Description
Verfahren zur Herstellung von Alkanolaminderivaten Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von neuen Alkanolaminderivaten, die ss-adrenergische Blockierungswirkung besitzen und da her für die Behandlung oder Prophylaxe von Herz krankheiten, z. B. Angina pectoris und Herzarrhyth mien, und für die Behandlung von erhöhtem Blutdruck und Phäochromocytom beim Menschen brauchbar sind.
Erfindungsgemäss werden neue Alkanolaminderivate der Formel:

worin Rl einen Alkylrest mit bis zu 12 Kohlenstoff atomen, der gegebenenfalls durch einen oder zwei der folgenden Substituenten: Hydroxylgruppen, Alkoxy- gruppen mit bis zu 5 Kohlenstoffatomen, Phenyl- und Phenoxygruppen substituiert ist, wobei die Phenyl- und Phenoxygruppen ihrerseits gegebenenfalls durch ein oder mehrere Chlor- oder Bromatome oder Methyl-, Äthyl-, Methoxy- oder Äthogruppen weiter substituiert sind, oder R1 einen Cycloalkylrest mit bis zu 8 Kohlen stoffatomen, R2 einen Alkanoyl-, Cycloalkancarbonyl-, Aroyl-, Aralkanoyl-, Aryloxyalkanoyl- oder Arylsulfo- nylrest, je mit bis zu 10 Kohlenstoffatomen, oder einen Halogenalkanoyl-,
Alkylsulfonyl- oder Alkoxy- carbonylrest, je mit bis zu 6 Kohlenstoffatomen, und R3 ein Halogenatom, den Cyanorest oder einen Alkyl- thio-, Cycloalkyl-, Alkanoyl- oder Alkoxycarbonylrest, je mit bis zu 6 Kohlenstoffatomen, oder den Phenyl- oder Phenoxyrest, welche gegebenenfalls durch ein oder mehrere Halogenatome oder Nitrogruppen oder Alkyl- oder Alkoxygruppen mit bis zu 4 Kohlenstoffatomen substituiert sind, oder einen Alkylrest mit bis zu 4 Koh lenstoffatomen,
welcher durch die Hydroxyl- oder Phe nylgruppe oder eine Alkoxygruppe mit bis zu 6 Kohlen stoffatomen oder ein oder mehrere Halogenatome sub stituiert ist, darstellen, sowie die Säureadditionssalze derselben hergestellt. Selbstverständlich soll die obige Definition von Alkanolaminderivaten alle möglichen Stereoisomeren und Gemische derselben umfassen.
Als geeignete Bedeutung von R1, wenn es einen gegebenenfalls substituierten Alkylrest darstellt, seien z. B. die folgenden Reste erwähnt: ein Alkylrest mit insbesondere 3, 4 oder 5 Kohlenstoffatomen, der vor zugsweise am α-Kohlenstoffatom verzweigt ist, z. B. der Isopropyl-, sek.-Butyl- oder tert.-Butylrest, wobei die ser Alkylrest gegebenenfalls durch einen oder zwei Substituenten substituiert sein kann, die aus Hydroxyl gruppen, Alkoxyresten mit bis zu 5 Kohlenstoffatomen, z. B. Methoxy- und Propoxyresten, und Phenyl- und Phenoxyresten, die ihrerseits gegebenenfalls durch ein oder mehrere Chlor- oder Bromatome oder einen oder mehreren Methyl-, Äthyl-, Methoxy- oder Äthoxyreste substituiert sein können, gewählt sind.
Eine spezifische Bedeutung von R1, wenn es einen gegebenenfalls sub stituierten Alkylrest darstellt, ist daher der Isopropyl-, sek.-Butyl-, tert.-Butyl-, 2-Hydroxy-1,1-dimethyläthyl- oder 1,1-Dimethyl-2-phenyläthylrest.
Als geeignete Bedeutung von R1, wenn es einen Cycloalkylrest darstellt, sei beispielsweise der Cyclo- propyl-, Cyclobutyl- oder Cyclopentylrest erwähnt.
