Description
Verfahren zur Herstellung von Aminoearbonylalkylverbindungen
Die Erfindung betrifft ein Verfahren zur Herstellung neuer N-substituierter und N,N-disubstituierter Aminocarbonylalkylverbindungen mit pharmakologischer Wirkung, z.B. einer entzündungshemmenden Wirkung und einer Wirkung gegen Arrhythmien.

Wie gefunden wurde, sind die Verbindungen der Formel I durch eine entzündungshemmende Wirkung und/oder eine Wirkung gegen Arrhythmien gekennzeichnet. Die folgenden Verbindungen zeigen z.B. besonders entzündungshemmende Wirkungen: 3-[4-(2-Hydroxyäthyl)- 1 -piperazinylcarbonylmethyl]-5- -chlor-2(3H)-benzothiazolinon, N-(2-Hydroxypropyl)-5-chlor-2-oxo-3 -benzothiazolin acetamid, N-(2-Hydroxyäthyl) -6-chlor-2-oxo-3 -benzothiazolin acetamid, N-[2-(2-Hydroxyäthoxy)-äthyl]-5-chlor-2-oxo-3-benzo thiazolinacetamid, N-(2-Hydroxypropyl)-7-chlor-2-oxo-3-benzothiazolin acetanilid, N-(2-Äthoxyäthyl)-5-chlor-2-oxo-3-benzothiazolin- acetamid und N-Methyl-5 -trifluormethyl-2-oxo-3 -benzothiazolin acetamid.
Eine relativ starke Wirkung gegen Arrhythmien zeigen z.B. die folgenden Verbindungen: N-[2-(N,N-Diäthylamino) -äthyl] -2-oxo-3 -benzothiazolin acetamid, N- [2-(N,N-Diäthylamino)-äthyl] -5-chlor-2-oxo-3 -benzo thiazolinacetamid, N-[3 -(N,N-Dimethylamino)-propyl] -2-oxo-3 -benzo thiazolinacetamid, N-[2-(N,N-Diäthylamino)-äthyl]-6-äthoxy-2-oxo-3 -benzothiazolinacetamid.
Das Verfahren gemäss der Erfindung ist dadurch gekennzeichnet, dass man eine Verbindung der Formel

Die Carboxyalkylverbindungen der Formel II (britische Patentschrift Nr. 862-226) oder ihre reaktionsfähigen Derivate, z.B. Salze, werden mit einer N-substituier ten oder N,N-disubstituierten Verbindung der Formel III umgesetzt. Beispiele für Salze von Verbindungen der Formel II sind die Metallsalze (z.B. das Natrium-, Kalium- oder Calciumsalz), Ammoniumsalze, Salze mit organischen Basen (z.B. Triäthylaminsalze) und dergleichen. Beispiele für reaktionsfähige Derivate von Verbindungen der Formel II sind die Säurehalogenide (z.B.
Säurechloride oder -bromide), die Säureanhydride (z.B.

Methylester, Äthylester, Cyanomethylester, p-Nitrophenylester, Pentachlorphenylester, 2,4,5- Trichlorphenylester, Propargylester, Carboxymethylthioester, Pyranylester, Methoxymethylester oder Phenylthioester) und die Säureazide. Wird die Verbindung der Formel II als solche verwendet. so muss man gewöhnlich die Umsetzung in Gegenwart eines Kondensationsmittels durchführen.

Die Kondensation wird gewöhnlich in einem inerten Lösungsmittel durchgeführt, z.B. in Aceton, Dioxan, Acetonitril, Chloroform, Äthylendichlorid, Tetrahydrofuran, Äthylacetat, Pyridin oder dergleichen. Erforderlichenfalls kann im Reaktionsgemisch eine basische Substanz (z.B.

Verbindungen der Formel Ig oder Ih, die in diese Kategorie fallen, können auch als Ausgangsverbindungen für diese Ausführungsform verwendet werden. Die Verbindungen der Formel Ii können durch Umsetzung mit einem Acylierungsmittel in die Verbindungen der Formel Ij übergeführt werden. Beispiele von geeigneten Acylierungsmitteln sind niedere Alkancarbonsäureanhydride (z.B. Essigsäureanhydrid, Propionsäureanhydrid oder Buttersäureanhydrid) und niedere Alkanoylhalogenide (z.B. Acetylchlorid, Acetylbromid, Propionylchlorid oder Butyrylchlorid). Die Reaktion kann auf die für Acylierungsverfahren übliche Weise ausgeführt werden.
