Die Erfindung bezieht sich'auf neue 2-Gaanidino-4-hetero- _, arylthiazole mit Aktivität als antisekretorische Mittel . und.-Hiatamin-Hp-Antagonisten und die somit bei der Verhütung und Behandlung von Magenübersäuerung und Geschwüren des Verdauungstraks brauchbar sind. '
Chronische Magen- und Zwölffingerdarmgeschwüre, zusammen als Geschwüre des Verdauungstraks bekannt ,x sind ein verbreitetes Leiden, für das eine Vielzahl von Behandlungen, einschließlich diätetische Maßnahmen, Arzneimitteltherapie und Chirurgie angewandt werden können, je nach der Schwere des Zustande.. Besonders wertvolle therapeutische Mittel, die zur Behandlung von Magenübersäuerung1 und Geschwüren des Verdauungstrakts brauchbar sind, sind die Hiatamin-Hp-Rezeptor-Antagonisten, die die Wirkung der physiologisch wirksamen( Verbindung Histamin an den Hp-3-ezeptorstellen im Tierkörper blockieren und somit die -Abscheidung von Magensäure hemmen. . - '

Das hierbei einzusetzende 2-R'-5-Rn-4-Acetyl-i:aidazol kann durch Bestrahlen von 1-Acetyl-2-Rl-5-Ri'-imidazol mit UV-Licht hergestellt werden. Andererseits können 2-H!-4- Acetyliiaidasole durch Umsetzen eines 3-Halogen-4-n-alko:cy-3-buten-2-ons, insbesondere der 3-Ghlor- oder 3-3rom-yerbindung, mit einem geeigneten R1 -substituierten -Ainidin der Pormel R1G(IiH2)IiH oder einem Salz: hiervon in,Gegenwart einer _ Base, wie einem Trialkylamin, vörz.iigaweise Triäthylamin,: in einem organischen Lösungsmittel, wie Aceton, Setrahydrofuran, Dioxan und dgl,, bei einer Temperatur von etwa 0 bis etwa.100 0G, vorzugsweise, bei Rückflußtemperatur in Tetrahydrofuran, hergestellt werden.
Die 3~Halogen-4-n-alkoxy-3-buten-2-on-Äu3gangsmaterialien können aus dem bekannten 3-Halogen-4-hydro3:y-3-buten-2-on durch Umsetzen mit einem Dialkylsulfat oder Diazoalkan in z, B., wäßrigem Tetrahydrofuran oder: D&oxan in Gegenwart einer achwachen.Base, wie einem Alkalimetallbicarbonat, bei einer Temperatur von etwa 0 bis etwa 60^ 0G, vorzugsweise etwa 15 bis etwa 30 0C, hergestellt werden. Andererseits kann das 3~Halogen-4-n-alkoxy-3-buten-2-on durch Erwärmen eines 3-Halogen-4-hydros:y-3-buten-2-ons auf Rückfluß mit einem geeigneten n-Alkanolj z, B, in' ^ol.uol-Lösung, hergestellt

Die pharmazeutisch annehmbaren Säureadditionssalze der neuen Verbindungen der formel I fallen auch unter die Erfindung. Die Salze werden leicht durch Zusammenbringen der freien Base mit einer geeigneten Mineral- oder organischen Säure entweder in wäßriger Lösung oder in einem geeigneten organischen Lösungsmittel hergestellt. Das feste Salz kann dann durch Ausfällung oder durch Einengen des Lösungsmittels erhalten werden« Die erfind ungsgeinäßen pharmazeutisch annehmbaren Säureadditionssalze umfassen, ohne hierauf au beschränken, das Hydrochlorid, Sulfat,. Biaulfat, Mesylat, Hitrat, Phosphat, Acetat, Lactat, Maleat, Pumarat, Gitrat, Tartrat, Succinat, Gluconat und dgl. Bevorzugte Salze sind das Hydrochlorid und DiJaydrochlorid. Wenn gewünscht, können die Verbindungen der -BOr-inel IA als freie Basen au's den Säureadditionssalzeh durch Behandeln mit einer geeigneten Base und anschließendes Extrahieren der freien Base mit einem geeigneten organischen Lösungsmittel hergestellt werden. ' \ · .
Die Verbindungen der formel IA und deren pharmazeutisch annehmbare Säureadditionssalze besitzen Aktivität als antisektretori3che Mittel und Histamin-Hp-Antagonisten und sind somit von therapeutischem ΐ/ert für die. Behandlung von Magenübersäuerung und Geschwüren des Verdauungstrakts. Für die Zwecke der vorliegenden Beschreibung und der Ansprüche soll der Begriff "Behandlung von Magenübersäuerung!l die Behänd-' lung von Geschwüren des Verdauungstrakts und anderer solcher durch Sekretion von ,Mägensäure verursachter oder erschwerter Zustände umfassen. Die Verbindungen können einem zu behandelnden Patienten nach einer Vielzahl herkömmlicher Ver-' abreichungswege, darunter oral und parenteral, verabreicht werden. Vorzugsweise werden die Verbindungen oral verabreicht.

