Besonders bevorzugte Verbindungen der obigen Formel sind solche, in denen R' Phenoxymethyl, Z Methansulfonyl und X p-Nitrobenzyloxy, 2,2,2-Trichloräthoxybenzyloxy oder Benzylhydroxy bedeuten oder worin R' Benzyl, Z Methansulfonyl und X p-Nitrobenzyloxy, 2,2,2-Trichloräthoxy, Ben-zyloxy oder Benzhydryloxy ist, oder R' Phenoxymethyl, Z Toluol-p-sulfonyl und X p-Nitrobenzyloxy oder 2,2,2-Trichloräthoxy bedeuten.
Die Zersetzung der Azetidinothiazolinverbindung mit wässriger Säure ist eine grundsätzlich neue Reaktion, um 4-Mercapto-3-carbonsäure AcyIamino-2-oxoazetidinderivate gemäss der folgenden Gleichung zu erhalten:

worin R' der Rest einer Acylgruppe R'CO; R" Wasserstoff oder eine Kohlenwasserstoffgruppe (z.B. Alkyl, Alkenyl, Ar-alkyl, Aryl), Acyl, abgeleitet von organischen oder anorganischen Säuren, Silyl, Sulfenyl oder eine andere einwertige Aminoschutzgruppe ist, gegebenenfalls substituiert durch eine Halogen-, Schwefel-, Sauerstoff-, Stickstoff-, Kohlenstoffoder Phosphorfunktion wie oben erwähnt.
Die Reaktion kann durchgeführt werden durch Behandlung eines Thiazolinazetidins (5) mit einer Säure und Wasser. Die Reaktion mit Wasser spaltet notwendigerweise den Thiazolinring auf, so dass ein 4-Mercapto und 3-Acylamino Azetidinring erhalten wird. Die bevorzugten Säuren sind Mineralsäuren (z.B. Salzsäure, Schwefelsäure, Salpetersäure, Phosphorsäure, Perchlorsäure, Chlorsäure), Sulfonsäuren (z.B. Alkansulfonsäure, Arylsulfonsäure, Aralkylsulfonsäure, insbesondere a-HalogenalkansuIfonsäure), a-Halogencarbon-säuren, Polycarbonsäuren, insbesondere Säuren mit einer Dissoziationskonstanten von mindestens 0,01. Spezielle Säuren sind starke Säuren z.B. Perchlorsäure, Trifluoressigsäure, Tri-chloressigsäure, Dichloressigsäure, Trifluormethansulfonsäure, Trichlormethansulfonsäure, Fluorborwasserstoff, Salzsäure, Flussäure, Bromwasserstoffsäure, Salpetersäure, Phosphorsäure, Benzolsulfonsäure, Toluolsulfonsäure, Brombenzolsul-fonsäure, Methansulfonsäure, Äthansulfonsäure, usw.
Die genannte Umsetzung kann in einem der oben genannten Lösungsmittel durchgeführt werden.
Besonders bevorzugte Lösungsmittel sind polare Lösungsmittel, die Wasser und Säure auflösen (z.B. Äther, Ketone, Alkohole, Amide, Sulfoxide, Wasser), Lösungsmittel die das Ausgangsprodukt (5) lösen (z.B. Halogenkohlenwasserstoffe, Ester, Äther, Ketone), und Mischungen derselben.
Wenn eine starke Säure verwendet wird, können Nebenreaktionen, z.B. Zersetzung des Azetidinonrings vorkommen. Die Ausbeute kann verbessert werden durch Auswahl der Reaktionsbedingungen, d.h. Konzentration, Temperatur, Re-5 aktionszeit, usw. Im allgemeinen verläuft die Umsetzung bei Zimmertemperatur genügend schnell, oft in 10 Minuten bis 1 Stunde um die gewünschte Verbindung in hoher Ausbeute zu erhalten.
Die Produkte sind instabil gegen Alkali und Oxidation. io Daher sollten diese Bedingungen bei den Reaktionen und der Aufarbeitung vermieden werden.
Die folgenden Ausführungsbeispiele erläutern die Erfindung im einzelnen.
15 Beispiel 1
Zu einer Lösung von 3-Phenoxymethyl-7-oxo-4-thia-2-,6--diazabicyclo[3,2,0]hept-2-en (200 mg) in einer Mischung von Methylenchlorid (8 ml) und Aceton (8 ml) wird eine 30% ige wässrige Lösung von Perchlorsäure (1,0 ml) zugefügt und die 20 Mischung 40 Minuten bei Zimmertemperatur gerührt. Nach Verdünnen mit einem Überschuss von Wasser wird die Reaktionsmischung mit Methylenchlorid extrahiert. Die extrahierte Lösung wird mit Wasser gewaschen, über Natriumsulfat getrocknet und eingeengt. Der weisse kristalline Rück-25 stand ist 4ß-Mercapto-3ß-phenoxyacetamido-2-oxoazetidin, Schmp. 137-138°C.

