In dem obigen Reaktionsschema bedeutet ABN eine Amino-oder substituierte Aminogruppe. Die genannte substituierte Amino-
gruppe kann Acylamino, Hydroxycarbylamino, Hydrocarbyliden-amino, Silylamino, Sulfenylamino oder ähnliche Schutzgruppen sein, die bis zu 20 Kohlenstoffatome enthalten, wie sie in der Cephalosporin oder Penicillinchemie verwendet werden.
5 Repräsentative Acylgruppen des genannten Acylaminos sind anorganische Acylgruppen wie Carbonsäureacyl (z.B. Alkoxycar-bonyl, Aralkoxycarbonyl oder Aryloxycarbonyl), Schwefelsäureacyl, Phosphorsäureacylgruppen (z.B. Dialkoxyphosphinyl, Dialkoxy-thiophosphonyl oder Alkoxyaminophosphoroyl) und organische io Acyle wie Alkanoyl, Cycloalkanoyl, Aralkanoyl, Aroyl, Alkylsul-fonyl, Arylsulfonyl oder Alkylphosphonyl. Diese Gruppen können wenn möglich in ihrer Struktur durch ein Heteroatom unterbrochen sein oder können ungesättigt oder substituiert sein, beispeilsweise durch Halogen (z.B. Fluor, Chlor oder Brom), eine Stickstoffunktion 15 (z.B. Amino, Hydrazin, Azido, Alkylamino, Arylamino, Acylamino, Alkylidenamino, Acylimino, Imino oder Nitro), eine Sauerstoffunk-tion (z.B. Hydroxy, Alkoxy, Aralkoxy, Aryloxy, Acyloxy oder Oxo), eine Schwefelfunktion (z.B. Mercapto, Alkylthio, Aralkylthio, Aryl-thio, Acylthio, Thiooxo, Sulfo, Sulfonyl, Sulfinyl, Alkoxysulfonyl, 20 oder Aryloxysulfinyl), eine Kohlenstoffunktion (z.B. Alkyl, Alkenyl, Aralkyl, Aryl, Carboxy, Carbalkoxy, Carbamoyl, Alkanoyl, Aroyl, Aminoalkyl, Aralkanoyl oder Cyano) oder eine Phosphorfunktion (z.B. Phospho oder Phosphoroyl). A und B können auch gemeinsam unter Bildung einer Diacylgruppe einer mehrbasischen Säure aufge-25 fasst werden (z.B. Phthaloyl, Pyridin-2,3-dicarbonyl, Maleoyl oder Succinoyl).
Bevorzugte Gruppen des genannten Acyls können die Acyle der Penicillinseitenkette sein (z.B. Phenylacetyl, Phenoxyacetyl, Hep-tanoyl) oder Gruppen, die umwandelbar sind in solche, die den End-30 Produkten antibakterielle Wirksamkeit verleihen (z.B. Wasserstoff, N-tert.-Butoxy-2-phenylglycinamido, a-( 1 -Carbomethoxy-1 -isopren-2-yl)amino-a-phenylglycyl, 4-Phenyl-2,2-dimethyl-5-oxo-1,3-imidazolidin-l-yl, a-Diphenylmethoxycarbonyl-a-phenyl-acetamido).
35 Die durch A und/oder B dargestellte Kohlenwasserstoffgruppe kann eine leicht entfernbare aliphatische Kohlenwasserstoffgruppe mit 1 bis 20 Kohlenstoffatomen sein (z.B. Alkyl, Alkenyl, Aralkyl oder andere aliphatische Kohlenwasserstoffgruppen) oder eine leicht entfernbare monocyclische aromatische Kohlenwasserstoffgruppe 40 (z.B. Phenyl oder Pyrimidyl). Diese Gruppen können wenn möglich in ihrer Struktur durch ein Heteroatom unterbrochen sein oder können ungesättigt oder substituiert sein (z.B. durch ein Halogenatom oder eine Stickstoff-, Sauerstoff-, Schwefel-, Kohlenstoff- oder Phosphorfunktion). A und B können auch gemeinsam eine zweiwertige 45 Kohlenwasserstoffgruppe bilden (z.B. Alkylen, Aralkylen, Alkyli-den, Aralkyliden, a-Halo- oder Alkoxyaraliden, Diarylmethyliden oder Cycloalkyliden), die wenn möglich durch ein Heteroatom unterbrochen sein können oder durch einen der oben genannten Reste substituiert sein oder ungesättigt sein können.
so Wenn die Gruppe A Acyl und die Gruppe B ein Kohlenwasserstoff ist, können sie mit dem Stickstoffatom in 7-Stellung zur Ausbildung eines cephemringes kombiniert sein, so dass sie eine cyclische Gruppe darstellen (z.B. einen 4-Oxo-3-imidazolidinylring).