Zum Nachweis der antiphlogistischen Wirkung der Verbindungen der Formel I kann ihre Hemmwirkung auf durch Formalin, Albumin oder Carraghenin bei der Ratte induzierte Schwellungen verwendet werden. Die antiarrhythmische Wirkung kann mittels des elektrisch oder durch Acetylcholin induzierten Vorhofflimmerns oder auf durch Lanatosid C hervorgerufene Arrhythmie gezeigt werden.
Aus diesen Versuchen kann entnommen werden, dass die Toxizität der Verbindungen der Formel I für ein kräftiges Mittel gegen Arrhythmien sehr niedrig ist. Sie können daher mit hohem Sicherheitsgrad in der Humanmedizin Verwendung finden.
Die Verbindungen der Formel I können zur Behandlung entzündlicher Zustände, die mit Schmerz, Rötung und/oder Schwellungen verbunden sind, infolge von Änderungen der Durchiässigkeit von Zellen und Gefässen, von Gefässerweiterung, vom Austreten flüssiger Blutbestandteile und Zellbestandteile und/oder der Rückbildung von zusammenhängenden Gewebezellen und der Bildung von Granulationsgewebe verwendet werden.
Sie sind ebenfalls bei der Behandlung bestimmter Arrhythmien brauchbar. Beispiele für die therapeutische Indikation sind die Behandlung von Vorhofflimmern, die Aufhebung von in Paroxysmen auftretender Vorhoftachycardie und die Beherrschung von ektopischen Herzkammerschlägen.
Die Verbindungen der Formel I können in üblicher Weise in üblichen Dosierungseinheiten oder mit den bekannten pharmazeutischen Trägern als entzündungshemmende Mittel und Mittel gegen Arrhythmie verabreicht werden. In Form therapeutischer Präparate können sie zusammen mit einem organischen od. anorganischen, für die enterale. parenterale oder örtliche Anwendung geeigneten pharmazeutischen Trägermaterial verwendet werden. Besonders vorteilhaft ist die orale Verabreichung in Form von Tabletten, Kapseln oder in flüssiger Form als Suspension, Lösung oder Emulsion. Bei der Tablettenherstellung können die bei üblichen therapeutischen Dosierungseinheiten verwendeten Binde- und Zerfallsmittel verwendet werden. Beispiele für Bindemittel sind Glucose, Lactose, Gummi arabicum, Gelatine, Mannit, Stärkepaste, Magnesiumtrisilikat und Talkum.
Beispiele für Mittel zur Förderung des Zerfalls sind Maisstärke, Keratin, kolloidales Siliciumdioxyd und Kartoffelstärke.
Bei der Verabreichung in flüssiger Form können die üblichen flüssigen Träger verwendet werden.
Die Dosierungseinheiten oder therapeutisch wirksamen Mengen der Verbindungen der Formel I in der Humanmedizin können in breiten Grenzen schwanken, etwa zwischen 6,5 mg und etwa 1,6 g oder mehr, z.B.
3,2 g. Die obere Grenze ist nur durch den gewünschten Wirkungsgrad und wirtschaftliche Gesichtspunkte bedingt. Zur oralen Verabreichung werden vorzugsweise etwa 65 mg bis etwa 650mg des Wirkstoffs je Dosierungseinheit verwendet. Aus Tierversuchen ergibt sich, dass die orale Verabreichung von etwa 65 bis etwa 650 mg 4mal täglich nach Bedarf eine bevorzugte tägliche Dosis darstellt. Die Dosierung des speziellen verwendeten Wirkstoffs kann jedoch je nach dem Alter des Patienten und dem gewünschten therapeutischen Effekt erheblich schwanken. Jede Dosierungseinheit der neuen therapeutisch wirksamen Verbindungen kann etwa 5 Gew.-% bis etwa 95 Gew.-% des Wirkstoffs, bezogen auf das Präparat, enthalten, wobei der Rest aus üblichen pharmazeutischen Trägern besteht.
