Den foreliggende opfindelse angår hidtil ukendte 17-substitu-erede 4-aza-5-o;-17/3-androsten-3-on-derivater, som har en 8(14)- eller 7(8)-dobbeltbinding og farmaceutiske midler indeholdende disse forbindelser.
5 Den klasse af steroide hormoner, der kendes som androgener, er ansvarlige for de fysiske karakteristika, der adskiller mænd fra kvinder. Af de adskillige organer, der producerer androgener, producerer testes disse hormoner i de største mængder. Centre i hjernen udøver primær styring af androgenproduktions-10 niveauet. Talrige fysiske manifestationer og sygdomstilstande er et resultat, når ineffektiv produktionsstyring resulterer i for høj androgenhormonproduktion. F.eks. er acne vulgaris, seboré, hirsutisme hos kvinder og godartet prostatisk hyper-trofi forbundet med forøgede androgenniveauer. Desuden er 15 tilfælde af mandlig mønsterskaldethed blevet forbundet med høje androgenniveauer.
Testosteron er det hovedandrogen, der udskilles af testes, og det er det primære androgene steroid i plasma hos mænd. Det er nu kendt, at 5-a-reducerede androgener er de aktive hormoner i 20 nogle væv såsom prostata og fedtkirtler. Cirkulerende testosteron tjener således som et prohormon for dihydrotestosteron (DHT), dets 5-a-reducerede analog i disse væv, men ikke i andre såsom muskel og testes. Steroid-5-a-reduktase er et NADPH-afhængigt enzym, der omdanner testosteron til DHT. Dette en-25 zyms vigtighed ved udviklingen til mand blev dramatisk understreget ved opdagelse af en genetisk steroid-S-a-reduktase-mangel i mandlige pseudohermafroditter, J. Imperato-McGinley et al., 1979, J. Steroid Biochem. 11:637-648.
Erkendelse af vigtigheden af forøgede DHT-niveauer i mange 30 sygdomstilstande har stimuleret mange forsøg på at syntetisere inhibitorer af dette enzym. Strukturerne af flere kendte steroid-5-ce-reduktaseinhibitorer er vist i tabel 1.

Secosteroidet (2) var den næste inhibitor, der blev beskrevet og har også vist anvendelighed som et affinitetsmærke 5 for 5-a-reduktase, B. Robaire et al., (1977), J. Steroid Bio-chem. 8:307-310. Diazoketonen (3) er blevet rapporteret somen kraftig, tidsafhængig inhibitor af steroid-5-a-reduktase, T.
R. Biohm et al., (1980), Biochem. Biophys. Res. Comm. 95:273-280, US patentskrift nr. 4.317.817. Forbindelse (4) er et ek-10 sempel på en gruppe 4-azasteroidinhibitorer af steroid-5-α-reduktase, der er beskrevet i US patentskrift nr. 4.377.584 og i T. Liang et al., (1983) J. Steroid Biochem. 19, 385-390. 6-methylensteroidet (5) har også vist sig at være en tidsafhængig inaktivator af steroid-5-a-reduktase, V. Petrow et al.

Der er også blevet beskrevet andre 5-a-reduktaseinhibitorer. I US patentskrift nr. 4.361.578 beskrives en klasse homosteroid-enzyminhibitorer. I de japanske patentskrifter J60146855-A og J60116657-A beskrives forskellige anilinderivater, der har 20 talrige aktiviteter, herunder 5-of-reduktasehæmmende aktivitet.
I japansk patentskrift nr. J60142941-A beskrives phenyl substituerede ketoner, der har 5-a-reduktasehæmmende aktivitet, og i europæisk patentskrift nr. EP173516-A beskrives forskellige phenylsubstituerede amider, der har tilsvarende aktivitet.
25 Shiseido refererede til tezpenderivater, der er aktive inhibitorer af steroid-5-α-reduktase, japansk patentskrift nr. J59053417-A.
Der kendes fra DK patentansøgning nr. 1525/79, DK patentansøgning nr. 859/85 og EP 200.859A1 forbindelser, som har en 5-a-30 reduktaseinhiberende virkning. Disse kendte forbindelser har dog ikke en 8(14)- eller 7 (8)-dobbeltbinding, som ved forsøg har vist sig at bibringe forbindelserne ifølge opfindelsen ændret konformation og bedre 5-a-reduktaseinhiberende aktivitet sammenlignet med en ellers identisk mættet forbindelse.