Die Silyl- (z.B. Trialkylsilyl) und Sulfenyl- (z.B. Phenylsulfenyl 55 oder o-Nitrophenylsulfenyl) gruppen, die A und/oder B bedeuten können, sind übliche Aminoschutzgruppen.

Andererseits können A und B zusammengefasst werden unter Ausbildung einer Diacylgruppe, abgeleitet von einer mehrbasischen Carbonsäure mit 4-12 Kohlenstoffatomen, Alkyliden mit 1-6 Kohlenstoffatomen oder Arylmethyliden mit 7-9 Kohlenstoffatomen.
Beispiele für die obigen Ar Gruppen sind Furyl, Thienyl, Pyrryl, Oxazolyl, Isoxazolyl, Oxadiazolyl, Oxatriazolyl, Pyrazolyl, Imidazo-lyl, Triazolyl, Tetrazolyl, Phenyl, Pyridyl, Pyrimidyl, Pyrazinyl, Pyri-dazinyl, Triazinyl und Dihydrophenyl, wobei jedes derselben gegebenenfalls substituiert sein kann durch Halogen, Alkyl mit 1-3 Kohlenstoffatomen, Hydroxy, Aminomethyl oder Alkoxy mit 1-3 Kohlenstoffatomen.
Die Carboxyschutzgruppe, dargestellt durch X kann bis zu 20 Kohlenstoffatome enthalten und kann eine Sauerstoffunktion enthalten wie beispielsweise Alkoxy mit 1-8 Kohlenstoffatomen (z.B. Methoxy, Äthoxy oder tert.-Butoxy), Aralkoxy mit 7-20 Kohlenstoffatomen (z.B. Benzyloxy, Methoxybenzyloxy, Nitrobenzyloxy, Di-phenylmethoxy oder Trityloxy), mono- oder bicyclisches Aryloxy (z.B. Phenoxy oder Naphthyloxy) oder Organometalloxy (z.B. Tri-methylzinnoxy oder Trimethylsilyloxy), organisches oder anorganisches Acyloxy mit bis zu 8 Kohlenstoffatomen, oder Metalloxy der Gruppen I, II oder III des Periodensystems (z.B. Natriumoxy, Kaliumoxy oder Magnesiumdioxy) oder X kann eine Schwefelfunktion enthalten wie beispielsweise Thiolester, Thiocarboxy oder ähnliche Gruppen, Stickstoffunktionen, wie Amide, Hydrazide, Azide oder ähnliche Gruppen oder X kann eine andere Carboxyschutzgruppe bedeuten.
Diese Gruppen können sofern möglich im Kern durch ein Heteroatom unterbrochen sein, können ungesättigt oder substituiert sein beispielsweise durch einen der oben genannten Substituenten (z.B. Stickstoff-, Sauerstoff-, Schwefel-, Kohlenstoff- oder Phosphorfunktionen, die oben genannt sind, oder Halogen). Bevorzugte Carboxy-schutzgruppen X sind solche, die Halogenalkylester mit 1-5 Kohlenstoffatomen bilden, Acylakylester mit 2-10 Kohlenstoffatomen, Al-koxyalkyl- oder Aminoalkylester mit 2-8 Kohlenstoffatomen, Aryl-ester oder Aralkylester mit 7-20 Kohlenstoffatomen, Ester mit einem Oxim mit 2-10 Kohlenstoffatomen, N-Alkoxyamid mit 1-5 Kohlenstoffatomen, das Imid des Sacharins, das Imid des Phthalimids, N,N'-Diisobutylhydrazid, Metallsalze oder Alkylaminsalze mit 1-6 Kohlenstoffatomen oder diesen gleichwirkende Gruppen (die vorstehend genannte Anzahl von Kohlenstoffatomen gelten für die Gruppen X).
Antibakteriell bervorzugte Carboxyschutzgruppen X sind solche, die die folgenden Ester bilden: Acyloxymethylester, Phenylacylester, Benzaldoximester, die N,N-Dimethylaminométhylester, ferner Alkalimetallsalze, Erdalkalimetallsalze, acylierte Erdalkalimetallsalze und andere diesen Gruppen gleichwirkende Gruppen. Bevorzugte Carboxyschutzgruppen X sind: tert.-Butoxybenzyloxy, Benzhydrylo-xy, p-Nitrobenzyloxy, p-Methoxybenzyloxy, 2,2,2-Trichloräthoxy und Alkalimetalloxy.