Unter pharmazeutischen Trägern sollen auch therapeutisch inaktive Materialien verstanden werden, die üblicherweise zusammen mit Dosierungseinheiten verwendet werden und Füllstoffe, Verdünnungsmittel, Bindemittel, Gleitmittel, Zerfallsbeschleuniger und Lösungsmittel mit umfassen. Natürlich ist es auch möglich, die reinen neuen Verbindungen ohne pharmazeutische Trägerstoffe zu verabreichen.
In den folgenden Beispielen wird die Herstellung besonders bevorzugter Verbindungen gemäss der Erfindung erläutert.
Beispiel 1
A) Ein Gemisch aus (2-Oxo-3-benzothiazolin-3-yl)- essigsäureäthylester (1,2 g), 2-(N,N-Diäthylamino)-äthylamin (1,0 g) und wasserfreiem Äthanol (1,0 g) wird 48 Stunden am Rückfluss erhitzt. Nach dem Abdestillieren des Äthanols wird der Rückstand mit verdünnter Salzsäure extrahiert. Der Extrakt wird unter Verwendung einer 10%igen Natriumhydroxydlösung alkalisch gemacht und mit Chloroform geschüttelt. Die Chloroformschicht wird mit Wasser gewaschen, getrocknet und eingeengt. Der Rückstand wird aus Äthanol umkristallisiert und ergibt farblose Kristallschuppen von N-[2-(N,N-Di äthylamino) - äthyl] -2-oxo-3 -benzothiazolinacetamid (2,0 g) vom F. 138,5 bis 139,50C.
Die Base wird mit Maleinsäure in üblicher Weise in das Maleat umgewandelt, das den F. 125 bis 1280hat.
B) Eine Lösung von 600 mg (2-Oxo-3-benzothiazolin -3-yl)essigsäureäthylester wird in überschüssigem 1-(2 -Hydroxyäthyl)-piperazin etwa 20 Stunden auf 1000C erhitzt. Nach dem Abkühlen wird eine grosse Menge Wasser zugefügt und das erhaltene Gemisch mit Chloroform extrahiert. Der Chloroformextrakt wird mit Wasser gewaschen und mit 10%iger Salzsäure geschüttelt. Die Salzsäureschicht wird mit Äther gewaschen, mit einer 20%igen Natriumhydroxydlösung alkalisch gemacht und mit Chloroform extrahiert. Der Chloroformextrakt wird mit Wasser gewaschen, getrocknet und eingeengt. Der Rückstand wird aus Äthanol kristallisiert und ergibt Kristalle von 3 -[4-(2-Hydroxyäthyl)- 1 -piperazinylcarbonyl methyl]-2(3H)-benzothiazolinon. F. des Maleats 197 bis
1980C.
C) Ein Gemisch aus 1,5 g (5-Trifluormethyl-2-oxo-3 -benzothiazolin-3-yl)essigsäureäthylester und 6,0g Morpholin wird unter Rühren 15 Stunden auf 1 100C erhitzt.
Nach dem Abkühlen wird das Reaktionsgemisch mit Chloroform versetzt und mit Wasser, verdünnter Salzsäure und Wasser in dieser Reihenfolge gewaschen. Die Chloroformlösung wird über Magneslumsulfat getrocknet und eingeengt. Der Rückstand wird aus Äthanol kristallisiert und liefert 0,65 g 3-Morpholirlocarbonylme- thyl-5-trifluromethyl-2(3H)-benzothiazolinon als weisse Nadeln vom F. 188 bis 190 C.
D) Eine Lösung von 4,0g (5-Chlor-2-oxo-3-benzo- thiazolin-3-yl)essigsäureäthylester in 1-(2-Hydroxyäthyl)- -piperazin wird 24 Stunden auf 100 C erhitzt. Nach dem Abkühlen wird das erhaltene Reaktionsgemisch mit Chloroform extrahiert. Der Chloroformextrakt wird mit Wasser gewaschen und mit 10%iger Salzsäure geschüttelt. Der Salzsäureextrakt wird mit Chloroform gewaschen, mit 10%iger Natriumhydroxydlösung alkalisch gemacht und mit Chloroform extrahiert. Der Chloroformextrakt wird mit Wasser gewaschen, über Magnesiumsulfat getrocknet und eingeengt. Das zurückbleibende öl (5,5 g) wird zur Kristallisation stehengelassen.