Den foreliggende opfindelse bygger på den opdagelse, at steroid- 5-α-reduktase inhiberes af 4-aza-5-a!-17-substitueret-an-drosten-3-on-forbindelser, der har en 8(14)- eller 7(8))-dobbeltbinding og eventuelt også har en 1(2)-dobbelt- binding.

For tiden foretrukne forbindelser ifølge opfindelsen og forbindelser, der anvendes i de farmaceutiske midler ifølge opfindelsen omfatter: 4-methyl-4-aza-5-a-8 (14) -pregnen-3-on- (20R) -20-carboxylsyre, 10 (20R) -hydroxymethyl-4-methyl-4-aza-5-a-8 (14) -pregnen-3-on og 4-methyl-4-aza-5-o;-8 (14) -androsten-3-on-17/3-N,N-diisopropyl-carboxamid.
Der kan ved fremstilling af forbindelserne ifølge opfindelsen anvendes hidtil ukendte mellemprodukter og hidtil ukendte 15 fremgangsmåder, som vil blive næmere beskrevet.
Opfindelsen vedrører ligeledes et farmaceutisk middel til anvendelse ved metoder til hæmning af S-a-reduktaseaktivitet i pattedyr, herunder mennesker, hvilke metoder omfatter intern indgift til et individ, der har behov derfor, af en effektiv 20 mængde af en 5-α-reduktaseinhiberende forbindelse ifølge opfindelsen.
Den foreliggende opfindelse angår således også farmaceutiske midler, der omfatter en farmaceutisk bærer og en forbindelse ifølge opfindelsen.

Ifølge skema I opløses forbindelse A i et egnet organisk opløsningsmiddel, såsom et C1_4alkyldihalogenid i en C1_4alka-nol, fortrinsvis methylenchlorid i methanol, afkøles til ca. minus 100°C til 30°C, fortrinsvis minus 78°C og behandles med 5 rutheniumdioxid/natriumperjodat, kaliumpermanganat eller fortrinsvis et overskud af ozon. Den resulterende opløsning renses med en inaktiv gas, såsom argon, idet den opvarmes til ca.
25°C til 50°C, fortrinsvis 40°C. 5 Fremstilling af forbindelser med formlen (I) ifølge skema II begynder ud fra de samme udgangsmaterialer som i skema I. Forbindelser med formlen (N) fremstilles ved omsætning af forbindelser med formlen (A) med et quinonoxidationsmiddel, såsom chloranil i et egnet organisk opløsningsmiddel, såsom t-buta-10 nol. Forbindelserne med formlen (N) omsættes derefter med en nukleofil thiolforbindelse, såsom thiophenol, butylmercaptan eller fortrinsvis thioeddikesyre, til fremstilling af forbindelser med formlen (O).
Thioacetylenonen med formlen (O) opløses derefter i et egnet 15 organisk opløsningsmiddel, såsom et C-j^alkyldihalogenid i en Ci_4alkanol, fortrinsvis methylenchlorid i methanol, afkøles til ca. minus 100°C til 0°C, fortrinsvis minus 78°C, og behandles med et overskud af ozon. Efter rensning med en inaktiv gas, såsom argon, opkoncentreres opløsningen og behandles med 20 en stærk base, såsom natriumhydroxid, til fremstilling af en rest. Resten opløses derefter i et egnet organisk opløsningsmiddel, afkøles til ca. minus 30°C til 30°C, fortrinsvis 0°C, og behandles, i nærværelse af en base, såsom natriumbicarbo-nat eller DBU, med et C-L_4alkylhalogenid, såsom methylchlorid, 25 ethylchlorid, methyljodid eller fortrinsvis diazomethan til opnåelse af en forbindelse med formlen (P).
Forbindelser med formlen (P) kombineres derefter med ammoniak eller en C^.galkylamin, der er udvalgt til at have den ønskede R1-substituent, og opvarmes til ca. 150°C til 200°C, for-30 trinsvis 180°C, til opnåelse af forbindelser med formlen (Q). Forbindelser med formlen (I) fremstilles derefter ved hydrogenering af forbindelser med formlen (P) under anvendelse af hydrogeneringsmidler, såsom palladium på carbon eller Raney-nikkel, fortrinsvis platandioxid, og hydrogeneringsopløsningsmid- 12 DK 168525 B1 ler, såsom egnede organiske opløsningsmidler. Som i skema I fremstilles forbindelser med formlen (I), hvor Δ7 er -CH=CH-, ved hydrogenering af forbindelser med formlen (Q) under anvendelse af Raney-nikkel og omhyggelig overvågning af reak-5 tionen for at minimere eller forhindre dannelse af forbindelser med formel (I), hvor Δ8 er -CH=CH-.
