher R eine niedere Alkanoyl-oder Alkoxycarbonylgruppe bedeutet und Ri die unter Formel I angegebene Bedeutung hat, werden anschliessend durch partielle Hydrolyse in Verbindungen der allgemeinen Formel I übergeführt. Die partielle Hydrolyse wird vorzugsweise in alkalischem Milieu, beispielsweise in methanolischer oder äthanolischer Natrium-oder Kaliumhydroxidlösung vorgenommen.
Ausgangsstoffe der allgemeinen Formel II, das 10,11-Dihydro-5H-dibenz [b, f] azepin-3-sulfonamid sowie das entsprechende 5-Acetylderivat, sind in der Literatur beschrieben (vgl. J. R. Geigy AG, belgische Patentschrift Nr. 616 134). Weitere Verbindungen von diesem Typus können analog hergestellt werden.
Die neuen Wirkstoffe oder pharmazeutisch annehmbaren Salze derselben werden vorzugsweise peroral verabreicht. Zur Salzbildung können anorganische oder organische Basen, wie beispielsweise Alkali-oder Erdalkalihydroxide, Carbonate oder Bicarbonate, Triätha- nolamin, Cholin, Nt-Dimethyl-oder N1-(ss-Phenyläthyl)- biguanid, verwendet werden. Die täglichen Dosen bewegen sich zwischen 50 und 2000 mg für erwachsene Patienten. Geeignete Doseneinheitsformen, wie Dragées, Tabletten oder Steckkapseln, enthalten vorzugsweise 25-500 mg eines erfindungsgemässen Wirkstoffes, und zwar 20 bis 80% einer Verbindung der allgemeinen Formel I.
Zur Herstellung von Dragées und Tabletten kombiniert man den Wirkstoff z. B. mit festen pulverförmi- gen Trägerstoffen, wie Lactose, Saccharose, Sorbit, Mannit ; Stärken, wie Kartoffelstärke, Maisstärke oder Amylopektin, ferner Laminariapulver oder Citruspulpenpulver ; Cellulosederivaten oder Gelatine, gegebenenfalls unter Zusatz von Gleitmitteln, wie Magnesiumoder Calciumstearat oder Polyäthylenglykolen, zu Tabletten oder zu Dragee-Kernen. Letztere überzieht man beispielsweise mit konzentrierten Zuckerlösungen, welche z. B. noch arabischen Gummi, Talk und/oder Titandioxid enthalten können, oder mit einem in leichtflüchtigen organischen Lösungsmitteln oder Lösungs mittelgemischen gelösten Lack.
Diesen tJberzügen kön- nen Farbstoffe zugefügt werden, z. B. zur Kennzeichnung verschiedener Wirkstoffdosen.
Die erwähnten Steckkapseln aus Gelatine enthalten den Wirkstoff vorzugsweise als Granulat, z. B. in Mischung mit Füllstoffen, wie Maisstärke und/oder Gleitmitteln, wie Talk oder Magnesiumstearat, und gegebenenfalls Stabilisatoren, wie Natriummetabisulfit (NagS2OD) oder Ascorbinsäure.
Die folgenden Vorschriften sollen die Herstellung von Tabletten und Dragees näher erläutern : a) 1000 g 1-(10, 11-Dihydro-5H-dibenz [b, f]-azepin- 3-yl-sulfonyl)-3-butyl-harnstoff werden mit 550 g Lactose und 292 g Kartoffelstärke vermischt, die Mischung mit einer wässrigen Lösung von 8,0 g Gelatine befeuchtet und durch ein Sieb granuliert.
Nach dem Trocknen mischt man 60,0 g Kartoffelstärke, 60,0 g Talk, 10,0 g Magnesiumstearat und 20,0 g kolloidales Siliciumdioxid zu und presst die Mischung zu 10000 Tabletten von je 200 mg Gewicht und 100 mg Wirkstoffgehalt, die gewünschtenfalls mit Teilkerben zur feineren Anpassung der Dosierung versehen sein können. b) Aus 1000 g Natrimusalz von 1- (10, 11-Dihydro- 5H-dibenz-[b, f] azepin-3-ylsulfonyl)-3-butyl-harnstoff, 379,0 g Lactose und der wässrigen Lösung von 6,0 g Gelatine stellt man ein Granulat her, das man nach dem Trocknen mit 10, 0 g kolloidalem Siliciumdioxid, 40,0 g Talk, 60,0 g Kartoffelstärke und 5,0 g Magnesiumstearat mischt und zu 10000 Dragee-Kernen presst.