Die Kristalle werden aus einem Gemisch aus 40ml Äthylacetat und 15ml Äthanol umkristallisiert und ergeben 3,2g farbloser Kristalle von 3-[4-(2-Hydroxyäthyl)-1-pi- perazinylcarbonylmethyl]-5-chlor-2(3H)-benzothiazolinon vom F. 159 bis 161 C.
Weitere Beispiele von Verbindungen, die in ähnlicher Weise hergestellt werden kömlen, sind: N-[2-(N,N-Diäthylamino) -äthyl] -5-chlor-2-oxo-3 -benzo thiazolinacetamid (F. 127 bis 128 C ; Maleat F. 119 bis

Beispiel 2
A) Ein Gemisch aus 5,04g 6-Chlor-2-oxo 3-benzotliiazolinessigsäure und 50 ml Thionylchlorid wird 2 Stunden am Rückfluss erhitzt und das Thionylchlorid abdestilliert. Das so erhaltene 6-Chlor-2-oxo-3-benzothiazolinacetylchlorid wird in einem Gemisch aus 60ml Benzol und 30ml Chloroform gelöst und die Lösung tropfenweise unter Kühlen im Eiswasserbad und Rühren zu einem Gemisch aus 12 ml Benzol, 3,4 g 2-Amino äthanol, 5;6 ml Wasser und 1,4 g Natriumcarbonat zugegeben. Das erhaltene Gemisch wird 40 Minuten bei Raumtemperatur gerührt und nach Zugabe von 70ml Wasser noch einige Minuten weiter gerührt.
Der Niederschlag wird abfiltriert, mit Wasser gewaschen und aus einem Gemisch aus Äthanol und Chloroform umkristallisiert und ergibt 2,0g weisse baumwollartige Kristalle von N-(2-Hydroxyäthyl)-6-chlor-2-oxo-3 -benzothiazolin- acetamid vom F. 222 bis 2230C.
B) Ein Gemisch aus 4,4 g 5-Chlor-2-oxo-3-benzo- thiazolinessigsäure und Thionylchlorid wird einige Stunden am Rückfluss erhitzt und das Thionylchlorid abdestilliert. Das erhaltene 5-Chlor-2-oxo-3-benzothiazolinylacetylchlorid wird in Benzol gelöst und die Lösung tropfenweise unter Kühlung in einem Eiswasserbad und Rühren zu einem Gemisch aus 4,0 g 2-(2-Aminoäthoxy) -äthanol, 2,2 g Natriumcarbonat, 12 mol Wasser und 40 ml Benzol zugegeben. Das erhaltene Gemisch wird 3 Stunden bei Raumtemperatur und 30 Minuten bei 600C gerührt.
Nach dem Abkühlen werden die ausgefallenen Kristalle abfiltriert und aus Wasser umkristallisiert und liefern 3,6 g farblose Schuppen von N-[2-(2-Hydroxy äthoxy)-äthyl]-5-chlor-2-oxo-3-benzothiazolinacetamid vom F. 133 bis 1340C.

Beispiel 3
A) Ein Gemisch aus 5,0 g 5-Chlor-2-oxo-3-benzothiazolinessigsäure und 50 ml Thionylchlorid wird 2 Stunden am Rückfluss erhitzt und das Thionylchlorid abdestilliert. Das so erhaltene 5-Chlor-2-oxo-3-benzothiazolinacetylchlorid wird in 50ml wasserfreiem Benzol gelöst und die erhaltene Lösung tropfenweise unter Kühlen auf 5 bis 100C und Rühren zu einem Gemisch aus 100 mol wasserfreiem Benzol, 2,3 g Aziridin, 5,03 Kaliumcarbonat und 15 ml Wasser gegeben. Bei der gleichen Temperatur wird das Reaktionsgemisch 10 Min.
gerührt und anschliessend mit 10%iger Natriumcarbonatlösung und dann mit Wasser gewaschen und über Magnesiumsulfat getrocknet. Nach Entfernen des Lö sungsmittels wird der Rückstand aus wasserfreiem Benzol umkristallisiert und ergibt 5,0g weisses körniges 5 -Chlor-3 -(1 -aziridinylcarbonylmethyl) -2(3H) -benzothiazo - linon vom F. 137 bis 1390C.