Forbindelser med formlen (A), udgangsmaterialerne i skema I og II, fremstilles ud fra de analoge 17-ketoforbindelser ved hjælp af kendte fremgangsmåder, såsom dem, der er beskrevet i 10 J. Fried og J. Edwards, Organic Reactions in Steroids Chemistry, Pub: Van Nostrand og Reinhold Company (1972). 17-keto-forbindelserae er tilgængelige og kan syntetiseres ved hjælp af kendte procedurer. Udgangsmaterialerne vælges således, at R2- og R3-grupperne i forbindelsen med formlen (A) er de sam-15 me som R2- og R3-grupperne i forbindelsen med formel (I) , der syntetiseres. Alternativt udvælges R2- og R3-grupperne i forbindelsen med formlen (A) således, at de kan omdannes ved kendte procedurer til R2- og R3-grupperne i målforbindelsen med formlen (I) ved yderligere trin i synteseprocessen. F.eks.
20 omdannes forbindelser med formel (I), hvor R3 er carboxylsyre, til de tilsvarende amider ved omsætning med aminer eller substituerede aminer via de tilsvarende syrechlorider. Tilsvarende fremstilles forbindelser med formlen (I), hvor R3 er CH3CHCOOH, ved oxidation af den tilsvarende alkohol.
25 Farmaceutisk acceptable syreadditionssalte af forbindelserne ifølge opfindelsen dannes, hvor det er hensigtsmæssigt, med stærke eller moderat stærke organiske eller uorganiske syrer i nærværelse af en basisk amin ved inden for fagområdet kendte metoder. F.eks. omsættes basen med en uorganisk eller organisk 30 syre i et vandblandbart opløsningsmiddel, såsom ethanol, med isolering af saltet ved fjernelse af opløsningsmidlet eller i et vandublandbart opløsningsmiddel, når syren er opløselig deri, såsom ethylether eller chloroform, idet det ønskede salt udskilles direkte eller isoleres ved fjernelse af opløsnings-35 midlet. Eksempler på syreadditionssaltene, der er omfattet af 13 DK 168525 B1 denne opfindelse, er maleat, fumarat, lactat, oxalat, methan-sulfonat, ethansulfonat, benzensulfonat, tartrat, citrat, hy-drochlorid, hydrobromid, sulfat, phosphat og nitrat. Farmaceutisk acceptable baseadditionssalte af forbindelser ifølge op-5 findelsen fremstilles ved kendte metoder, omfattende ikke-tok-siske alkalimetal- og jordalkalimetalsalte, f.eks. calcium-, natrium- og kaliumsalte, ammoniumsalte og salte af ikke-tok-siske organiske baser såsom triethylamin, butylamin, piperazin og (trihydroxymethyl) methylamin.

Fordi forbindelser med formlen (I) inhiberer steroid-5-a-re-duktaseaktivitet, har de terapeutisk anvendelighed til behand-5 ling af sygdomme og tilstande, hvor formindskelser i DHT-aktivitet giver den ønskede terapeutiske virkning. Sådanne sygdomme og tilstande omfatter acne vulgaris, seboré, hirsutisme hos kvinder, prostatasygdomme såsom godartet prostatisk hy-pertrofi og mønsterskaldethed hos mænd. Styrken hos flere for-10 bindeiser ifølge opfindelsen blev undersøgt for styrke til at inhibere human steroid-5-α-reduktase under anvendelse af væv fra humane hyperplastiske prostata (hyperplastic human prostates) . Til bestemmelse af styrke til at inhibere det humane enzym anvendtes følgende procedure: 15 Frosne humane prostata blev optøet og hakket i små stykker (<5 mm3) . Vævet blev homogeniseret i 3 til 5 volumener 20 mM kali-umphosphat, pH-værdi 6,5, puffer indeholdende 0,33M saccharose, 1 mM dithiothreitol og 50 μΜ NADPH med en Brinkmann Poly-tron (Sybron Corporation, Westbury, New York). Opløsningen 20 blev underkastet lydbehandling i 3 til 5 minutter med en Soni-fier (Branson Sonic Power Co.) efterfulgt af håndhomogenisering i en glas-til-glas-Dounce-homogenisator (Kontes Glass Company, Vineland, New Jersey).