Als geeignete Bedeutung von R2 sei als Alkanoyl- rest, beispielsweise der Formyl-, Acetyl-, Propionyl-, Butyryl-, Valeryl-, Pivaloyl-, Pentanoyl-, Hexanoyl- oder Heptanoylrest, als Halogenalkanoylrest, beispiels weise der Chloracetylrest, als Cycloalkancarbonylrest, beispielsweise der Cyclopropancarbonyl- oder Cyclo- hexancarbonylrest, als Aroylrest, beispielsweise der Benzoyl-, p-Methylbenzoyl- oder p-Chlorbenzoylrest, als Aralkanoylrest, beispielsweise der Phenylacetylrest, als Aryloxyalkanoylrest, beispielsweise der Phenoxy- acetylrest, als Alkansulfonylrest,
beispielsweise der Me- thansulfonyl- oder Äthansulfonylrest, als Arylsulfonyl- rest, beispielsweise der Benzolsulfonyl- oder Toluol-p- sulfonylrest, oder als Alkoxycarbonylrest, beispielsweise der Äthoxycarbonylrest, genannt.
Als geeignete Bedeutung von R3, wenn es ein Halo- genatom darstellt, sei z. B. das Fluor-, Chlor-, Brom- oder Jodatom genannt.
Eine geeignete Bedeutung von R3, wenn es einen Alkylthio-, Cycloalkyl-, Alkanoyl- oder Alkoxycar- bonylrest darstellt, ist z. B. der Methylthio-, Äthylthio-, Cyclohexyl-, Acetyl-, Propionyl-, Methoxycarbonyl- oder Äthoxycarbonylrest.
Als geeignete Bedeutung von R3, wenn es einen Phenyl- oder Phenoxyrest darstellt, der gegebenenfalls durch ein oder mehrere Halogenatome oder eine oder mehrere Nitrogruppen oder Alkyl- oder Alkoxygruppen mit je bis zu 4 Kohlenstoffatomen, z. B. Methyl- oder Methoxyreste, substituiert ist, sei beispielsweise der p-Tolyloxyrest genannt.
Eine geeignete Bedeutung von R3, wenn es einen substituierten Alkylrest darstellt, ist beispielsweise der Hydroxymethyl-, Methoxymethyl-, n-Butoxymethyl-, Trifluormethyl-, Benzyl- oder α-Phenyläthylrest.
Geeignete Säureadditionssalze der erfindungsgemäss erhältlichen Alkanolaminderivate sind z. B. von anorga nischen Säuren abgeleitete Salze, beispielsweise Hydro- chloride, Hydrobromide, Phosphate oder Sulfate, oder von organischen Säuren abgeleitete Salze, beispielsweise Oxalate, Lactate, Tartrate, Acetate, Salicylate, Citrate, Benzoate, ss-Naphthoate, Adipate oder 1,1-Methylen- bis-(2-hydroxy-3-naphthoate), oder von sauren synthe tischen Harzen, z. B. sulfonierten Polystyrolharzen, z. B. Zeo-Karb 225 (Markenprodukt), abgeleitete Harze.
Eine besonders bevorzugte Gruppe von erfindungs gemäss erhältlichen Alkanolaminderivaten sind die Ver bindungen der Formel:

worin R1 den Isopropyl- oder tert.-Butylrest und R6 einen Alkylrest mit bis zu 6 Kohlenstoffatomen bedeu ten, sowie die Säureadditionssalze derselben.
Spezifische erfindungsgemäss erhältliche Alkanol- aminderivate werden im folgenden in den Beispielen 1 und 2 beschrieben. Unter diesen Derivaten werden die jenigen Verbindungen bevorzugt, die die oben zuletzt angegebene Formel haben, wobei a) R1 den Isopropylrest und entweder R6 den Methylrest und R3 das Fluor-, Chlor- oder Bromatom oder den Methylthio- oder den Phenylrest oder R6 den Äthylrest und R3 das Fluor-, Chlor-, Brom- oder Jod- atom oder den Phenyl-, p-Tolyloxy-, Methoxymethyl-, Benzyl-, α
-Phenyläthyl-, Cyclohexyl- oder Propionylrest oder R6 den n-Propylrest und R3 den p-Tolyloxyrest oder R6 dien n-Butyl- oder n-Hexylrest und R3 das Brom atom bedeuten oder b) R1 den tert.-Butylrest und entweder R6 den Methylrest und R3 das Chloratom oder R6 den Athylrest und R3 das Fluor-, Chlor-, Brom- oder Jodatom oder den Methylthio-, Phenoxy-, Hydroxymethyl-, Trifluor- methyl-, Benzyl-, α
-Phenyläthyl-, Methoxycarbonyl- oder Cyanorest oder R6 den n-Propyl-, n-Pentyl- oder Phenyl rest und R3 das Bromatom bedeuten oder c) R1 den sek.-Butyl-, 2-Hydroxy-1,1-dimethyläthyl- oder Cyclopentylrest, R6 den Äthylrest und R3 das Brom atom bedeuten, sowie die Säureadditionssalze derselben.