Das durch die Formel wiedergegebene Hai kann sein: Chlor, Brom, Iod oder Fluor, wobei Chlor und Brom am meisten bevorzugt sind.
Der Thiolsubstituent R kann ein solcher sein, der ohne nachteilige Wirkung auf den anderen Teil des Moleküls vor oder während der Cyclisierung entfernbar ist. Er kann eine Acylgruppe sein, wie tert.-Butoxycarbonyl, Carbobenzoxy, Cyclopropylmethoxycarbonyl, Cyclopropyläthoxycarbonyl, 2,2,2-Trichloräthoxycarbonyl, 2-Methansulfonyläthoxycarbonyl; 1-Alkoxy oder Acyloxyalkylgrup-pen enthaltend 2-10 Kohlenstoffatome (z.B. Methoxymethyl, Äthoxymethyl, Acetoxymethyl, 1-Benzoyloxyäthyl), mono- oder dicyclische aromatische Thiogruppen (z.B. Thiadiazolylthio, Thiazo-lylthio, Benzothiazolylthio, Phenylthio, o-Nitrophenylthio, Naph-thylthio) und ähnliche Gruppen.
Die erfindungsgemässe oxidative Aufspaltung von ungesättigten Bindungen erfolgt gemäss dem folgenden Reaktionsschema:

Die Ozonoxidation ist insbesondere weniger von Seitenreaktionen begleitet und ist daher wegen ihrer milden Reaktionsbedingungen vorzuziehen. Die Ozonoxidation erfolgt durch Einführen von Ozon unter Ausbildung eines Ozonids des Ausgangsmaterials und anschliessende Umsetzung mit einem Reduktionsmittel, das das gebildete Ozonid reduktiv aufspalten kann. Besonders bevorzugte Lösungsmittel sind Alkane, Halogenalkane, Äther, Alkanoatester, Alkansäuren, Alkohole, aromatische Kohlenwasserstoffe, insbesondere eine Mischung von Chloralkanen und Alkoholen z.B. Methanol und
Äthanol. Die Reaktionsmedien können Metall und Säure sein (Zink-und Essigsäure, Eisen und Salzsäure, usw.), Schwefeldioxid oder Sulfit (Natriumsulfit, Kaliumbisulfit usw.), dreiwertige Phosphorverbindungen (Phosphitester, Phosphitsalze usw.), Ferrocyanid, Sulfide 5 (Dialkylsulfid z.B. Dimethylsulfid, aromatische Sulfide z.B. Di-phenylsulfid, Dibenzylsulfid), Wasserstoff (in Gegenwart von Raney-Nickel, Platin, Palladium usw.), Borhydride oder deren Komplexe (Natriumborhydride usw.), komplexe Aluminiumhydride (Lithium-aluminiumhydrid usw.), Hydrazine und andere Reaktionsmittel. Bildung des Ozonids verläuft bei einer Temperatur von weniger als — 80°C oder höher als Zimmertemperatur, jedoch bei einer niedrigen Temperatur, bei der die Wirksamkeit des Ozons hoch ist und Nebenreaktionen unterdrückt werden. Ein Überschuss von Ozon kann aus dem Reaktionsmedium durch Einführen von Sauerstoff, Stickstoff, Luft usw. in die Reaktionsmischung entfernt werden. Die Reduktionsmittel können in die Reaktionsmischung, enthaltend das Ozonid als solches, oder nach Einengung der Mischung zugefügt werden. In eine Lösung von Benzyl-a-(3-phenoxyméthyl-7-oxo-4-thia-2,6-diazabicyclo-[3,2,0]-hept-2-en-6-yl)-a-isopropenylacetat(4,22g) in Dichlormethan und Methanol (5:1) wird ozonisierter Sauerstoff eingeführt bis die blaue Farbe der Lösung nicht mehr verschwindet. Dann wird die Lösung mit Dimethylsulfid gemischt, mit Wasser gewaschen, getrocknet und eingeengt. Der erhaltene Rückstand wird chromatographisch über Silikagel, enthaltend 10% Wasser, gereinigt und ergibt Benzyl-a-(3-phenoxyméthyl-7-oxo-4-thia-2,6-diazabi-cyclo-[3,2,0]-hept-2-en-6-yl)-a-( 1 -hydroxyäthyliden)acetat (2,98 g). Ansbeute: 70,28%.