Der opnåedes prostatapartikler ved differentiel centrifugering 25 ved 600 eller 1000 x g i 20 minutter og 140.000 x g i 60 minutter ved 4°C. Den pellet, der opnåedes ved centrifugering ved 140.000 x g, blev vasket med 5 til 10 vævsvolumener af pufferen beskrevet ovenfor og gencentrifugeret ved 140.000 x g. Den resulterende pellet blev suspenderet i 20 mM kalium-30 phosphatpuffer, pH-værdi 6,5, indeholdende 20% glycerol, 1 mM dithiothreitol og 50 μΜ NADPH. Den suspenderede partikelopløsning blev lagret ved minus 80°C.
En konstant mængde [14C]-testosteron (52 til 55 mCi/mmol, New 15 DK 168525 B1
England Nuclear, Boston, MA) i ethanol og varierende mængder af den potentielle inhibitor i ethanol blev anbragt i prøverør og opkoncentreret til tørhed i en SAVANT Speed Vac. Til hvert rør blev sat puffer, 20 μΐ 10 mM NADPH og en portion prostata-5 partikelopløsning til et slutvolumen på 0,5 ml 50 mM natriumcitrat, pH-værdi 5,0. Efter inkubation af opløsningen ved 37°C i 20 til 30 minutter blev reaktionen standset ved tilsætning af 4 ml ethylacetat og 0,25 μιηοΐ af hver af testosteron, di-hydrotestosteron, androstandiol og androstandion som bærere.
10 Det organiske lag blev fjernet til et andet prøverør og inddampet til tørhed i en Speed Vac. Resten blev opløst i 20 til 30 μΐ chloroform, anbragt i pletter på en individuel bane af en 20 x 20 cm i forvejen med kanaler forsynet silicagel-TLC-plade (Si 250R-PA, Baker Chemical) og fremkaldt to gange med 15 acetone: chloroform (1:9). Det radiokemiske indhold i båndene af substratet og produkterne blev bestemt med BIOSCAN Imaging Scanner (Bioscan, Inc., Washington D.C.). Procentsatsen af udvundet radioaktivt mærke omdannet til produkt blev beregnet, hvor ud fra enzymaktiviteten blev bestemt. Alle inkubationer 20 blev udført således, at der ikke forbrugtes mere end 12% af substratet (testosteron).
De eksperimentelt opnåede data blev EDB-tilpasset til en lineær funktion ved at afsætte den reciprokke værdi af enzymaktiviteten (1/hastighed) mod den variable inhibitorkoncentration 25 (M. Dixon, 1973), Biochem. J., 55, 170). Ved at antage, at den steroide inhibitor er en kompetitiv inhibitor mod testosteron, kan en værdi for inhiberingskonstanten (Ki) beregnes ud fra ligning 1: ^ = (B/A) /S/Kjh + 1) ligning 1 30 hvor B er afskæringen på aksen 1/hastighed, A er liniens hældning, S er substratkoncentrationen (testosteron), der anvendtes i eksperimentet, og er Michaelis-Menton-konstanten for substratet (testosteron), bestemt i et separat eksperiment til at være 4,5 μΜ.

Tabel II viser resultaterne af den ovenstående afprøvning og viser, at de testede forbindelser ifølge opfindelsen er kraftige inhibitorer af human steroid-a-reduktase.
Visse forbindelser ifølge opfindelsen blev også testet for de-5 res in vivo-styrke til at inhibere steroid-5-of-reduktaseakti-vitet. Charles River CD-hanrotter, der var 48 dage gamle og vejede ca. 200 g, fik indgivet forbindelserne ifølge opfindelsen, forbindelserne 1) og 2), vist i tabel III, opløst i pro-pylenglycol og fortyndet i normal saltopløsning. Forbindelser-10 ne blev indgivet oralt i doser på 5, 10 og 20 mg/kg. Seks timer efter indgift af forbindelsen blev dyrene aflivet, ventral prostata udskåret og testosteron (T) - og DHT-niveauer målt ved hjælp af følgende procedure.