Im allgemeinen werden sie oral in Dosen zwischen etwa .0,1 and 20 mg/kg Körpergewicht des zu behandelnden Patienten pro 'Tag verabreicht, vorzugsweise zu etwa 0,2 bis 2,5 mg/kg pro 'lag. Wird parenterale Verabreichung gewünscht, können diese Verbindungen in Dosen zwischen etwa 0,1 und 1,0 mg/kg Körpergewicht des zu behandelnden Patienten pro Tag gegeben werden. Doch wird ^e nach dem Zustand des behandelten Patienten und der speziell angewandten Verbindung eine gewisse Schwankung in der Dosierung notwendigerweise eintreten. ".>.
Die Verbindung- kann- allein oder in Kombination mit., pharmazeutisch annehmbaren Trägern oder Verdünnungsmitteln verabreicht werden, entweder in einer Einzeldosis oder in mehreren Bösen« Geeignete pharmazeutische,'!rager umfassen inerte Verdünnungsmittel oder füllstoffe, sterile wäßrige Lösungen, und verschiedene organische Lösungsmittel. Die durch Kombinieren der neuen Verbindungen der formel Ia oder ihrer Salze und pharmazeutisch annehmbarer Träger gebildeten Pharmazeutika sind leicht in einer Vielzahl von Dosierungsformen, wie Tabletten, Pulver, Kapseln, Pastillen, Sirupen u.: dgl., zu verabreichen. Diese Pharmazeutika können, wenn gewünscht, zusätzliche Bestandteile, wie Geschmacks- oder Aromastoffe, Bindemittel, izsipienten und dgl.-, enthalten. So können für: orale Verabreichung Tabletten mit verschiedenen iixzipienten, wie Hatriumcitrat, zusammen mit verschiedenen den Zerfall fördernden Mitteln, wie Stärke, Alginsäure und bestimmten komplexen Silikaten," zusammen mit Bindemitteln, wie Polyvinylpyrrolidon, Saccharose, Gelatine und·Akazienharz, verwendet werden. Außerdem sind häufig Gleitmittel, wie Magnesiumstearat, Hatriuinlaury !sulfat und Talkum, für Tablettierungszwecke nützlich, Feste-1 Mittel ..

"enn wäßrige Suspensionen oder Elixieren für orale Verabreichung gewünscht werden, kann der·notwendige aktive Bestandteil mit verschiedenen Süßungsmittel oder Aromatiaierungsmitteln-, ν färbenden Materialien oder 'Farbstoffen und, wenn gewünscht, eraulgierenden Mitteln oder suspendierenden Mitteln, zusammen mit Verdünnungsmitteln, wie Nasser, Äthanol, Propylengly.kol, Glyzerin oder Kombinationen hiervon, kombiniert werden. . - - .' ,. .
Vorzugsweise werden die erfindungsgernäßen neuen Verbindungen oral in Sinheitsdosisform verabreicht, d. h. als eine einzelne körperlich selbständige Dosierungseinheit mit einer, - geeigneten Menge" der aktiven Verbindung in· Kombination mit. einem pharmazeutisch annehmbaren Träger oder Verdünner. Beispiele für solche ^inheitsdosierungsformen sind Tabletten oder Kapseln mit etwa 5 bis 1000 mg des aktiven Bestandteils, wobei die Verbindung der Pornel IA etwa 10 bis 30 % des Gesamtgewichts der Dosierungseinheit ausmacht. :
für parenterale Verabreichung können Lösungen, der Verbindungen der formel IA in sterilen wäßrigen Lösungen, s. B. wäßrigem Propylenglykol, Natriumchlorid, Dextrose oder Natrium-bicaronatlösungen, verwendet werden,.Solche Lösungen sollten, wenn nötig, geeignet gepuffert sein,.und das flüssige Verdünnungsmittel zuerst mit genügend Salzlösung oder Glukose isotonisch gemacht sein. Die Herstellung geeigneter steriler flüssiger Medien für parenterale Verabreichung ist dem Fachmann auf dem Gebiet gut bekannt, . ·.." ,