Beispiel 2
Zu einer Lösung von a-[3-substituiert-(R1)-7-oxo-4-thia-35 -2,6-diazabicyclo [3,2,0] hept-2-en-6-yl] -a-substituiert-(R2)--essigsäureester (R3) (5) in einem Lösungsmittel wird eine Säure zugefügt und die Mischung bei einer bestimmten Temperatur eine bestimmte Zeit lang gerührt. Darauf wird die Reaktionsmischung mit Wasser verdünnt und mit Methylen-40 chlorid extrahiert. Die Extraktlösung wird mit Wasser gewaschen, über Natriumsulfat getrocknet und eingeengt, so dass ein gewünschter a-[4-Mercapto-3-substituierter-amino-(RICONH-)-2-oxoazetidin-l-yl]-a-substituierter (R2)essig-säureester (R3) (6) erhalten wird.
45 Tabelle III gibt die Reaktionsbedingungen und Tabelle IV die physikalischen Konstanten des Produktes (6) an. In Tabelle III bedeutet die rohe Ausbeute das Gewicht der Produkte. Gemäss Analyse durch Dünnschicht-Chromatogramm und NMR-Spektrum sind sie im wesentlichen rein. Einige so von ihnen wurden durch Kristallisation gereinigt.
Bei den Reaktionen Nr. 3 und 6 wurden die betreffenden Verbindungen in niedrigen Ausbeuten erhalten zusammen mit einer grossen Menge des Ausgangsproduktes und Nebenprodukten.

Beispiel 3
Zu einer Lösung p-Nitrobenzyl-a-[3-phenoxymethyl-7--oxo-4-thia-2,6-diazabicyclo[3,2,0]hept-2-en-6-yl]-a-isopro-penylacetat (200 mg) in Tetrahydrofuran (5 ml) wird Oxalsäure (200 mg) und Wasser (0,5 ml) zugefügt und die Mischung bei Raumtemperatur 3 Stunden gerührt. Das Chroma-togramm der Reaktionsmischung zeigt die Gegenwart von p-Nitrobenzyl-a-[mercapto-3-phenoxyacetamido-2-oxoazet-idin-l-yl]-a-isopropenylacetat und das Ausgangsmaterial an.
Beispiel 4
Nach ähnlichen Verfahren wie Beispiel 2 werden die folgenden Verbindungen hergestellt:
(1) 4-Mercapto-3-thienylacetamido-2-oxo-1 -acetylazetidin aus 6-Acetyl-3-thienylmethyl-7-oxo-4-thia-2,6-diazabi-cyclo [3,2,0] hept-2-en;
(2) 4-Mercapto-3-benzamido-2-oxo-l-trifluoracetylazetidin aus 6-Trifluoracetyl-3-phenyl-7-oxo-4-thia-2,6-diazabi-cyclo[3,2,0]hept-2-en;
(3) 4-Mercapto-3-acetamido-2-oxo-l-methylazetidin aus 3,6-Dimethyl-7-oxo-4-thia-2,6-diazabicyclo[3,2,0]hept--2-en;
(4) 4-Mercapto-3-(«-phenyl-a-chloracetamido)-2-oxo-l-carb-äthoxycarbonylazetidin aus 6-Carbäthoxycarbonyl-3-phe-nylchlormethyl-7-oxo-4-thia-2,6-diazabicyclo[3,2,0]hept--2-en;
(5) a-[4-Mercapto-3-formamid-2-oxoazetidin-l-yl]-a-isopro-
pylidenessigsäure aus a-[7-oxo-4-thia-2,6-diazabicyclo-[3,2,0]hept-2-en-6-yl]-a-isopropylidenessigsäure; und (6) 4-Mercapto-3-benzylthiocarbonylamino-2-oxo-l-p-toluoi-45 sulfonylazetidin aus 3-Benzylthio-6-p-toluolsulfonyl-7--oxo-4-thia-2,6-diazabicyclo[3,2,0]hept-2-en.
Beispiel 5
Zu einer Lösung von p-Nitrobenzyl-a-[3-phenoxymethyl-50-7-oxo-4-thia-2,6-diazabicyclo[3,2,0]hept-2-en-6-yl]-a-[2--brom-l-(morpholin-4-yl)äthyliden]acetat (580 mg) in Tetrahydrofuran (10 ml) wird bei — 10°C eine 60%ige wässrige Lösung von Perchlorsäure (1,5 ml) zugefügt und die Mischung 30 Minuten gerührt. Die Reaktionsmischung wird mit 55 Wasser verdünnt und mit Methylenchlorid extrahiert Der Extrakt wird mit Wasser gewaschen, über wasserfreiem Natriumsulfat getrocknet, eingedampft und ergibt einen hellgelben Schaum (512 mg). Der Schaum wird chromatographisch über Silicagel, enthaltend 10% Wasser (50 g) gereinigt und 60 scheidet sich aus den mit einer Mischung von Benzol und Äthylacetat (1:1) eluierten Fraktionen aus: p-Nitrobenzyl-a--[3-phenoxymethyl-7-oxo-4-thia-2,6-diazabicyclo[3,2,0]hept--2-en-6-yl]-a-(2-brom-l-hydroxyäthyliden)acetat (Schaum: 207 mg; Ausbeute: 40%).