Prostatavæv blev udskåret, gjort parat, vejet, hakket og va-15 sket med phosphatpuffer. Vævet blev derefter homogeniseret i phosphatpuffer og ekstraheret ved tilsætning af ethylacetat og blanding på en omløbsmixer i 45 minutter. Ethylacetatet blev af dampet, og resten blev genopløst i ethanol og blev centrifugefiltreret under anvendelse af 0,45 μΜ filtrerpapir. Kompo-20 nenteme blev derefter adskilt under anvendelse af HPLC med omvendt fase til opsamling af T- og DHT-fraktioner. Fraktionerne blev reduceret til tørhed og genopløst i standard-T/DHT-analysepuffer, der fås fra Amersham. 15 I tabel III sammenlignes de resultater, der opnås i dyrene, der er behandlet med forbindelserne ifølge opfindelsen med dem, der opnås i bærerbehandlede kontroldyr. Som dataene viser, gav hvert af de undersøgte dosisniveauer væsentlige formindskelser i prostata-DHT-niveauer og forøgede således T/DHT-20 forholdene. Forbindelserne ifølge opfindelsen er derfor kraftige steroid-5-a-reduktaseinhibitorer in vivo.
Tidsforløbet for in vivo 5-a;-reduktaseinhibering, der fremkaldes af forbindelserne ifølge opfindelsen, blev også undersøgt. Under anvendelse af fremgangsmåder, som svarer til dem, der 25 anvendes til opnåelse af de data, der er vist i tabel III, fik rotter indgivet 4-methyl-4-aza-8 (14)-androsten-3-on-17/3-N,N-diisopropylcarboxamid, og ventralprostatavæv blev udskåret til T- og DHT-bestemmelser på flere tidspunkter efter indgift. Som dataene i tabel IV viser, observeredes ved sammenligning med 30 kontroller signifikante forskelle i DHT-niveauer 1, 2, 4 og 8 timer efter indgift af forbindelsen.

Forbindelserne med formlen I inkorporeres i passende dosisformer, såsom kapsler, tabletter eller injicerbare præparater.
15 Der anvendes faste eller flydende farmaceutiske bærere. Faste bærere omfatter stivelse, lactose, calciumsulfatdihydrat, terra alba, saccharose, talkum, gelatine, agar, pektin, akacie, magnesiumstearat og stearinsyre. Flydende bærere omfatter sirup, jordnøddeolie, olivenolie, saltopløsning og vand. Tilsva-20 rende kan bæreren eller fortyndingsmidlet omfatte ethvert materiale med forlænget frigivelse, såsom glycerylmonostearat eller glyceryldistearat, alene eller sammen med en voksart. Mængden af fast bærer varierer meget, men vil fortrinsvis være fra ca. 25 mg til ca. 1 g pr. dosisenhed. 1 Når der anvendes en 25 flydende bærer, vil præparatet foreligge i form af en sirup, eleksir, emulsion, blød gelatinekapsel, steril, injicerbar væske, såsom en ampul, eller en vandig eller ikke-vandig væskesuspension.
De farmaceutiske præparater fremstilles ved at følge konven-30 tionelle farmaceutiske metoder, der involverer blanding, granulering og komprimering, om nødvendigt, til tabletf omer, eller blanding, fyldning og opløsning af bestanddelene, som det 20 DK 168525 B1 er passende, til opnåelse af de ønskede orale eller parente-rale produkter.

15 Metoden til hæmning af steroid-5-areduktaseaktivitet i pattedyr, herunder mennesker, omfatter intern indgift til et individ med behov for en sådan inhibering af en effektiv steroid-5-a-redukateseinhiberende mængde af en forbindelse med formlen I.
20 De følgende eksempler illustrerer fremstilling af forbindelser med formlen (I) og farmaceutiske midler indeholdende disse forbindelser.
Eksempel 1 (20R) -hydroxymethvl-4-methyl-4-aza-5-Q'-8 (14) -prectnen-3-on 25 (i) (2OR) -20-hvdroxvmethvl-4-precmen-3-on
Til en opløsning af (20R) -3-oxopregn-4-en-20-carboxaldehyd (100 g, 305 mmol, kommercielt tilgængeligt fra Upjhon) i 1500 ml ethanol og 250 ml tetrahydrofuran afkølet til ca. 5°C blev portionsvis sat natriumborhydrid (3,35 g, 89 mmol) for at hol- 21 DK 168525 B1 de temperaturen under 15°C. Efter 20 minutters forløb tilsattes eddikesyre (100 ml) , og blandingen blev opkoncentreret til en sjappet masse (slush) på en rotationsfordamper. Remanensen blev derefter taget op i chloroform, successivt vasket med 5 vand og saltopløsning og derefter tørret over natriumcarbonat og opkoncentreret til opnåelse af (20R)-20-hydroxymethyl-4-pregnen-3-on som et hvidt, fast stof (100 g, 100%) .