B) 0,7 g wie oben hergestelltes 5-Chlor-3-(1-aziridinylcarbonylmethyl)-2(3H)-benzothiazolinon werden zu
1,5 g N,N-Dicyclohexylamin zugegeben und das erhaltene Gemisch 20 Stunden auf 1 100C erhitzt. Man lässt das Reaktionsgemisch abkühlen. Die ausgefallenen Kristalle werden abfiltriert, mit Benzol und danach mit Petroläther gewaschen und aus Äthanol umkristallisiert und ergeben 0,5 g weisse Kristalle von N-[2-(N,N-Dicyclohexylamino)-äthyl] - 5-chlor-2-oxo-3-benzothiazolin- acetamid vom F. 209 bis 2100C.

Beispiel 4
A) In ein Bombenrohr aus Stahl werden 1,0 g N-(2 -Chloräthyl)-5-chlor-2-oxo-3 -benzothiazollnacetamid und 1,62g N-Methylanilin eingebracht und 5,5 Stunden auf 900C erhitzt.
Anschliessend werden 10%ige Salzsäure und Äthylacetat zugegeben und die Äthylacetatschicht mit 10%iger Salzsäure extrahiert. Der Salzsäureextrakt und die Salzsäureschicht werden vereinigt, mit Äthylacetat gewaschen und anschliessend mit 10%iger Natriumcarbonatlösung alkalisch gemacht. Die ausgefallenen Kristalle werden mit Chloroform extrahiert. Der Chloroformextrakt wird mit Wasser gewaschen, über wasserfreiem Magnesiumsulfat getrocknet und eingeengt.
Der ölige Rückstand wird aus Petroläther kristallisiert und aus Äthanol umkristallisiert und ergibt 0,45 g weisse Kristalle von N-[2-(N-Methylanilino)-äthyl]-5-chlor-2- -oxo-3-benzothiazolinacetamid vom F. 155 bis 1570C.
B) 3,0 g N-(2-Chloräthyl)-2-oxo-3-benzothiazolinacetamid und 4,75 g N-Methylanilin werden in ein Bombenrohr aus Stahl gegeben und 5,5 Stunden auf 1 100C erhitzt. Das Reaktionsgemisch wird wie oben weiter behandelt. Die erhaltenen blass braunen Kristalle (6,7g) werden aus Äthanol umkristallisiert und liefern 2,5 g weisse Nadeln von N-[2-(N-Methylanilino)-äthyl]-2-oxo -3-benzothiazolinacetamid vom F. 143 bis 1450C.

Zu einer Lösung von 0,45 g 3-(1-Piperazinylcarbonylmethyl)-5-chlor-2(3H)-benzothiazolinon in 30ml Methanol wird 0,25 g Propylenoxyd zugegeben und das erhaltene Gemisch 5 Stunden auf 50 C erwärmt, während welcher Zeit zweimal 0,3 g Propylenoxyd zugefügt wird.
Das Reaktionsgemisch wird unter vermindertem Druck eingeengt. Das als Rückstand erhaltene Öl wird aus Petroläther kristallisiert und aus einem Gemisch von Äthylacetat und Petroläther umkristallisiert und ergibt 0,43 g farblose Nadeln von 3-[4-(2-Hydroxypropyl)-1-piperazi- hylcarbonylmethyl]-5-chlor-2(3H)-benzothiazolinon vom F. 112 bis 115 C.