Diese werden anschliessend mit einem konzentrierten Sirup aus 533,5 g krist. Saccharose, 20,0 g Schellack, 75,0 g arabischem Gummi, 250 g Talk, 20 g kolloida- lem Siliciumdioxid und 1,5 g Farbstoff überzogen und getrocknet. Die erhaltenen Dragées wiegen je 240 mg und enthalten je 100 mg Wirkstoff.
Die nachfolgenden Beispiele erläutern die Herstellung der neuen Verbindungen der allgemeinen Formel I und von bisher nicht beschriebenen Zwischenprodukten näher, stellen jedoch keineswegs die einzige Ausführungsform derselben dar. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel 1 a) 31,6 g 5-Acetyl-10,11-dihydro-5H-dibenz [b, f]- azepin-3-sulfonamid (vgl. J. R. Geigy AG, belgische Patentschrift Nr. 616134) werden in 25 ml 5n Natronlauge und 200 ml Dioxan unter Rühren gelöst und mit 8,5 g Propylisocyanat versetzt. Eine leicht exotherme Reaktion setzt ein. Nachdem diese abgeklungen ist, wird das Reaktionsgemisch eine Stunde bei 80 gerührt, anschliessend auf 20 gekühlt und im Vakuum eingeengt. Man löst den Rückstand in Wasser, filtriert die Lösung und giesst sie auf eiskalte 2n Salzsäure. Das Rohprodukt fällt aus. Es wird abgesaugt, mit Wasser gewaschen, mit 2n Ammoniak versetzt, der Rückstand abfiltriert und die Ammoniaklösung auf eiskalte 2n Salzsäure gegossen.
Der rohe 1- (5-Acetyl-10, 11-di- hydro-5H-dibenz [b, f] azepin-3-yl-sulfonyl)-3-propyl- harnstoff fällt aus. Das Rohprodukt wird abgesaugt, mit Wasser gründlich gewaschen und im Vakuum bei 60 getrocknet. b) 40,1 g roher 1-(5-Acetyl-10, 11-dihydro-5H- dibenz [b, f] azepin-3-ylsulfonyl)-3-propyl-harnstoff wer den in 400 ml Athanol gelöst und mit 20,0 g Natriumhydroxid versetzt. Man kocht das Gemisch 3 Stunden unter Rückfluss, wobei das Natriumsalz von 1- (10, 11 Dihydro-5H-dibenz [b, f] azepin-3-yl-sulfonyl)-3-propylharnstoff auskristallisiert. Das Natriumsalz wird abfiltriert, mit Athanol nachgewaschen, aus 50 % igem Athanol (Volumprozente) umkristallisiert und in 300 ml warmem Wasser gelöst.
Diese Lösung giesst man auf 2n Salzsäure. Der freie, rohe 1- (10, 11-Dihydro-5H- dibenz [b, f]'azepin-3-yl-sulfonyl)-3-propyl-harnstoff fällt aus. Das Rohprodukt wird abgesaugt, gründlich mit Wasser gewaschen und aus Athanol umkristallisiert, worauf die reine Verbindung nach vorangehendem Sintern bei 183-185 schmilzt.
Beispiel 2 a) Analog Beispiel la stellt man, ausgehend von 5 Acetyl-10,11-dihydro-5H-dibenz [b, f] azepin-3-sulfon- amid die folgenden Zwischenprodukte her : al mit Isopropylisocyanat den 1- (5-Acetyl-10, 11- dihydro-5H-dibenz [b, f] azepin-3-ylsulfonyl)-3- isopropyl-harnstoff (Rohprodukt) ; a'mit Butylisocyanat den amorphen 1- (5-Acetyl-
10,11-dihydro-5H-dibenz [b, f] azepin-3-ylsulfonyl)-
3-butyl-harnstoff vom Zersetzungspunkt 100-105 ; mit sek.
Butylisocyanat den 1- (5-Acetyl-10, 11- dihydro-5H-dibenz [b, f] azepin-3-ylsulfonyl)-
3-sek. butyl-harnstoff (1/3 Mol Kristallwasser ent haltend) vom Smp. 118-120 ; a ; mit Isopentylisocyanat den 1-(5-Acetyl-10, 11- dihydro-5H-dibenz [b, f] azepin-3-ylsulfonyl)-3- isopentyl-harnstoff vom Smp. 106-110 ;

Beispiel 3
27,4 g 10,11-Dihydro-5H-dibenz [b, f] azepin-3-sulfonamid (vgl. J. R. Geigy AG, belgische Patentschrift Nr. 616 134) und 5,6 g fein pulverisiertes Kaliumhydroxid werden in 250 ml Dioxan gelöst und mit 9,9 g Butylisocyanat eine Stunde unter Rückfluss gekocht.