(ii) Methyl-(20R)-20-hydroxymethyl-5-oxo-3.5-secopreqnan-3-oat 10 Den ovennævnte enon (100 g, 303 mmol) blev opløst i 1000 ml dichlormethan og 500 ml methanol, afkølet til minus 78°C og behandlet med et overskud af ozon. Den resulterende opløsning blev renset med argon, idet den blev opvarmet til 40°C. Opløsningen blev derefter opkoncentreret til en viskos, farveløs 15 olie, taget op i 500 ml dimethylacetamid og omrørt i mørke under argon med 50 g natriumbicarbonat og 50 ml methyljodid i 48 timer. Reaktionsblandingen blev derefter hældt i 4 /il kold saltopløsning og ekstraheret med dichlormethan. De organiske ekstrakter blev omhyggeligt vasket med vand og saltopløsning 20 og derefter tørret over natriumsulfat og opkoncentreret til en gul-rød-olie. Denne olie blev filtreret gennem 1000 g silica-gel med 25% ethylacetat i hexan til efter opkoncentrering at give ketoesteren methyl- (20R) -20-hydroxymethyl-5-oxo-3,5-seco-pregnan-3-oat som en let gul, viskos olie (71,8 g, 65%).
25 (iii) Methyl-(20R)-20-hvdroxymethvl-5-oxo-3.5-seco-6-preanen- 3-oat
Til ketoesteren ovenfor (71,8 g, 197 mmol) i 1500 ml ethylacetat blev sat phenylselenylchlorid (45 g, 235 mmol) , og den resulterende mørke opløsning blev omrørt under argon i 3 timer.
30 Opløsningen blev derefter vasket med vand, mættet natriumbicarbonat, vand og saltopløsning og tørret over kaliumcarbonat. Ethylacetatopløsningen blev derefter afkølet til 15°C og behandlet med 30% hydrogenperoxid (65 ml), idet temperaturen 22 DK 168525 B1 blev holdt under 30°C. Efter 1,5 times forløb blev opløsningen vasket med vand, mættet natriumcarbonat, vand og saltopløsning og tørret og opkoncentreret til et gult skum (65 g) . Søjlekromatografi (silica, 25% ethylacetat i hexan) gav esterenonen 5 methyl-(20R) -20-hydroxymethyl-5-oxo-3,5-seco-6-pregnen-3-oat som en farveløs, viskos olie (3 g, 32%).
(iv) (20R)-hydroxymethvl-4-methyl-4-aza-5.7-precmadien-3-on
Til den viskose esterenon ovenfor (1,67 g, 4,6 mmol) blev sat 10 ml ethylenglycol. Methylamin (3,5 g) blev derefter boblet 10 ind i glycolen, i hvilket tidsrum esterenonen opløstes. Opløsningen blev langsomt opvarmet til 180°C i 20 minutter før afkøling til omgivelsernes temperatur. Den resulterende mørke opløsning blev fortyndet med koldt vand og omhyggeligt ekstraheret med chloroform. De kombinerede ekstrakter blev vasket 15 med vand og saltopløsning og tørret og opkoncentretet til et brunt skum (1,74 g), der blev kromatograferet (silica, 20% ethylacetat i hexan) til opnåelse af dienamidet (20R)-hydroxymethyl -4-methyl-4-aza-5,7-pregnadien-3-on som et hvidt, fast stof (380 mg, 24%) .
20 (v) (20R) -hvdroxymethvl-4-methvl-4-aza-5-Q!-8 (14) -precmen-3-on
Dienamidet ovenfor (500 mg, 1,38 mmol) i 100 ml ethylacetat indeholdende 1% ethanol blev hydrogeneret ved 25°C og l atmosfære over 1,3 g 10% Pd på carbon i 3 timer. Opløsningen blev filtreret for at fjerne katalysator og opkoncentreret til et 25 hvidt, fast stof. Omkrystallisation fra ethylacetat gav 300 mg (20R)-hydroxymethyl-4-methyl-4-aza-5-a-8(14)-pregnen-3-on som et hvidt, krystallinsk, fast stof, smeltepunkt (ukorrigeret) 219-220°C.