Ein Gemisch aus 500 mg 3-(1-Piperazinylcarbonyl- methyl)-5-chlor-2(3H)-benzothiazolinon, 400 mg wasser- freiem Kaliumcarbonat. 300 mg 2-Hydroxyäthylbromid und 20 ml wasserfreiem Äthanol wird 5 Stunden am Rückfluss erhitzt. Das Reaktionsgemisch wird unter vermindertem Druck eingeengt und der Rückstand mit Chloroform extrahiert. Die Chloroformschicht wird über Magnesiumsulfat getrocknet und eingeengt. Der Rückstand wird aus einem Gemisch aus Äthylacetat und Äthanol kristallisiert und liefert 370 mg Kristalle von 3-[4-(2 -Hydroxyäthyl)-1-piperazinylcarbonylmethyl]-5-chlor -2(3H)-benzothiazolinon vom F. 159 bis 160 C.

Beispiel 7
Ein Gemisch aus 1,42 g 3-[4-(2-Hydroxyäthyl)-1-piperazinylcarbonylmethyl]-5-chlor-2(3H)-benzothiazolinon.
1,4 g Kaliumcarbonat und 40 ml Chloroform wird unter Kühlen innerhalb von ca. 30 Minuten tropfenweise mit einem Gemisch aus 0,8 g Acetylchlorid und 5 ml Tetrahydrofuran versetzt. Das erhaltene Gemisch wird während 6 Stunden bei Raumtemperatur gerührt. Das Reaktionsgemisch wird mit Wasser gewaschen, getrocknet und eingeengt und ergibt 3-[4-(2-Acetyloxyäthyl)-1-piperazinylcarbonylmethyl]-5-chlor-2(3H)-benzothiazolino als farbloses öl. Das öl wird mit Maleinsäure behandelt, und das erhaltene Maleat wird aus Äthanol kristallisiert, wobei farblose Schuppen vom Schmelzpunkt 199,5 bis 2000C erhalten werden.
Andere in ähnlicher Weise erhältliche Verbindungen sind beispielsweise: 3-{4-[2-(Isobutyryloxy)-propyl]- 1 -piperazinylcarbonyl mothyl}-5-chlor-2(3H)-benzothiazolinon (Smp. 123 bis 1250C), 3- (4-[2-(Isobutyryloxy) -äthyl]-l -piperazinylcarbonyl- methyl)5-chlo r-2(3H) -benzothiazolinon (Smp. 111 bis 1 130C), 3-{4-[2-(Acetyloxy)-propyl] 1 -piperazinylcarbonylme thyl} -5-chlor-2(3H)-benzothiazolinon-hydrochlorid (Smp. 248 bis 2490C), 3-(Acetyloxyäthylaminocarbonylmethyl)-5-chlor-2(3H) -benzothiazolinon (Smp. 1440C).
Beispiel 8
A) Ein Gemisch aus 0,5g (6-Chlor-3-methyl-2-oxo -1-benzimidazolin-3-yl)essigsäureäthylester und 2,5 g 2 -(N,N-Diäthylamino)-äthylamin wird 20 Stunden auf 1 100C erhitzt. Das Reaktionsgemisch wird in 100 ml Chloroform gelöst und mit Wasser gewaschen. Die Chloroformschicht wird mit 10%iger Salzsäure extrahiert.
Die Salzsäureschicht wird mit Chloroform gewaschen.
mit 10%iger Natrimuhydroxydlösung alkalisch gemacht und mit Chloroform extrahiert. Der Chloroformextrakt wird mit Wasser gewaschen, über wasserfreiem Magnesiumsulfat getrocknet und unter vermindertem Druck eingeengt. Der Rückstand wird mit Petroläther gewaschen und aus einem Gemisch aus Benzol und Petrol äther krigtallisiert und ergibt 0,2g weisse Kristalle von
N-[-2-(N,N-Diäthylamino)-äthyl]-6-chlor-3-methyl-2-oxo -1-benzimidazolinacetamid vom F. 124 bis 1260C.