Das Verfahren gemäss der Erfindung zur Herstel lung der oben definierten Alkanolaminderivate ist da- durch gekennzeichnet, dass man eine Verbindung der Formel:

in welcher R1, R2 und R3 obige Bedeutung besitzen und R5 einen hydrogenolysierbaren Rest darstellt, oder ein Säureadditionssalz davon hydrogenolysiert.
Eine geeignete Bedeutung von R5 ist z. B. ein α-Arylalkylrest, beispielsweise der Benzylrest. Die Hy- drogenolyse kann beispielsweise durch katalytische Hy drierung, z. B. Hydrierung in Gegenwart eines Palla dium-auf-Kohle-Katalysators, in einem inerten Ver dünnungsmittel oder Lösungsmittel, z. B. Äthanol oder wässrigem Äthanol, bewirkt werden. Das Verfahren kann durch das Vorhandensein eines sauren Katalysa- tors, z. B. Salzsäure oder Oxalsäure, beschleunigt oder zu Ende geführt werden.
Das verwendete Ausgangsmaterial kann durch Acy- lierung der entsprechenden Verbindung, worin R2 Was serstoff darstellt, mit einem von einer Säure der Formel R2-OH abgeleiteten Acylierungsmittel erhalten werden. Ein geeignetes Acylierungsmittel ist z. B. ein Acyl- halogenid oder Säureanhydrid oder die Säure selbst in Gegenwart eines Kondensationsmittels, z. B. eines Carbodiimides.
Das Ausgangsmaterial kann aber auch durch Umsetzung einer Verbindung der Formel:

in welchen R1, R5 und Y obige Bedeutung besitzen, er halten werden.
Die erfindungsgemäss erhältlichen Alkanolaminderi- vate können durch Umsetzung mit einer Säure in be kannter Weise in die Säureadditionssalze übergeführt werden.
Wie oben angegeben, sind die erfindungsgemäss er hältlichen Alkanolaminderivate wertvoll für die Behand lung oder Prophylaxe von Herzkrankheiten. Ferner be- sitzen einige der erfindungsgemäss erhältlichen Alkanol- aminderivate eine selektive ss-adrenergische Blockie rungswirkung. Verbindungen, welche diese selektive Wirkung zeigen, besitzen einen grösseren Grad von spezifischer Wirkung bei der Blockierung der Herz-ss- Rezeptoren als der ss-Rezeptoren in den peripheren Blutgefässen und Bronchenmuskeln.
Daher kann bei einer solchen Verbindung eine solche Dosis gewählt werden, dass die Verbindung zwar die chronotrope Herzwirkung eines Brenzkatechinamins [z. B. von Iso- prenalin, d. h. 1-(3,4-Dihydroxyphenyl)-2-isopropyl- aminoäthanol] blockiert, aber die durch Isoprenalin her vorgerufene Entspannung des glatten Luftröhrenmus kels oder die periphere gefässerweiternde Wirkung des Isoprenalins nicht blockiert. Wegen dieser selektiven Wirkung kann eine dieser Verbindungen vorteilhafter weise zusammen mit einem sympathikomimetischen Bronchodilatator, z. B.
Isoprenalin, Orciprenalin, Adre nalin oder Ephedrin, zur Behandlung von Asthma und anderen die Luftwege schädigenden Krank heiten verwendet werden, da die selektiv wirksame Ver bindung im wesentlichen die unerwünschte stimulie rende Wirkung des Bronchodilatators auf das Herz hemmt, aber die erwünschte therapeutische Wirkung des Bronchodilatators nicht behindert.