Eksempel 2 30 4-methyl-4-aza-5-o?-8 (14) -precmen-3-on- (2OR) -20-carboxylsyre;
Til en opløsning (20R) -hydroxymethyl-4-methyl-4-aza-5-Q!-8 (14) - 23 DK 168525 B1 pregnen-3-on (200 mg, 0,56 mmol), fremstillet i eksempel l, i 15 ml acetone og 1 ml tetrahydrofuran blev dråbevis sat Jones reagens, indtil en rød farve varede ved. Der tilsattes derefter isopropanol for at standse overskud af oxidationsmiddel.
5 Opløsningen blev dekanteret fra de gummiagtige chromsalte, op-koncentreret og delt mellem dichlormethan og vand. Saltene blev opløst i vand og ekstraheret med dichlormethan. Eksempel 3 15 4-methyl-4-aza-5-a?-8 (14) -androsten-3-on-17i8-N.N-diisopropyl- carboxamid.
(i) Methylandrost-4.6-dien-3-on-176-carboxylat
Methyl-androst-4-en-3-on-17/?-carboxylat (10 g, 30,3 mmol, fremstillet i overensstemmelse med Rasmusson et al., J. Med.
20 Chem. 17, 1960 (1984) og chloranil (8,95 g, 36,4 mmol) i 700 ml t-butanol blev opvarmet under tilbagesvaling 3,5 timer og derefter afkølet og filtreret. Filtratet blev opkoncentreret, og remanensen taget op i 700 ml chloroform og vasket successivt med 4 x 150 ml vand, 3 x 150 ml mættet natriumbicarbonat, 25 3 x 150 ml 5% natriumhydroxid og 3 x 150 ml saltopløsning, tørret over natriumsulfat og opkoncentreret til opnåelse af methylandrost-4,6-dien-3-on-17/6-carboxylat som et mørkt skum (7,0 g, 70%) .
(ii) Methyl-7-thioacetyl-androst-4-en-3-on-176-carboxylat 30 Dienonen ovenfor (7,0 g, 21,3 mmol) og thioeddikesyre (15 ml) blev opvarmet ved tilbage svaling i 2,5 timer. Overskud af thi- DK 168525 Bl 24 oeddikesyre blev derefter fjernet i vakuum, og remanensen blev opløst i 100 ml ethylacetat og vasket med vand, mættet natri-umbicarbonat og saltopløsning, tørret og opkoncentreret til opnåelse af methyl-7-thioacetylandrost-4-en-3-on-17/?-carboxy-5 lat som en mørk olie (8,46 g, 98%).
(iii) Dimethvl-3.5-seco-6-androsten-5-on-3.17fl-dicarboxvlat
Thioacetylenonen ovenfor (17,0 g, 42,1 mmol) i 100 ml dichlor-methan og 50 ml methanol ved minus 78°C blev behandlet med overskud af ozon. Den resulterende opløsning blev renset med 10 argon, opvarmet til omgivelsernes temperatur og opkoncentreret. Remanensen blev opløst i 100 ml methanol og 20 ml 10% natriumhydroxid og opvarmet til tilbagesvaling i 3 timer. Opløsningen blev derefter syrnet med saltsyre, opkoncentreret og delt mellem chloroform og vand. Det vandige lag blev yderlige-15 re ekstraheret med chloroform, og de kombinerede organiske lag blev vasket med saltopløsning, tørret og opkoncentreret. Remanensen blev taget i diethylether med nok dichlormethan til at bevirke opløsning, afkølet til 0°C og behandlet med en æterisk opløsning af overskud af diazomethan. Opkoncentrering af den 20 resulterende opløsning gav 10,5 g af en gul olie, der blev kromatograferet (silica, 30% ethylacetat i hexan) til opnåelse af forbindelsen dimethyl-3,5-seco-6-androsten-5~on-3,17/3-dicarboxylat (5,0 g, 33%) som en farveløs olie.