B) Ein Gesnisch aus 0.3 g (5-Chlor-3-methyl-2-oxo-1 -benzimidazolin-3-yl)essigsäuteäthylester und 1,0 ml 1 -(2-Hydroxyäthyl)-piperazin wird 15 Stunden auf 1 100C erhitzt. Das Reaktionsgemisch wird mit Äthylacetat versetzt, mit Wasser gewaschen und mit 10%iger Salzsäure extrahiert. Der Salzsäureextrakt wird mit Chloroform gewaschen und mit 1 0%iger Natriumhydroxydlösung unter Eiskühlung alkalisch gemacht. Die alkalische Lösung wird mit Chloroform extrahiert und der Chloroformextrakt über Magnesiumsulfat getrocknet und unter vermindertem Druck eingeengt. Der Rückstand wird aus Petroläther kristallisiert und aus Äthylacetat umkristallisiert und liefert 0,3 g farblose Nadeln von 1-[4-(2-Hy droxyäthyl) - 1 - piperazinylcarbonylmethyl]-5-chlor-3-methyl-2-oxo-benzimidazolin vom F. 173 bis 1740C.
Andere in ähnlicher Weise erhältliche Verbindungen sind z.B.

Das Reaktionsgemisch wird in 100 ml Chloroform gelöst und mit Wasser gewaschen. Die Chloroformschicht wird mit 10%iger Salzsäure extrahiert.Die Salzsäureschicht wird mit Chloroform gewaschen.mit 10%iger Natrimuhydroxydlösung alkalisch gemacht und mit Chloroform extrahiert. Der Chloroformextrakt wird mit Wasser gewaschen, über wasserfreiem Magnesiumsulfat getrocknet und unter vermindertem Druck eingeengt. Der Rückstand wird mit Petroläther gewaschen und aus einem Gemisch aus Benzol und Petrol äther krigtallisiert und ergibt 0,2g weisse Kristalle von N-[-2-(N,N-Diäthylamino)-äthyl]-6-chlor-3-methyl-2-oxo -1-benzimidazolinacetamid vom F. 124 bis 1260C.B) Ein Gesnisch aus 0.3 g (5-Chlor-3-methyl-2-oxo-1 -benzimidazolin-3-yl)essigsäuteäthylester und 1,0 ml 1 -(2-Hydroxyäthyl)-piperazin wird 15 Stunden auf 1 100C erhitzt. Das Reaktionsgemisch wird mit Äthylacetat versetzt, mit Wasser gewaschen und mit 10%iger Salzsäure extrahiert. Der Salzsäureextrakt wird mit Chloroform gewaschen und mit 1 0%iger Natriumhydroxydlösung unter Eiskühlung alkalisch gemacht. Die alkalische Lösung wird mit Chloroform extrahiert und der Chloroformextrakt über Magnesiumsulfat getrocknet und unter vermindertem Druck eingeengt. Verfahren nach Patentanspruch I, dadurch gekennzeichnet, dass man erhaltene Verbindungen der Formel EMI15.4 in der R7 und R8 Wasserstoff oder niedere Alkylreste sind, mit einem Alkylenoxyd der Formel EMI15.5 in der Rfi Wasserstoff oder ein niederer Alkylrest ist, unter Bildung von Verbindungen der Formel EMI15.6 umsetzt.2. Verfahren nach Patentanspruch I, dadurch gekennzeichnet, dass man erhaltene Verbindungen der Formel EMI15.7 in der R7 und R8 Wasserstoff oder niedere Alkylreste sind, mit einem Halogenalkanol der Formel X'''-A''-OH in der X''' ein Halogenatom und A'' eine niedere Alkylengruppe ist, unter Bildung von Verbindungen der Formel EMI15.8 umsetzt.3. Verfahren nach Patentanspruch I, dadurch gekenn- zeichnet, dass man erhaltene Verbindungen der Formel EMI15.9 in der B eine Gruppe der Formel EMI16.1 und A"' einen niederen Alkylenrest bedeuten, mit einem Acylierungsmittel unter Bildung von Verbindungen der Formel EMI16.2 umsetzt.PATENTANSPRUCH II Verwendung von nach dem Verfahren gemäss Patentanspruch I erhaltenen Verbindungen der Formel I, in welcher entweder R2 und R3 zusammen mit dem N Atom eine 1 -Aziridinylgruppe, oder R2 Wasserstoff und R3 eine Halogenalkylgruppe bedeutet, zur Herstellung entsprechender Verbindungen mit einer Seitenkette der Formel EMI16.3 worin R2 Wasserstoff bedeutet, dadurch gekennzeichnet, dass man besagte Verbindungen mit einem Amin der Formel EMI16.4 umsetzt.