Es wird erwartet, dass die bevorzugten Verbindun gen Menschen bei einer oralen Dosis zwischen 20 mg und 600 mg täglich in Einzeldosen im Abstand von 6 bis 8 Stunden oder bei einer intravenösen Dosis zwi schen 1 mg und 20 mg täglich verabreicht werden. Bevorzugte orale Dosisformen sind Tabletten oder Kap seln, die zwischen 10 und 100 mg, vorzugsweise 10 oder 40 mg, des Wirkstoffes enthalten. Bevorzugte intravenöse Dosierungsformen sind sterile wässrige Lö sungen der Alkanolaminderivate oder der nicht toxi schen Säureadditionssalze derselben, welche Lösungen zwischen 0,05 und 1 % (Gewicht/Volumen) Wirkstoff, insbesondere 0,1 % (Gewicht/Volumen) Wirkstoff ent halten.
Die Erfindung wird erläutert durch die folgenden Beispiele, in welchen die Teile gewichtsmässig angege ben sind: <I>Beispiel 1</I> Eine Lösung von 6 Teilen 1-(2-Chlor-4-propionamidophenoxy)- 3-N-benzyl-N-tert.-butylamino-2-propanol in 100 Teilen Äthanol wird in Gegenwart von 0,5 Teil eines 5 % igen Palladium-auf-Kohle-Katalysators mit Wasserstoff geschüttelt, bis die Absorption von Wasser stoff aufhört. Das Gemisch wird filtriert und das Filtrat zur Trockene eingedampft.
Der Rückstand wird aus einem Gemisch aus Äthylacetat und Petroläther (Siede bereich 60 bis 80 C) kristallisiert, und auf diese Weise wird 1-(2-Chlor-4-propionamidophenoxy)- 3-tert.-butylaminopropan-2-ol vom Schmelzpunkt 139,5 bis 141 C erhalten.
Das als Ausgangsmaterial verwendete 1-(2-Chlor-4-propionamidophenoxy)- 3-N-benzyl-N-tert.-butylamino-2-propanol kann folgendermassen hergestellt werden: Ein Gemisch aus 98 Teilen N-Benzyl-tert.-butyl- amin und 63 Teilen Epichlorhydrin wird während 4 Stunden auf 95 bis 100 C erhitzt und dann während 17 Stunden bei Umgebungstemperatur gehalten. Das Gemisch wird während 10 Minuten mit 100 Teilen 20 % iger wässriger Natriumcarbonatlösung geschüttelt, mit 150 Teilen 35 % iger wässriger Natriumhydroxyd lösung versetzt und das Gemisch während 3 Stunden gerührt und unter Rückfluss erhitzt. Das Gemisch wird mit Äther extrahiert und die ätherische Schicht über Kaliumcarbonat getrocknet und zur Trockene einge dampft.
Der ölige Rückstand wird unter vermindertem Druck destilliert, und auf diese Weise wird 1,2-Epoxy-3-(N-benzyl-N-tert.-butylamino)-propan vom Siedepunkt 118 bis 120 C/1,5 mm erhalten. Eine Lösung von 3 Teilen 2-Chlor-4-propion-amidophenol, 3,3 Teilen 1,2-Epoxy-3-(N-benzyl-N-tert.-butylamino)-propan und 0,6 Teil Natriumhydroxyd und 100 Teilen Äthanol wird während 5 Tagen unter Rückfluss erhitzt. Die Lösung wird eingedampft, bis nur 1/4 des ursprüngli chen Volumens übrig ist, und dann in 250 Teile Wasser gegossen und das Gemisch mit Äther extrahiert. Der ätherische Extrakt wird getrocknet und zur Trockene eingedampft, und auf diese Weise wird als Rückstand 1-(2-Chlor-4-propionamidophenoxy)- 3-(N-benzyl-N-tert.-butylamino)-2-propanol erhalten.
<I>Beispiel 2</I> Das in Beispiel 1 beschriebene Verfahren wird unter Verwendung anderer Ausgangsprodukte wieder holt, wobei man die in der folgenden Tabelle angeführ ten Verbindungen erhält.