Die Aktivität der erfindungsgemäßen Verbindungen als antisekr.etorische Mittel und als Histamin-H2-Antagonismen kann nach pharmakologisciien Standardteats ermitteis werden, dazu gehört z. B, (1) die Messung ihres. Eine Lösung von 0,50'g (4,5 mMol) 4-Acetyliinidazo'l in 10 ml Methanol wurde bei Raumtemperatur gerührt, und 10 Tropfen 48%iger Bromwasserstoffsäure wurden zugesetzt» !lach 15 min Rühren bei Raumtemperatur wurden 50 ml' absoluter Äther augesetzt und der anfallende niederschlag durch filtrieren, gesammelt und getrocknet, um 0,54 S des1 Hydrobromids, Schmp, 214 0C (Zers.) ,zu ergeben. 2,3 g (9,86 mMol) 1-(2-Methyl-4-imidazolyl)-2-bromäthanon-Hydrobromid wurden in 10 ml.Wasser gelöst. Gesättigte IJatriumbicarbonatlösung-wurde bis zum pH 10 zugesetzt und der anfallende Peststoff durch !filtrieren gesammelt und mit 15 ml Wasser gewaschen. Die getrocknete freie. Base wurde in 50 ml Aceton auf Rückfluß erwärmt, Zu der rückflußkochenden klaren Äcetonlösung wurden 1,2 g (9,36 πώϊοΐ) .Amidinothionarnstoff gegeben. Eine Lösung von 1,57 g (12,6 miiol) 5-^cetyl-4-methylimidazol in 15 ml.konz. Bromwasserstoffsäure wurde auf 50 0G erwärmt und. eine Lösung, von 2,Q1 g (12,6 mMol) Brom in 15 al konz. Bromwasserstoff-säure-über. 0,75 h zugesetzt, Das Gemisch, wurde 1,25 h bei 50 0C gerührt, dann' e:.inge,engt ,·. Der Rückstand wurde mit Isopropylalkohol "verrieben und der weiße Feststoff durch filtrieren gesammelt« Dies ergab 2,78 g C78 %) 1-(4-Metiiyl-5-iiiiidazolyl)-2-bromäthanon-H:/drobromid: MR (D6DMSO) S : 9,40 (s, 1H); 4,87. (s, 2H); 2,65 (s, 3H)... Dieses Material wurde durch 15 min Rühren in 50 ml gesättigter latriumbioarbonatlösung in die freie Base umge- ; wandelt, um nach dem filtrieren und Trocknen 1-(4-Methyl-' 5-imidazolyl)-2-bromäthanon als; weißes.Pulver zu liefern,

Lösung von 6,9 g (0,05 Mol) 1-Acetyl-2,4-dimethylimidazol in 700 ml trockenem i'etrahydrofuran v/arde in einem Quarzkolben mit einer kurzwelligen UV-Lichtquelle1 (254 ^m) 40 h photolysiert. Dieses Material wurde über Toluol bei · Hochvakuum zu einem.hellgelbbraunen Feststoff getrocknet, Schmp, 208-209 0G unter Zersetzung, NMR (DMSO-dg) ifV 7,16. s, 1H); 6,83 (breites s, 4H); 6,68 (s, 1H); 4,45 (a, 2H)1 3,46 (aehr breites s, 1Ή). Wirkstoff oder Träger wurde intravenös 90 min nach dem Beginn der Pentagastrin-Infusion in einer Dosis von 1 mg/kg gegeben« Die der Magensäuresekretion entgegenwirkenden .Effekte wurden durch Vergleich der niedrigsten Säureabgabe nach Wirkstoff-Verabreichung.mit der mittleren Säureabgabe unmittelbar vor dem Wirkstoff errechnet.
Die erhaltenen Ergebnisse zeigten, daß die Verbindungen,, der Beispiele.3» 3, 12, 16 und 18 die Magensäuresekretion' um mindestens 15 % bei- einer Dosis von 1mg/kg hemmten.

Die Histamin-Hg-Antagonisten-Aktivität der erfindungsgemäßen ^erbindungen wurde nach folgender Arbeitsweise bestimmt:" .· . '
Meerschweinchen wurden durch einen Schlag auf den Kopf rasch getötet, das Hera entnommen und ^die rechten Vorhöfe freigelegt. Die Ergebnisse sind' in Dosis-Verhältnissen ausgedrückt, das Verhältnis von.2ur iraielung der Hälfte der maximalen Anregung'' in" Gagenwart und Abwesenheit -des Antagonisten erforderlichen Histamin-Konzentrationen, und diescM& bare Dissoziationskonstante des. Claims (2)
- oder eines pharmazeutisch annehmbaren·Säureadditions--'aa-lses hiervon, worin Z ΐϊΗ, ϊ GH oder G-GH^, R' iv'a stöff,. Hydroxymethyl, Alkyl ait 1 bis 6 Kohlenstoffatomen oder -(CH2),-Ar, η eine ganze Zahl von 2 bis 4 ,und Ar Phe'nyl oder Phenyl mono substituiert mit Chlor, .Brom, Fluor, Alkyl ait 1 bis 3 Kohlenstoffatomen oder Alkoxy mit 1 bis 3 Kohlenstoffatomen ist, . gekennzeichnet dadurch, daß eine Verbindung der Formelworin R^ Halogen ist, mit H-Amidinothioharnstoff wenn gewünscht, die erhaltene Verbindung der Formel'IA mit einer' pharmazeutisch annehmbaren Säure zu "einem pharmazeutisch annehmbaren ^aIs umgesetzt wird.
- 2. Verfahren nach Punkt 1, gekennzeichnet dadurch,'daß R' Wasserstoff, Hydrosymethyl oder Methyl ist,.