Aus den Fraktionen kann auch p-Nitrobenzyl-a-[3-phen-oxyacetamido-4-mercapto-2-oxoazetidin-l-yl]-a-(2-brom-l-hy-droxyäthyliden)acetat als Nebenprodukt isoliert werden.
Das oben genannte Hauptprodukt (84 mg) wird in Tetra-hydrofuran (2 ml) gelöst, mit 2N-Salzsäure (0,2 ml) gemischt und 30 Minuten bei 0°C und sodann 1 Stunde bei Zimmertemperatur stehengelassen. Die Reaktionsmischung wird mit Wasser verdünnt und mit Methylenchlorid extrahiert. Der Extrakt wird mit Wasser gewaschen, über Natriumsulfat getrocknet und eingedampft. Der Rückstand (75 mg) kann 5 durch IR- und NMR-Spektroskopie als p-Nitrobenzyl-7-phen-oxyacetamido-3-oxocepham-4-carboxylat identifiziert werden.

Verfahren zur Herstellung einer Verbindung der Formel (6)R'CONH ^SHN*R" IrOfJÌ.^R"(6)]S /(5)worin R' der Rest einer Acylgruppe R'CO und R" Wasserstoff oder eine gegebenenfalls substituierte Kohlenwasserstoffgruppe, Acyl, abgeleitet von organischen oder anorganischen Säuren, Silyl, oder Sulfenyl ist, dadurch gekennzeichnet, dass eine Verbindung der Formel (5)R«X100 R"N' \f(5)-N^r«in Gegenwart von Wasser mit einer Säure behandelt wird.
- 2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass die Säure eine Dissoziationskonstante von mindestens 0,001 hat.
- 3. Verfahren gemäss Anspruch 1, dadurch gekennzeichnet, dass die Säure eine der folgenden ist: Perchlorsäure, Tri-fluoressigsäure, Trichloressigsäure, Dichloressigsäure, Tri-fluormethansulfonsäure, Trichlormethansulfonsäure, Tetra-fluorborwasserstoffsäure, Chlorwasserstoffsäure, Bromwasserstoffsäure, Fluorwasserstoffsäure, Salpetersäure, Phosphorsäure, Benzolsulfonsäure, Toluolsulfonsäure, Brombenzol-sulfonsäure, Methansulfonsäure oder Äthansulfonsäure.
- 4. Verfahren gemäss Anspruch 1, worin R" eine Gruppe der folgenden Formel istT r^CH2HalCOXworin Hai Halogen, X eine Hydroxy- oder Carboxyschutz-gruppe und Y Piperidino, Morpholino oder eine disubsti-tuierte Aminogruppe mit 2-20 Kohlenstoffatomen ist.in Gegenwart von Wasser mit einer Säure behandelt wird.Repräsentative Acylgruppen R'CO sind anorganische 15 Acylgruppen wie Carbonsäureacyl (z.B. Alkoxycarbonyl, Aralkoxycarbonyl oder Aryloxycarbonyl), Schwefelsäureacyl, Phosphorsäureacylgruppen (z.B. Dialkoxyphosphinyl, Di-alkoxythiophosphonyl oder Alkoxyaminophosphoroyl) und organische Acyle wie Alkanoyl, Cycloalkanoyl, Aralkanoyl, 20 Aroyl, Alkylsulfonyl, Arylsulfonyl oder Alkylphosphonyl. Diese Gruppen können, wenn möglich, in ihrer Struktur durch ein Heteroatom unterbrochen sein oder können ungesättigt oder substituiert sein, beispielsweise durch Halogen (z.B. Fluor, Chlor oder Brom), eine Stickstoff-Funktion (z.B. Ami-25 no, Hydrazin, Azido, Alkylamino, Arylamino, Acylamino, Alkylidenamino, Acylimino, Imino oder Nitro), eine Sauerstoff-Funktion (z.B. Alkyl, Alkenyl, Aralkyl,Aryl, Carboxy, Carbalkoxy, Carbamoyl, Alkanoyl, Aroyl, Aminoalkyl, Aralkanoyl oder Cyano), oder eine Phosphorfunktion (z.B. Phospho oder Phosphoroyl). A und B können 35 auch gemeinsam unter Bildung einer Diacylgruppe einer mehrbasischen Säure aufgefasst werden (z.B. Phthaloyl, Pyri-din-2,3-dicarbonyl, Maleoyl oder Succinoyl).Bevorzugte Gruppen des genannten Acyls können die Acyle der Penicillinseitenkette sein (z.B. Phenylacetyl, Phen-oxyacetyl, Heptanoyl) oder Gruppen, die umwandelbar sind in solche, die den Endprodukten antibakterielle Wirksamkeit verleihen (z.B. Wasserstoff, N-tert.-Butoxy-2-phenylglycin-amido, a-(l-Carbomethoxy-l-isopren-2-yl)amino-a-phenyl-glycyl, 4-Phenyl-2,2-dimethyl-5-oxo-l,3-imidazolidin-l-yl, a-Diphenylmethoxycarbonyl-a-phenyl-acetamido).Besonders bevorzugte Verbindungen (I) gemäss dem er-findungsgemässen Verfahren sind die der folgenden Formel4045