(iv) Hvdroxyethyl-4-methyl-4-aza-5.7-androstadien-3-on-17/3- 25 carboxvlat
Til enonesteren ovenfor (5,0 g, 13,8 mmol) og 50 ml ethylen-glycol blev sat methylamin (10 g), hvorefter enonesteren blev opløst. Den resulterende opløsning blev derefter opvarmet over en 90 minutters periode til 180°C og holdt på denne temperatur 30 i 20 minutter før afkøling til omgivelsernes temperatur. Den resulterende mørke opløsning blev fortyndet med koldt vand og omhyggeligt ekstraheret med chloroform. De kombinerede organiske ekstrakter blev derefter vasket med vand og saltopløsning, 25 DK 168525 B1 tørret og opkoncentreret til en mørkebrun olie. Kromatografi (silica, 1:1 ethylacetat/hexan) gav dienamidet hydroxyethyl- 4-methyl-4-aza-5,7-androstadien-3-on-17/?-carboxylat (1,5 g, 29%) som et offwhite, fast stof.
5 (v) Hvdroxvethvl-4-methvl-4-aza-5a!-8 (14) -androsten-3-on-17fl- carboxvlat
Dienamidet ovenfor (1,5 g, 4,02 mmol) i 20 ml 10% ethanol i ethylacetat blev hydrogeneret over 1,5 g palladiumdioxid ved omgivelsernes temperatur og l atmosfære i 3 timer. Katalysa-10 toren blev derefter fjernet ved filtrering, og opløsningen blev opkoncentreret til opnåelse af hydroxyethyl-4-methy1 - 4 -aza-5a-8 (14) -androsten-3-on-17jS-carboxylat som et hvidt, fast stof (1,5 g).
(vi) 4-methvl-4-aza-5tt-8 (14Ϊ -androsten-3-on-17fl-carboxvlsvre 15 Hydroxyethylesteren ovenfor (1,5 g, 4,0 mmol) og kaliumcarbonat (3 g) i 150 ml 10:1 methanol-vand blev opvarmet til tilbagesvaling i 12 timer. Den resulterende blanding blev opkoncentreret, fortyndet med vand, syrnet og omhyggeligt ekstraheret med chloroform. Chloroformen blev afdampet, og remanensen 20 blev kromatograferet (silica, 5% methanol i dichlormethan) til opnåelse af 4-methyl-4-aza-5a-8 (14)-androsten-3-on-lj8-carbo-xylsyre (0,75 g, 57%) som et hvidt, fast stof.
fviii4-methvl-4-aza-5a-8(14)-androsten-3-on-17fi-carboxvlsvre-chlorid 25 En opløsning af syren ovenfor (250 mg, 0,75 mmol) i toluen (8 ml) blev behandlet med 0,5 ml oxalylchlorid. Efter 2 timers forløb blev de flygtige materialer fjernet ved 1 mm Hg og efterlod en remanens af 4-methyl-4-aza-5a-8 (14) -androsten-3-on-carboxylsyrechlorid.

En opløsning af ovennævnte syrechlorid (283 mg, 0,75 mmol) i 8 ml tetrahydrofuran blev behandlet med 1 ml diisopropylamin 5 natten over. Opløsningen blev fortyndet med vand og ekstraheret omhyggeligt med di chl orme than. Ekstrakterne blev tørret og opkoncentreret til et hvidt, fast stof. Præparativ HPLC (silica, 10% isopropylalkohol i hexan) efterfulgt af omkrystallisation i diethylether gav 4-methyl-4-aza-5-a-8(14)-androsten-3-10 on-17/3-N, N-diisopropylcarboxamid (100 mg), smeltepunkt (ukor- rigeret) 185-190°C.
Eksempel 4 (20R) -hydroxymethyl-4-methyl-4-aza-5-o?-7-preanen-3-on
Aktiv Raney-nikkel-W-2-katalysator (500 mg), som var blevet 15 vasket med vand, ethanol og ethylacetat, blev suspenderet i en opløsning af (20R) -hydroxymethyl-4-methyl-4-aza-5,7-pregnadi-en-3-on (115 mg, 0,33 mmol), fremstillet som i eksempel 1, i 15 ml ethylacetat og omrørt under 1 atmosfære hydrogen i 20 timer. Katalysatoren blev derefter fjernet ved filtrering, og 20 filtratet blev opkoncentreret til et hvidt, fast stof. Kromatografi (silica, 6:3:1 cyklohexan-acetone-ethylacetat) gav 56 mg af udgangsmaterialet, (20R)-hydroxymethyl-4-methyl-4-aza- 5,7-pregnadien-3-on, og 46 mg (2OR)-hydroxymethyl-4-methyl-4-aza-5-Q!-7-pregnen-3-on som hvide nåle (omkrystalliseret fra 25 ethylacetat), smeltepunkt 196-199°C.