Claims (1)
- PATENTANSPRUCH Verfahren zur Herstellung von Alkanolaminderi- vaten der Formel: EMI0005.0005 und deren Säureadditionssalzen, worin R1 einen Alkyl rest mit bis zu 12 Kohlenstoffatomen, der gegebenen falls durch einen oder zwei der folgenden Substituenten:Hydroxylgruppen, Alkoxygruppen mit bis zu 5 Koh lenstoffatomen, Phenyl- und Phenoxygruppen substi tuiert ist, wobei die Phenyl- und Phenoxygruppen ihrer seits gegebenenfalls durch ein oder mehrere Chlor- oder Bromatome oder Methyl-, Äthyl-, Methoxy- oder Äthoxygruppen weiter substituiert sind, oder R1 einen Cycloalkylrest mit bis zu 8 Kohlenstoffatomen, R2 einen Alkanoyl-, Cycloalkancarbonyl-, Aroyl-, Aralkanoyl-, Aryloxyalkanoyl- oder Arylsulfonylrest, je mit bis zu 10 Kohlenstoffatomen oder einen Halogenalkanoyl-, Alkylsulfonyl- oder Alkoxycarbonylrest, je mit bis zu 6 Kohlenstoffatomen, und R3 ein Halogenatom, den Cyanorest oder einen Alkylthio-, Cycloalkyl-, Alkanoyl- oder Alkoxycarbonylrest, je mit bis zu 6 Kohlenstoff atomen, oder den Phenyl- oder Phenoxyrest,welche gegebenenfalls durch ein oder mehrere Halogenatome oder Nitrogruppen oder Alkyl- oder Alkoxygruppen mit bis zu 4 Kohlenstoffatomen substituiert sind, oder einen Alkylrest mit bis zu 4 Kohlenstoffatomen, welcher durch die Hydroxyl- oder Phenylgruppe oder eine Alk- oxygruppe mit bis zu 6 Kohlenstoffatomen oder ein oder mehrere Halogenatome substituiert ist, darstellen, dadurch gekennzeichnet, dass man eine Verbindung der Formel: EMI0005.0008 in welcher R1, R2 und R3 obige Bedeutung besitzen und R5 einen hydrogenolysierbaren Rest darstellt, oder ein Säureadditionssalz davon einer Hydrogenolyse unter wirft. UNTERANSPRÜCHE 1. Verfahren gemäss Patentanspruch, dadurch ge kennzeichnet, dass in Formel II R5 für den Benzylrest steht. 2.Verfahren gemäss Patentanspruch oder Unter anspruch 1, dadurch gekennzeichnet, dass man die Hydrogenolyse durch katalytische Hydrierung in einem inerten Verdünnungs- oder Lösungsmittel durchführt. 3. Verfahren gemäss Unteranspruch 2, dadurch ge kennzeichnet, dass man die Hydrogenolyse durch Hy drierung in Gegenwart eines Palladium/Kohle-Kataly- sators und in Äthanol durchführt. 4. Verfahren gemäss Patentanspruch oder Unter anspruch 1, 2 oder 3, dadurch gekennzeichnet, dass man die Hydrogenolyse in Gegenwart eines sauren Kataly- sators durchführt. 5.Verfahren gemäss Patentanspruch oder Unter anspruch 1, dadurch gekennzeichnet, dass man eine Verbindung der Formel EMI0006.0000 in welcher R5 obige Bedeutung besitzt, R1 einen Isopro- pyl- oder tert-Butylrest, R6 einen Alkylrest mit bis zu 6 Kohlenstoffatomen und R3 das Fluor-, Chlor-, Brom- oder Jodatom oder den Methylthio-, Äthylthio-, Cyclo- hexyl-, Methoxycarbonyl-, Äthoxycarbonyl-, Phenyl-, Phenoxy-, p-Tolyloxy-, Hydroxymethyl-, Methoxyme- thyl-, n-Butoxymethyl-, Trifluormethyl-, Benzyl-,a- Phenäthyl- oder Cyanorest bedeutet, einer Hydrogeno- lyse unterwirft. 6. Verfahren gemäss Patentanspruch, dadurch ge- kennzeichnet, dass man erhaltene Verbindungen durch Umsetzung mit anorganischen oder organischen Säuren oder mit sauren synthetischen Harzen in die entspre chenden Salze überführt.