Dann kühlt man das Reaktionsgemisch ab und dampft es im Vakuum ein. Den Rückstand nimmt man in 300 ml heissem Wasser auf, filtriert von wenig Unlös- lichem ab, kühlt das Filtrat und giesst es unter Rühren auf 2n Salzsäure. Das Rohprodukt fällt aus ; es wird abfiltriert, mit Wasser nachgewaschen und in 2n Ammoniaklösung aufgenommen. Die Ammoniaklösung wird von wenig Unlöslichem abgetrennt und wiederum auf 2n Salzsäure gegossen. Das Rohprodukt fällt aus ; es wird abfiltriert, gründlich mit Wasser gewaschen und aus Aceton umkristallisiert. Der erhaltene, reine 1- (10, 11-Dihydro-SH-dibenz [b, f] azepin-3-ylsulfonyl)-3-butyl- harnstoff schmilzt bei 183,5-185,5 .
Dieses Harnstoffderivat wird wie folgt in das Natriumsalz übergeführt : 37,3 g der erhaltenen Verbindung werden in 400 ml Athanol, das 5 g pulverisiertes Natriumhydroxid enthält, unter Rückfluss erhitzt. Dann wird das Reaktionsgemisch abgekühlt und das ausgefallene Natriumsalz des 1- (10, 11-Dihydro-5H-dibenz- [b, f] azepin-3-ylsulfonyl)-3-butyl-harnstoffs abfiltriert und im Vakuum getrocknet.
Beispiel 4
Aus 27,4 g 10,11-Dihydro-5H-dibenz [b, f] lazepin-3- sulfonamid und 5,6 g pulverisiertem Kaliumhydroxid in 250 ml Dioxan erhält man analog Beispiel 3 folgende Endprodukte : a) mit 8,5 g Propylisocyanat den 1- (10, 11-Dihydro-
5H-dibenz [b, f] azepin-3-ylsulfonyl)-3-propyl harnstoff vom Smp. 183-183,5 (aus Athanol) ; b) mit 8,5 g Isopropylisocyanat den 1- (14, 11-Dihydro-
5H-dibenz [b, f] azepin-3-ylsulfonyl)-3-isopropyl harnstoff vom Smp. 196-197 ; c) mit 9,9 g sek.
Butylisocyanat den 1- (10, 11-Dihydro-
5H-dibenz [b, f] azepin-3-ylsulfonyl)-3-sek. butyl harnstoff vom Smp. 184-185 (aus Methanol) ; d) mit 11,3 g Isopentylisocyanat den 1- (10, 11-Di- hydro-5H-dibenz [b, f] azepin-3-ylsulfonyl)-3- isopentyl-harnstoff vom Smp. 170,5-171,5 (aus Methanol) ;
e) mit 12,5 g Cyclohexylisocyanat den 1- (10, 11-Di hydro-5H-dibenz [b, f] azepin-3-ylsulfonyl)-3-cyclo hexyl-harnstoff vom Smp. 186-188 , und f) mit 16,1 g Octahydro-1, 2,4-methenopentalen-5-yl isocyanat den l-(10, 11-Dihydro-5H-dibenz [b, f]- azepin-3-ylsulfonyl)-3- (octahydro-1, 2,4-metheno pentalen-5-yl)-harnstoff vom Smp. 182 (aus
Essigsäureäthylester).

- PATENTANSPRUCH Verfahren zur Herstellung von neuen Sulfonylharnstoffen der allgemeinen Formel I, EMI4.1 in welcher Ri eine Alkyl-oder Alkenylgruppe von höch- stens 10 Kohlenstoffatomen, eine Cycloalkyl-oder Cycloalkenylgruppe von höchstens 12 Kohlenstoff- atomen, oder eine Polycycloalkylgruppe von höchstens 11 Kohlenstoffatomen bedeutet, sowie ihre Salze mit anorganischen oder organischen Basen, dadurch gekennzeichnet, dass man eine Verbindung der allgemeinen Formel II, EMI4.2 in welcher R2 Wasserstoff oder einen Acylrest bedeutet, mit einem Isocyanderivat der allgemeinen Formel III, Ri-N=C=O (III) oder mit einem reaktionsfähigen Ester einer Carbaminsäure der allgemeinen Formel IV,EMI4.3 in welchen Ri die unter Formel I angegebene Bedeutung hat, umsetzt, nötigenfalls das Reaktionsprodukt zur Abspaltung eines an den Ringstickstoff gebundenen Acylrestes partiell hydrolysiert und gewünschtenfalls eine erhaltene Verbindung der allgemeinen Formel I mit einer anorganischen oder organischen Base in ein Salz überführt.