Beispiel 2:
In eine Lösung von Methyl-a-(4-acetylthio-3-phthalimido-
2-oxazetidin-l-yl)-a-isopropenylacetat (200 mg) in Methylenchlorid (10 ml) wird ein Überschuss von Ozon bei — 5°C eingeführt. Nachdem das Gas bei seinem Austritt ein Kaliumiodid-Stärkepapier anfärbt, wird die Lösung auf 2 ml eingeengt, mit einer Lösung von Natriumborhydrid (50 mg) in Methanol (10 ml) gemischt und 30 min. gerührt. Die Reaktionsmischung wird eingeengt, in Methylenchlorid gelöst, mit Wasser gewaschen, getrocknet und eingedampft. Der Rückstand wird aus einer Mischung von Methylenchlorid und Äther umkristallisiert und ergibt Methyl-a-(4-acetylthio-
3-phthalimido-2-oxoazetidin-1 -yl)-a-( 1 -hydroxyäthyliden)acetat (157 mg). Ausbeute: 78%. Schmelzpunkt: 178-183°C.
Beispiel 3:
In eine Lösung von a-(4-Acetylthio-3-phthalimido-2-oxoazetidin-l-yl)-a-isopropenylessigsäure (100 mg) in Methanol (10 ml) wird bei 0°C ein Überschuss von Ozon eingeführt. Nachdem das Gas am Austritt ein Kaliumiodid-Stärkepapier anfärbt, wird durch die Lösung Schwefeldioxidgas durchgeleitet. Die Reaktionsmischung wird konzentriert und der erhaltene Rückstand in einer wässrigen Lösung von Natriumbicarbonat gelöst, mit Äther gewaschen, mit Salzsäure neutralisiert und mit Methylenchlorid extrahiert. Der Extrakt wird mit Wasser gewaschen, getrocknet, eingedampft und ergibt a -(4-Acetylthio-3-phthalimido-2-oxoazetidin-l-yl)-a-acetylessigsäure (64 mg). Ausbeute: 63%. Schaum.

Beispiel 4:
In eine gerührte Lösung von 2,2,2-Trichloräthyl-a -[4-(2-benzothiazolyl)dithio-3-phenoxyacetamido-2-oxoazetidin-l-yl]-a-isopropenylacetat (1,76 g) in einer Mischung von Methylenchlorid (70 ml) und Methanol (18 ml) wird unter Kühlung mit Trockeneis/Aceton Ozon eingeführt, bis die Lösung hellblau erscheint. Nach Einführung von Stickstoffgas wird die Reaktionsmischung mit Schwefeldioxidgas behandelt, bis die Lösung hellgelb erscheint (ungefähr 30 s) und wird dann konzentriert. Der erhaltene Rückstand wird mit Methylenchlorid extrahiert, mit Salzlösung gewaschen, getrocknet und eingeengt, so dass ein pulverförmiger Rückstand (1,35 g) erhalten wird. Die chromatographische Reinigung des Rückstandes über Silikagel (30 g) ergibt 2,2,2-Trichloräthyl-a -[4-(2-benzothiazolyl)dithio-3-phenoxyacetamido-2-oxoazetidin-l-yl]-a-(l-hydroxyäthyliden)acetat (1,09 g, Ausbeute: 62,0%, Schmelzpunkt 130-131°C) und 2,2,2-Trichloräthyl-a -[4-(2-benzothiazolyl)dithio-3-phenoxyacetamido-2-oxoazetidin-lyl]-a-(l,l-dimethoxyäthyl)-acetat (0,30 g). Ausbeute: 15,6%.
Beispiel 5:
In eine Lösung von a-[4-substituierter (R2) thio-3-substituierter-amino (R1)-2-oxoazetidin-l-yl]-a-isopropenylacetatester (R3) in einem Lösungsmittel wird unter Kühlung Ozon eingeführt. Nachdem das Gas an dem Ausgang Kaliumiodid-Stärkepapier blau färbt, wird die Zuführung von Ozon unterbrochen und der Überschuss des

Ozons durch Einführung von Stickstoffgas entfernt. Zu der Reaktionsmischung wird ein Reduktionsmittel zugegeben und 10 bis 30 min reagieren gelassen. Nach der Reduktion wird das Lösungsmittel abgedampft und der Rückstand umkristallisiert, oder die Reaktionsmischung durch eine Schicht von Silikagel filtriert und das
Filtrat eingedampft, wobei a-(4-substituierter-thio-3-substituierter-amino-2-oxoazetidin-1 -yl)-a-( 1 -hydroxyäthyliden)essigsäureester erhalten wird.
Die Reaktionsbedingungen sind in Tabelle i und die physikalischen Konstanten der Produkte in Tabelle ii angegeben.
